ABSTRACT

After rice is processed into parboiled rice, it becomes a food with low glycemic index.Due to the effect of parboiled rice-preparing process on the rice, the changed starch structure and digestibility have induced a reduced hyperglycemic response to rice.After changes in starch structure, direct chain starch content increases with the result in digestive rate decrease.Furthermore, the proportion of the slowly digesting-starch(SDS)in the starch, and the content of the resistant starch(RS)go up, which induces an decreased digestive rate.In addition, parboiled rice also confers greater protection against oxidative stress.It also contains high fat and protein with strong sense of satiety, which reduces the level of blood glucose after eating.The parboiled rice is suitable for patients with diabetes and for the individuals with high risk for diabetes, and can also be applied to patients with hypertension, obesity and other metabolic diseases.

ABSTRACT

Rice germplasms collected from different regions could be used as valuable resources for the future breeding programme. For the utilization of such collections, knowledge about the level and distribution of genetic diversity among these collections will facilitate the breeder. In this study, we report the phenotypic correlation, biochemical quality parameters and population genetic analysis of 35 rice accessions including 34 aromatic rice from different countries and a nonaromatic, Nagina 22, a well-known drought resistance variety. Further biochemical quality analysis, gel consistency test, molecular diversity analysis with 55 simple sequence repeat markers, population structure analysis and pair wise FST analysis were also conducted to assess the genetic diversity. The collected rice genotypes showed significant variability in different agronomic traits, i.e. spikelet per panicle, branches per panicle etc. Results obtained from the above tests demonstrated the importance of regional genetic studies for understanding the diversification of aromatic rice in Asian and African rice.

ABSTRACT

OBJECTIVE: To study the large/small granule number ratio, granule morphology, crystal structure, amylopectin and amylose ratio of five kinds of medicinal starches. METHODS: Five kinds of medicinal starches were separated and characterized by self-assembled gravitational field-flow fractionation (GrFFF) instrument and the proportion of large and small starch granules was calculated according to the results. Optical microscopy and scanning electron microscopy were applied to characterize the morphological property of starch. FT-IR and X-ray diffraction were used to obtain the starch crystalline structure and crystallinity. The contents of amylose and amylopectin were determinate by double wavelength method. RESULTS: The large/small granule number ratios of the five kinds of medicinal starches had obvious difference. The size difference was obvious and the shape was different. The change trends of the order and the amorphous structure of different kinds of starches were the same as that of its relative crystallinity; the higher the amylopectin content, the lower the amylopectin/amylopectin was, the better the crystallinity was. CONCLUSION: GrFFF can be used to characterize and separate large and small starch granules, and the results are in agreement with that of other methods.

ABSTRACT

Background: The enzymes utilized in the process of beer production are generally sensitive to higher temperatures. About 60% of them are deactivated in drying the malt that limits the utilization of starting material in the fermentation process. Gene transfer from thermophilic bacteria is a promising tool for producing barley grains harboring thermotolerant enzymes. Results: Gene for α-amylase from hydrothermal Thermococcus, optimally active at 75­85°C and pH between 5.0 and 5.5, was adapted in silico to barley codon usage. The corresponding sequence was put under control of the endosperm-specific promoter 1Dx5 and after synthesis and cloning transferred into barley by biolistics. In addition to model cultivar Golden Promise we transformed three Slovak barley cultivars Pribina, Levan and Nitran, and transgenic plants were obtained. Expression of the ~50 kDa active recombinant enzyme in grains of cvs. Pribina and Nitran resulted in retaining up to 9.39% of enzyme activity upon heating to 75°C, which is more than 4 times higher compared to non-transgenic controls. In the model cv. Golden Promise the grain α-amylase activity upon heating was above 9% either, however, the effects of the introduced enzyme were less pronounced (only 1.22 fold difference compared with non-transgenic barley). Conclusions: Expression of the synthetic gene in barley enhanced the residual α-amylase activity in grains at high temperatures.

