ABSTRACT

Objective:To explore the effects of Alpha-2-macroglobulin-rich serum (A2MRS) on knee post-traumatic osteoarthritis (PTOA).Methods:The knee PTOA models were constructed by transection of the anterior cruciate ligament (ACL) in 80 SD male rats, aged 2 months and weighing from 250 to 300 g, which were randomized into 4 groups ( n=20): a high dose group (A2MRS containing 20 μg/μL A2M administered), a low dose group (A2MRS containing 10 μg/μL A2M administered), a positive control group (normal saline administered), and a blank control group (the knee joint cut pseudooperatively and normal saline administered). HE, toluidine blue staining, safranine O staining, modified Mankin scoring and Osteoarthritis Research Society International (OARSI) scoring were conducted to evaluate and compare the therapeutic effects of A2MRS on the knee PTOA among the 4 groups. Results:The rat cartilage was thinner with patchy and cracked surface, and the chondrocytes were reduced and distributed unevenly in the positive control group, compared with the blank control group. The modified Mankin score (3.89±0.93) and OARSI score (10.05±0.72) in the positive control group were significantly higher than those in the blank control group (0.67±0.07 and 3.10±0.29) ( P<0.05). The rat cartilage was thicker with basically complete and crack-free surface, and the chondrocytes were increased and distributed more evenly in the high dose group and the low dose group, compared with the positive control group. The modified Mankin scores (1.33±0.50 and 1.56±0.53) and OARSI scores (6.30±0.64 and 4.75±0.66) in the high dose group and the low dose group were significantly lower than those in the positive control group ( P<0.05). However, there were no such differences between the high dose group and the low dose group ( P>0.05). Conclusion:A2MRS effectively delays the pathological process of knee PTOA.

ABSTRACT

Objective To investigate the core fucosylated alpha 2-macroglobulin(LCA-α2M) level in patients with hepatocellular carcinoma (HCC), liver cirrhosis (LC) and chronic hepatitis B (CHB), and explore its diagnostic value in HCC. Methods A total of 193 HCC patients,104 LC patients and 71 HC patients in Shanghai Eastern Hepatobiliary Surgery Hospital and 45 CHB patients in Changzheng Hospital from January 2013 to December 2016 were included for retrospective study. The method for detecting LCA-α2M was set up, and then the levels of serum α2M and LCA-α2M in each group were detected. The diagnostic value of LCA-α2M for HCC was evaluated by receiver operating characteristic (ROC) analysis. Result The level of LCA-α2M/α2M × 100(LCA-α2M%) was significantly higher in HCC patients[31.25(26.61-35.42)] than that in LC patients [26.00(22.30-30.64)], CHB patients[26.23 (23.86-31.86)] and healthy controls[20.29(17.35-22.60)] (H values were 5.626, 3.388 and 10.942, respectively, P<0.05). The area under the receiver operating characteristic curve (AUC) of LCA-α2M%for identifying HCC was 0.768 (0.725-0.808). Combined α-fetoprotein(AFP) and LCA-α2M%, the area under the ROC curve was 0.890(0.856-0.919). For AFP negative HCC patients, the sensitivity of LCA-α2M%was 77.42%(24/31). Conclusion LCA-α2M% has some values in assistant diagnosis of HCC, and could improve the detection of AFP negative HCC patients.

ABSTRACT

Objective To investigate the relationship between the expression level of serum zinc α2 glycoprotein (ZAG) and type 2 diabetes mellitus (T2DM) complicated with hyperuricemia (HUA).Methods A total of 220 patients with T2DM and normal uric acid (group A),100 patients with T2DM complicated with hyperuricemia (group B),and 80 cases of normal healthy people (group C) were randomly selected.The clinical and biochemical indexes and expression levels of serum ZAG among three groups were determined and compared.The relationship between expression level of serum ZAG and related indexes was analyzed with Pearson correlation and multiple linear stepwise regression analysis.Results The serum ZAG levels showed significant differences among groups A,B and C [(58.89 ± 23.17)mg/L vs (86.42 ± 25.36) mg/L vs (40.65 ± 16.48) mg/L] (P < 0.05).Pearson correlation analysis showed that serum ZAG level was positively correlated with body mass index (BMI),triglyceride (TG),glycosylated hemoglobin (HbAlc),creatinine (Cr),urea nitrogen (BUN),and serum uric acid (SUA) (P < 0.05),and negatively correlated with high density lipoprotein cholesterol (HDL-C) (P < 0.05).Multiple linear stepwise regression analysis showed that BMI,TG and HDL-C were independent factors influencing serum ZAG level.Conclusions Serum ZAG level is significantly higher in patients with T2FM,especially in patients complicated with HUA.The increased level may be related to obesity and dyslipidemia,showing that serum ZAG may be involved in the pathogenesis of T2DM complicated with HUA.
