ABSTRACT

Objective@#To investigate the detection of HIV/AIDS cases in medical institutions in Tianjin Municipality from 2005 to 2021, so as to provide insights into optimization of HIV antibody testing strategies in medical institutions.@*Methods@#All data pertaining to individuals receiving HIV antibody tests in medical institutions in Tianjin Municipality from 2005 to 2021 were collected from the information systems in medical institutions and the HIV/AIDS Prevention and Control Information System of Chinese Disease Prevention and Control Information System. The positive rate of HIV antibody and epidemiological characteristics of HIV/AIDS cases in medical institutions were analyzed.@*Results@#The number of HIV antibody tests increased from 86 102 person-times in 2005 to 1 369 345 person-times in 2021 in medical institutions in Tianjin Municipality, with annual increase by 18.88%. The positive rate of HIV antibody increased from 2.44/104 to 2.85/104 (P<0.05), with annual increase by 2.79/104. Both the proportion of the number of HIV antibody tests and the number of HIV/AIDS cases detected by medical institutions in the whole city appeared a tendency towards a rise (P<0.05). A high positive rate of HIV antibody was seen in sexually transmitted disease clinics (53.69/104) and other testing services (12.25/104), and the highest positive rate of HIV antibody was seen in individuals at ages of 20 to 39 years (5.40/104), while the positive rate of HIV antibody was higher in men (5.84/104) than in women (0.53/104) (P<0.05). A total of 3 610 HIV/AIDS cases were detected in medical institutions in Tianjin Municipality from 2005 to 2021, with a median age of 38 (interquartile range, 24) years. Of all HIV/AIDS patients, there were 3 277 men (90.78%), and men who have sex with men was the predominant transmission route (63.46%), while the proportion of late identification of HIV/AIDS cases was 65.60%. In addition, the proportion of timely detection for the first CD4+T lymphocyte count testing increased from 23.81% in 2005 to 71.54% in 2021 (P<0.05).@*Conclusions@#The number of HIV antibody tests and positive rate of HIV antibody appeared a tendency towards a rise in medical institutions in Tianjin Municipality from 2005 to 2021. The HIV/AIDS cases were predominantly identified in sexually transmitted disease clinics and other testing services, and were predominantly men.

ABSTRACT

OBJECTIVE@#The prevalence and related factors of serum anti-HCV in different regions and hospitals have not been studied extensively in China. We used routine screening data to determine the prevalence of HCV antibody in hospital patients, evaluate the epidemic trend of hepatitis C and formulate screening strategies.@*METHODS@#Patient information and HCV antibody testing results were collected from January 2017 to December 2019 in 77 HCV sentinel hospitals in China. Univariate and multivariate logistic regression was used to determine the characteristics and associations.@*RESULTS@#HCV antibody prevalence rates were distinct among patients in different departments, with a range of 0.33%-6.93%. Patients who were admitted to the liver disease-related departments (a OR = 10.76; 95% CI, 10.27-11.28), Internal Medicine (a OR = 2.87; 95% CI, 2.75-3.00), and Department of Surgery (a OR = 1.95; 95% CI, 1.87-2.04), were more likely to be tested for HCV antibody positive. HCV antibody prevalence was associated with patients aged 45 years and older (a OR = 2.74; 95% CI, 2.69-2.80), testing in infetious disease hospitals (a OR = 2.33; 95% CI, 2.26-2.40) and secondary hospitals (a OR = 1.72; 95% CI, 1.69-1.75). Patients in sentinel hospitals of the Northeast (a OR = 12.75; 95% CI, 12.40-13.11), the Central (a OR = 1.65; 95% CI, 1.61-1.70), and the West (a OR = 1.78; 95% CI, 1.73-1.83) China had higher HCV prevalence than those who were in the Eastern coastal area.@*CONCLUSION@#Those who were over 45 years old and saw doctors for liver diseases, and invasive diagnosis and treatment should be referred to HCV antibody testing.

ABSTRACT

【Objective】 To analyze the results of different methods for reactive samples screened by the enzyme linked immunosorbent assay (ELISA) against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in blood donors. 【Methods】 From March to April 2020, a total of 8 632 blood samples in Shenzhen were screened for SARS-CoV-2 total antibodies (TAb, including IgG, IgM, IgA) in plasma using ELISA(PC group), the antibody reactivity samples and their follow up plasma samples (FC group), and samples of disease control group(DC group) from January to April 2020 were detected using the following methods: 1) ELISA method for detecting IgG, IgM, and (or without detection) TAb; 2) pseudovirus neutralizing antibody test(pVNT); 3) western blot (WB) of SARS-CoV-2 antibody. The negative control group(NC group) from February to April 2020 performed ELISA and WB testing. 【Results】 Among the 34 total antibody positive samples, 2 were positive for pVNT test, and the total antibody, IgG and WB in the initial screening and tracking testing were positive. Thereafter, it was determined to be confirmed positive. The other 2 cases were positive for pVNT test, while the samples with positive WB results were in the follow-up stage. The TAb, IgG, and pVNT results did not conform to the dynamic evolution of antibodies, and cannot be determined as confirmed positive. 【Conclusion】 The infection status of antibody reactivity samples screened by SARS-CoV-2 ELISA can be judged by the logic of pVNT, WB and the dynamic change of antibody.

