ABSTRACT

To study the effect of 50% ethanol extract of Bougainvillea xbuttiana on the enzymatic activity, cell via bility and cytokine production provoked by the venom of Bothrops jararaca in macro - phages. Three assays were used to study the effects of B. xbuttiana extract on the damage pro - duced by B. jararaca : Enzymatic activity was detected by measuring the proteoly tic and phos - pholipase A2; macrophages cytotoxicity was determined by the MTT method; levels of cytokine were evaluated using ELISA and a biological assay. After treatment with 300 µg/mL B. xbuttiana extract for 30 min, the proteolytic and phospholipase A2 activities of the venom were reduced to 95 and 61%, respectively. In macrophages cultures treated with B. xbuttiana extract combined with venom, the production of TNF - α, IL - 6 and IFN - γ was reduced, whereas IL - 10 was potenti - ated. Our results support the potential effect of the B. xbuttiana extract as a complementary therapy against the toxicity caused by the venom of B . jararaca snakes


Estudiar el efecto del extracto etanólico al 50% de Bougainvillea xbuttiana sobre la actividad enzimática viabilidad celular y producci ón de citoquinas provocada por el veneno de Bothrops jararaca en macrófagos Se utilizaron tres ensayos para estudiar los efectos del extracto de B. xbuttiana sobre el daño producido por B. jararaca : Se detectó actividad enzimática mediante la medición del proteolítico y fosfolipasa A2; la citotoxicidad de los macrófagos se determinó por el método MTT; Los niveles de citoquinas se evaluaron utilizando ELISA y un ensayo biológico. Después del tratamiento con 300 µg/mL de extracto de B. xbuttiana durante 30 mi n, las actividades proteolíticas y de fosfolipasa A2 del veneno se redujeron a 95 y 61%, respectivamente. En cultivos de macrófagos tratados con extracto de B. xbuttiana combinado con veneno, la producción de TNF - α, IL - 6 e IFN - γ se redujeron, mientras que IL - 10 se potenció. Nuestros resultados apoyan el efecto potencial del extracto de B. xbuttiana como terapia complementaria frente a la toxicidad provocada por el veneno de B. jararaca .

ABSTRACT

Background: The bioactive peptides derived from snake venoms of the Viperidae family species have been promising as therapeutic candidates for neuroprotection due to their ability to prevent neuronal cell loss, injury, and death. Therefore, this study aimed to evaluate the cytoprotective effects of a synthetic proline-rich oligopeptide 7a (PRO-7a; <EDGPIPP) from Bothrops jararaca snake, on oxidative stress-induced toxicity in neuronal PC12 cells and astrocyte-like C6 cells. Methods: Both cells were pre-treated for four hours with different concentrations of PRO-7a, submitted to H2O2-induced damage for 20 h, and then the oxidative stress markers were analyzed. Also, two independent neuroprotective mechanisms were investigated: a) L-arginine metabolite generation via argininosuccinate synthetase (AsS) activity regulation to produce agmatine or polyamines with neuroprotective properties; b) M1 mAChR receptor subtype activation pathway to reduce oxidative stress and neuron injury. Results: PRO-7a was not cytoprotective in C6 cells, but potentiated the H2O2-induced damage to cell integrity at a concentration lower than 0.38 μM. However, PRO-7a at 1.56 µM, on the other hand, modified H2O2-induced toxicity in PC12 cells by restoring cell integrity, mitochondrial metabolism, ROS generation, and arginase indirect activity. The α-Methyl-DL-aspartic acid (MDLA) and L-NΩ-Nitroarginine methyl ester (L-Name), specific inhibitors of AsS and nitric oxide synthase (NOS), which catalyzes the synthesis of polyamines and NO from L-arginine, did not suppress PRO-7a-mediated cytoprotection against oxidative stress. It suggested that its mechanism is independent of the production of L-arginine metabolites with neuroprotective properties by increased AsS activity. On the other hand, the neuroprotective effect of PRO-7a was blocked in the presence of dicyclomine hydrochloride (DCH), an M1 mAChR antagonist. Conclusions: For the first time, this work provides evidence that PRO-7a-induced neuroprotection seems to be mediated through M1 mAChR activation in PC12 cells, which reduces oxidative stress independently of AsS activity and L-arginine bioavailability.(AU)

