ABSTRACT

Pharmacognostical evaluation of leaves of Adiantum lunulatum is done for identification in field and for differentiation from other species of Adiantum. Methods: A detailed literary work of drug studied and its pharmacognostical, phytochemical and analytical study including powder microscopy and taste determination of leaves with suitable instruments. Results: Macroscopic evaluation reveals the pinnacles are half moon shaped, alternate, sub-opposite or opposite in apical portion of the frond, having lobes with minor incisions. In microscopic structure the two surfaces of pinna are covered by epidermis and epidermal cells are wavy in outline. The opening and closing of stomata will be regulated by guard cells, whereas there is absence of stomata on epidermis. The vascular cells are lined by an endodermal tissue. Transverse section of petiole showed that it is externally covered by an epidermis lined by cuticle on its outer wall which renders the petiole to be shining. Below the epidermis is a fairly thick hypodermis made up Sclerenchyma cells of 4-5 layers in thickness. They are dead cells compact in arrangement and look dark in colour. Powder microscopy of leaves reveals shows few Tracheids of the veins and the Parenchyma cells of the leaves. The Sori are very prominent structures occurring at the leaf margin, they are visible characteristically under the microscope. The Sori contain number of Sporangia each Sporangium is lined by cells with U shaped thickening having their outer wall thin in nature. Conclusion: This study will help in Authentication of Hamspadi plant.

ABSTRACT

OBJECTIVE:To establish the fingerprint of Adiantum capillusveneris from different producing areas ,and to conduct chemometric analysis and content determination of differential components ,so as to provide reference for quality control of A. capillusveneris . METHODS :HPLC-DAD combined with Similarity Evaluation System of TCM Chromatogramtic Fingerprint (2004 A edition )were used to establish fingerprint of 19 batches of A. capillusveneris from different producing areas (S1-S19). Common peaks were confirmed and their similarities were evaluated. Chemometric analysis methods such as cluster analysis , principle component analysis (PCA),orthogonal partial least squares discriminant analysis (OPLS-DA)were used to evaluate the quality of 19 batches of A. capillusveneris from different producing areas ,screen the differential components ,and determine the contents of some differential components. RESULTS :Among 19 batches of A. capillusveneris from different producing areas ,22 common peaks were confirmed ;peak 9 was chlorogenic acid ,peak 17 was quercetin- 3-O-β-D-glucopyranoside,peak 20 was kaempferol-3-O-rutoside;the similarity of 19 batches of sample were 0.677-0.962. Through cluster analysis ,it was found that S 7 and S 10 were clustered into one category ;S15 and S 18 were clustered into one category ;and S 1-S6,S8,S9,S11-S14,S16,S17 and S 19 were clustered into one category. PCA and OPLS-DA found that S 7 and S 10 were clustered into one category ;S15 were clustered into one category ;S18 were clustered into one category ;S1-S6,S8,S9,S11-S14,S16,S17 and S 19 were clustered into one category. Chlorogenic acid ,quercetin-3-O-β-D-glucopyranoside,kaempferol-3-O-rutoside and chemical composition represented by peak 14 were the differential components of the〔2017〕2841); medicinal material. Among 19 batches of A. capillusveneris , average contents of chlorogenic acid and quercetin- 3-O-β-D- glucopyranoside and kaempferol- 3-O-rutoside were 0.10-4.25, 0.31-7.11,0.61-12.00 mg/g,respectively. Four differential components affecting the quality of A. capillusveneris are screened , and the contents of chlorogenic acid , quercetin-3-O-β-D-glucopyranoside and kaempferol-3-O-rutoside in A. capillusveneris from different producing areas were significantly different.