ABSTRACT

OBJECTIVE@#To investigate the causes of ineffectiveness of platelet transfusion with monoclonal antibody solid phase platelet antibody test (MASPAT) matching in patients with allogeneic hematopoietic stem cell transplantation and explore the strategies of platelet transfusion.@*METHODS@#A case of donor-specific HLA antibodies (DSA) induced by transfusion which ultimately resulted in transplantation failure and ineffective platelet transfusion with MASPAT matching was selected, and the causes of ineffective platelet transfusion and platelet transfusion strategy were retrospectively analyzed.@*RESULTS@#The 32-year-old female patient was diagnosed as acute myeloid leukemia (high risk) in another hospital with the main symptoms of fever and leukopenia, who should be admitted for hematopoietic stem cell transplantation after remission by chemotherapy. In the course of chemotherapy, DSA was generated due to platelet transfusion, and had HLA gene loci incompatible with the donor of the first transplant, leading to the failure of the first transplant. The patient received platelet transfusion for several times before and after transplantation, and the results showed that the effective rate of MASPAT matched platelet transfusion was only 35.3%. Further analysis showed that the reason for the ineffective platelet transfusion was due to the missed detection of antibodies by MASPAT method. During the second hematopoietic stem cell transplantation, the DSA-negative donor was selected, and the matching platelets but ineffective transfusion during the primary transplantation were avoided. Finally, the patient was successfully transplanted and discharged from hospital.@*CONCLUSIONS@#DSA can cause graft failure or render the graft ineffective. For the platelet transfusion of patients with DSA, the platelet transfusion strategy with matching type only using MASPAT method will miss the detection of antibodies, resulting in invalid platelet transfusion.

ABSTRACT

Objective@#To investigate the prevalence and influencing factors of repeated HIV antibody tests among men who have sex with men (MSM) in Wuhan City, so as to provide insights into the development of interventions against AIDS among MSM.@*Methods@#MSM with self-reported homosexual behaviors in a community studio in Wuhan City were recruited using a convenient sampling method from January to April 2019, and participants' demographic features, sexual behaviors, HIV antibody tests and recognition of HIV antibody tests were collected using questionnaires. Factors affecting repeated HIV antibody tests were identified using a multivariable logistic regression model.@*Results@#Totally 300 valid questionnaires were recovered, with a mean age of (31.06±10.24) years. Among all the participants, 196 participants had the first homosexual behaviors over 18 years of age (65.33%), 107 participants were insertive partners (35.67%), and 125 participants received sex-transmitted diseases (STDs) tests in the past six months (41.67%). The mean score for recognizing the risk of HIV infection was 12.41±3.09, for HIV antibody test self-efficiency was 17.07±2.12, and for perceived social supports was 17.42±2.41. A total of 287 respondents received HIV antibody tests (95.67%), including 192 participants receiving repeated HIV antibody tests (64.00%). Multivariable logistic regression analysis showed that age of >18 years for the first homosexual sex behavior (OR=0.404, 95%CI: 0.223-0.734), receiving STDs tests in the past six months (OR=3.896, 95%CI: 2.145-7.076), sex role as receptive partners or both receptive and insertive partners (OR=0.502, 95%CI: 0.275-0.917), satisfying with HIV antibody test services (OR=2.955, 95%CI: 1.311-6.660), and high score for HIV antibody test self-efficiency (OR=1.149, 95%CI: 1.005-1.314) were factors affecting repeated HIV antibody tests among MSM.@*Conclusions@#The detection of repeated HIV antibody tests was 64.00% among MSM in Wuhan City in 2019, and age for the first homosexual behavior, STDs tests, sex role, evaluation of HIV antibody test services and self-efficiency of HIV antibody tests may be factors affecting repeated HIV antibody tests among MSM.

ABSTRACT

Background: Different diagnostic tools could improve early detection of coronavirus disease 2019 (COVID-19). A number of antibody-based serological point-of-care tests have been developed to supplement real-time reverse transcriptase polymerase chain reaction (RT-PCR)-based diagnosis. This study describes the validity of an antibody test, namely the immunoglobulin G (IgG)/immunoglobulin M (IgM) Rapid Test Cassette® (BNCP ­ 402 and BNCP402), manufactured by Spring Healthcare Services. Methods: A prospective cohort validation study was undertaken at Chris Hani Baragwanath Academic Hospital between 16 July 2020 and 12 August 2020. A total of 101 patients admitte as COVID-19 cases under investigation were included in the study. They were divided into two categories depending on time since symptom onset: testing performed within seven days (early cohort) and after seven days (late cohort). The rapid antibody test was compared to the RT-PCR. Results: Overall, the test has a sensitivity and specificity of 85.2% and 80.0%, respectively, for a combination of IgG and IgM. Sensitivity and specificity of IgG testing alone were 81.5% and 85%. Sensitivity improved for testing with increasing time from symptom onset; however, specifity was not significantly different. Conclusion: The study data adds to the body of evidence that because of relatively low sensitivity and specificity, there is a limited role for antibody-based point-of-care testing in the acute phase of COVID-19 infection, as was the case with this IgG/IgM Rapid Test Cassette (BNCP ­ 402 and BNCP402). There may exist a role for such testing in patients recovered from prior COVID-19 infection or in seroprevalence studies; however, additional evaluations at later timepoints from symptom onset are required.

