ABSTRACT

Aim: The study investigated the anti-hyperglycemic and anti-hyperlipidemic potentials of methanol extracts of Piper guineense and Aframomum melegueta leaves with a view to utilizing the plants in the treatment and management of cardiovascular disorders. Methodology: Twenty-eight healthy albino rats were randomly divided into seven equal groups: Group I received normal saline (2 ml/kg bwt); Group II received a single dose of alloxan(150 mg/kg bwt) intraperitoneally; Group III received alloxan (150 mg/kg bwt) + glibenclamide (5 mg/kg bwt);Group IV received alloxan (150 mg/kg bwt) +PG (200 mg/kg bwt); Group V received alloxan (150 mg/kg bwt) + PG (400 mg/kg bwt); Group VI received alloxan (150 mg/kg bwt) + AM 200 (mg/kg bwt); Group VII received alloxan (150 mg/kg bwt) + AM (400 mg/kg bwt). The blood glucose level was determined before and after treatment with the extracts. The lipid: (total cholesterol (TC), triglycerides (TG), high density lipoprotein (HDL) and low density lipoprotein (LDL) were estimated using the Randox diagnostic kits. Results: The results revealed that alloxan was able to induce hyperglycemia at 150 mg/kg bwt and post-treatment with P. guineense and A. melegueta at 200 mg/kg and 400 mg/ kg bwt were able to significantly lower the blood glucose level which was quite apparent in AM treated groups. Also, the extracts at 200 mg/kg and 400 mg/kg were able to bring a significant (p < 0.05) reduction in TC, TG and LDL concentrations when compared to the alloxan treated group with the highest reduction in AM treated groups. Conclusion: These results revealed that the methanol extract of P. guineense and A. melegueta elicited anti-hyperglycemic and anti-hyperlipidemic potentials of the extracts with the highest effect observed in A. melegueta treated rats.

ABSTRACT

OBJECTIVE@#Hypothyroidism has been claimed to generate sexual dysfunctions such as ejaculatory disorders. Aframomum melegueta is an aphrodisiac plant with pro-ejaculatory properties. We investigated the protective effects of aqueous extract (AE) and methanolic extract (ME) of A. melegueta on the ejaculatory function of hypothyroid male rats.@*METHODS@#Forty sexually experienced male rats were partitioned into 8 groups (5 rats per group) and treated for 28 d as follows: Group 1, Control; Group 2, propylthiouracil (PTU, 10 mg/kg) + distilled water (DW, 10 mL/kg); Group 3, PTU + 5% Tween 80 (10 mL/kg); Group 4, PTU + bromocriptine (6 mg/kg); Group 5, PTU + AE (20 mg/kg); Group 6, PTU + AE (100 mg/kg); Group 7, PTU + ME (20 mg/kg), and Group 8, PTU + ME (100 mg/kg). On days 0, 7, 14 and 28 of treatment, each male rat was paired with primed receptive female for measurement of ejaculatory latency time (ELT) and post-ejaculatory interval (PEI) for 1.5 h. On day 29, each male rat was urethane-anesthetized and the spinal cord was transected. Thereafter, following urethral/penile stimulations and intravenous injection of dopamine, contractions of the bulbospongiosus muscles and the intraseminal pressure were registered. After these recordings, blood was collected through the catheterization of abdominal artery and plasma was used for thyroid-stimulating hormone (TSH), prolactin and testosterone assays.@*RESULTS@#PTU-induced hypothyroidism was characterized by a significant elevation (P < 0.001) of plasmatic TSH and prolactin levels, but a decline (P < 0.001) in plasmatic testosterone, compared to untreated group. ELT, PEI, contractions of the bulbospongiosus muscles and the intraseminal pressure were also altered by PTU treatment. On the contrary, A. melegueta extracts elevated testosterone (AE, 100 mg/kg, P < 0.01; ME, 100 mg/kg, P < 0.05) and decreased prolactin (AE, 100 mg/kg, P < 0.05; ME, 20 mg/kg, P < 0.05) levels, compared to corresponding controls. With regard to DW + PTU group, prolactin concentration was lowered (P < 0.05) in rats administered with bromocriptine. Treatment with A. melegueta extracts significantly prevented the lengthening of ELT (P < 0.05) and PEI (P < 0.001). Hypothyroid state also altered the fictive ejaculation by increasing the latency and decreasing the number and frequency of bulbospongiosus muscle contractions. There was also a decrease in the intraseminal pressure. These alterations were significantly (P < 0.05) alleviated in plant extract-treated groups.@*CONCLUSION@#This study highlighted the ejaculatory disturbance of hypothyroidism in male rats and its prevention with A. melegueta extracts.

