ABSTRACT

Objective@#To investigate the prevalence of Ureaplasma urealyticum (UU) infection in infertile men, its influence on routine semen parameters and the distribution of antisperm antibody (AsAb) and its types in infertile patients with UU infection.@*METHODS@#We detected the positive rate of UU infection, semen parameters, and the distribution of AsAb and its types in 662 infertile men and 25 normal fertile male controls followed by comparison of the obtained data between the two groups of subjects.@*RESULTS@#The positive rate of UU infection was significantly higher in the infertile men than in the normal controls (52.87% [350/662] vs 16.00% [4/25], χ2 = 11.68, P 0.05) and AsAb IgA, IgM and IgG in the seminal plasma, or in the percentages of serum AsAb IgM (16.9% vs 20.5%, χ2 = 1.22, P >0.05) and IgG (32.7% vs 28.9%, χ2 = 0.99, P >0.05) except in that of serum AsAb IgA (23.6% vs 17.0%, χ2 = 4.03, P <0.05).@*CONCLUSIONS@#The UU infection rate is high in infertile males, which decreases the semen volume, total sperm count, motile sperm concentration and percentage of progressively motile sperm and increases the positive rate of serum AsAb IgA.

ABSTRACT

Objective@#To investigate the influence of different procedures of testicular sperm retrieval on the levels of serum inhibin B (INHB), antisperm antibodies (AsAb), follicle-stimulating hormone (FSH), and testosterone (T) in patients with azoospermia.@*METHODS@#We randomly assigned 210 azoospermia patients to receive testicular sperm extraction (TESE, n = 50), testicular sperm aspiration (TESA, n = 56), testicular fine needle aspiration (TEFNA, n = 64), or microscopic TESE (micro-TESE, n = 40). We measured the levels of serum INHB, FSH, and T and the positive rate of AsAb before and at 1 and 3 months after surgery.@*RESULTS@#Compared with the baseline, the levels of serum FSH at 1 and 3 months after surgery showed no statistically significant differences in the TESE ([8.51 ± 4.34] vs [8.76 ± 3.07] and [7.24 ± 3.32] IU/L, P >0.05), TESA ([7.70 ± 2.72] vs [7.90 ± 4.57] and [8.04 ± 3.65] IU/L, P >0.05), TEFNA ([6.04 ± 3.17] vs [6.08 ± 2.70] and [6.10 ± 3.32] IU/L, P >0.05), or micro-TESE group ([6.59 ± 2.74] vs [6.89 ± 1.78] and [6.75 ± 2.57] IU/L, P >0.05); the positive rate of AsAb (IgM) was significantly increased at 1 month in the TESE (0.00 vs 14.00%, P 0.05).@*CONCLUSIONS@#Different procedures of testicular sperm retrieval have different impacts on the testicular function and AsAb in patients with azoospermia.

ABSTRACT

HSP110 functions to protect cells, tissues, and organs from noxious conditions. Vasectomy induces apoptosis in the testis; however, little is known about the reason leading to this outcome. The aim of the present study was to evaluate the expression and function of HSP110 in mouse testis after vasectomy. Following bilateral vasectomy, we used fluorescent Terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) to detect apoptosis, Western blotting and immunohistochemistry to examine HSP110 expression and localization. Serum antisperm antibody (AsAb) and testosterone were measured by Enzyme-linked immunosorbent assay (ELISA) and radioimmunoassay, respectively. Expression of endoplasmic reticulum stress (ERS) sensors and downstream signaling components was measured by Reverse Transcription-Polymerase Chain Reaction (RT-PCR), and the phosphorylation of eIF2 and JNK was detected by Western blotting. Vasectomy induced morphologic changes, increased apoptosis in the testis, increased serum AsAb, and decreased testosterone levels. After vasectomy, ORP150 mRNA level was increased first and then decreased, Bcl-2 was decreased, and the expression of HSPA4l, GRP78, GADD153, PERK, ATF6, IRE-1, XBP-1s, Bax, Bak, and caspases and the phosphorylation of eIF2 and JNK were increased. We present that an ER stress-mediated pathway is activated and involved in apoptosis in the testis after vasectomy. HSPA4l and ORP150 may play important roles in maintaining the normal structure and function of testis.

ABSTRACT

Objective To investigate the value of detections of anti‐nuclear antibody (ANA) combined with anti‐cardiolipin anti‐body (ACA) ,anti‐sperm antibody (AsAb) and anti‐beta 2 glycoprotein I (β2 GPI) antibody in the diagnosis of female infertility dis‐ease .Methods A total of 187 female cases of infertility (infertility group) were detected serum ANA and AsAb by the indirect im‐munofluorescence assay ,and ACA andβ2 GPI antibody by the enzyme linked immunosorbent assay (ELISA) .ANA ,ACA ,AsAb andβ2 GPI antibody also were detected in 80 females cases of normal fertility (normal group) .Results Among 187 cases of female infer‐tility ,ANA positive rate was 18 .1% (34/187) and which in the normal group was 2 .5% (2/80) .The ACA positive rate was 22 .3%(43/187) in the infertility group and 5 .0% (4/80) in the normal group ;the AsAb positive rate was 18 .7% (35/187) in the infertil‐ity group and 3 .8% (3/80) in the normal group ;theβ2 GPI positive rate was 20 .3% (38/187) in the infertility group and 3 .8% (3/80) in the normal group;the differences between the two groups had statistical significance (P<0 .05) .Conclusion Infertility is closely correlated with the in vivo existence of ANA ,ACA ,AsAb andβ2 GPI antibody ,the joint detection is conducive to find the e‐tiology of infertility and improve the clinical diagnosis rate .