Background/Introduction@#As the world face health-system shocks from COVID-19, Obstetrician-Gynecologists become perplexed by the uncertainties these bring to the vulnerable pregnant and gynecologic population. The country's capacity for diagnosis via RTPCR consists of only a tiny proportion of the population. With the intent of coming up with a less expensive fast point of care test kits, antibody-based lateral flow assays were developed. @*Aims and Objectives@#Determine the diagnostic accuracy of a lateral flow immunoassay for the detection IgM/IgG antibodies to SARS-CoV2 using RT-PCR as gold standard among symptomatic and asymptomatic high risk OBGYN population. @*Materials and Methods@#This is a multicenter cross-sectional prospective study performed on 147 asymptomatic and symptomatic high risk OBGYN patients who underwent both RTPCR and RAT. Test results were entered using a two by two table to compute for the sensitivity (Sn), specificity (Sp), positive and negative predictive value (PPV/NPV), likelihood ratios (LR) comparing RT-PCR with IgM/IgG using Medcalc statistical software. @*Results@#The RAT for IgG/IgM was not found to be sensitive in both groups. It was able to identify only one of the five patients who had COVID-19 based on RT-PCR. Moreover, the (PPV) was found to be only 20% since only one patient tested positive in the RAT for IgM/IgG that was also positive in the RT-PCR. The (LR+ and LR-) for the symptomatic group was close to 1 implying a slightly higher probability of a true positive compared to that of a false positive test and a negative test result given the absence of the disease respectively. (Sp) and (NPV) of the RAT for IgM/IgG is high for both groups. This means that RAT for IgM/IgG does well in identifying patients who truly do not have COVID-19. @*Conclusion@#With a very low sensitivity of 5% in this study, RAT for COVID-19 cannot be used for screening purposes.

ABSTRACT

Objectives@#The study was done to determine the diagnostic accuracy of rapid antibody tests (RATs) in detecting SARS-CoV-2 infection among asymptomatic employees returning to work. The study also aimed to determine the prevalence of asymptomatic RT-PCR-confirmed COVID-19 infection among these workers. @*Methods@#The study was a retrospective cohort of asymptomatic employees involved in the manufacturing, marketing, and sales of pharmaceutical, consumer, and animal products in the Philippines. They underwent screening for COVID-19 using both rapid antibody and RT-PCR tests as part of the return-to-work company policy from June 1 to July 31, 2020. The sensitivity and specificity of the RATs were computed using RT-PCR as the reference standard. The prevalence of RT-PCR positive COVID-19 infections among asymptomatic returning workers was also determined. @*Results@#Asymptomatic workers (n=5,585) were simultaneously tested for COVID-19 using RAT and RT-PCR. The sensitivity of a positive IgG and IgM was 23.9 (95% CI: 22.8 – 25.0), while the specificity was 93.3 (95% CI: 92.6 - 93.9). The prevalence of RT-PCR identified COVID-19 infection among asymptomatic workers was 4.9% (95% CI: 4.4 – 5.5). @*Conclusion@#The utility of RATs as a screening tool to detect asymptomatic COVID-19 individuals is limited due to its low sensitivity compared to RT-PCR.

ABSTRACT

Abstract Toxoplasma gondii is one of the most important protozoa parasites worldwide. Although many seroprevalence studies have been performed in domestic and wild species, data on the cumulative incidence and the spatial distribution of T. gondii in animals are extremely scarce. In the present study, dogs from Botucatu municipality, São Paulo state, were followed for one year and their blood samples were collected on three moments: days 1, 180, and 360. The sera were submitted to the immunofluorescence antibody test (IFAT) to detect IgG antibodies to T. gondii. Age and sex were compared with IFAT results through statistical tests. Spatial analysis was used to detect clusters of seropositive dogs. Among the 350 dogs that were seronegative on day 1, 53 became seropositive in subsequent samplings; thus, cumulative incidence was 15.1% exposed dogs/year. Age and sex were not associated with serological results. The spatial analysis revealed that seropositive dogs were distributed in all the studied areas, with a significant cluster in a zone with poor sanitary conditions and low socioeconomic status. T. gondii is frequent and widely distributed in the urban area of Botucatu, and impoverished areas are possibly associated with high levels of environmental contamination by this parasite.