ABSTRACT

La espectroscopia de infrarrojo cercano NIRS (por sus siglas en inglés) se emplea para el control de calidad de los alimentos. La determinación de amilosa en el arroz es un carácter heredable que correlaciona con la calidad del grano, importante en los programas de fitomejormaiento de arroz. El presente trabajo, establece la viabilidad de evaluar el contenido de amilosa en el arroz con la técnica NIRS. Inicialmente, se determinó entre arroz paddy o integral, cuál es el más apropiado para evaluar amilosa empleando NIRS. Se tomaron espectros a las 540 materiales de arroz procedentes del germoplasma del Fondo Latinoamericano para Arroz de Riego FLAR, y posteriormente se hizo análisis de componentes principales y análisis de regresión múltiple en cada tipo de arroz. El arroz integral fue el que obtuvo mejores resultados estadísticos. En el desarrollo de la calibración se empleó como referencia otra calibración en NIRS basada en harina de arroz con un R2=0.9 de calibración y R2= 0.78 de validación, y se empleó Winisi para desarrollar la quimiometría. Se utilizó mínimos cuadrados parciales MPLS y una matemática de 2,1,1,2; longitud de onda de1100-2492,2, lo que arrojó un R2≥0.68 de validación externa,aceptable para caracterizar las generaciones tempranas, con aproximadamente el 83% de aciertos. Los programas de fitomejoramiento del FLAR evalúan 15.000 materiales en el año en las generaciones tempranas, lo que significa US$ 3.600 dólares y 45 días menos que evaluar con la calibración de harina de arroz, siendo la que actualmente se emplea en el FLAR(AU)


The determination of amilosa in the rice is an inheritable character that it correlates with the quality of the grain, importantly breeder rice. The present work, it establishes the viability of evaluating the content of amilosa in the rice with the technology NIRS. Initially, paddy or integral decided between rice, which is most adapted to evaluate amilosa using NIRS. Spectra took to 540 materials of rice of the germplasms banks of the Latin American Found for Irrigated Rice. FLAR, and later there was done analysis of principal components and analysis of multiple regression in every type of rice. The brown rice was the one that most better statistical results. In the development of the calibration another calibration used as reference in NIRS based on flour of rice with a R2 = 0.9 of calibration and R2 = 0.78 of validation, and Winisi was used to develop the chemometrie. There was in use square minimums partial MPLS and a mathematics of 2,1,1,2; wavelength de1100-2492,2, which a R2 = 0.68 of external, acceptable validation to characterize the early generations, with approximately 83 % of successes. The programs breeding of the FLAR evaluate 15.000 materials in the year in the early generations, which means US$ 3.600 dollars and 45 days less that to evaluate with the calibration of flour of rice, being the one that nowadays is used in the FLAR(AU)

ABSTRACT Effects of leached amylose (AM) and amylopectin (AP) on textural and morphological properties of cooked rice were investigated separately by replacing cooking liquid with AM and AP separated from pouring cooking liquid. The pouring of cooking water reduced the hardness (from 28.45 to 19.42N) and stickiness (from 1.74 to 1.19N·s) significantly. However, the addition of AM and AP enhanced the hardness (27.63N) and stickiness (1.71N·s).Scanning electron microscopy show that the leached short-chain AM entered the surface hollows in the cooked rice after water evaporation. Meanwhile, the Long-chain AM cross-linked to formed a three-dimensional network structures, which covered on the filled hollows. This distribution led to a harder texture of cooked rice. The leached AP absorbed water and swelledto form masses. Atthe gelatinization temperature,theAPmassesagglomeratedtoformafilmlayer,whichcoveredtheunevenstructure, the thicker and smoother film contributed to the sticky texture.

ABSTRACT

Genetic divergence was assessed among 23 genotypes of basmati rice representing different regions of India on the basis of yield and quality characteristics utilizing Mahalanobis D2 analysis. Based on the genetic distance (D2 values), the rice genotypes were grouped into six clusters. Of the six clusters formed cluster VI consisted of maximum eight genotypes followed by cluster V accommodating six genotypes, cluster I with four genotypes, cluster III and IV with three genotypes each and cluster II with one genotype. The results indicated that there was some degree of similarity of genotypes clustered together on the basis of their origin. However, the pattern of distribution of some genotypes from different eco-geographical regions was found random, indicating that geographical diversity and genetic diversity were not related. The maximum intra cluster distance was observed for the cluster III. The highest genetic divergence was observed between the clusters IV and I exhibiting wide diversity. The genotypes representing cluster VI are more yielding combined with excellent cooking quality. Among different traits, plant height, kernel length, elongation ratio and amylose content had maximum contribution towards total divergence may be used as selection parameters in segregating generations. Intelligent selection of genotypes from the clusters may be used as potential donors for future hybridization programmes to develop varieties and hybrids of high yield without much compromise on quality.

