ABSTRACT

@#ObjectiveTo investigate the relationship between the degenerative mechanisms of lumbar intervertebral disc (LID) and apoptosis.MethodsThe total RNAs were isolated from human LID tissues. Both the mRNAs from the degeneration and normal LID were reversely transcribed to the cDNAs. The cDNAs were labeled with the incorporations of fluorescent dUTP, for preparing the hybridization probes. The mixed probes were then hybridized to the cDNA microarray. After high-stringent washing, the cDNA microarray was scanned for the fluorescent signals and analyzed by computer image analysis. The apoptotic status and the expression of Bcl-2 and Bax in 12 cases of degenerative LID and 10 cases of normal LID were detected with TdT-mediated dUTP-biotin nick end labeling (TUNEL) and immunohistochemistry methods.ResultsAmong the 4096 targets, there were 10 genes related to apoptosis. The expression related to Bax protein gene was up-regulated and it was down-regulated for Bcl-2 protein. In group of normal LID, the average apoptotic index (AI) was (24.897±3.620); percentage of Bcl-2 positive cells was (31.440±4.150)%; percentage of Bax positive cells was (29.372±2.588)%, average optical density (OD) values of positive particles were (0.183± 0.010 ), ( 0.203 ±0.012) and (0.169±0.005) respectively. In group of degenerative LID, the average AI was (49.232±3.440); percentage of Bcl-2 positive cells was (18.239±2.470)%; percentage of Bax positive cells was (52.349±3.764)%; average OD values of positive particles were (0.152±0.003), (0.310±0.008) and (0.262±0.014) respectively. There were significantly differences in AI and expressions of Bcl-2 and Bax proteins between normal LID and degenerative LID (P<0.05).ConclusionCell apoptosis plays an important role in the process of LID degeneration. Both Bcl-2 and Bax take part in the occurrence and progression of LID.

ABSTRACT

Objective To observe the vascular collagen remodeling, apoptosis and Bcl-2/Bax of vascular smooth muscle cells in carotid artery on SHR . Methods 12 SHR treated with Irbesartan ,12 SHR without treated Irbesartan ,12 wistar rats were normotensive control group. The vascular volume fraction of Collagen were determined by picrosirius red staining .The expression of the proteins Bcl-2 and Bax were determined by Immunhistochemia,VSMC apoptosis was identified by in situ TDT-mediated dUTP nick end labeling(TUNEL). Results Significant Fibrosis exists in carotid artery of SHR. After 20 week's treating, the vascular volume fraction of Collagen and the ratio of wall/cavity were descended, but weren't parallel. The expression of the protein Bcl-2 and APOI were higher than the subjects of normotensive control, its reduced after treating; The expression of the proteins Bax was similar with control, it increased after treating. Conclusions The results suggest that vascular collagen remodeling exist, its maybe related with apoptosis , the proteins Bcl-2 and Bax of VSMC. The Irbesartan can treat it.

ABSTRACT

This study was undertaken to know the extent of apoptosis, expression of bcl-2 and proliferating cell nuclear antigen (PCNA) in uterine cervical intraepithelial neoplasia (CIN; 15 cases) and invasive carcinoma (27 cases) and to evaluate them as a prognostic marker. Apoptosis was analysed by using the in situ apoptosis detection kit and bcl-2 and PCNA were detected by the immunohistochemical method. The results were as follows: Apoptotic indices (AI) in the invasive carcinoma (mean: 4.3) were 10-times higher than that in the CIN (mean: 0.43). Bcl-2 was expressed 60% of the cases in the dysplastic cells of the CIN II and CIN III, 33.3% of cases in the invasive carcinoma and not expressed in the CIN I except basal cells. The expression of the PCNA was increased by the grades of CIN and was strong in invasive carcinoma. The mean survival time of the patient with invasive carcinoma was significantly decreased in the higher AI index (above 4.3) than in the lower AI index (below 4.3). There was no significant correlation between the extent of apoptosis and the expression of bcl-2. According to the above results, AI are able to be used as an independent prognostic marker in the invasive cervical carcinoma, and bcl-2 and PCNA have an important role in the tumorigenesis of uterine cervical carcinoma.

