ABSTRACT

The objective of this research was to evaluate the response of wheat plants to the application of possible elicitor compounds against Bipolaris sorokiniana pathogen. This response was measured through the quantification of antioxidant enzymes, malondialdehyde and flavonoids, evaluation of the severity of brown spot disease and productivity in wheat, greenhouse and field crops. The treatments consisted of suspensions of endophytic fungi Aspergillus japonicus and Trichoderma tomentosum, salicylic acid, acibenzolar-S-methyl and fungicide. In the field trials, in 2015 and 2016, the development of the disease was lower and productivity was higher in all treatments, with emphasis on the fungicide. However, endophytic fungi suspensions demonstrated potential as growth promoters, disease severity reducers and protective antioxidant response activators, as they promoted significant increase in superoxide dismutase, catalase, glutathione and flavonoid enzymes.(AU)


O objetivo desta pesquisa foi avaliar a resposta de plantas de trigo diante da aplicação de possíveis compostos elicitores perante o patógeno Bipolaris sorokiniana. Tal resposta foi examinada por meio da quantificação de enzimas antioxidantes, malondialdeído e flavonoides, da análise da severidade da doença mancha marrom e da produtividade na cultura do trigo, em casa de vegetação e em campo. Os tratamentos consistiram em suspensões de fungos endofíticos Aspergillus japonicus e Trichoderma tomentosum, ácido salicílico, acibenzolar-S-metil e fungicida. Nos ensaios em campo, em 2015 e 2016, o desenvolvimento da doença foi menor e a produtividade foi superior em todos os tratamentos, com destaque para o fungicida. No entanto, as suspensões de fungos endofíticos demonstraram potencial como promotores de crescimento, redutores da severidade da doença e ativadores de resposta antioxidante protetora, pois promoveram o aumento significativo das enzimas superóxido dismutase e catalase, das glutationas e flavonoides.(AU)

ABSTRACT

Objective: To study the chemical structure and activity of secondary metabolites of the endophytic fungus Bipolaris sorokiniana A606 isolated from Pogostemon cablin. Methods: The compounds were isolated and purified from the ethyle acetate extract of the fungal fermentation broth by silica gel, reverse phase silica gel, Sephadex-LH20, HPLC, and so on. Their structures were identified by extensive spectroscopic analysis. The compounds were evaluated for their cytotoxic activities against human tumor cell lines by the SRB assay and their antibacterial activities against bacteria were evaluated by the filter paper method. Results: Two compounds were isolated from the fermentation broth extract of the strain A606 and identified as (3S*,4aS*,6aS*,12bS*)-3-(2-hydroxypropan-2-yl)-6a,12b-dimethyl-9-[(2R*,4R*)-4-methyl-3-oxohexan-2-yl]-1,2,3,4a,5,6-hexahydropyrano [3,2-a]xanthene-8,11-dione (1) and precochlioquinol D (2). Conclusion: Compound 1 is a new compound, named 11-hydroxy-12,13-dehydrocochlioquinone B. Compound 2 is isolated from the species of genus Bipolaris for the first time. Compound 1 shows the cytotoxic and antibacterial activities.

ABSTRACT

Barley (Hordeum vulgaris L.) is the second most important winter crop in Southern Brazil. The excessive rainfall in this region during the crop-growing season increases the frequency and intensity of foliar fungal diseases. The research aimed to determine the damage function equations (DFE) for the multiple pathosystem of barley brown spot and powdery mildew based on the relationship between grain yield and diseases intensity at different 'BRS Cauê' cultivar growth stages (GS) during 2009 and 2010 growing seasons in Southern Brazil. The experiments were arranged in a randomized complete block design with nine treatments and four replicates. The disease gradients were generated by strobilurins and triazols fungicides rates and number of applications on barley cv. Cauê. The fungicide applications and disease incidence and severity assessments were performed at the 22, 31, 39, 45 and 56 plant GS. The DFE were obtained by variance analysis and linear regression between grain yield and diseases intensity. Significant and negative DFE were obtained and the damage coefficients (DC) varied from 29.48 to 100.08 (2009) and from 36.08 to 113.57kg ha-1 (2010) for incidence, and from 219.5 to 6,276.6 (2009) and 102.3 to 5,292.5kg ha-1 (2010) for severity. The largest damage coefficients were obtained when diseases assessments were made on GS 22 and 31 on both growing seasons evaluated. DFE were used to calculate the economic damage threshold (EDT) as a criterion to indicate the fungicide application moment to control the diseases in cultivars similar to 'BRS Cauê' in Southern Brazil.


A cevada (Hordeum vulgaris L.) é a segunda mais importante cultura de inverno no Sul do Brasil. Nessa região, o excesso de chuvas durante a estação de crescimento da cultura favorece o aumento na frequência e intensidade de doenças foliares. O trabalho objetivou determinar as equações de função de dano (EFD) para o patossistema múltiplo mancha-marrom e oídio da cevada pela relação entre rendimento de grãos e intensidade foliar das doenças em diferentes estádios fenológicos da cultivar de cevada 'BRS Cauê', nas safras 2009 e 2010 no sul do Brasil. O delineamento foi em blocos casualizados com nove tratamentos e quatro repetições. Os tratamentos foram constituídos de diferentes doses e número de aplicações dos fungicidas triazóis e estrobilurinas para gerar os gradientes de intensidade das doenças. As aplicações de fungicidas e as avaliações da incidência e severidade foliar foram realizadas nos estádios fenológicos (EF) 22, 31, 39, 45 e 56. As equações foram obtidas pela análise de variância e regressão linear entre rendimento de grãos e intensidade das doenças. As EFD foram significativas e negativas e os coeficientes de danos variaram de 29,48 a 100,08kg ha-1 (2009) e 36,08 a 113,57kg ha-1 ( 2010) para a incidência e de 219,5 a 6.276,6kg ha-1 ( 2009) e 102,3 a 5.292,5kg ha-1 (2010). Os maiores coeficientes de dano foram obtidos nos estádios EF 22 e 31em ambas as safras. As EFD foram usadas para calcular o limiar de dano econômico, que é um critério que indica o correto momento de aplicação de fungicidas para o controle das doenças em cultivares similares a 'BRS Cauê' no sul do Brasil.