ABSTRACT

O efeito de doses em cobertura de adubação nitrogenada foi avaliado sobre o rendimento do beneficiamento, a incidência de grãos gessados e barriga branca, os teores de proteína e de amilose nos grãos polidos da variedade de arroz irrigado IRGA 422CL. Utilizando a ureia como fonte de nitrogênio (N), os tratamentos constituíram-se de doses crescentes distribuídas em cobertura na primeira aplicação (0, 40, 63, 80 e 120kg ha-1 de N), na parcela principal (10x5m), e na segunda aplicação (0, 25, 50 e 75kg ha-1 de N), na subparcela (2,5x5m). Assim, foram obtidos 20 tratamentos com variação de 0 a 195kg ha-1 de N aplicado durante o cultivo. As doses em cobertura de adubação nitrogenada influenciaram quadrática e positivamente o rendimento de grãos inteiros (R²=0,75) e negativamente a percentagem de grãos quebrados (R²=-0,89), não apresentando efeitos significativos na percentagem de casca + farelo. As doses de nitrogênio influenciaram de forma linear positiva sobre o teor de proteína (R²=0,81) e linear negativa sobre o teor de amilose (R²=-0,66), nos grãos polidos da variedade de arroz irrigado IRGA 422CL.


The effect of nitrogen (N) fertilization levels was assessed by the industrial yield, the incidence of chalky and white belly grains as well as protein and amylose content of IRGA 422CL cultivar polished grains. The experiment was consisted with N levels applied in coverage using urea as a N source. The first application (0, 40, 63, 80 and 120kg ha-1 of N) was carried out on a main plot (10x5m) and the second application (0, 25, 50 and 75kg ha-1 of N) on a subplot (2,5x5m). Therefore, 20 treatments were obtained varying from 0 to 195kg ha-1 of N applied during cultivation. The nitrogen levels presented a positive quadratic influence on the yield of whole grains (R²=0.75) and a negative influence on the percentage of broken grains (R²=-0.89), not demonstrating significant effects on the percentage of rice hulls + bran. The nitrogen levels influenced protein content in a positive linear way (R²=0.81) and amylose content in a negative linear way (R²=-0.66), in IRGA 422CL cultivar flood-irrigated polished grains.

ABSTRACT

The enantioseparations of 38 racemates on Chiralcel OD, Chiralpak AD, Chiralpak IA and(S, S)-Whelk-01 were presented by HPLC. Those enantiomers come from the amines, alcohols, ethers, ketones, aromatic derivatives, heterocyclic compounds, amide acids and medicines etc. With the mobile phase of n-hexane)/isopropanol(90∶ 10, V/V), n-hexane/isopropanol/trifluoracetic acid(90∶ 10∶ 0.2, V/V) or n-hexane/isopropanol/triethylamine(90∶ 10∶ 0.2, V/V), over 70% enantioseparations were obtained for OD, AD and IA columns). The order of enantioseparation selectivity for four columns was OD>AD>IA>(S,S)-Whelk-01, and among those columns there was a big chiral discriminating complementarity. This investigation was useful for choosing chiral columns to separate chiral compounds.