Resumo Toxoplasma gondii é um dos parasitas mais importantes mundialmente. Embora muitos estudos de soroprevalência tenham sido realizados em espécies domésticas e selvagens, dados sobre a incidência cumulativa e distribuição espacial de T. gondii em animais são extremamente escassos. No presente estudo, cães do município de Botucatu, estado de São Paulo, foram acompanhados por um ano, e suas amostras de sangue foram colhidas em três momentos: dias 1, 180 e 360. O soro foi submetido ao teste de imunofluorescência indireta (RIFI), para detectar anticorpos IgG para T. gondii. A idade e o sexo foram comparados com os resultados da RIFI por meio de testes estatísticos. Dos 350 cães soronegativos no dia, 1,53 se tornaram soropositivos nas amostragens subsequentes; portanto, a incidência cumulativa foi de 15,1% de cães expostos/ano. A idade e o sexo não tiveram associação com os resultados da sorologia. A análise espacial demonstrou que os cães soropositivos se distribuíram em todas as áreas compreendidas pelo estudo, com uma concentração significativa em uma região com condições sanitárias precárias e baixos níveis socioeconômicos. T. gondii é frequente e amplamente distribuído na área urbana de Botucatu, e regiões empobrecidas podem estar associadas a maiores níveis de contaminação ambiental por esse parasita.

ABSTRACT

Abstract Neospora caninum is an obligate intracellular parasite that can infect many domestic and wild animals, including birds. These animals are important sources for monitoring of environmental contamination, as they could become infected through sporulated oocysts; however, the real role of birds in the biological cycle of N. caninum remains uncertain. This study aimed to determine the prevalence of anti-N. caninum antibodies, evaluate associated factors, detect the parasite by molecular testing of free-range chickens from Brazil, and evaluate different techniques for its serological diagnosis. Blood samples of 366 chickens from 25 farms were collected for serological assays. The indirect fluorescent antibody test (IFAT) and the indirect enzyme-linked immunosorbent assay (ELISA) were used to detect anti-N. caninum antibodies. Chickens that tested seropositive by IFAT had their brain tissues and a pool of organs (heart, lung, and liver) submitted to PCR for molecular detection of the parasite. Out of 366 chickens, 65 (17.8%) and 163 (44.6%) were seropositive by IFAT and ELISA, respectively. Brain tissues (n=60) and the pools of organs (n=65) were negative in the PCR. Our results showed a high prevalence of antibodies in free-range chickens and that IFAT is the more sensitive technique for the detection of anti-N. caninum antibodies.


Resumo Neospora caninum é um parasita intracelular obrigatório que pode infectar vários animais domésticos e silvestres, incluindo as aves. Esses animais são importantes fontes para o monitoramento da contaminação ambiental, uma vez que eles podem se infectar por meio da ingestão de oocistos. Porém, o real papel das aves no ciclo biológico do N. caninum ainda é incerto. Este estudo teve como objetivos determinar a prevalência de anticorpos anti-N. caninum, avaliar fatores associados, detectar o parasita por meio de teste molecular em galinhas de vida livre do Brasil e avaliar diferentes técnicas para o diagnóstico sorológico. Amostras de sangue de 366 galinhas de 25 fazendas foram coletadas para testes sorológicos. A reação de imunofluorescência indireta (RIFI) e o ensaio de imunoabsorção enzimática (ELISA) foram utilizados para detectar anticorpos anti-N. caninum. As amostras de tecidos (cérebro, coração, pulmão e fígado) de galinhas soropositivas na RIFI foram coletadas e submetidas à PCR para detecção molecular do parasita. Das 366 galinhas, 65 (17,8%) e 163 (44,6%) foram soropositivas pela RIFI e ELISA, respectivamente. Nenhuma amostra foi positiva na PCR. Esses resultados demonstram uma elevada prevalência de anticorpos em galinhas de vida livre e que a RIFI é uma técnica mais sensível para a detecção de anticorpos anti-N. caninum.

