ABSTRACT

Abstract The aim of this study was to evaluate different low-cost culture media for biomass production of 3 potential probiotic L. salivarius strains, which could be destined to broilers at farms. Different formulated media based on whey permeate (WP) supplemented with nitrogenous sources were evaluated: yeast extract (YE), whey hydrolysate (WH) and MnSO..H.O (Mn), MgSO..7H.O (Mg). The growth of each strain in the formulated media and the cost was compared with their growth and cost in commercial medium (MRS). L. salivarius DSPV008P did not grow adequately in any of the formulated media. On the other hand, addition of YE and Mn in the formulated media increased L. salivarius DSPV002P and L. salivarius DPSV011P growth. In contrast, WH and Mg addition increased the L. salivarius DSPV002P biomass only. L. salivarius DSPV011P was the only strain that had similar growth performance in MRS as in the selected medium: WP + YE 8g/l + Mn. In this sense, L. salivarius DSPV011P reached a biomass of 9.22 Log (CFU/ml) in the selected formulated medium, with a low-cost growth medium 12 times less than in MRS. Although the effect of supplements added to the culture medium on kinetic parameters are strain dependent, L. salivarius DSPV011P is the strain with the best technological characteristics, capable of growing in a medium based on a by-product of the dairy industry supplemented with YE and Mn and at a much less cost than in MRS medium.


Resumen El objetivo de este trabajo fue evaluar diferentes medios de cultivo de bajo costo para la producción de biomasa de 3 cepas potencialmente probióticas de L. salivarius, las cuales podrían ser destinadas a pollos parrilleros en las granjas. Para ello se evaluaron diferentes formulaciones basadas en permeado de suero de queso (WP) suplementado con fuentes nitrogenadas: extracto de levadura (YE) e hidrolizado de suero (WH) y MnSO..H.O (Mn), MgSO..7H.O (Mg). El crecimiento de las cepas en estas formulaciones y el costo económico fue comparado con el crecimiento y costo en el medio de cultivo comercial (MRS). L. salivarius DSPV008P no creció adecuadamente en ninguno de los medios evaluados. Por otro lado, la adición del YE y Mg al medio mejoró el desarrollo microbiano de L. salivarius DSPV002P y L. salivarius DPSV011P. El agregado de WH y Mn solo tuvo un efecto positivo en el incremento de la biomasa de L. salivarius DSPV002P. L. salivarius DSPV011P fue la única cepa que desarrolló la misma cantidad de biomasa en MRS y en el medio seleccionado WP + YE 8 g/L + Mn. L. salivarius DSPV011P logró un desarrollo de biomasa de 9.22 Log (UFC/ml) en el medio seleccionado con un costo económico 12 veces menor que en MRS. Aunque el efecto de los suplementos añadidos al medio de cultivo sobre los parámetros cinéticos depende de la cepa, L. salivarius DSPV011P es la cepa con mejores características tecnológicas, capaz de crecer en un medio a base de un subproducto de la industria láctea suplementado con YE y Mn y a un costo mucho menor que en MRS.

ABSTRACT

Benthic microalgae have the natural capacity to adhere to a diversity of fixed submerged substrates to form biofilms, which have important roles not only in natural ecosystems, but also in aquaculture systems. An experimental investigation was performed to assess the biofilm-forming capacity of two microalgae (Navicula incerta and Navicula sp.) on three different substrates (plastic net, fabric, and wood) under controlled temperature and light conditions. The substrates were arranged on curtains suspended from a wood stick, into plastic aquariums (45 L in capacity) filled with filtered marine water enriched with F/2 medium. The trial was carried out until the exponential growing phase of the microalgae was reached. After that, the incorporated biomass was gravimetrically calculated, and its biochemical composition was determined by standard methods. The greatest amount of incorporated dry matter was observed for Navicula sp. on fabric and the lowest was observed for wood. The highest number of cells associated with the biofilm was obtained for Navicula sp. on the plastic net (1.24 x 109 cells/m2), while the lowest was recorded for Navicula sp. on the wood (1.43 x 108 cells/m2). Significant differences in organic matter were found among the substrates, with the highest values for N. incerta on the fabric (3.22 g/m2) and the lowest for Navicula sp. on the wood (0.02 g/m2). The best biochemical profiles among the formed biofilms were observed for N. incerta on the plastic net and Navicula sp. on the fabric. The plastic net was considered the best substrate because of the stability of the biofilm and the easiness of harvesting the biomass.