ABSTRACT

The present study was undertaken to explore the ameliorative potential of aqueous extract of Aframomum melegueta (AM) on cadmium-induced hepatic damage in rats. Toxicity was induced by daily administration of 200 mg/L cadmium: Cd (Cd as CdCl2) in the animals’ main drinking water for 21 days. Lipid peroxidation (LPO), catalase (CAT), glutathione peroxidase (GPx) and superoxide dismutase (SOD) activities were determined in the liver while total protein, albumin, direct bilirubin and total bilirubin concentration as well as alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were monitored in the serum and histological examination was carried out. Exposure to cadmium resulted in various alterations in all the evaluated parameters. Treatment with AM (200 or 400 mg/kg bw) extract showed a significant (P <0.05) reversal effect that mitigated the deleterious effect of cadmium. Results of the histological examination also support the above findings. The results suggest that aqueous extracts of A. melegueta when administered orally, could ameliorate cadmium-associated oxidative stress in male Wistar rats in a dose dependant manner via its free radical-scavenging mechanism which could be linked to the synergetic effects between the bioactive constituents present in the extract.

ABSTRACT

The effect of methanolic extract of Alligator pepper (Aframomum melegueta) (AME) on the serum creatinine, blood urea nitrogen (BUN) and electrolytes were investigated in Wistar albino rats. The extraction was done by cold maceration of pulverized seeds in absolute methanol for 48 hours. The extract was filtered and concentrated in vacuo in a rotary evaporator at 40ºC. Twenty four male albino Wistar rats were randomly assigned into four groups of six animals each. Group A served as the control and received 10 ml/kg of 5 % Tween 20 while Group B-D received 100, 200 and 400 mg/kg respectively, of the AME for 21 days. Twenty four hours later, blood was collected from the rats through direct cardiac puncture. The separated serum was used to determine the serum creatinine, BUN and electrolyte assay. The extract produced no significant (p > 0.05) difference in mean serum creatinine, BUN and electrolytes level in the treated rats when compared to the control group rats. In conclusion the extract did not exhibit any adverse effect on the kidney at the doses used and for the period of experiment.

ABSTRACT

This present study was performed to evaluate the hepatoprotective effect of Aframomum melegueta seeds, used as spicy in the traditional conditions. The hepatoprotective activity of the seeds of A. melegueta at 100 and 150 mgEq.pp/kg, administered orally, was assessed using carbon tetrachloride-induced (CCl4) liver damage (1 ml/kg bw 1:1 CCl4 and corn oil). The results obtained from this study show that A. melegueta produced a significant decrease in serum transaminases and alkaline phosphatase. At the dose of 150 mg Eq.pp/kg, A. melegueta induced a significant increase in serum total proteins. Liver and serum antioxidant potentials were also significantly increased. Those results indicate that A. melegueta seeds protect the liver against toxicity induced in this study.

ABSTRACT

Aframomum melegueta (Zingiberaceae) seeds are used in West Africa, as a remedy for variety of ailments such as stomach ache, snakebite, diarrhea and anti-inflammatory properties. The hypoglycaemic effects of crude leaf extract of Aframomum melegueta on the treatment of alloxan induced diabetes in male rats and non-diabetic rats (control) were examined in this study. Results obtained from the experiment showed that the elevated blood glucose level caused by oral administration of 250 mg / kg body weight of alloxan was reduced significantly (p < 0.01) by oral administration of Aframomum melegueta leaf extract doses of 50, 100 and 200 mg/kg with the exception of 20 mg/kg when compared to control groups. The non-diabetic groups that received the extract showed reduction in blood sugar level as the dose increases when compared to their control group. There was a final weight gain and organ restoration for both the diabetic and non-diabetic rats after treatment when compared with their controls. This study showed that the extract have hypoglycemic and prophylactic effects.