ABSTRACT

Abstract Snake envenomation is a public health problem, and while serum therapy prevents death, the local effects of venoms can lead to amputations or morbidities. Thus, alternative treatments deserve attention. In this study, we tested eight derivatives of 1,2,3-triazole against some toxic activities of Bothrops jararaca venom. The derivatives were synthesized, and their structures analyzed by infrared and nuclear magnetic resonance. After that, the ability of compounds to inhibit hemolysis, coagulation, proteolysis, hemorrhaging, edema, and lethal activities of B. jararaca venom was investigated. The derivatives were incubated with B. jararaca venom (incubation protocol), administered before (prevention protocol) or after (treatment protocol) injecting venom into the mice. Then, hemorrhaging assay occurred. As a result, most of the derivatives inhibited the activities, even if they were incubated, injected before or after B. jararaca venom. However, the derivatives TRI 07 and TRI 18 seemed to be the most efficient in impairing hemorrhaging. The derivatives showed a low drug score of toxicity based on an in silico technique. Therefore, the derivatives fulfilled physicochemical and biological requirements to become drugs, and they may be a brand new initiative for designing antivenom molecules to complement antivenom therapy to efficiently block tissue necrosis or any other local effects.

ABSTRACT

Lack of complete genomic data of Bothrops jararaca impedes molecular biology research focusing on biotechnological applications of venom gland components. Identification of full-length coding regions of genes is crucial for the correct molecular cloning design. Methods: RNA was extracted from the venom gland of one adult female specimen of Bothrops jararaca. Deep sequencing of the mRNA library was performed using Illumina NextSeq 500 platform. De novo assembly of B. jararaca transcriptome was done using Trinity. Annotation was performed using Blast2GO. All predicted proteins after clustering step were blasted against non-redundant protein database of NCBI using BLASTP. Metabolic pathways present in the transcriptome were annotated using the KAAS-KEGG Automatic Annotation Server. Toxins were identified in the B. jararaca predicted proteome using BLASTP against all protein sequences obtained from Animal Toxin Annotation Project from Uniprot KB/Swiss-Pro database. Figures and data visualization were performed using ggplot2 package in R language environment. Results: We described the in-depth transcriptome analysis of B. jararaca venom gland, in which 76,765 de novo assembled isoforms, 96,044 transcribed genes and 41,196 unique proteins were identified. The most abundant transcript was the zinc metalloproteinase-disintegrin-like jararhagin. Moreover, we identified 78 distinct functional classes of proteins, including toxins, inhibitors and tumor suppressors. Other venom proteins identified were the hemolytic lethal factors stonustoxin and verrucotoxin. Conclusion: It is believed that the application of deep sequencing to the analysis of snake venom transcriptomes may represent invaluable insight on their biotechnological potential focusing on candidate molecules.(AU)

Pathogenesis of Bothrops envenomations is complex and despite numerous studies on the effects of this snake venom on various biological systems, relatively little is known about such effects on the male reproductive system. In the present study, the toxicological outcomes of the low molecular weight fraction (LMWF) of B. jararaca snake venom - containing a range of bioactive peptides - were investigated on the dynamics and structure of the seminiferous epithelium and 15P-1 Sertoli cells viability. Methods: LMWF (5 µg/dose per testis) venom was administered in male Swiss mice by intratesticular (i.t.) injection. Seven days after this procedure, the testes were collected for morphological and morphometric evaluation, distribution of claudin-1 in the seminiferous epithelium by immunohistochemical analyses of testes, and the nitric oxide (NO) levels were evaluated in the total extract of the testis protein. In addition, the toxicological effects of LMWF and crude venom (CV) were analyzed on the 15P-1 Sertoli cell culture. Results: LMWF induced changes in the structure and function of the seminiferous epithelium without altering claudin-1 distribution. LMWF effects were characterized especially by lost cells in the adluminal compartment of epithelium (spermatocytes in pachytene, preleptotene spermatocytes, zygotene spermatocytes, and round spermatid) and different stages of the seminiferous epithelium cycle. LMWF also increased the NO levels in the total extract of the testis protein and was not cytotoxic in concentrations and time tested in the present study. However, CV showed cytotoxicity at 10 μg/mL from 6 to 48 h of treatment. Conclusions: The major finding of the present study was that the LMWF inhibited spermatozoa production; principally in the spermiogenesis stage without altering claudin-1 distribution in the basal compartment. Moreover, NO increased by LMWF induce open of complexes junctions and release the germ cells of the adluminal compartment to the seminiferous tubule.(AU)