ABSTRACT

O objetivo desse trabalho foi determinar a ocorrência de anticorpos IgG para Neospora caninum bem como avaliar os fatores de risco associados à infecção em rebanhos ovinos do estado de Sergipe, Brasil. Foram coletadas, nos anos de 2011 e 2012, 1200 amostras de sangue de ovinos oriundos de sessenta propriedades distribuídas em três mesorregiões do estado para pesquisa de anticorpos para N. caninum pela Reação de Imunofluorescência Indireta (RIFI) utilizando-se como ponto de corte de 50 e as amostras diluídas na base 2. Os dados de 34 variáveis estudadas foram obtidos a partir de questionários aplicados aos proprietários e analisados para se determinar a frequências absolutas e relativas e análise dos fatores de risco pelo teste Qui-quadrado de Pearson (p≤0,05). A ocorrência de ovinos soropositivos para N. caninum foi de 39,83% (478/1200). Em relação às mesorregiões a ocorrência de animais e propriedades positivas foi de, respectivamente, 55,88% (380/680) e 88,24% (30/34) na Leste; 21,42% (60/280) e 42,85% (6/14) no Agreste e 15,83% (38/240) e 41,67% (5/12) no Sertão. Os títulos de anticorpos variaram de 50, representando 96,02% (459/478) das amostras soropositivas, a 6400 (1/478). Dentre as variáveis significantes, na analise multivariada, que foram consideradas com fatores de risco para a infecção pelo N. caninum estavam propriedades localizadas na mesorregião Leste (p=0,000, OR=4,64, IC95%=3,36-6,41), presença de fonte de água parada e corrente (p=0,000 OR=2,03, IC95%=1,41-2,92), ausência de quarentena (p=0,000 OR=2,71, IC95%=2,08-3,53), não utilização de esterqueiras (p=0,000 OR=3,14, IC95%=2,45-4,02), criações com finalidade de subsistência (p=0,000 OR=4,99, IC95%=3,15-7,92), de reprodução (p=0,002, OR=1,74, IC95%=1,22-2,49), presença de cães (p=0,000 OR=2,74, IC95%=1,73-433) e circulação de animais silvestres nos rebanhos (p=0,000 OR=3,45, IC95%=2,44-4,87). Os resultados evidenciam a ocorrência de N. caninum em rebanhos ovinos sergipanos, demonstrando o manejo e a localização dos rebanhos no estado como importantes fatores de risco.(AU)


The aim of this study was to determine the occurrence of antibodies IgG against Neospora caninumand evaluate the risk factors associated with the infection in ovine herds, State of Sergipe, Brazil. Blood samples (n=1200) were collected from sheep raised in 60 sheep run located in the three mesoregions of the State of Sergipe, between 2011 e 2012. Antibodies were investigated by Indirect Immunofluorescence Antibody Test of which cutoff point was 50 and positive samples were diluted in base 2 until the last positive titer. Data from 15 variables was obtained from questionnaires given to farmers. Absolute and relative frequencies were determined and the risk factors were analyzed by Pearson's Qui-Square test (p≤0,05). The occurrences of serum reactive sheep were 39.83% (478/1200). The occurrences of positive sheep and sheep run were 55.88% (380/680) and 88.24% (30/34) in the Eastern region; 21.42% (60/280) e 42.85% (6/14) in dry region and 15.83% (38/240) e 41.67% (5/12) in the backwoods respectively. Antibody titers ranged from 50 (n=459), represented 96.02% (459/478) of seropositive samples to 6400 (1/478). Among the significant variables in the multivariate analysis were considered risk factors for infection with N. caninum were, sheep run located in Eastern region (p=0.000, OR=4.64, CI95%=3.36-6.41); standing and running water sources (p=0.000 OR=2.03, CI95%=1.41-2.92), absence of quarantine (p=0.000, OR=2.71, CI95%=2.08-3.53), absence of dunghill (p=0.000, OR=3.14, CI95%=2.45-4.02), presence of dogs (p=0.000, OR=2.74, CI95%=1.73-433), presence of wild animals (p=0.000, OR=3.45, CI95%=2.44-4.87) and subsistence (p=0.000, OR=4.99, CI95%=3.15-7.92) or reproduction (p=0.002, OR=1.74, CI95%=1.22-2.49) livestock were important risk factors. Our results highlight the occurrence ofN. caninumin the ovine herds from State of Sergipe. Management and location of sheep runs were important risk factors associated to the infection.(AU)

BACKGROUND: Automated analyzer-based nontreponemal serological tests for syphilis (STS) have been used for several decades. OBJECTIVE: In this study, we evaluated serological responses to treatment and proposed clinical guidelines for automated STS. METHODS: This retrospective cohort study analyzed human immunodeficiency virus-negative syphilis patients who were diagnosed with automated rapid plasma reagin (auto RPR) tests as a nontreponemal STS, and who also received the fluorescent treponemal antibody-absorption test as a confirmatory test. The ratio of auto RPR values after treatment against those at baseline was defined as the auto RPR ratio for the analysis of the serological response to treatment. The cutoff value for reliable seroreversion prediction was assessed with receiver-operating-characteristic curves. RESULTS: Overall, 89.7% of participants (78/87) seroreverted and 10.3% of participants (9/87) remained serofast during the two-year follow-up period. We were unable to describe trends in the changes among auto RPR values within six months after treatment because of high variation. All of the patients who had an auto RPR ratio ≥1.0 after six months continuously had positive serologic results during their 24-month follow-up and were classified as a serofast group. The receiver-operating-characteristic curves revealed a 25% reduction in auto RPR values nine months after treatment and predicted seroreversion with a sensitivity of 96.2% and a specificity of 100%. CONCLUSION: The most important primary checkpoint for syphilis treatment response is an increase in automated nontreponemal STS six months after treatment. Thus, we recommend monitoring the treatment response with an auto RPR.