ABSTRACT

Objective To investigate the involvement of apoptosis in the lesions of patients with psoriasis. Methods The apoptosis was detected with terminal deoxynucleotidyl transferase mediated dUTP biotin nick end labeling (TUNEL), and the expression of p53, PCNA and apoptosis suppressing protein bcl 2 was assessed with immunoperoxidase technique in psoriatic lesions and normal skin. Results A large number of keratinocytes showing biochemical and morphologic features of cells undergoing apoptosis were observed in all the suprabasal layers of the psoriatic epidermis. The plaques from all patients analysed showed marked increase in the number of PCNA positive cells in the middle and basal keratinocytes, and a dramatic reduction in the number of bcl 2 positive cells in the basal cell layer. Conclusion The increased apoptosis of keratinocytes in the lesions of psoriasis might be a homeostatic mechanism to the hyperplasia of cells.

ABSTRACT

Objective:To investigate the mechanisms of Mitoxantrone(MTN) induced apoptosis in human acute myloid leukemia HL 60 cells. Methods:Exposured log phase growth HL 60 cells to different Mitoxantrone concentrations for different time respectively and then test the inhibitive effect by modified tertrozalium salt(MTT) assay.Morphologic evidence for apoptosis of MTN induced on HL 60 cells was determined by transmission electron microscope. The detection of MTN induced internucleosamal DNA fragmentation by agurose gele lectrophoresis. The effects on HL 60 cells proteins(Bcl 2,Bax) implicated in apoptosis were determined by Flow cytoetric.Results:(1)The antiproliferative effects were observed following exposure micromilligram level of MTN.It shows a very substantial differences in the dose and duration dependency of the observed antiproliferative and cytotoxic effects.(2)The HL 60 cells treated by MTN are observed apoptosis characteristic morphological changes such as complete cell membrane, nuclear fragmention apoptosic bodies and found about 200 bp trapezoidal belt with DNA electrophoresis.(3)The protein test results of Bcl 2 and Bax indicate that when the time of MTN treating HL 60 cells is the same,Bcl 2 protein and the ratio of Bcl 2 protein and Bax protein(Bcl 2/Bax) descend along with the ascending concentration of MTN.Bax protein has no connection with the concentration of MTN.When the concentration of MTN treating HL 60 cells are the same. The ratio of Bcl 2/Bax descends as the time of MTN treating HL 60 cells is prolonged. Conclusion:MTN has the function of inducing apoptosis on HL 60 cells with descent of Bcl 2 and Bcl 2/Bax.The regulation pathway of Bcl 2 may be a mechanism of MTN induced apoptosis on HL 60 cells.

ABSTRACT

Objective:To explore the change of the expression of apoptotic regulatory protein bcl 2 and bax in genetic hypertension and heart hypertrophy To evaluate the anti apoptotic effect of Valsartan Methods:Thirty SHRs were divided equally into two groups One group was treated with Valsartan 〔20 mg/(kg?d)〕, the other with placebo, with normal Wistar rats as controls The observation period was from 8 week old to 16 week old Expression of bcl 2 and bax in left ventricular myocytes was studied by the techniques of immunohistochemistry and Western blot Results:The expression of bax in SHR+placebo was markedly higher than that of Wistar or SHR+Valsartan, and so was the ratio of bax/ bcl 2 The expression of bcl 2 was similar in the three groups Conclusion:These data suggest that in response to chronic arterial pressure overload , cardiomyocyte specific apoptosis contributes to the transition from compensatory hypertrophy to decompensation Thus, apoptosis may be involved in the pathogenesis of congestive heart failure And Valsartan may reverse such decompensation by inhibiting the expression of bax