ABSTRACT

Snake venom phospholipases A2 (PLA2s) have been reported to induce myotoxic, neurotoxic, hemolytic, edematogenic, cytotoxic and proinflammatory effects. This work aimed at the isolation and functional characterization of a PLA2 isolated from Bothrops jararaca venom, named BJ-PLA2-I. Methods and Results: For its purification, three consecutive chromatographic steps were used (Sephacryl S-200, Source 15Q and Mono Q 5/50 GL). BJ-PLA2-I showed acidic characteristics, with pI~4.4 and molecular mass of 14. 2 kDa. Sequencing resulted in 60 amino acid residues that showed high similarity to other Bothrops PLA2s, including 100% identity with BJ-PLA2, an Asp49 PLA2 previously isolated from B. jararaca venom. Being an Asp49 PLA2, BJ-PLA2-I showed high catalytic activity, and also inhibitory effects on the ADP-induced platelet aggregation. Its inflammatory characterization showed that BJ-PLA2-I was able to promote leukocyte migration in mice at different concentrations (5, 10 and 20 µg/mL) and also at different response periods (2, 4 and 24 h), mainly by stimulating neutrophil infiltration. Furthermore, increased levels of total proteins, IL-6, IL-1 ß and PGE2 were observed in the inflammatory exudate induced by BJ-PLA2-I, while nitric oxide, TNF-α, IL-10 and LTB4 levels were not significantly altered. This toxin was also evaluated for its cytotoxic potential on normal (PBMC) and tumor cell lines (HL-60 and HepG2). Overall, BJ-PLA2-I (2.5-160 µg/mL) promoted low cytotoxicity, with cell viabilities mostly varying between 70 and 80% and significant values obtained for HL-60 and PBMC only at the highest concentrations of the toxin evaluated. Conclusions: BJ-PLA2-I was characterized as an acidic Asp49 PLA2 that induces acute local inflammation and low cytotoxicity. These results should contribute to elucidate the action mechanisms of snake venom PLA2s.(AU)

ABSTRACT

Os elementos químicos, em sua maioria, encontram-se em ciclos bioquímicos e geoquímicos fechados e em concentrações que não causam efeitos nocivos aos organismos. Contudo, ações antrópicas aceleradas promovem alterações ambientais, como o aumento no aporte de contaminantes. Com essas informações, o presente trabalho buscou caracterizar a presença aos elementos traços em duas espécies de serpentes comuns na região sudeste, Boa constrictor e Bothrops jararaca. Os exemplares de B. constrictor (n=18) foram provenientes de atropelamentos ocorridos no trecho da Rodovia ES-060 do Km 0 ao Km 67,5. Os exemplares de B. jararaca (n=18) foram capturados por fazendeiros na zona rural da região serrana do Espírito Santo. Foram analisados 1 grama de fragmento de rim, de espécimes necropsiados. Os rins coletados foram digeridos com mistura ácida (HNO3;HCl; 1:1) a 300oC, 40min e a quantificação de microelementos (Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb e Zn) foi realizada por espectrometria de emissão óptica com plasma indutivamente acoplado (ICP OES). Não foram observadas diferenças significativas entre machos e fêmeas, exceto para Cromo nas B. constrictor (p=0,03), com média de 1,6595 nas concentrações de Cr nas fêmeas e 0,2896 em machos. Contudo, quando foram comparadas as diferentes espécies de serpentes, diferentes concentrações foram encontradas, para todos os elementos determinados, com destaque para o Ferro com uma concentração de 106,2mg/g em B. constrictor e 120,3mg/g em B. jararaca, provavelmente por esses animais virem de áreas próximas à portos de minério de ferro. A concentração de Zinco em B. constrictor foi de 1261,8mg/g e em B. jararaca foi de 28,4mg/g. O presente estudo indica que as serpentes analisadas, que habitam as regiões da Grande Vitória e serrana do Espírito Santo apresentam elevadas concentrações dos microelementos Zn e Fe.(AU)