Las microalgas bentónicas tienen la capacidad natural de adherirse a diversos sustratos fijos sumergidos para formar biopelículas, las cuales tienen roles importantes no solo en ecosistemas naturales sino también en sistemas de producción acuícolas. Se llevó a cabo una investigación experimental para evaluar la capacidad formadora de biopelículas de dos microalgas bentónicas (Navicula incerta y Navicula sp.) en tres diferentes sustratos (malla plástica, tela y madera), bajo condiciones controladas de temperatura y luz. Los sustratos fueron arreglados a manera de cortinas suspendidas de un tubo de PVC dentro de acuarios de plástico (45 L de capacidad) con agua marina enriquecida con el medio F/2. El experimento se llevó hasta que la fase de crecimiento exponencial de la microalga fue alcanzada. Posteriormente la biomasa incorporada fue calculada gravimétricamente, y su composición bioquímica fue determinada por métodos estándar. La mayor cantidad de materia seca se observó para N. incerta en el sustrato de tela y la menor se encontró en el de madera. El mayor número de células asociadas a la biopelícula fue registrado para Navicula sp. en malla plástica (1.24 x 109 cel/m2), mientras que el menor se encontró para Navicula sp. en madera (1.43 x 108 cels/m2). Diferencias significativas en cuanto a materia orgánica se encontraron entre los sustratos y las especies, con valores más altos para N. incerta en tela (3.22 g/m2) y más bajos para Navicula sp. en madera (0.02 ± 0.05 g/m2). Los mejores perfiles bioquímicos para las biopelículas correspondieron a las formadas por N. incerta sobre malla plástica y Navicula sp. sobre tela. La red de plástico se consideró el mejor sustrato debido a la estabilidad de la biopelícula y la facilidad para cosechar la biomasa.

ABSTRACT: The water-driven AquaCrop model to simulate yield response has been calibrated and validated for soybean cultivated under different water levels irrigation in Matopiba region, Brazil. The crop was submitted to seven irrigation treatments during the dry season and a dry treatment in the rainy season. The model was parameterized and calibrated by using soybean yield data collected at field level. Model performance was evaluated by using the following statistical parameters: prediction error (Pe), Nash-Sutcliffe efficiency index (E), coefficient of determination (R2), mean absolute error (MAE), root mean square error normalized (RMSEN) and Willmott's index (d). The statistical analyses of the AquaCrop model calibrated for the Matopiba region disclosed error acceptable for yield prediction of soybean grown under tropical climate conditions. Results also indicated that the C2 soybean cultivar is more resistant to water stress than the C1 soybean grown in the Matopiba region, Brazil. In the treatments when the crop was well supplied with water, at least in one phase, the yield was greater than those with drought stress at last in one phase.


RESUMO: O modelo AquaCrop foi calibrado e validado para simular a produtividade da soja cultivada sob diferentes lâminas de irrigação na região de Matopiba, Brasil. A cultura foi submetida a sete tratamentos de irrigação durante a estação seca e um tratamento de sequeiro na estação chuvosa. O modelo foi parametrizado e calibrado utilizando dados de produtividade de soja coletados diretamente em campo. O desempenho do modelo foi avaliado utilizando os seguintes parâmetros estatísticos: erro de predição (Pe), índice de eficiência de Nash-Sutcliffe (E), coeficiente de determinação (R2), erro médio absoluto (EMA), raiz quadrática do erro médio normalizado (EQEMN) e índice de Willmott (d). As análises estatísticas do modelo AquaCrop calibrado para a região de Matopiba apresentaram erros aceitáveis na predição da produtividade de soja cultivada sob condições climáticas tropicais. Os resultados também indicaram que a cultivar de soja C2 é mais resistente ao estresse hídrico do que a soja C1 cultivada na região de Matopiba, Brasil. Nos tratamentos durante os quais a cultura foi bem abastecida com água, em pelo menos em uma fase, a produtividade foi maior, em vista do estresse hídrico que ocorre em uma fase.