ABSTRACT

Adiantum capillus-veneris, commonly known as maidenhair fern belongs to family Pteridaceae, has traditionally been used in various medicinal preparations as demulcent, expectorant, emmenagogue, diuretic etc. in the form of oil, paste, decoction and powder. It has also prominent role in hair growing and has anti-microbial, anti-inflammatory, anti-diabetic, anti-nociceptive and antioxidant properties of therapeutic interest. This study aimed to investigate the in vitro cytotoxic activity of fractions of ethanolic extract isolated from the aerial part of A. capillus-veneris against some human cancer cell lines such as colon (HCT-116), lung (A549), breast (MCF-7) and pancreatic (MIA PaCa-2) and tumor cell proliferation/inhibition was assessed using MTT assay. The in vivo anticancer activity of hexane fraction was also evaluated against murine Ehrlich ascites carcinoma (EAC) model. The results confirmed that all the fractions of ethanolic extract exhibited promising in vitro inhibition of tumor cell proliferation when tested against different human cancer cell lines. Among all, hexane fraction proved to be more effective having IC50 values 21.72, 22.67, 26.25 μg/mL, for HCT- 116, A-549, MCF-7, respectively, but chloroform fraction revealed to be more cytotoxic against Mia-PACA-2 having IC50 value 14.72 μg/mL. Higher cytotoxic activity is found to be associated with lower IC50 values. The findings showed that all five fractions exhibited dose-dependent killing capabilities in various human derived cancer cell lines at 48 h of treatment. Hexane fraction was found to inhibit tumour growth development by 16.95%, 41.12% and 82.07% at 50, 100 and 200 mg/kg body weight, respectively. Additionally, this fraction was predicted to be non-toxic at the tested doses. The findings indicate that A. capillus-veneris herb is an antineoplastic agent and suggest that further studies evaluating the isolation of active antitumor compounds from A. capillus-veneris and their mechanism(s) of action are necessary.

ABSTRACT

Desiccation-tolerance (DT) is the ability to lose virtually all free intracellular water and then recover normalphysiological functions. The phenomenon of DT is present in higher plants as well as few Pteridophytes whichwithstand drying up to 10%–20% of their dry weight and still resume metabolism following rehydration.The present investigation was carried to understand the biochemical adaptations against desiccation andalso subsequent rehydration in Adiantum raddianum (AR), an omnipresent fern in the Devarayanadurgaforest region of Tumakuru district of Karnataka, India. Detached fronds from healthy AR fern were fullyhydrated, subsequently desiccated, and rehydrated under in vitro conditions. During the desiccation process,the relative water content (RWC) of detached fronds decreased to as low as 16% after 4 hours with intenseinward curving. Upon rehydration, the RWC of the desiccated fronds regained 85% of the initial water contentwithin 4 hours. The rehydrated fronds almost showed the original morphology. Furthermore, physiologicalactivities of antioxidant enzymes such as superoxide dismutase, peroxidase, catalase, glutathione reductase,lipid peroxidation, and proline increased during desiccation but sucrose and starch content showed differentialresponse. The result obtained in this study reveals that AR has desiccation-tolerant properties.

ABSTRACT

Depression is one of the most common psychiatric disorders with a prevalence of 15%-25%. Monoamine aminotransferases, in particular, norepinephrine, serotonin, and dopamine, change in the brain of depressed people. Adiantum capillus-veneris is one of the species of the maidenhair fern genus that have traditionally been used to treat cough, cold symptoms, and local hair loss.In this experimental study, white male rats weighing 250-300 g were assigned to 5 groups of 10 each; group 1: Receiving normal saline; groups 2-4: Receiving A.capillus-veneris extract at 50, 100, and 200 mg/kg, respectively; and group 5: Receiving fluoxetine at 10 mg/kg. Chronic unpredictable stress (CUS) was induced by 3-week exposure to chronic stress. The forced swim test and plus maze were used to assess depression and anxiety, respectively. Malondialdehyde (MDA) levels and antioxidant capacity in the serum and brain were measured. Treatment with A.capillus-veneris extract at 200 mg/kg significantly reduced the duration of immobility. In the group given extract at 200 mg/kg, a significant increase in the number of open arm entries was observed when compared to the control group. A.capillus-veneris extract at 50, 100, and 200 mg/kg resulted in a significant increase in the time spent in the open arm. A.capillus-veneris extract reduced MDA levels and increased antioxidant levels of serum and brain in rat. A.capillus-veneris has significant antidepressant and anti-anxiety effects in rat, probably due to its antioxidant and anti-inflammatory activities.

ABSTRACT

The study aims at investigating the ecological environment Adiantum reniforme var. sinense of in Three Gorges Reservoir region, and providing a reference basis for the protection of resources and artificial cultivation of A. reniforme var. sinense. By using the method of investigation, field survey and experimental analysis, the vegetation, natural geographical environment, climate, soil nutrients of A. reniforme var. sinense were studied and analyzed. The survey found that A. reniforme var. sinense distribution area reduced fast in Three Gorges region, a lot of distribution has diminished and vanished due to excessive digging, currently only in 3 towns of Wanzhou there exist 4 wild distribution areas. The growth of A. reniforme var. sinense needs an environment with low altitude, steep slope and thin soil, northeast slope, canopy height and warm and humid climate characteristics, and the soil in distribution has the characteristics of high organic matter, available nitrogen, available potassium, and low available phosphorus content.