ABSTRACT

O objetivo do trabalho foi avaliar o efeito dos fungicidas difenoconazole e carboxin + thiram aplicados em sementes de trigo cultivares 'CD 104' e 'OR Mirante', inoculadas com Cochliobolus sativus. O delineamento experimental foi inteiramente casualizado, em esquema fatorial 2x3 (2 cultivares e 2 fungicidas + testemunha sem controle) em sete repetições. O fungo foi isolado e reproduzido em meio de cultura BDA (batata-dextrose-ágar) e, posteriormente, multiplicado em sementes de trigo pré-cozidas, constituindo a fonte de inóculo. As avaliações foram do número de lesões na folha basal de cinco plantas por vaso aos 22, 25, 29, 31, 35 e 38 dias após a semeadura (DAS) e aos 41 dias foram avaliados o comprimento (cm), massa fresca (g) e seca (g) da parte aérea e das raízes. A partir das avaliações do número de lesões, também foi calculada a área abaixo da curva de progresso da doença (AACPD), cujos dados foram submetidos à análise de variância e as médias comparadas pelo teste de Tukey, a 5% de probabilidade. A cultivar 'CD 104' foi mais suscetível ao patógeno do que 'OR Mirante'; difenoconazole foi eficiente no controle da doença na parte aérea, enquanto carboxin + thiram se igualou à testemunha; os dois fungicidas interferiram negativamente no desenvolvimento das plantas, uma vez que as médias de comprimento, massa fresca e seca da parte aérea e das raízes obtidas das sementes tratadas é inferior à média das sementes não tratadas.


The objective of this research was to evaluate the effect of fungicides difenoconazole and carboxin + thiram applied in seed wheat cultivars 'CD 104' and 'OR Mirante', inoculated with Cochliobolus sativus. The experimental design was completely randomized in a 2x3 factorial design (2 cultivars and 2 fungicides + untreated control) in seven replications. The fungi was isolated and reproduced in PDA culture medium (potato-dextrose-agar) and subsequently multiplied in seeds of wheat pre-cooked and is the source of inoculum. The evaluations were the number of lesions in the basal leaf of five plants per pot at 22, 25, 29, 31, 35 and 38 days after sowing (DAS), and at 41DAS were evaluated the length (cm), fresh weight (g) and dry weight (g) of shoot and root. From the evaluations of the number of lesions was also calculated the area under the disease progress curve (AUDPC). The datas were subjected to analysis of variance and means were compared by Tukey test at 5% probability. The 'CD 104' cultivar was more susceptible to the pathogen than 'OR Mirante'; difenoconazole was efficient to control the disease as carboxin + thiram equaled the witness; both fungicides interfered negatively on plant development, since the average length, fresh weight and dry shoot and root obtained from treated seed is lower than the untreated seeds.

ABSTRACT

Bipolaris sorokiniana is a phytopathogenic fungus causing diseases of cereal crops such as common root rot, the leaf spot disease, seedling blight, and black point of the grain. Random-amplified polymorphic DNA (RAPD) assay was used to investigate the genetic diversity of 20 isolates collected from different cultivars in wheat-producing regions in Brazil. Seventy primers, with random nucleotide sequences, were tested. Reproducibility to amplify the genomic DNA of isolates was found for 30 of the 70 primers tested, generating between 1 and 17 fragments ranging from 0.35 to 2.0 kb (average size). The degree of similarity between samples was calculated through simple association and the dendrogram was assessed using the unweighted pair group method with arithmetical average. After the RAPD analyses 19 isolates were closely grouped, having a similarity coefficient of >or= 78%. Isolate I017 showed very low similarity coefficients, ranging between 38 and 46%. The RAPD analyses provided important information as to the degree of genetic variability and the relationship between the isolates investigated, revealing polymorphism and establishing electrophoretic profiles useful to characterize the phytopathogen.

ABSTRACT

The main aim of this study is to evaluate the efficiency of two biologically active compounds (Strom and F-760) in control of wheat root rot disease and its causal organisms. Fusarium graminearum, F. oxysporum, F. solani and Bipolaris sorokiniana were used as target organisms. In vitro, the two compounds showed fungicidal effect on all investigated pathogens resulted in suppression of radial growth and mycelial dry weight of them. Under greenhouse conditions, treatment of wheat grains with either Strom or F-760 before cultivation significantly reduced the percent of disease distribution as well as the mean disease rating of plants in both seedling and flowering stages. Fresh and dry weights of plants as well as water maintenance capacity were increased as the result of applying these compounds as seed dressing. Also data showed that the membrane stability of plants was injured as a result of infection with all investigated organisms, while this injury was alleviated when F-760 and Strom were applied. The K+ efflux and the leakage of UV-absorbing metabolites was stimulated with fungal infection. However, F-760 and Storm treatment partially retarded the stimulatory effect on leakage of K+ and UV-absorbing metabolites of fungal infected plants. On the other side, the fungal infection had inhibitory effects on pigment fractions (chlorophyll a, b, and carotenoids) biosynthesis in wheat leaves. This retarding effect was partially or completely alleviated as the grains were treated with the applied compounds.
