ABSTRACT

Introduction: In oral smear, a small condensed mass of sex chromatin usually located just inside the nuclearmembrane of the nucleus is called Barr body. The study of Barr bodies is advantageous for sex determination bypresence or absent. Oral mucosal smear of 150 students (79 female students and 71 male students in the agegroup of 17 to 32 years) from P.D.U. Govt. medical college, Rajkot were selected with aim to study oral mucosalcells for presence of sex chromatin in oral mucosal smear and to measure efficacy of oral smear in determinationof sex by presence of sex chromatin during 2012 to 2014.Method: Oral smear was prepared for sex determination by scrapping buccal mucosa with wooden spatula andobtained turbid suspension, these were smeared on a glass slide, fixed by mixture of Methanol and Glacialacetic acid in the ratio of 3:1 for 10 min and stained by Carbol Fuschin for 15-20 min at room temperature, aftertaking informed written consent from students. Smears were examined with Compound light uniocular microscopeunder 100x magnification (Oil immersion), cells and nuclei were easily identified.Observations and Results: One hundred cells were counted in each slide. Mean value of Barr body positive cellsin male was 0.647 and Mean value of Barr body positive cells in female was 35.215 and range of percentage ofBarr body positive cells in male was 0-5% and range of percentage of Barr body positive cells in female was 0-55%. Presence of sex chromatin in oral mucosal cell of female was statistically significant. (P value < 0.05).Conclusion: Mean value of Barr body positive cells in male was 0.647 ± 1.148, Mean value of Barr body positivecells in female was 35.215 ± 10.28, range of percentage of Barr body positive cells in female was 0-55% and rangeof percentage of Barr body positive cells in male was 0-5%. Presence of sex chromatin in oral mucosal cell offemale was statistically significant. (P value<0.05) Mean values of sex chromatin positive oral mucosal cells ofmale was lower than mean value of sex chromatin positive oral mucosal cells in female, that is the supportingfact that sex chromatin is present in female in higher frequency. Sex chromatin can be used as simple and easilyavailable method for sex determination.

ABSTRACT

The need to identify bodies that are found as a result of disappearances with a diversity of causes, illegal burials and massive disasters, represent a wide percentage of dentistry practice on forensic research. The following study determined the performance of Barr Body Test, in fibroblasts of healthy teeth, under different conditions of burial (in vitro) with variations in pH, humidity and salinity in terms of general accuracy and sensitivity for men and women. Analyzed sample considered 47 dental pulps, taken from teeth under burial conditions during a period of a month. From dental pulps samples, 265 histological cuts valid for this study, were obtained, which were observed with an optical microscope under conventional H/E staining. Results showed a 98.9% of well-diagnosed cases, which correspond to the overall accuracy of the method. Sensitivity for men was 97.5% and 100% for women, over the analyzed sample. In low humidity conditions, 3 samples of badly diagnosed cases in men were observed, with a group accuracy of a 90%, with a sensitivity of 25% for men and 100% for women. The present study establishes that based on these results, the performance of Barr Body Test in fibroblasts, proposed for healthy pulp teeth, is not affected by burial conditions in terms of pH (acid-alkaline), salinity (high-low) and high humidity.


La necesidad de identificar cuerpos que resultan como consecuencia de desapariciones de causas variadas, inhumaciones ilegales y desastres masivos representa un porcentaje amplio en el quehacer odontológico en un escenario de investigación forense. El presente estudio determinó el rendimiento de la prueba diagnóstica de observación del cuerpo de Barr en células de la pulpa de dientes sanos, sometidos a distintas condiciones de enterramiento (in vitro) con variación de pH, humedad y salinidad en términos de exactitud general y sensibilidad para hombres y mujeres. La muestra analizada consideró 47 pulpas dentales, extraídas de dientes sometidos a condiciones de enterramiento durante un mes. De las pulpas dentarias se obtuvieron 265 cortes histológicos válidos para el estudio, los cuales mediante la tinción convencional H/E, fueron observados al microscopio óptico. Los resultados arrojaron un 98,9% de casos bien diagnosticados, que correspondió a la exactitud general del método. La sensibilidad para hombres fue de 97,5% y para mujeres de 100% sobre el total de la muestra analizada. Las condiciones de pH (ácido y alcalino), salinidad (alta y baja) y alta humedad presentaron una exactitud de grupo de 100%, con una sensibilidad para hombres y mujeres de 100%. En la condición de baja humedad se observaron 3 muestras de hombres mal diagnosticadas con una exactitud de grupo de 90% y sensibilidad para hombres de 25% y para mujeres de 100%. A la luz de los resultados, el presente estudio establece que el rendimiento de la prueba diagnóstica de observación del cuerpo de Barr en fibroblastos, propuesto para pulpas de dientes sanos, no se afecta con las condiciones de enterramiento propuestas bajo pH ácido ­ alcalino, salinidad alta ­ baja y humedad alta.

ABSTRACT

Determination of sex is useful in Forensic Medicine. Barr bodies can be seen hair root sheath cells. The presence of barr bodies indicates that sex of the person is female. They are inactive X chromosome. The Barr bodies are also known as sex chromatin. Hair can present in the crime scene. Hair is trace evidence. The stain used was aceto-orcein. The percentage of barr bodies was found to be 28-49 %. The study was carried out for eight months and it was found that barr bodies persisted for eight months. It can be present anywhere at the crime scene. Hair of the accused can be present in hands of victim due to cadaveric spasm. Hair of victim can be found on the clothes of the accused. There is average fall in percentage of barr bodies with the passage of time due to the start of decomposition changes in the root sheath cells.

ABSTRACT

Buccal mucosal smears of 100 female patients of carcinoma of breast were compared with 100 controls matched accordingly. The frequency of Barr bodies was significantly lower in carcinoma of breast patients (menstruating and menopausal women) P < 0.001 when compared with controls indicating reactivation of the inactive X chromosome.