Devido aos escassos trabalhos brasileiros sobre a estrutura dos grânulos de amido, o objetivo desta revisão foi demonstrar os avanços atuais na elucidação da arquitetura e da estrutura química desse polímero. Muitas pesquisas sobre a avaliação da relação existente entre a estrutura molecular do amido e seu comportamento em algumas propriedades físico-químicas sugerem que diversas características estruturais, tais como teor de amilose, distribuição de comprimento das cadeias de amilopectina e grau de cristalinidade no grânulo, estejam intimamente relacionadas a eventos associados à gelatinização e retrogradação, como inchamento do grânulo, lixiviação de amilose e/ou amilopectina, perda da estrutura radial (birrefringência), supra-molecular (cristalinidade) e molecular e recristalização. O foco principal desta revisão foi a compilação de estudos a fim de obter melhor entendimento da estrutura e das características dos componentes do amido e sua relação com propriedades físico-químicas, principalmente no que diz respeito à gelatinização e retrogradação, as quais apresentam grande importância nutricional e tecnológica.


Due to poor Brazilians work on the structure of starch granules, the purpose of this review was to demonstrate the advances in current understanding of architecture and chemical structure of this polymer. Many studies evaluating the relationship between the molecular structure of starch and its behavior in some physicochemical properties, suggest that several structural features, such as amylose content, amylopectin chain length distribution and the degree of crystallinity in the granule, are closely related to the events associated with gelatinization and retrogradation as starch granule swelling, amylose and/or amylopectin leaching, loss of radial structure (birefringence), supra-molecular (crystallinity) and molecular order, and recrystallization. The main focus of this review was to make a compiler of studies in order to gain better understanding of the granule structure and characteristics of starch components and physicochemical properties (gelatinization and retrogradation) that could play a significant role in improving the quality of different food products.

ABSTRACT

O amido é uma importante fonte energética podendo representar mais de 60% da ingestão calórica na dieta humana. A fração do amido, que é resistente à digestão enzimática, é denominada de amido resistente. Na parboilização de arroz ocorre a retrogradação do amido, com a formação do amido resistente tipo 3 (retrogradado), que apresenta efeitos benéficos à saúde, por atuar como prébiótico. Neste estudo, três amostras de arroz com alta, média e baixa amilose foram selecionadas, com o objetivo de avaliar o efeito de arroz convencional e parboilizado na glicemia de ratos Wistar. As amostras com alta e média amilose foram encharcadas durante 6h a 65°C e a amostra com baixa amilose durante 7h a 70°C, posteriormente foram autoclavadas em pressão 0,7 kgf.cm-2 durante 10 minutos. Utilizouse 6 ratos Wistar, machos, adultos por tratamento. Foram elaboradas 7 dietas experimentais, formuladas de acordo com AIN-93M, dieta controle e, dietas com substituição da fonte de carboidratos por arroz convencional e parboilizado. Determinou-se amido resistente nas dietas e a glicemia foi monitorada por leitura de glicofita, com sangue da parte distal da cauda do rato. Para a curva glicêmica as medidas de glicemia foram feitas em jejum, e ao longo de 90 minutos. Os resultados indicam que não foram encontradas diferenças significativas entre as dietas formuladas com arroz de alta, média e baixa amilose, parboilizado ou convencional na glicemia de jejum e pós-prandial de ratos Wistar.


The effect of parboiled rice on glycemia in Wistar rats. Starch is an important energy source and can represent more than 60% of the calories of the human diet. The starch fraction resistant to enzymatic digestion is called resistant starch. When rice is parboiled, the starch retrogrades with the formation of type 3- resistant starch (retrograded), which presents beneficial effects on the health, since it acts as a prebiotic. In the present study three types of rice were selected, with high, medium and low amylose contents, with the objective of evaluating the effects of conventional and parboiled rice on glycemia in Wistar rats. The samples with high and medium amylose contents were soaked for 6 h at 65ºC, and the low amylose sample for 7 h at 70ºC. The samples were subsequently autoclaved for 10 minutes at 0.7kgf.cm-2. Six male Wistar rats were used for each treatment. Seven experimental diets were elaborated, formulated according to AIN-93M, the control diet and diets substituting the carbohydrate source with conventional or parboiled rice. Resistant starch was determined in the diets and glycemia monitored using glucose paper strips, the sample being blood obtained from the distal part of the rat’s tail. For the glycemic curve, glycemia was measured in the fasting state and during 90 minutes post-prandial. The results indicated there were no significant differences between the diets formulated with high, medium and low amylose, parboiled or conventionally prepared, with respect to fasting or post-prandial glycemia in Wistar rats.

