ABSTRACT

OBJECTIVE@#To evaluate the therapeutic effects of acupoint autohemotherapy (A-AHT) on 1-chloro-2,4-dinitrobenzene (DNCB)-induced atopic dermatitis (AD) in mice focusing on regulating T helper 1/T helper 2 (Th1/Th2) immune responses.@*METHODS@#Thirty BALB/c mice were divided into 5 groups by a random number table, including normal control (NC), AD model (AD), A-AHT, sham A-AHT (sA-AHT), and acupoint injection of normal saline (A-NS) groups, 6 mice per group. Mice were challenged by DNCB for the establishment of experimental AD model. On the 8th day, except for the NC and AD groups, the mice in the other groups received management once every other day for a total of 28 days. For the A-AHT and sA-AHT groups, 0.05 mL of autologous whole blood (AWB) was injected into bilateral Zusanli (ST 36) and Quchi (LI 11) and sham-acupoints (5 mm lateral to ST 36 and LI 11), respectively. The A-NS group was administrated with 0.05 mL of normal saline by acupoint injection into ST 36 and LI 11. Dermatitis severity for dorsal skin of mice was determined using the Severity Scoring of Atopic Dermatitis (SCORAD) every week. The total immunoglobulin E (IgE), interleukin-4 (IL-4), and interferon-gamma (IFN-γ) cytokine levels in serum were examined by enzyme-linked immunosorbent assay (ELISA). Spleen Th1/Th2 expression were analyzed via flow cytometry and immunohistochemical assay was used to detect T-box expressed in T cell (T-bet) and GATA-binding protein 3 (GATA3) expressions in skin lesions of mice.@*RESULTS@#Compared with the AD group, both A-AHT and sA-AHT reduced the SCORAD index and serum IgE level (P<0.05 or P<0.01); A-AHT, sA-AHT and A-NS down-regulated serum IL-4 level and upregulated IFN-γ/IL-4 ratio (P<0.05 or P<0.01); A-AHT regulated the Th1/Th2 shift specifically and increased the related transcription factors such as T-bet expression and T-bet/GATA3 ratio (P<0.05).@*CONCLUSION@#A-AHT showed significant effectiveness on the AD model mice, through regulating Th1/Th2 immune responses.

ABSTRACT

Acupoint autohemotherapy at bilateral Zusanli (ST36) and Xuehai (SP10) was used to treat a 26-year-old female patient who had suffered from recalcitrant atopic eczema (AE) for five years. The treatment was applied at a frequency of once per week for the first month, followed by a three-month period of once every other week. At the end of treatment, the patient's AE symptoms were entirely resolved, and by the end of a six-month follow-up her immunoglobulin E level had returned to the normal range. Further, there was no relapse of AE symptoms during the six-month follow-up. Therefore, we hypothesized that after the repeated treatments the local inflammatory reaction induced by autologous blood injection triggered a local immune response, followed by a systemic immune response after the repeated treatment, finally leading to the anti-inflammation and immunomodulation effects. This case suggests that acupoint autohemotherapy could be used as an effective complementary treatment for recalcitrant AE, especially in cases where other treatments have failed. Further comparative studies are needed to corroborate the value and mechanisms of this therapy.

ABSTRACT

Abstract Transmissible Venereal Tumor (TVT) is a common canine neoplasia; it is mainly located in the genital mucosa. This pathology is highly contagious and can be transmitted by direct contact cell transplantation from a diseased to a non-diseased mucosa. It is primarily diagnosed by cytology and its treatment is chemotherapy being the vincristine sulfate the medicine of choice. The Vincristine has a high rate of effectiveness and low recidivism; however, this alternative has important side effects. The treatment with Vincristine could cause primarily digestive upset, hematological, and nervous diseases. The objective of this work is to determine the efficacy and performance of alternative therapies for the treatment of TVT, firstly, with an autohemoterapy protocol, and using an immunotherapy treatment with yatrenic acid and casein, which acts in the animal's immune system and causes the same organism to eradicate the tumor.