Abstract The aims of the present study were to serosurvey dogs, horses, and humans highly exposed to tick bites for anti-Borrelia burgdorferi s.l. antibodies, identify tick species present, and determine risk factors associated with seropositivity in a rural settlement of Paraná State, southern Brazil. Eighty-seven residents were sampled, along with their 83 dogs and 18 horses, and individual questionnaires were administered. Immunofluorescence antibody test (IFAT) was performed on serum samples and positive samples were subjected to western blot (WB) analysis. Anti-B. burgdorferi antibodies were found in 4/87 (4.6%) humans, 26/83 (31.3%) dogs, and 7/18 (38.9%) horses by IFAT, with 4/4 humans also positive by WB. Ticks identified were mostly from dogs and included 45/67 Rhipicephalus sanguineus, 21/67 Amblyomma ovale, and 1/67 A. cajennense sensu lato. All (34/34) horse ticks were identified as A. cajennense s.l.. No significant association was found when age, gender, or presence of ticks was correlated to seropositivity to Borrelia sp. In conclusion, although anti-Borrelia antibodies have been found in dogs, horses and their owners from the rural settlement, the lack of isolation, molecular characterization, absence of competent vectors and the low specificity of the commercial WB kit used herein may have impaired risk factor analysis.


Resumo Os objetivos do presente estudo foram realizar um levantamento sorológico de cães, cavalos e humanos altamente expostos a picadas de carrapatos para anticorpos anti-B. burgdorferi s.l., identificar as espécies de carrapatos presentes, e determinar os fatores de risco associados a soropositividade em um assentamento rural do Estado do Paraná, sul do Brasil. Oitenta e sete residentes foram amostrados junto com seus respectivos 83 cães e 118 cavalos e questionários individuais foram aplicados. O teste de imunofluorescência indireta (IFI) foi realizado nas amostras sorológicas e as positivas foram submetidas a análise por western blot (WB). Anticorpos anti-B. burgdorferi foram detectados em 4/87 (4,6%) humanos, 26/83 (31,3%) cães e 7/18 (38,9%) cavalos pela IFI, com 4/4 humanos também positivos pelo WB. Os carrapatos identificados foram em sua maioria de cães e incluíram 45/67 Rhipicephalus sanguineus, 21/67 Amblyomma ovale e 1/67 A. cajennense sensu lato. Todos (34/34) carrapatos dos cavalos foram identificados como A. cajennense s.l.. Não foram observadas diferenças estatísticas entre idade, sexo ou presença de carrapatos e soropositividade para Borrelia sp. Em conclusão, embora anticorpos anti-Borrelia tenham sido encontrados em cães, equinos e seus proprietários do assentamento rural, a ausência de isolamento, caracterização molecular, ausência de vetores competentes e baixa especificidade do kit comercial de WB utilizado podem ter limitado a análise de fatores de risco.

ABSTRACT

The aim of this study was to examine the intra-uterine exposure to Sarcocystis spp. antigens, determining the number of foals with detectable concentrations of antibodies against these agents in the serum, before colostrum ingestion and collect data about exposure of horses to the parasite. Serum samples were collected from 195 thoroughbred mares and their newborns in two farms from southern Brazil. Parasite specific antibody responses to Sarcocystis antigens were detected using the indirect immunofluorescent antibody test (IFAT) and immunoblot analysis. In 84.1% (159/189) of the pregnant mares and in 7.4% (14/189) of foals we detected antibodies anti-Sarcocystis spp. by IFAT. All samples seropositive from foals were also positive in their respective mares. Serum samples of seropositive foals by IFAT, showed no reactivity on the immunoblot, having as antigens S. neurona merozoites. In conclusion, the intra-uterine exposure to Sarcocystis spp. antigens in horses was demonstrated, with occurrence not only in mares, but also in their foals, before colostrum ingestion these occurrences were reduced.


O objetivo deste estudo foi avaliar a exposição intrauterina ao Sarcocystis spp., para determinar o número de potros que possuem concentrações detectáveis de anticorpos contra esses agentes no soro, antes da ingestão do colostro, por meio da coleta de dados sobre a exposição a esses protozoários nos equinos. Amostras de soro foram coletadas de 195 éguas puro-sangue e seus respectivos potros recém-nascidos, em duas fazendas localizadas na região Sul do Brasil. Os testes utilizados na detecção de anticorpos específicos para o Sarcocystis foram a reação de imunofluorescência indireta (RIFI) e análise por meio de immunoblot. Pela RIFI, em 84,1% (159/189) das éguas e em 7,4% (14/189) dos potros foram detectados anticorpos anti-Sarcocystis spp. Todas as amostras soropositivas dos potros também foram positivas para suas respectivas mães. As amostras de soro dos potros soropositivos na RIFI, não apresentaram reatividade no immunoblot, tendo como antígenos merozoítos de S. neurona. Em conclusão, foi demonstrada a exposição intrauterina de Sarcocystis spp. em equinos, com ocorrência em éguas, porém, em seus respectivos potros, antes da ingestão de colostro a ocorrência foi reduzida.