ABSTRACT

The genetic relationships between amylose content (AC) and appearance quality traits of indica rice (Oryza sativa L.) were investigated using conditional analysis and unconditional analysis in present experiment. The results from the unconditional analysis indicated that AC of rice positively correlated with brown rice (BR, i.e., dehulled but unmilled rice) length (BRL), width (BRW) and thickness (BRT), but was negatively correlated with the ratio of length to width (RLW). The conditional analysis showed that weight of brown rice (WBR) negatively affected the genetic relationships between AC and the appearance quality traits of rice except between AC and BRW, while the genetic relationships between AC and most appearance quality traits were negatively affected by protein content (PC). However, these influences were not apparent due to the impact of WBR or PC on the most covariance components of the different genetic systems between AC and the appearance quality traits. The conditional analysis showed that it was possible to improve AC while significantly reduce BRL and BRT under maintaining WBR. Furthermore, AC could be improved when BRL was reduced under maintaining PC, but BRW and BRT could be significantly increased.

This study investigated the effects of high amylose starch (HAS) consumption on gut functions in male Sprague-Dawley rats. Experimental animals were fed an diet containing HAS for 4 weeks (0, 125, 250, 500 g/kg diet). Stool weights, transit time, the pH of cecum, Bifidobacterium growth, short chain fatty acid production, and prostaglandin E2 production in colon mucus were measured. HAS intake did not affect body weight gain or food efficiency ratio during experimental period. There were no significant differences in kidney weight, epididymal fat pad weights or spleen weights, but the weights of the liver and thymus were significantly lower in the HAS100 group. The length of the large intestine, the weights of the cecum wall and cecum contents, and stool weights significantly increased through HAS intake. But transit time was not affected by the experimental diet. Although Bifidobacterium growth in the cecum increased through the HAS intake dose dependently, there were significant differences in the HAS50 and HAS100 groups. HAS intake increased the production of short chain fatty acid in the cecum contents. In particular, acetate and butyrate concentrations grew significantly. And the production of prostaglandin E2 in the colon mucus significantly decreased through HAS intake. These results demonstrate that high amylose starch intake significantly improves gut function.

Objective:To explore the functional mechanism for therapeutic effects of angelica amylose in mice and rats with adjuvant-induced arthritis (AIA) and its effect of abirritation.Methods:Adjuvant arthritis was induced in either mice or rats.The effect of angelica amylose on rats’ arthritis and on mouse writhing response caused by acetic acid were observed.Tripterygium wilfordii was used as the positive drug for control.Results:Angelica amylose had obvious inhibition on mice writhing response and rats’ adjuvant-induced arthritis.Conclusion:Angelica amylose has effects of abirritation and anti-AIA.

ABSTRACT

Objective To study the medicinal plants of Dioscorea nipponica, the physicochemical pro-perties of starch in D. nipponica were investigated by means of various analytical methods. Methods D. nipponica starch was characterized by scanning electron microscope (SEM), X-ray diffraction, granule size analysis, DSC, and Brabender Viscograph system. Results Compared with tapioca and potato starch, the morphology of D. nipponica starch showed smaller particles, oval shaped, and dissimilar granules in size. The crystal type of D. nipponica starch was C-type pattern. The amylose content in D. nipponica starch was 26.3%. The starch separated from D. nipponica showed the highest transition temperature and intermediate enthalpy of gelatinization between potato and tapioca. According to the viscosity measurement with Brabender viscograph, D. nipponica starch exhibited lower peak viscosity, higher setback and lower breakdown viscosity. Conclusion D. nipponica starch is obtained from D. nipponica. Significant differences from D. nipponica and other tuber starches in physicochemical properties are obtained due to their geographical origin.

Objective: To compare amylose content of five varieties of Cordyceps Militaris sample to supply a basis for devoloping and urilizing amylose of Cordyceps Militaris. Methods: Phenol sulfuric acid procedure was used.Results: There is great difference in amylose in the case of cross comparision but not obvious in sequence reference.. Conclusion: Amylose chiefly exists in mycelium.