ABSTRACT

OBJECTIVE@#Autologous blood acupoint injection has practiced for treatment of various diseases, but its therapeutic mechanism remains unknown. This study aimed to explore the histological changes in the acupoint area after blood injection.@*METHODS@#Blood samples (0.1 mL) collected from donator mice was injected in the recipient mice at the acupoint of left Zusanli (ST- 36). At 0, 6, 12, 24, 48, 72 and 96 h after the injection (3 mice at each time point), peripheral blood and muscular tissue were collected from the injection site for blood cell classification and histological study with HE staining.@*RESULTS@#White blood cells and lymphocytes tended to increase but platelets tended to decrease at the acupoint after blood injection, but these changes were not statistically significant. Within 24 h after blood injection, acute inflammation occurred at the acupoint, and the injected blood components were infiltrated by neutrophils; a small number of monocytes were observed at the injection site, where red blood cells gradually disappeared. From 24 to 96 h, the injected area showed chronic inflammation with infiltration by mononuclear cells, macrophages, and lymphocytes, and the number of fibroblasts increased and neutrophils decreased gradually over time; the local muscle fibers were destroyed, and proliferation of fibrous tissue could be seen.@*CONCLUSIONS@#Blood injection at the acupoints can mimic the process of hematoma absorption in the muscular tissue, and local inflammation is initiated to engulf and clear the blood components, which may be the basis for the therapeutic effects induced by acupoint stimulation.

ABSTRACT

Although packed red blood cells transfusions are important for treating patients with sickle cell anemia, this intervention may lead to immunological disturbs, such as alloimmunization by erythrocyte antigens. Approximately 50% of patients with sickle cell anemia receive blood transfusions during their life span, and about 5 to 10% of them require a chronic transfusion scheme. The red blood cell alloimmunization is a serious but common transfusion reaction. This condition could lead to delayed hemolytic transfusion reactions, contributing to increase comorbidities of the disease. Important measures to prevent these complications in patients are the use of previously phenotyped red blood cells, in addition to the phenotyping of red blood cells from the acceptor patient, determining the correct phenotypic profile and enabling the choice of red blood cells with corresponding antigens to the patient to be transfused. Extensive prophylactic red blood cell genotyping to select donors for patients receiving repeated transfusions over a long period of time is a compelling application of DNA-based blood typing. This is particularly relevant for patients with sickle cell disease where the rate of alloimmunization is high.

ABSTRACT

Ozone (O3) gas discovered in the mid-nineteenth century is a molecule consisting of three atoms of oxygen in a dynamically unstable structure due to the presence of mesomeric states. Ozone therapy has been utilized and heavily studied for more than a century. Its effects are proven, consistent, safe and with minimal and preventable side effects. Medical O3 is used to disinfect and treat disease. Mechanism of actions is by inactivation of bacteria, viruses, fungi, yeast and protozoa, stimulation of oxygen metabolism, activation of the immune system. Medication forms in a gaseous state are somewhat unusual, and it is for this reason that special application techniques have had to be developed for the safe use of O3. In local applications as in the treatment of external wounds, its application in the form of a transcutaneous O3 gas bath has established itself as being the most practical and useful method, for example at low (sub-atmospheric) pressure in a closed system guaranteeing no escape of O3 into the surrounding air. Ozonized water, whose use is particularly known in dental medicine, is optimally applied as a spray or compress. Diseases treated are infected wounds, circulatory disorders, geriatric conditions, macular degeneration, viral diseases, rheumatism/arthritis, cancer, SARS and AIDS.

ABSTRACT

The increasingly frequent practice of autohemotherapy entails a potential risk to the health of individuals since it is scientifically unproven. There are practically no clinical studies showing the efficacy and safety of this procedure; only experimental research with questionable results from studies on human beings and animals. Over the last years, the sanitary surveillance area has expanded its preventive and risk control actions based upon a precautionary philosophy in the private as well as the public sphere. By observing the theoretical and practical reference points of the "four Ps" (prevention, protection, precaution and prudence), in accordance with the epistemology developed within Intervention Bioethics, sanitary surveillance actions are based upon legal administrative policing powers. These powers are understood to be the Public Administration's competence to set conditions on and restrict the use and benefit of goods, activities and individual rights, on behalf of the common well being of people or the State itself. The Brazilian State, through its sanitary surveillance policing powers at the three levels of government, has intervened in the practice of autohemotherapy. Considering the responsible State intervention in situations of collective interest, vulnerability and susceptibility, the present study proposed to analyze the practice of autohemotherapy in the light of sanitary surveillance actions and their relationship with the "four Ps" of Intervention Bioethics.