BACKGROUND: Since 2001, the Korean Red Cross has performed malaria antibody test for blood donors in malaria-risk areas to prevent transfusion-transmitted malaria. However, due to insufficient sensitivity and specificity the malaria antibody assay is not considered an efficient screening method. Therefore, we have considered discontinuing malaria antibody testing for blood donors. METHODS: We analyzed the results of malaria antibody test from 2001 to 2014 utilizing data from the Blood Information Management System of the Korean Red Cross. RESULTS: Among 16,650,812 donations tested from 2001 to 2014, 50,143 donations (0.30%) showed positive results. However, there was no truly infected case at the time of donation. The positive rate among blood donations was between 34 and 39 per 10,000 in 2001, but between 9 and 10 per 10,000 in 2014. There was no interregional disparity in the positive rate of blood donations. CONCLUSION: Korea is in a malaria elimination phase and malaria antibody testing in limited areas is not effective, therefore we propose discontinuing the malaria antibody test.

Objective:To explore blood type distribution of newborns hemolytic disease ( HDN ) caused by maternal and neonatal blood type incompatibility and analyze the value of hemolysis three trials in the diagnosis of HDN.Methods:Hemolysis three trials of type O or Rh negative maternal cord blood samples and hyperbilirubinemia of the newborn blood samples from January 2014 to 2016 were detected by micro-column gel test cards.Then the results were statistically analyzed.Results:(1) There were 918 cases of maternal and neonatal blood type incompatibility in all 1350 cases.569 cases were detected HDN positive with the rate of 62%( 569/918).Among 569 cases,the positive rate of direct anti-globulin test,free antibody test and antibody released test were 27.9%(159/569),86.5%(492/569) and 100% respectively.There was statistical difference of the combination of direct anti-globulin test negative,free antibody test positive and antibody released test positive compared with other combinations ( P<0.05 ).( 2 ) There was statistical difference of HDN positive rate between ABO 73.8%(551/747) and Rh 10.5%(18/171)in 918 cases of blood type incom-patibility.(3)There was statistical difference between A positive rate of 80%(280/350) and B positive rate of 68.3%(271/397) in 747 cases of ABO incompatibility.(4)There was statistical difference among RhD positive rate of 17.7%(14/79),RhE positive rate of 6.8%(4/59) and RhC positive rate of 0(0/33).Conclusion: Antibody released test was the most sensitive test in hemolysis three trials to diagnose HDN.The probability of HDN positive caused by maternal and neonatal ABO blood type incompatibility was significantly higher than Rh.The probability of HDN positive with type A newborns was significantly higher than type B.The probability of HDN positive caused by RhD blood type incompatibility was significantly higher than RhE and RhC.

Neospora caninum is a protozoan parasite that causes abortion and important economic losses in cattle worldwide. The accurate diagnosis of neosporosis is essential for management and control measures. The aims of this study were: i) to evaluate the performance of an in-house enzyme-linked immunosorbent assay based on the 38 kDa native antigen (p38-ELISA) to diagnose bovine neosporosis in Argentina using a well- characterized local sera panel from experimentally infected and naturally exposed cattle and ii) to compare the diagnostic performance and agreement of three N. caninum serological tests: p38-ELISA, indirect fluorescence antibody test (IFAT) and immunoblotting (IB) using the same sera panel. Serum samples testing either positive or negative by IFAT and IB were considered "Relative Standards of Comparison" (RSC) and used for p38-ELISA evaluation. Receiver operating characteristics analysis revealed that p38-ELISA was highly accurate (area under the curve= 0.982) according to RSC with a cut-off index of 0.0905. Relative sensitivity and specificity of p38-ELISA were 97.8 % and 99.5 %, respectively and agreement between RSC and p38-ELISA was almost perfect (k= 0.97). The evaluation and performance comparison of serological tests were performed according to the definition of gold standard based on the decision of the "majority of tests". All tests displayed high sensitivity and specificity values (greater than 95 %); and excellent agreement. This study describes the accurate performance of p38-ELISA evaluated locally and the highly accurate diagnostic performance of the studied tests for the detection of anti-N. caninum antibodies in cattle from Argentina.