Chemical elements are mostly involved in biochemical and geochemical cycles in concentrations which are harmless for organisms. However, accelerated human actions promote environmental changes such as the increase in contaminant intake. With this information, this study aimed to characterize the presence of trace elements in two snake species common in Southeastern Brazil, Boa constrictor and Bothrops jararaca. Copies of B. constrictor (n=18) were from pedestrian accidents which occurred on Highway stretch ES-060 from Km 0 to Km 67.5. Copies of B. jararaca (n=18) were captured by farmers in a rural mountainous area of Espíirito Santo State, Brazilo. They were analyzed 1 kidney fragment grass, autopsied specimens. Harvested kidneys were digested with acid mixture (HNO3 e HCl 1:1) at 300°C, 40 min and quantification of microelementos (Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb and Zn) were determined by spectrometric optical emission with inductively coupled plasma (ICP OES). significant differences between males and females were observed, except for Chrome in B. constrictor (p=0.03), with an average of 1.6595 in Cr concentration of females and 0.2896 in males. However, when different species of snakes were compared, different concentrations were found for all elements, especially iron with a concentration of 106.2mg/g in B. constrictor and 120.3mg/g in B. jararaca; probably these animals came from areas near iron ore ports. The concentration of zinc in B. constrictor was 1261.8mg/g and in B. jararaca 28,4mg/g. This study indicates that the analyzed snakes, which inhabit the regions of Greater Victoria and the mountainous region of the State of Espírito Santo have high concentrations of the microelements Zn and Fe.(AU)

Abstract Bradykinin-potentiating peptides (BPPs) are molecules discovered by Sergio Ferreira who found them in the venom of Bothrops jararaca in the 1960s that literally potentiate the action of bradykinin in vivo by, allegedly, inhibiting the angiotensin-converting enzymes. After administration, the global physiological effect of BPP is the decrease of the blood pressure. Due to this interesting effect, one of these peptides was used by David Cushman and Miguel Ondetti to develop a hypotensive drug, the widely known captopril, vastly employed on hypertension treatment. From that time on, many studies on BPPs have been conducted, basically describing new peptides and assaying their pharmacological effects, mostly in comparison to captopryl. After compiling most of these data, we are proposing that snake BPPs are modular peptidic molecules, in which the combination of given amino acid blocks results in the different existing peptides (BPPs), commonly found in snake venom. We have observed that there would be mandatory modules (present in all snake BPPs), such as the N-terminal pyroglutamic acid and C-terminal QIPP, and optionalmodules (amino acid blocks present in some of them), such as AP or WAQ. Scattered between these modules, there might be other amino acids that would complete the peptide, without disrupting the signature of the classical BPP. This modular arrangement would represent an important evolutionary advantage in terms of biological diversity that might have its origins either at the genomic or at the post-translational modification levels. Regardless of the modules origin, the increase in the diversity of peptides has definitely been essential for snakes success on nature.

Abstract Background Snakes of the genus Bothrops, popularly known as pit vipers, are responsible for most cases of snakebite in Brazil. Within this genus, Bothrops jararacussu and B. jararaca deserve special attention due to the severity of their bites and for inhabiting densely populated areas. Regarding the treatment of snakebites by Bothrops jararacussu, questions have been raised about the effectiveness of the specific bothropic antivenom in neutralizing myotoxic effects; however, there are no accurate data for humans. Thus, the development of a differential diagnostic kit for this species would be of great interest because it provides, for healthcare professionals, a tool that would allow us to determine whether the accident was caused by B. jararacussu or other species of the genus. It would also make it possible to evaluate the specificity of the treatment and to provide data for epidemiological studies. Methods First, we produced a species-specific polyclonal antibody a potential biomarker of Bothrops jararacussu venom against bothropstoxin-I (BthTx-I), which is also found in smaller quantities in the venoms of B. jararaca from southern Brazil. Results Polyclonal antibodies against bothropstoxin-I could be separated into several species-specific immunoglobulins. Then, aiming to develop a system of safe and standardized immunoassay, we produced monoclonal antibodies. Seven hybridomas were obtained. Five of them were specific to the venom of B. jararacussu and two recognized the venom of B. jararaca from the southeastern population. The use of monoclonal antibodies also made it possible to differentiate B. jararacussu from B. jararaca venom obtained from the southern population. Analyzing the reactivity of monoclonal antibodies against other bothropic venoms, we found mAb Bt-3 to be more specific than others for B. jararacussu venom. Conclusions These results show the potential of BthTx-I for producing monoclonal antibodies that differentiate between B. jararacussu and other Bothrops species venoms.

