ABSTRACT

Objective To explore the effects and mechanisms of nesfatin-1 injection in the central nucleus of the amygdale(CeA) on exploratory behavior in rats.Methods Fifty rats were divided into 5 groups randomly : nesfatin-1 low dose group, nesfatin-1 high dose group, SHU9119 group, SHU9119+nesfatin-1 low dose group and control group.Drugs were administrated via CeA to examine behavioral changes of rats by elevated plus maze model test and nesfatin-1 mRNA expression in CeA of anxiety rats (10 rats).Results Compared with the control group,anxiety behaviors of rats in nesfatin-1 low dose group and nesfatin-1 high dose group were improved remarkably,showing that the open arm entries were significantly reduced (OE, t=4.16-12.87, P<0.01), open arm entry proportion was decreased remarkably (OE%, t=2.39-4.39, P<0.01-0.05), and time proportion in open arm was decreased significantly (OT%, t=5.43-20.55, P<0.01),which presented dose dependent.Compared with the control group, anxiety degree of rats in nesfatin-1 low dose group and nesfatin-1 high dose group significantly enhanced, showing that the rats head dipping reduced obviously (HD, t=6.97-16.73, P<0.01) ,while rearing was increased significantly (RE, t=6.60-13.25, P<0.01) ,which was presented dose dependent.Melanocortin receptor antagonist SHU9119 can partly eliminate nesfatin-1 induce-anxiety (t =2.11-3.08, P< 0.01-0.05).Real-time PCR results showed that nesfatin-1 mRNA expression of anxiety model rats was significantly increased (t =3.40-5.58, P<0.05).Conclusion Amygdala nesfatin-1 can increase anxiety and fear of rats,which may be associated with melanocortin systems.

ABSTRACT

Objective To review psychopathic individuals' moral judgments and the underlying neural mechanisms in order to provide cognitive basis and the corresponding intervention for their socially deviant behavior.Methods Literatures were searched in Academic Search Premier,Science Direct,Highwire,PubMed and Wanfang database by July 2015.Index strategy:AB [psychopathy OR psychopathic] AND AB [moral judgments OR moral reasoning].Forty-seven articles including six in Chinese were chosen based on their abstracts and key words.Results 33 papers were adopted finally including 2 in Chinese.Most of the papers involved representative empirical studies within five years.Conclusion Compared with non-psychopathic individuals,psychopathic individuals are inclined to consider moral transgressions as more acceptable and are more inclined to make utilitarian moral judgments.Their impairment in moral judgments is associated with dysfunction in specific brain regions such as amygdale,dorsolateral prefrontal cortex,and ventromedial prefrontal cortex,and dysfunction in the basic brain system.Psychopathic individuals' moral judgment impairment needs to be examined within the frame of affect and their moral reasoning processes should be investigated in the future.

ABSTRACT

The comorbidity of pain and depression is common. Both disorders might share common neuroanatomical and molecu-lar mechanisms. Recent studies have found that the brain-de-rived neurotrophic factor( BDNF) ,which plays an important role in the process of pain-depression comorbidity, has gradually be-come a hot topic and target of treatment. The article mainly sum-marizes the mechanism underlying comorbid pain and depres-sion,as well as the significance of blood BDNF in diagnosis and treatment of pain and depression.

ABSTRACT

Objeaive To explore the effects of the function of hypothalamus-pituitary-adrenal (HPA) axis on emotional development and motor coordination ability by studying the influences of early environment on long-term behaviors of developing rats.Methods Forty-five neonatal rats were randomly divided into 3 groups:the enriched environment group (EE group),the isolated environment group (IE group) and the normal environment group (NE group),with 15 cases in each group.There were 15 rats nurtured separately in EE group,IE group or NE group.The rat motor coordination abilities were tested by straight walking test on postnatal day 28 (P28) and the emotional behaviors were evaluated by handling test at P29.The levels of serum corticosterone(CORT) of rats were detected by using emission immunology method.The morphology of neural cells in rat brain was examined by adopting HE staining.Results In the straight walking test,the motor coordination abilities of rats in EE group > NE group > IE group(F =49.30,P < 0.000).In the handing test,the scores of reaction for stressful stimuli and painful stimuli in IE group were (1.14 ± 0.36,1.93 ±0.83) < (1.47 ±0.64,2.60 ±0.91) in NE group < (2.07 ±0.73,3.43 ±0.65) in IE group (all P < 0.01).The base serum CORT levels were (32.56 ± 9.05) μg/L in EE group > (24.96 ± 6.19) μg/L in NE group > (15.53 ± 6.78) μg/L in IE group (F =19.71,P < 0.001).After a lower-dose adrenocorticotropic hormone stimulation test,the concentration of CORT was (18.24 ± 5.53) μg/L in EE group < (21.71 ± 6.05) μg/L in NE group < (28.09 ±6.51) μg/L in IE group (F=10.25,P <0.000).The HE staining showed the number of neural cells in striaturn and amygdale of rats were[(72.80 ±6.81) cell/HP,(73.53 ±6.14) cell/HP] in EE group > [(65.67 ± 8.98) cell/HP,(61.47 ±6.57) cell/HP] in NE group > [(53.33 ± 6.84) cell/HP,(48.13 ± 6.53) cell/HP] in IE group (all P < 0.001).Conclusions Early environment can affect emotional reaction,motor behavior and brain development of immature rats by regulating HPA axis function.Early isolation environment can lead to HPA axis dysfunction,which can make nerve cell damage and emotion and motor coordination ability dysfunction.On the contrary,early enriched environment can moderate the hyperactivity of HPA axis after stress and promote brain development and ameliorate long-term behavioral development.