Neospora caninum es un parásito protozoo responsable de abortos y pérdidas económicas en bovinos. La realización de un diagnóstico serológico preciso y con resultados comparables obtenidos por diferentes pruebas contribuye al manejo de este problema y a encarar medidas de control. Los objetivos del presente trabajo fueron los siguientes: 1) evaluar en Argentina una prueba de enzimoinmunoensayo in-house con el antígeno nativo de 38 kDa de N. caninum (ELISA-p38) para el diagnóstico de la neosporosis bovina, utilizando un panel de sueros locales bien caracterizados, procedentes de bovinos infectados de modo experimental o naturalmente expuestos; 2) comparar el desempeño y establecer el nivel de concordancia de tres pruebas serológicas para la detección de N. caninum, ELISA-p38, inmunofluorescencia indirecta (IFI) e inmunoblot (IB), con el mismo panel de sueros. Los sueros que resultaron positivos o negativos a IFI e IB fueron considerados como estándares relativos de comparación (ERC) para evaluar la prueba de ELISA-p38. El análisis de característica operativa del receptor determinó que la prueba de ELISA-p38 fue altamente precisa (área bajo la curva= 0,982) usando el punto de corte 0,0905. La sensibilidad y especificidad relativa del ELISA-p38 fue 97,8 % y 99,5 %, respectivamente, con una concordancia casi perfecta (k= 0,97) respecto del ERC. La comparación del desempeño de las pruebas se realizó usando como gold standard el criterio de la decisión de la "mayoría de las pruebas". Las pruebas exhibieron altos valores de sensibilidad y especificidad (mayores del 95 %) y excelente concordancia. Este trabajo describe un buen desempeño de la prueba de ELISA-p38 evaluada localmente y adecuada performance diagnóstica de las pruebas serológicas analizadas para la detección de anticuerpos anti N. caninum en bovinos de Argentina.

INTRODUCTION: The present study was designed to assess the occurrence of co-infection or cross-reaction in the serological techniques used for detecting the anti-Leishmania spp., -Babesia canis vogeli and -Ehrlichia canis antibodies in urban dogs from an area endemic to these parasites. METHODS: The serum samples from dogs were tested for the Babesia canis vogeli strain Belo Horizonte antigen and Ehrlichia canis strain São Paulo by immunofluorescence antibody test (IFAT) and by anti-Leishmania immunoglobulin G (IgG) antibody detection to assess Leishmania infection. We used the following four commercial kits for canine visceral leishmaniasis: ELISA, IFAT, Dual Path Platform (DPP) (Bio Manguinhos(r)/FIOCRUZ/MS) and a rK39 RDT (Kalazar Detect Canine Rapid Test; Inbios). RESULTS : Of 96 serum samples submitted to serological assays, 4 (4.2%) were positive for Leishmania as determined by ELISA; 12 (12.5%), by IFAT; 14 (14.6%) by rK39 RDT; and 20 (20.8%), by DPP. Antibodies against Ehrlichia and Babesia were detected in 23/96 (23.9%) and 30/96 (31.2%) samples, respectively. No significant association was identified between the results of tests for detecting Babesia or Ehrlichia and those for detecting Leishmania (p-value>0.05). CONCLUSIONS: In the present study, we demonstrated co-infection with Ehrlichia or Babesia and Leishmania in dogs from Minas Gerais (Brazil); we also found that the serological tests that were used did not cross-react. .

Toxoplasmosis and neosporosis have been recognized as economically important diseases with considerable impact on the livestock industry. Little is known concerning the occurrence of Toxoplasma gondii and Neospora caninum in sheep from Tocantins state, Brazil. Here, we investigated antibodies against these parasites and associated factors in 182 sheep from Araguaína, Santa Terezinha do Tocantins, Arguianópolis and Palmeiras do Tocantins districts, Tocantins. Sheep sera were assayed for T. gondii and N. caninum IgG antibodies by indirect fluorescence antibody test (IFAT), using cut-off point at a dilution of 1:40 and 1:25 respectively. The prevalence of seropositive animal for T. gondii was 13.74% and 13.74% for N. caninum. None of the characteristics studied including reproductive problems, presence of cats, presence of dogs and veterinary care (p>0.05) was associated with occurrence of T. gondii or N. caninum infection. Only breed was identified as associated factor for the occurrence of toxoplasmosis in sheep (p<0.05). The present study is the first report on serum occurrence of T. gondii and N. caninum in sheep from the state of Tocantins, Brazil.


Toxoplasmose e Neosporose são reconhecidas por doenças economicamente importantes com impacto considerável na indústria pecuária. Pouco se sabe sobre a ocorrência de Toxoplasma gondii e Neospora caninum em ovelhas do estado do Tocantins, Brasil. Foram investigados a ocorrência de anticorpos contra estes parasitos e fatores associados em 182 ovelhas das cidades de Araguaína, Santa Terezinha do Tocantins, Arguianópolis e Palmeiras do Tocantins, Tocantins. Os soro das ovelhas foram testados para anticorpos IgG anti-T. gondii e anti-N. caninum pela Reação de Imunofluorescência Indireta (RIFI), usando os pontos de corte na diluição de 1:40 e 1:25, respectivamente. A prevalência de animais soropositivos para T. gondii foi de 13.74% e para N. caninum, 13.74%. Nenhuma das características estudadas incluindo problemas reprodutivos, presença de gatos, presença de cães e cuidados veterinários (p>0.05) foram associadas com a ocorrência infecção por T. gondii ou N. caninum. Somente raça foi identificada como fator associado à ocorrência de toxoplasmose em ovelhas (p<0.05). O presente trabalho é o primeiro relato da ocorrência sérica de T. gondii e N. caninum em ovelhas do estado do Tocantins, Brasil.