ABSTRACT

Background: The yeast strain IB09 was isolated from the gut of Calosoma sp. (Carabidae, Coleoptera, Insecta) that were collected in the central Amazon rainforest. First, tolerance of the strain to ethanol and heat was tested. Then, IB09 was cultivated in a medium using sugarcane bagasse hemicellulosic hydrolyzate as a carbon source, and cell growth (OD600), specific growth rate (uMAX, h-1), biomass yield (Y B, g.g-1) and relative sugar consumption (RSC, percent) were evaluated. Taxonomic identification was determined by sequencing the ITS1 region of IB09 and comparing it to sequences obtained from the GenBank database (NCBI). Results: IB09 showed both ethanol tolerance and thermotolerance. Relative sugar consumption indicated that IB09 was able to perform saccharification of sugarcane bagasse hemicellulosic hydrolyzate, increasing the total reducing sugar concentration by approximately 50 percent. The μMAX value obtained was 0,20, indicating that cell growth was slow under the assessed conditions. Biomass yield was 0,701 g per g of consumed sugar, which is relatively high when compared with other findings in the literature. After 120 hrs of cultivation, 80,1 percent of total reducing sugar had been consumed. Sequencing of the ITS1 region identified IB09 as Trichosporon mycotoxinivorans. Conclusion: This is the first report to document this species in the central Amazon rainforest at this host. Trichosporon mycotoxinivorans has great biotechnological potential for use in the saccharification of sugarcane bagasse hemicellulosic hydrolyzate and for biomass production with this substrate as carbon source.

ABSTRACT

The effect of soil polluted by diesel oil on the germination of seeds and the growth of Schinus terebinthifolius Raddi seedlings was analyzed at different times after contamination of the soil. The experiments were conducted under greenhouse conditions, with four treatments and five repetitions. The four treatments included: soil contaminated 30 (T30), 90 (T90) or 180 (T180) days before planting as well as a non-polluted soil (T0) (control). Soil saturated to 50 percent of its maximum retention capacity (MRC) was contaminated with diesel oil at a rate of 92.4 mL per kg. The germination rate and germination speed index (GSI) were significantly affected only in T30. The development of the plants was affected significantly in all the treatments, with reductions of biomass and eophyll area. It could be concluded that diesel oil significantly affected the germination, GSI and seedling growth of S. terebinthifolius, but the toxic effect decreased over the time.

ABSTRACT

Response surface methodology (RSM) was employed to study the effects of various medium components on biomass production by Lactobacillus rhamnosus E/N. This strain is commonly used in the pharmaceutical and food industries due to its beneficial effect on the human gut and general health. The best medium composition derived from RSM regression was (in g/l) glucose 15.44, sodium pyruvate 3.92, meat extract 8.0, potassium phosphate 1.88, sodium acetate 4.7, and ammonium citrate 1.88. With this medium composition biomass production was 23 g/l of dry cell weight after 18 h of cultivation in bioreactor conditions, whereas on MRS the yield of biomass was 21 g/l of dry cell weight. The cost of 1 g of biomass obtained on MRS broth was calculated at the level of 0.44 € whereas on the new optimal medium it was 25 percent lower. It may be concluded then, that the new medium, being cheaper than the control MRS allows large scale commercial cultivation of the L. rhamnosus strain. This study is of relevance to food industry because the possibility to obtain high yield of bacterial biomass is necessary step in manufacturing of probiotic food.

Introducción: Artemisia annua L es una planta medicinal de gran interés y es importante precisar los aspectos agrícolas que garanticen su adecuado cultivo. Objetivos: determinar la influencia del cultivo a pleno sol y a la sombra y de la cosecha a diferentes horas y edades de la planta en la producción de biomasa y de los principios activos. Métodos: se utilizaron canteros a plena exposición solar y con determinada disminución de la luz para estudiar las condiciones de cultivo y en las investigaciones del momento de cosecha, en la recolección a diferentes horas del día: 9:00 am, 26 ºC; 11:00 am, 30 ºC; 1:00 pm, 32 ºC; 3:00 pm 33 ºC se seleccionaron plantas cultivadas a pleno sol, de 6 meses de edad; en el otro experimento se evaluó en plantas procedentes de semilleros establecidos en 2 fechas: 5 de noviembre y 8 diciembre, la recolección de la parte aérea a diferentes edades: 5, 6 y 7 meses. Se valoró la producción de biomasa, el contenido de artemisinina y adicionalmente el de aceite esencial y el de alcanfor. La extracción del aceite esencial se realizó por hidrodestilación y la de artemisinina y alcanfor por cromatografía gaseosa, la determinación de la presencia de estos dos componentes se realizó por cromatografía en capa delgada y además se valoraron los parámetros farmacognósticos: sustancias solubles en agua y en alcohol. Resultados: se encontraron pocas diferencias en cuanto al rendimiento de la parte aérea en las dos formas de cultivo, pero sí se obtuvo mayor contenido de artemisinina, aceite esencial y alcanfor en el cultivo a pleno sol. Introduction: Artemisia annua L is medicinal plant of great interest, and therefore it is important to specify exactly the agricultural elements supporting its proper growing. Objectives: to determine the influence of growing this plant in the sun and in the shade and of harvesting at different hours and plant ages in the production of biomass and active principles. Methods: flowerbeds were placed in the sun and in dimmed light conditions in order to study the growing conditions. In the research conducted at the time of harvest, plants grown in the sun, of 6 months of age and at various day hours: 9:00 am at 26 ºC; 11:00 am at 30 ºC; 1:00 pm at 32 ºC, and 3:00 pm at 33 ºC were harvested. In the other experiment, the harvest of the aerial parts of the plants coming from seedbeds created on November 5th and on December 8th and aged 5, 6 and 7 months was evaluated. The production of biomass, the content of artemisinin and additionally that of the essential oil and camphor were assessed. The extraction of the essential oil using hydrodistillation and that of artemisinin and the camphor by gas chromatography, the detection of the these two components was made by thin-layer chromatography and also the pharmacognostic parameters such as water- and alcohol-soluble substances were evaluated. Results: there were little differences in aerial parts yielding in the two growing methods but there was high content of artemisinin, essential oil and camphor in plants grown in the sun. ABSTRACT

