ABSTRACT

OBJECTIVE To investigate the regulatory effects of couplet medicinals of Atractylodes macrocephala-Aucklandia lappa on gut microbiota and short-chain fatty acids (SCFAs) in the diarrhea-type irritable bowel syndrome (IBS-D) rats with spleen deficiency. METHODS The IBS-D rat model with spleen deficiency was induced by intragastric administration of Senna alexandrina combined with restraint stimulation. The model rats were divided into model group, positive control group (pinaverium bromide 1.5 mg/kg), A. macrocephala-A. lappa low-dose, medium-dose and high-dose groups (0.7, 1.4, 2.8 g/kg), with 6 rats in each group. Another 6 healthy rats were taken as the blank control group. The blank control group and the model group were given normal saline intragastrically, and other groups were given relevant drug liquid intragastrically, once a day, for consecutive 14 days. The general characteristics of rats and fecal water content were observed, and intestinal sensitivity [evaluating by abdominal wall withdrawal reflex (AWR) threshold] and the intestinal propulsion rate were determined. The serum levels of 5- hydroxytryptamine(5-HT)and SP were detected, and the pathological changes of colon tissue were observed; the protein expressions of 5-HT-3 receptor(5-HT3R), 5-HT4R and 5-HT transporter(SERT) in colon tissue of rats were detected. 16S rRNA sequencing was performed for the feces of rats in blank control group, model group and A. macrocephala-A. lappa high-dose group; the contents of acetic acid, propionic acid and butyric acid in the feces of the rats were determined. RESULTS Compared with the model group, the body weight after 7 and 14 days of medication, fecal water content, AWR threshold, and the protein expressions of 5-HT4R and SERT in colon tissue were increased significantly in the A. macrocephala-A. lappa medium-dose and high-dose groups (P<0.05 or P<0.01); serum contents of 5-HT and SP, intestinal propulsion rate (except for A. macrocephala-A. lappa medium-dose group), the protein expression of 5-HT3R in colon tissue were decreased significantly (P<0.01); diarrhea relief, mental state recovery, and partially recovery of the structure of colon tissue were all found; moreover, the diversity and species number of gut microbiota were reduced in A. macrocephala-A. lappa high-dose group and the content of butyric acid in fecal samples was significantly reduced (P<0.05). CONCLUSIONS The compatibility of A. macrocephala and A. lappa can improve intestinal motility and sensitivity of IBS-D model rats with spleen deficiency, and alleviate diarrhea. This may be related to improving changes in intestinal microbiota structure, reducing 5-HT expression and butyric acid content, and increasing 5-HT4R and SERT expression.

ABSTRACT

The medicinal part of Aucklandia lappa (Asteraceae) is its dried root, which is one of the commonly used Chinese medicinal materials. Here we reviewed sesquiterpene lactones isolated from A. lappa over the past ten years in the following aspects of pharmacological activities, pharmacokinetics, toxicology, structure–activity relationship. Pharmacological activities consist of anti-cancer, anti-inflammatory activity, anti-immunity activity, anti-oxidant activity, antimicrobial activity, spasmolytic activity and so on. The extractive, showing similar pharmacokinetics parameters, may exert their various biological activities by the interaction of their α-methylene-γ-butyrolactone moiety with the thiol groups of biomacromolecules through Michael-addition. However, the poor aqueous solubility, non-selective binding as a Michael acceptor at undesired targets limited clinical translation of this class. In order to evaluate the potential effect of the extractive applied in clinical trial, the present review outlines information on pharmacological activities, pharmacokinetics, toxicology, and structure–activity relationship, as well as the future research directions of the extractive for further development and utilization of A. lappa.

ABSTRACT

Objective:To explore the synergistic effect of arbuscular mycorrhizal(AM) fungi mixed inoculation on the growth, physiological and biochemical characteristics, root biomass and terpenoid component accumulation of Aucklandia lappa seedlings,so as to provide a reference for the combination and application of the dominant complementary effect mycorrhizal fungi. Method:The effect of different AM fungi combined with inoculation on the root mycorrhizal infection rate, plant growth, physiological and biochemical characteristics, root biomass, costunolide and dehydrocostus lactone of A. lappa seedlings were determined by pot inoculation at room temperature. Result:It was found that AM fungi could form good mycorrhizal symbiosis with the roots of A. lappa. The formation of mycorrhizal symbiosis system could increase the chlorophyll content of A. lappa leaves, increase the activities of catalase(CAT),peroxidase (POD),superoxide dismutase (SOD),reduce the content of malondialdehyde(MDA),and promote photosynthesis of A. lappa. Compared with CK group,AM fungus treatment could significantly promote the accumulation of costunolide and dehydrocostus lactone, and the accumulation of its metabolites, costunolide and dehydrocostus lactone, into roots during the symbiotic cultivation of A. lappa seedlings, indirectly improving the quality of medicines and yield of alantolactone. Conclusion:Inoculation of AM fungi can improve the root mycorrhizal viability, increase the absorption of nutrients and promote the growth of woody incense. The mixed inoculation treatment of S2,S4 and S5 had the best mycorrhizal effect in artificial cultivation, and the growth and medicinal quality of A. lappa were the best, which provided technical support for the application and popularization of A. lappa mycorrhizal biotechnology.

ABSTRACT

OBJECTIVE: To investigate the effects of Lindera aggregate of stir-baking with vinegar-Aucklandia lappa on gastric emptying and gastrointestinal hormones in functional dyspepsia liver depression and qi stagnation (FDLDQS) model rats. METHODS: Totally 60 SD rats were randomly divided into blank group (n=10) and modeling group (n=50); FDLDQS model was induced by chronic stress restraint or food deprivation or excessive fatigue in modeling group. After modeling, model rats were randomly divided into model group (normal saline), L. aggregate of stir-baking with vinegar group (1.62 g/kg, calculated by crude drug), A. lappa group (1.62 g/kg, calculated by crude drug), L. aggregate of stir-baking with vinegar-A. lappa group (1 ∶ 1, m/m,1.62 g/kg, calculated by crude drug) and mosapride group (positive control, 1.35 mg/kg), with 10 rats in each group. Administration groups were given relevant medicine intragastrically once a day; blank group and model group were given constant volume of normal saline intragastrically, for consecutive 14 d. 2 h after last medication, gastric emptying rate and small intestinal propulsion rate of rats in each group were measured by phenol red content method. After HE staining, the morphological changes of antrum tissue were observed under microscope. The contents of motilin (MTL), gastrin (GAS) and cholecystokinin (CCK) in serum were determined by ELISA method. RESULTS: Compared with blank group, gastric emptying rate and small intestinal propulsion rate of rats were decreased significantly in model group (P<0.01); the serum contents of MTL and GAS were decreased significantly (P<0.01), while the content of CCK was increased significantly (P<0.01). No organic damage was found in all model groups. Compared with model group, L. aggregate stir-baking with vinegar group and A. lappa group, gastric emptying rate and small intestinal propulsion rate of rats were increased significantly in L. aggregate of stir-baking with vinegar-A. lappa group and mosapride group (P<0.05 or P<0.01); serum contents of MTL and GAS were increased significantly (P<0.01), while the content of CCK was decreased significantly (P<0.01). CONCLUSIONS: L. aggregate of stir-baking with vinegar-A. lappa can accelerate gastric emptying and small intestinal propulsion rate of FDLDQS rats, increase serum contents of MTL and GAS but decrease the content of CCK; its effects are better than that of them alone.