ABSTRACT

Aim To explore the effects of acupuncture and sinomenine on the withdrawal symptoms,on the spontaneous discharge and synaptophysin expression of amygdala of heroin-addicted rats.Methods 75 SD rats were randomly and equally divided into 5 groups consisted of control,heroin-addicted,acupuncture,sinomenine and combined treatment group.The model of heroin-addicted rats were established with heroin subcutaneous injection,which in acupuncture,sinomenine and combined group were subsequently punctured at 3 acupoint(Neiguan,Baihui and Zusanli),intramuscular injected with sinomenine and treated with combined methods(acupuncture and sinomenine)respectively.The withdrawal symptoms of rats in different group were evaluated,and then the spontaneous discharges and synaptophysin expression of amygdala were detected with electrophysiological and immunohistochemical technology respectively.Results The heroin-addicted rats were successfully established,being suggested by typical withdrawal symptoms induced by naloxone(P<0.01 vs control),which were alleviated in the 3 treatment groups(acupuncture,sinomenine and combined ) with varying degree(P<0.01 vs heroin group).The electrophysiological results suggest that the forms of low-frequency and single irregular discharges in amygdala were increase,bunchy/cluster and complex discharges were decreased(P<0.05 vs control),which were reversed apparently in sinomenine and combined treatment groups(P<0.05 vs heroin group),closed to control.The immunohistochemical results show that expression of synaptophysin in amygdala was increased obviously(P<0.01 vs control),which was decreased in the 3 treatment groups(acupuncture,sinomenine and combined),P<0.01 vs addicted group.Conclusion Treatments of acupuncture and sinomenine administration can alleviate withdrawal symptoms of heroin-addicted,and rectify the abnormal spontaneous discharges and expression of synaptophysin in amygdala of heroin-addicted rats for certain degree,especially with combined treatments.

ABSTRACT

Objective To explore the possible mechanism of over expression of P-glycoprotein(Pgp) in brain tissue and peripheral blood in refractory epileptic rats with amygdale kindled by electricity and kainic acid,and compare the expression tendency of Pgp in brain tissue and peripheral blood of the different ways to establish refractory epileptic rats models.Methods The models of refractory epileptic rats were made by electrical kindling and kainic kindling in amygdale.Group A was for amygdale-electrical-kindling rats(n=12),group B was for amygdale-kainic-kindling rats(n=12),group C was implanted electrode without kindling and Carbamazepine was given by gavage(n=8),group D was injected saline in amygdale and Carbamazepine was given by gavage(n=8),group E was implanted electrode without kindling and saline was given by gavage(n=8) and group F was injected saline in amygdale and saline was given by gavage(n=8).The expression of Pgp in brain tissue and peripheral blood was detected by immunohistochemistry in each group.Results The expression of Pgp in brain tissue and peripheral blood of rats in group A and B were both much higher than those in the other groups and there were statistical differences(all P

ABSTRACT

Objective To observe the characteristics and onset time of seizures caused by kindling of bilateral amygdale, and to discuss its mechanism.Methods 40 adult Wistar rats were randomly assigned into bilateral amygdala kindling group and unilateral amygdala kindling group. The models were made according to Goddard's method. Results All the rats in bilateral amygdala kindling group developed stage Ⅴ convulsions after a mean of 20.9 stimulations.12 rats of which showed spontaneous seizure discharges. But in unilateral amygdala kindling group, the successful kindling rate was 60% after a mean of 8.9 stimulations. ABSTRACT

Objective To estimate the effect of the NO on the medial amygdaloid nucleus(Me), we studied the origins of the NOS positive terminals in the Me. Methods Noergic afferent projection to Me was identified by a combined NADPH-d histochemical staning and retrograde CTb immunocytochemical method after microinjecting CTb into Me. Results The double labeled of neurons (NOS and CTb) were located in dorsal raphe nucleus, locus ceruleus, basolateral amygdaloid nucleus, parabrachial nucleus, ventrolateral part of periaqueductal gray.Conclution The NADPH-d positive terminals in the Me originates from the aforementioned nucleus, and may relate to the function of the Me.