The impact of elevated CO2 concentration on the growth, photosynthesis and biomass production was investigated in three oat (Avena sativa L) cultivars viz. Kent, JHO-822 and JHO-851 by growing under three environmental conditions i.e. elevated CO2 at 600 ± 50 μ mol mol-1 (C600), OTC with ambient CO2 (COTC) and under open field condition (Ca). Plant height and leaf area increased in the elevated CO2 grown plants. JHO-822 attained maximum height under C600 followed by Kent and JHO-851. The specific leaf mass (SLM) and specific leaf area (SLA) were also influenced significantly when the plants were grown under C600. Kent showed highest SLM under C600 corresponding lower value of SLA. The accumulation of soluble protein in the oat leaves decreased under C600 except JHO-822 where marginal increase in soluble protein was recorded under C600. JHO-822 showed an increase in Chl orophyll a, b and total in C600 over Ca, whereas other two cultivars did not follow any specific trend in the pigment accumulation. Our results confirmed that the net phosynthetic rate (PN) increased by 37% in Kent followed by JHO 822 under elevated CO2 over the control. This strong association of PN with gs was evidenced by a positive significant correlation (r=0.885**). A clear stimulatory effect at elevated CO2 was detected in all the cultivars in term of green and dry matter production than at ambient CO2 and COTC. A large increase in PN in the present investigation was accompanied by relatively small decrease in gs, which limits the water loss through transpiration rate. The elevated CO2 induced changes in gs and reduction in transpiration.

ABSTRACT

Growth of Spirulina maxima was studied in three types of culture conditions with four replicates each: 1) manual aeration with natural sunlight; 2) manual aeration with artificial light; and 3) constant aeration with an aquarium compressor and artificial light. After 185 days of incubation, growth declined in the first two treatments, while in the third treatment, higher growth was observed with average optical density of 3.7 against 1.8 and 1.9 in the first and second treatment, respectively. This was probably due to the fact that under constant aeration the salts were suspended avoiding the crystallization what could cause a decrease in the availability of the necessary nutrients for the growth. Also, the constant stirring allowed all the cells to receive the same amount of light promoting photosynthesis and consequently, a larger growth and characteristic green coloration. Culture with constant aeration under artificial light should be used for S. maxima cultivation because, besides reducing labor hours, it could be a more effective method for improving the economic income.


O crescimento de Spirulina máxima foi estudado em três condições de cultivo: 1) agitação manual com iluminação natural; 2) agitação manual com iluminação artificial e; 3) agitação constante feita por compressor de aquário e iluminação artificial. O maior crescimento foi observado nesta última condição, onde após 185 dias foram observados valores médios de densidade ótica de 3,7 enquanto, valores de 1,8 e 1,9 foram obtidos no primeiro e segundo experimento, respectivamente. Diferentemente do ocorrido com os outros tratamentos, não foi observado declínio no crescimento após 185 dias, o que foi atribuído ao fato de que, sob constante agitação os sais ficam suspensos e não cristalizam, fato que acarretaria diminuição da disponibilidade de nutrientes necessários ao crescimento. Também a agitação constante permite que todas as células recebam a mesma iluminação, promovendo fotossíntese e consequentemente um maior crescimento e coloração verde característica. Conclui-se que o emprego do compressor de aquário é não somente viável, mas recomendável, pois além de diminuir a mão de obra ainda se mostrou mais eficaz, melhorando o rendimento.