Abstract Introduction Envenomation by Bothrops snakes can produce local pain, edema, hemorrhage and myonecrosis. However, standard antivenom therapy is generally ineffective in neutralizing these effects so that alternative methods of treatment have been investigated. In experimental animals, low-level laser therapy (LLLT) attenuates the local effects of Bothrops venoms, but the benefits of LLLT on muscle function after envenomation are unclear. In this study, we examined the influence of LLLT on the contractile activity of mouse skeletal muscle injected with venom from Bothrops jararaca, the principal cause of snakebite in southeastern Brazil. Methods Twenty-seven male mice were used. Mice were injected with venom (40 μg in 50 μl) in the right anterior tibialis muscle, after which the muscle tendon was exposed, connected to an isometric transducer and subjected to a resting tension of 1 g. A bipolar electrode was attached to the tibial nerve for electrical stimulation. The mice were randomly allocated to five groups: A – Control (n = 3), B – Venom 3 h (n = 6), C – Venom 9 h (n = 6), D – Venom + Laser 3 h (n = 6), E – Venom + Laser 9 h (n = 6). Results The two groups that received LLLT post-venom showed improved muscle contraction and contracture in relation to muscle treated with venom alone. Conclusion These results indicate that LLLT can improve muscle function after damage induced by B. jararaca venom.

Although ultrastructural characteristics of mature neuroglia in the central nervous system (CNS) are very well described in mammals, much less is known in reptiles, especially serpents. In this context, two specimens of Bothrops jararaca were euthanized for morphological analysis of CNS glial cells. Samples from telencephalon, mesencephalon and spinal cord were collected and processed for light and transmission electron microscopy investigation. Astrocytes, oligodendrocytes, microglial cells and ependymal cells, as well as myelin sheaths, presented similar ultrastructural features to those already observed in mammals and tended to maintain their general aspect all over the distinct CNS regions observed. Muito embora as características ultraestruturais da neuróglia madura do sistema nervoso central (SNC) sejam bem descritas em mamíferos, muito pouco é conhecido em répteis, especialmente em serpentes. Neste contexto, dois espécimes de Bothrops jararaca foram eutanasiados para análise morfológica das células gliais presentes no SNC. Amostras de telencéfalo, mesencéfalo e medula espinhal foram coletadas e processadas para investigação por microscopia de luz e eletrônica de transmissão. Astrócitos, oligodendócitos, células microgliais e ependimárias, bem como bainhas de mielina, apresentaram características ultraestruturais similares àquelas já observadas em mamíferos e tenderam a manter seu aspecto geral pelas diferentes regiões observadas no SNC. ABSTRACT