ABSTRACT

ObjectiveTo examine the level of antisperm antibody (ASAb),antiendometrial antibody (EMAb) and anti-mulerian hormone(AMH) in serum of women with infertility and to provide a reliable basis for prediction,diagnosis and treatment of infertility.Methods Two hundred cases of women with infertility visiting our hospital between May.2010 and May.2011 were chosen as the infertility group and 100 cases of women with fertility experience visiting our hospital at the same period with the infertility group were chosen as the controlgroup.We examined the presence of ASAband EMAb and the levelsof AMHof the participants.ResultsIn the infertility group,the total positive rate of serum ASAb was 27.5% (55/200) and the positive rates of ASAb-IgG,ASAb-IgM and ASAb-IgA were 11.5 % (23/200),22.5 % (45/200) and 9.5 %(19/200) respectively.While in the control group,the rates were 6.0% (6/100),1.0% (1/100),0 and 2.0%(2/100) respectively ( x2 =5.33,5.37,5.41,4.05 ;P < 0.05 ).The total positive rate of EMAb was 148.5%(97/200),and the positive rates of EMAb-IgG and EMAb-IgM were 13.5% and 32.5% respectively,and EMAb-IgG + EMAb-IgM positive rate was 5.5% (11/20).These parameters were significantly higher than those in the control group [0,1.0% (1/100),3.0% (3/100),0,x2 =5.01,5.24,5.16,5.33 ;P <0.01 ].There was significantly difference on the level of AMH between the experimental group and the control group [ (5.39 ±1.42) μg/L vs.(2.55 ± 1.27 ) μg/L,t =5.39,P < 0.01 ].Significant correlation was found between ASAb and EMAb( x2 =6.27,P =0.03) by correlation analysis.ConclusionThe level of AMH and the positive rates of ASAb and EMAb are higher in women with infertility than in normal people. Detection of ASAb,EMAb and AMH are important in in finding cause for infertility and taking appropriate measures to treat infertility

ABSTRACT

Objective To observe the curative effect of the combined action of Compound Xuanju Capsule and Prednisone on oligospermatism and antisperm antibody due to yang deficiency of kidney. Methods 136 patients were randomly divided into two groups: a treated group, which was taken Compound Xuanju Capsule and Prednisone, and a control group, which was taken Prednisone and Tamoxifen Citrate Tablet. Both groups received a 3-month therapy. The conditions of semen and antisperm antibody between the two groups were observed before and after the treatment. Results The total effective rate of the treated group was 90.94% (61 in 67 cases), and in which 11 cases were pregnant (16.41%). While the total effective rate of the control group was 63.07% (41 in 65 cases), and in which 2 cases were pregnant (3.08%).The clinical efficacy of the treated group was better than that of the control group. (P<0.05) After the treatment, sperm density and motility of two groups were both evidently increased, (P<0.05, P<0.01) and the treated group increased much more obviously (P<0.05). Conclusion The combined action of Compound Xuanju Capsule and Prednisone can improve the curative effect on the treatment for male infertility from oligospermatism and antisperm antibody due to yang deficiency of kidney.

ABSTRACT

Objective To research the possible influence of operation and human chorionic gonadotropin(HCG) therapy on the development of antisperm antibody (AsAb) and serum levels of estradiol (E_2) and testosterone (T) of cryptorchidism boys after orchiopexy. Methods Forty cryptorchidism boys were studied who were divided into HCG treatment group and without HCG treatment group. Twenty boys who underwent inguinal operation were studied as the operation control group and 20 normal boys as the control group. The serum were collected for detection AsAb, E_2,and T. Results There were no significant difference in serum levels of E_2 and T among the three groups(P 0.05 ). Conclusions The level of AsAb is associated with the destroy of blood barrier and the disorder of immune modulation and endocrine. Treatment with HCG has no effect on AsAb level.