Este trabalho objetivou a purificação parcial, por precipitação com sulfato de amônio (SA) e cromatografia de filtração em gel (CFG), de compostos presentes no decocto de Adiantum capillus-veneris (avenca) eficientes na indução de fitoalexinas em mesocótilos de Sorghum bicolor sorgo. Decocto de A. capillus-veneris a 1% (peso seco/volume) foi precipitado com concentrações de SA variando de 0 a 100% (em intervalos de 20%), e essas frações foram submetidas à CFG. Para o decocto não precipitado foram obtidos nove picos proteicos e um pico glicídico com massas moleculares variando de 0,61 à 0,01 KDa. Para a precipitação fracionada obteve-se: na fração 0-20% dois picos proteicos (menores que 0,01 KDa) e dois glicídicos com concentração de açúcares variando de 4,1 a 17,5 µg mL-1; na fração 20-40% três picos proteicos (111,5 à 0,98 KDa) e cinco glicídicos (11,3 a 73,7 µg de açúcares mL-1); na fração 40-60% dois picos proteicos (111,5 à 0,09 KDa) e dois glicídicos (5,6 a 7, 5 µg de açúcares mL-1); na fração 60-80% seis picos proteicos (menores que 0,02 KDa) e dois glicídicos (16,5 a 51,3 µg de açúcares mL-1); e na fração 80-100% três picos proteicos (menores que 0,09 KDa). Mesocótilos de sorgo foram tratados com as frações provenientes da CFG, além do decocto a 1%, acibenzolar-S-metil (ASM) (125 mg L-1 do i.a. como eliciador de referência) e tampão fosfato de sódio 10 mM pH 6,0. O pico proteico II (0,09 KDa) do decocto não precipitado induziu fitoalexinas, 6,68% superior a ASM. Entre os precipitados, a fração 60-80% de SA induziu 76% mais que ASM. Dessa forma, pôde-se obter frações proteicas e/ou glicídicas indutoras de fitoalexinas em sorgo de maneira superior ao extrato (decocto) do qual é originária, indicando o potencial dessas moléculas para trabalhos futuros sobre indução de resistência.


This study aimed to partially purify the compounds present in decoction of Adiantum capillus-veneris, which are efficient in the induction of phytoalexins in sorghum mesocotyl, by ammonium sulphate (AS) fractionation and gel filtration chromatography (GFC). The decoction of A. capillus-veneris at 1% (weight/volume) was precipitated with AS at the concentration of 0-20%, 20-40%, 40-60%, 60-80% and 80-100%, and these fractions were subjected to GFC. For the decoction not precipitated with AS, nine protein peaks and one carbohydrate peak were obtained with molecular weights ranging from 0.61 to 0.01 KDa. For the AS precipitation, we obtained: for the fraction 0-20%, two protein peaks (0.01 KDa) and two carbohydrate peaks with concentration of sugars ranging from 4.1 to 17.5 µg of sugar mL-1; for the 20-40%, three protein peaks (0.98 to 111.5 KDa) and five carbohydrate peaks (11.3 to 73.7 µg sugar mL-1); for the 40-60%, two protein peaks (0.09 to 111.5 KDa) and two carbohydrate peaks (5.6 to 7.5 µg of sugar mL-1); for the 60-80%, six protein peaks (lower than 0.02 KDa) and two carbohydrate peaks (16.5 to 51.3 µg of sugar mL-1); and for the 80-100%, three protein peaks with molecular weight equivalent to 0.09 KDa. The sorghum mesocotyls were treated with GFC fractions, decoction (1%), acibenzolar-S-methyl (ASM) (125 mg L-1 a.i. as elicitor reference) and sodium phosphate buffer (10 mM, pH 6.0). The protein peak II (0.09 KDa) from the decoction not precipitated was effective in inducing phytoalexin, exceeding in 6.68% the ASM. Among the fractions, the one with 60-80% of AS increased in 76% the induction of phytoalexin compared to ASM. According to the results, we could obtain protein and/or carbohydrate fractions capable of inducing phytoalexins in sorghum better than the decoction from which they are derived from, showing the potential of these molecules for future research studies on the induction of resistance.