Background Snakebite is considered a neglected tropical disease by the World Health Organization. In Brazil, about 70% of the envenomation cases are caused by Bothrops snakes. Its venom may provoke hemorrhage, pain, necrosis, hemolysis, renal or cardiac failure and even death in victims. Since commercial antivenom does not efficiently neutralize the local toxic effects of venoms, natural products have been tested in order to provide alternative or complementary treatment to serum therapy. Therefore, the present study aimed to evaluate the ability of the seaweed Plocamium brasiliense and its active derivatives to neutralize hemorrhagic, edematogenic, hemolytic, coagulant and proteolytic activities of B. jararaca venom. Methods Specimens of P. brasiliense were collected in Rio de Janeiro state, Brazil, dried and submitted to oil extraction using four solvents of increasing polarities, n-hexane (HEX), dichloromethane (DCM), ethyl acetate (ETA) and hydroalcoholic solution (HYD). The solvents were evaporated, yielding HEX, DCM, ETA and HYD extracts. Further, all extracts were dissolved in dimethylsulfoxide. In addition, two monoterpenes (8-bromo-3,4,7-trichloro-3,7-dimethyl-1E, 5E-octadiene and 1,8-dibromo-3,4,7-trichloro-3,7-dimethyl-1E, 5E-octadiene) and a cholesterol fraction were isolated from the extract of P. brasiliense prepared in hexane. Algal samples were incubated for 30 minutes with B. jararaca venom, and then tested for lethality; hemorrhagic, edematogenic, hemolytic, coagulant and proteolytic effects. Results Most of the algal extracts inhibited the toxic effects with different potencies. The DCM extract was the most effective, since it inhibited all types of toxic activity. On the other hand, the HYD extract failed to inhibit any effect. Detection of Leptospira by PCR had not yet been described in snakes. This study investigated, by microscopic agglutination test (MAT) and PCR, the presence of antibodies to Leptospira spp. and Leptospira spp., respectively, in venomous and non-venomous wildlife and captivity snakes. All snakes were divided into three groups to be compared: Group 1 (wildlife snakes - WS); Group 2 (snakes in intensive captivity - IC), and Group 3 (collective semi-extensive captivity -CC). Of the 147 snakes studied, 52 (35.4 percent) were positive for leptospirosis by MAT, 8 (15.4 percent) belonging to Group 1 (WS), 34 (65.4 percent) to Group 2 (IC) and 10 (19.2 percent) to Group 3 (CC). Jararaca (Bothrops jararaca) presented the highest average titer (66.7 percent, N=22/33) among the three group studied, and Hardjo prajtino was the most prevalent serovar (88.5 percent, N=46/52), with titers varying from 100 to 3200. Leptospira interrogans was revealed by PCR in kidney and liver of caiçaca (Bothrops moojeni) and jararaca-pintada (Bothrops pauloensis), showing 100 percent and 93 percent identity respectively. Future studies should be carried out for better understanding of the role of snakes as a reservoir of Leptospira in nature.


A detecção de Leptospira pela técnica de PCR não havia sido descrita em serpentes. Este estudo investigou pelo teste de aglutinação microscópica (MAT) e PCR, a presença de anticorpos anti-Leptospira spp. e Leptospira spp., respectivamente, em serpentes peçonhentas e não peçonhentas de vida livre e de cativeiro. As serpentes foram divididas em três grupos para comparação: Grupo 1 (serpentes recém-chegadas da natureza - WS); Grupo 2 (serpentes em regime de cativeiro intensivo -IC) e Grupo 3 (serpentes em regime de cativeiro coletivo semi-extensivo - CC). Do total de 147 serpentes estudadas, 52 (35,4 por cento) foram positivas para leptospirose pelo MAT, as quais 8 (15,4 por cento) pertenciam ao Grupo 1 (WS), 34 (65,4 por cento) ao Grupo 2 (IC) e 10 (19,2 por cento) ao Grupo 3 (CC). Das espécies estudadas, a jararaca (Bothrops jararaca) apresentou maior soropositividade (66,7 por cento, N=22/33). O sorovar mais prevalente foi o Hardjo prajtino (88,5 por cento, N=46/52) e os títulos variaram de 100 a 3200. Leptospira interrogans foi revelada por PCR nos rins e no fígado de caiçaca (Bothrops moojeni) e de jararaca-pintada (Bothrops pauloensis), mostrando 100 por cento e 93 por cento de identidade, respectivamente. Futuros estudos devem ser realizados para melhor compreensão do papel das serpentes como reservatório de leptospiras na natureza.