ABSTRACT

【Objective】To explore the correlation of antisperm immune infertility(AII) in females with chronic genital tract inflammation and to observe the therapeutic effect of Chinese medicine of Qingshi Huayu Mixture(QHM) and Yishen Huoxie Prescription Ⅰ(YHPⅠ) for AII.【Methods】Fifty-eight AII patients were screened from 200 infertility patients,and were randomized into three groups.Group A(N=18) was treated with YHPⅠ,group B(N=20) with QHM and group C(N=20) with western medicine.Three months constituted one treatment course.The incidence of positive antisperm antibody(AsAb+)combined with chronic genital tract inflammation,therapeutic effect,pregnancy rate,the content of serum AsAb,as well as the changes of lymphocyte subtypes,natural killer(NK) and interleukin 6(IL-6) were observed.The correlation of the above indexes with serum AsAb content was also analyzed.【Results】(1) The incidence of positive antisperm antibody(AsAb+)combined with chronic genital tract inflammation: There were 96(48%)with AsAb+ in 200 patients,and 62(64.58%) with AsAb+ in 122 combined with chronic genital tract inflammation;there were 34(35.42%)AsAb+ patients in 78 without inflammation,the difference of AsAb+ incidence was significant between the inflammation patients and the non-inflammation patients(P0.05).There were 2 in 5 cured patients of group A,2 in 7 of group B and 1 in 7 of group C being pregnancy.(3) Content of serum AsAb: serum AsAb content was decreased in the three groups after treatment,the difference being significant as compared with that before treatment(P0.05).(4) The changes of lymphocyte subtypes,NK and IL-6: CD8+ was increased,CD4+/CD8+ and NK were decreased in groups A and B,the differences being significant as compared with those before treatment and those in group C after treatment(P

Objective To explore the effect of acupuncture combined with Guishen Decoction (GD) on serum antisperm antibody (ASAb) level of rat models with immunological infertility. Methods Active immunization was used to establish rat models with positive serum ASAb. ABSTRACT

The aim of this investigation was to determine the incidence of sperm antibodies in serum using indirect immunobead test (IBT) in 113 patients of scrotal disease. The results of this survey showed that 17.7% of 113 men had antisperm antibodies of IgG and/or IgA class bound to the surface of at least 20% of motile spermatozoa of normal donor. There were positive immunobead test in 16 of 36(44.4% ) vasovasostomy patients, 1 of 18 testicular trauma, 1 of 15 primary infertility, 1 of 4 obstructive infertility, 1 of 7 hydrocele. There were no statistical differences in sperm count, sperm morphology between the groups of patients with positive or negative IBT results. As for the immunoglobulins, the most frequent class was IgG, and in terms of morphological sites, tail binding was most frequent. The results of this investigation therefore suggest that the IBT is an excellent test for sperm antibodies in scrotal disease.

ABSTRACT

Immunologic factors including antisperm antibodies play a significant role in pathogenesis of 10-20% of unexplained infertility cases. and any event or circumstance that would breach the blood-testis barrier may result in the formation of antisperm antibodies. Using an immunobead test we studied the occurrence of circulating antisperm antibodies in twelve patients who had undergone surgical treatment for testicular rupture due to blunt scrotal trauma. On the follow-up ranging from 6 to 14 months after surgical treatment only one patient was found to have circulating antisperm antibodies but his semen remained normal in spite of the presence of circulating antisperm antibodies.

ABSTRACT

Objective To determine the levels of anti-Toxoplasma gondii antibody (ATAb) and antisperm antibody (AsAb) in serum samples and interferon-?(IFN-?) in supernatants of peripheral blood cell culture in females with infertility, and to analyze their correlation. Methods ELISA was applied to determine and compare the ATAb and AsAb in serum samples of 182 women with infertility and those of 94 women with normal pregnant history. The level of IFN-? in supernatants of peripheral blood cell culture was also measured and compared by ELISA. Results The positive rates of serum ATAb and AsAb in the cases of infertility were 15.38% and 18.13% respectively, which were significantly higher than those of women with normal pregnant history( 6.38% and 5.32%). The supernatant IFN-? level in infertile women was significantly higher than that of women with normal pregnant history (0.970?0.493) ?g/L vs (0.531?0.274) ?g/L(P

ABSTRACT

An extensive variety of methods has been used to detect antisperm antibodies in infertile individuals, and this reflects a concern about the immunological validity, interpretation and standardization of the tests. Comparisons of different methods using the same test materials have shown little correlation between results. The purpose of this Study is to compare the results of three methods. ELISA, gelatin agglutination test and sperm immobilization test-using the same test materials based on recommendations from WHO workshop. The results are as follows: 1. Ten normal controls showed negative reactions in all the 3 tests. Out of 34 patients, the positive sera were noted in 23 (67.6%) on ELISA test, 20 (58.8%) on gelatin agglutination test and 18 (52.9%) on sperm immobilization test. 2. Fifteen (44.1 %) out of 34 patients showed positive reactions in all the 3 tests, and 26 (76.5%) out of 34 patients showed positive reaction in one or more tests. 3. The titers of the antisperm antibodies were higher in the following orders; ELISA, gelatin agglutination test and sperm immobilization test. Therefore, it seems to be possible to increase the detectability of the antisperm antibodies, if more than one tests are impolyed.

ABSTRACT

We studied antisperm antibody by the use of the tray agglutination test in sera of 37 vasectomized males and 65 infertile males Antisperm antibody was found in 27 out of 37 vasectomized males (73%). Antibody titer was more than 1:32 in majority of the patients (25/27). The most common agglutination pattern was tail-to-tail agglutination (70.5 %). Antisperm antibody was found in 7 out of 17 obstructive azoospermia (41.2 %), and 3 out of 42 idiopathic infertility (7.1 %). But none was found in 6 patients with varicocele.