ABSTRACT

El pasto Vidal (Bothriochloa saccharoides) podría ser una buena opción forrajera en sistemas silvopastoriles del trópico, ya que tolera una alta cobertura arbórea, sin embargo, esta gramínea no ha sido suficientemente estudiada. Este estudio tuvo como objetivo analizar indicadores agronómicos y otras características de B. Sacharoides bajo dos niveles de sombra simulada: 30%; 50% y total exposición a la radicación solar 0%. La producción de materia seca del Bothriochloa saccharoides en época húmeda fue de 3.47, 3.0 y 5.56 y, para la época seca, de 2.33, 2.44 y 2.5 ton/ha para las coberturas de 50%, 30% y control, respectivamente; no hay diferencias significativas (p <= 0.05) para las coberturas, lo cual sugiere un potencial del pasto para su uso en sistemas con cobertura arbórea. Estos resultados demuestran un comportamiento productivo similar en las diferentes coberturas, lo cual lo hace un pasto tolerante a la sombra y una buena opción para la utilización en sistemas silvopastoriles.


Silver Bluestern (Bothriochloa saccharoides) grass could be is a good feeding option managed under tropical silvopastoral systems, becausesince it is tolerant to a high degree of tree coverage; however, this grass has not been sufficiently studied. This study aimed to analyze agronomic indicators and other characteristics from of Bothriochloa saccharoides under two levels of simulated shade: 30%; 50% and total exhibition to sun radiation (0% of shade). Production of dry matter of the Bothriochloa saccharoides, duringin the humid season was 3.47, 3.0 and 5.56 ton/ha, and induring the dry season it was 2.33, 2.44 and 2.5 ton/ha, for the simulated coverage of 50%, 30% and control test, respectively. There are not significant differences (p <= 0.05) between treatments; it, which suggests a potentiality from use for this grass to use it in pastures with trees. These results demonstrated a similar productive performance under different simulated shade conditions, that which makinges it a tolerant grass to the shade and a good option in silvopastoral systems.

ABSTRACT

The objective of this study was to compare density, vegetative cover, basal area, height growth, aboveground biomass production, frequency, floristic composition, and species diversity of understory vegetation growing in Hungarian oak (Quercus frainetto Ten.), Oriental beech (Fagus orientalis L.) and a mixed stand of both species (90% Hungarian oak and 10% Oriental beech) with the same crown closure. Understory vegetation was sampled with a quadrat of 0.5 m x 1 m along the 25 m-long transect lines in each stand. A total of seventeen perennial plant species from thirteen different families were found in the three stand types and all of them were present in Hungarian oak stand, eleven in Oriental beech stand, and seven in the mixed stand of Hungarian oak and Oriental beech trees. Results showed that stand types had a statistically significant effect on density, vegetative cover, basal area, height growth, biomass production, frequency, floristic composition, and species diversity of understory vegetation. Total understory vegetation density was the highest for Hungarian oak stand with about 136 plants m-2 and the lowest for the mixed stand of Hungarian oak and Oriental beech with 44 plants m-2. In addition, the Hungarian oak stand had the most diverse understory vegetation with about 4.3 different plant m-2, followed by the Oriental beech stand with 3.7 plants m-2, and the mixed stand of the Hungarian oak and Oriental beech with 2.7 plants m-2. Species density and diversity were the highest in Hungarian oak stand whereas aboveground biomass production and height growth of understory vegetation were highest in the mixed stand.

ABSTRACT

This paper deals with variations in biomass and in the production of Eichhornia crassipes (water hyacinth) in a eutrophic tropical reservoir. Net production was assessed by both a traditional method and a demographic approach. The initial biomass ranges from 96 g DW.m-2 to a maximum of 2027 g DW.m-2. Net production estimates varied greatly according to which method was used to calculate them (traditional: 20.6 ton DW.hectare-1.year-1, demographic: 226.9 ton DW.year-1).


Este estudo mostra as variações da biomassa e da produção de Eichhornia crassipes em um reservatório eutrófico tropical. A produção líquida foi estimada por intermédio do método tradicional e do demográfico. A biomassa inicial de 96 g PS.m-2chegou ao máximo de 2.027 g PS.m-2 A produção líquida variou de acordo com o método utilizado para sua estimativa (tradicional: 20,6 ton PS. ha-1.ano-1, demográfico: 226,9 ton PS.ano-1).