Adiantum capillus-veneris L., Pteridaceae, is a fern reputed to have numerous applications in traditional medicine in the treatment of leprosy, animal bites, thyroid dysfunction and musculoskeletal disorder.The proposed study was aimed to investigate the potency of the fern Adiantum capillus-veneris on the thyroid dysfunction - hypothyroidism in terms of determining thyroid gland weight, thyroid peroxidase activity, estimation of iodine in urine, concentrations of total thyroxine (T4), triiodothyronine (T3) and thyroid stimulating hormone (TSH) in serum. The weight of thyroid gland was decreased, while thyroid peroxidase activity, serum T4 (p < 0.01) and serum T3 levels (p < 0.01) were increased in animals treated with the ethanol plant extract (500 mg/kg); but serum TSH level decreased significantly (p < 0.01) when compared with hypothyroid control animals. The SOD, Catalase and glutathione antioxidant enzyme levels and lipid peroxidation were measured.The results of lipid peroxidation and antioxidant enzyme assay indicated that the level of malondialdehyde was significantly lowered and the levels of antioxidant enzymes were significantly increased when treated with ethanol extract of Adiantum capillus (500 mg/kg). Polyphenolic compounds viz., quercetin and gallic acid were determined by HPTLC analysis. With the outlined results, Adiantum capillus could be used for the regulation of hypothyroidism.

Genus Adiantum L., Pteridaceae, is an important fern used in traditional systems of medicine. Different species of Adiantum are known as avenca in Brazil; hansraj/hanmspadi in India; maiden hair fern in English. The present study aims to develop the quality control parameters of four similar looking Adiantum species viz. A. capillus-veneris L., A. lunulatum Burm. f., A. peruvianum Klotzsch, and A. venustum D. Don. Standard methods for macro-microscopic evaluation, physico-chemical parameters and HPTLC were used for authentication and identification. The salient distinctive characters under the microscope are the presence of slightly wavy elongated epidermal cells in A. capillus-veneris; epidermal cells strongly wavy in A. lunulatum; star shaped epidermal cells and mixed spores of regular and irregular shaped in A. peruvianum; stomata on both the surfaces of pinnule, absence of spinulus spores only in A. venustum. In addition, rachis anatomy showed different cellular and stellar characteristics as identifying characters of aforesaid four Adiantum species. Physico-chemical parameters and HPTLC finger print profiles along with stigmasterol and lupeol play significant role for the quality evaluation of raw drugs. The above finding will serve the purpose of quality control and assurance for the future studies.

Damage to cells caused by free radicals has been implicated in the disease progression of at least 50 diseases that is cancer, cardiovascular disease, renal dysfunction and other. So many factors contribute to oxidative stress. Hence, the present study was designed to investigate the potential nephroprotective activity of 250mg/kg and 500mg/kg. Ethanolic extract of Adiantum capillus-veneris dried fronds against Cisplatin induced oxidative stress caused in male Wistar rats. Acute nephrotoxicity was induced by i.p. injection of Cisplatin (7 mg/kg of body weight (b.w.)). Administration of ethanol extract at dose level of 500 and 250 mg/kg (b.w.) to Cisplatin-intoxicated rats (toxic control) for 14 days attenuated the biochemical and histological signs of nephrotoxicity of Cisplatin in dose-dependent fashion. Ethanol extract at 500 mg/kg decreased the serum level of creatinine and urea as compared to the toxic control group. The ethanol extract of Adiantum capillus-veneris at 500 mg/kg (b.w.) exhibited significant and comparable nephroprotective potential. The statistically (one-way-ANOVA followed by Dunnet’s test) processed results suggested the positive action of Adiantum capillus-veneris Cisplatin-induced nephropathy.

ABSTRACT

To study the leaves of Adiantum philippense L. for their antioxidant, cytotoxicity and thrombolytic activities and to perform phytochemical evaluation. Methods: In-vitro antioxidant activity of extract was studied using DPPH radical scavenging, reducing power, total phenol and total flavonoid content determination assays. The cytotoxic activity was determined using brine shrimp lethality bioassay, thrombolytic activity by clot disruption and phytochemical potential by qualitative analysis. Results: The antioxidant activity of the extracts was found promising. The reducing power of this crude extract increase with the increase of concentration; IC50 values of DPPH scavenging activity was (140.00±0.86) μg/mL as compared to ascorbic acid [IC50 (130.00±0.76) μg/mL];Total phenol and total flavonoids content were (148.26±0.24) mg/mL and (163.06±0.56) mg/mL respectively. In cytotoxicity assay the LC50 values of the sample was (106.41±0.78) μg/mL where as for standard vincristin sulphate was (08.50±0.24) μg/mL as a positive control and the extract shows (12.86±1.02)% clot lytic whereas standard streptokinase shows (30.86±0.44% clot lytic activity in thrombolytic assay. The phytochemical evaluation indicates the presence of chemical constituents including carbohydrates, alkaloids, saponins, glycosides, flavonoids. Conclusions: This study shows that the methanol extract of leaves of Adiantum philippense L. has bioactivity but further compound isolation is necessary to confirm the activities of individual compounds.