Objective - Bothrops jararaca snake is involved in almost 90% of all reports in the State of São Paulo, Brazil. Little is known about the effect of maternal exposure to B. jararaca snake venom [BjV] on fetal development. This study was designed to investigate the effect of a moderate dose of the venom (1.2 mg/kg sc on either gestation day GD5 or GD12), in pregnant mice and their offspring. Methods - In dams, during pregnancy, it was observed the body weight gain, food and water consumptions. In the last day of pregnancy, dams were submitted to a cesarean and the reproductive performance was measured. Thus, the fetuses body weight, the number of live and dead fetuses as wells as the external, visceral or skeletal alterations were assessed. Results - Results showed that the venom injection on GD5 did not change the dams weight and reproductive parameters, the fetuses weight, but it was observed high incidence of skeletal anomalies such as incomplete skull ossification and supernumerary ribs relative to controls. Dams treated in GD11 showed decreased food ingestion in the day after treatment. Their offspring presented a high incidence of skeletal anomalies such as vertebrae anomalies, sternebrae anomalies and incomplete skull ossification, which might be a sign of craniostenosis, than controls. Conclusions - In conclusion, subcutaneous administration of 1.2mg/kg BjV to pregnant mice either at GD4 or GD12 produced subtle maternal toxicity but a clear fetotoxicity. Whether these observations represent a reaction to treatment and, if so, the underlying mechanisms and their toxicological impact remain to be examined further in future studies.


Objetivo - O veneno de Bothrops jararaca está envolvido em quase 90% dos casos de envenenamento do Estado de São Paulo, Brasil. Pouco se sabe sobre o efeito no desenvolvimento fetal quando no caso de envenenamento materno. Este estudo examinou, em camundongas prenhes e sua prole, os efeitos do envenenamento por uma dose moderada do veneno de B. jararaca (1,2 mg/kg sc no 5° (GD5) e 11°(GD11) da gestação. Métodos - Anotou-se durante a gestação o ganho de peso corporal e o consumo de água e comida materno; no último dia de gestação, as mães foram submetidas a uma cesariana e a performance reprodutiva foi avaliada. Para tanto, anotou-se o peso dos fetos, o número de fetos vivos e mortos, assim como examinou-se a presença de alterações externas, esqueléticas, e viscerais. Resultados - Os resultados mostraram que a injeção do veneno no GD5 não alterou o peso materno, o consumo de água e ração, o peso dos fetos e os parâmetros reprodutivos, tendo sido observada alta incidência de anomalias esqueléticas tais como ossificação do crânio incompleta e costelas supranumerárias em relação aos controles. O envenenamento no GD11 promoveu decréscimo na ingestão de alimentos no dia subsequente ao tratamento. Neste caso, a prole apresentou alta incidência de anomalias esqueléticas tais como anomalias vertebrais, do esterno e ossificação incompleta do crânio, ou seja, craniostenose. Conclusões - O envenenamento moderado pelo veneno da B. jararaca em camundongas prenhas tanto no GD5 como no GD11 produziram efeitos sutís ao nível materno, porém nos fetos, apareceram severas alterações. Estudos futuros deverão ser feitos para entender os mecanismos subjacentes a este envenenamento durante a gestação.

Estudos prévios demonstraram que as atividades biológicas do veneno da serpente Bothrops jararaca sofrem significantes modificações ontogenéticas. Neste estudo é apresentada uma análise comparativa do proteoma, peptidoma e transcriptoma da glândula de veneno de filhotes e adultos de B. jararaca, correlacionando os resultados obtidos com algumas características funcionais dos venenos. Venenos de 694 filhotes de até duas semanas de idade e 110 adultos, provenientes do Estado de São Paulo foram extraídos e liofilizados para as análises proteômicas/peptidômicas e funcionais. O mRNA de glândulas de veneno de 20 filhotes e 10 adultos foi obtido para a contrução de bibliotecas de cDNA e a análise de Expressed Sequence Tag (ESTs). Demonstramos que a atividade hemorrágica é similar para os venenos de filhotes e adultos, enquanto que o veneno de adultos é discretamente mais letal para camundongos; entretanto, o veneno de filhotes mostrou-se extremamente mais letal para aves, uma característica que pode garantir proteção contra potenciais predadores nas fases iniciais de vida da espécie. A atividade coagulante do veneno de filhotes é cerca de 10 vezes mais alta que aquela verificada para o veneno de adultos e é atribuída sobretudo à atividade de metaloproteinases. Essas diferenças nas atividades funcionais se refletiram nos diferentes perfis verificados por eletroforese bidimensional e identificação de spots de proteínas por digestão tripsínica in-gel seguida de análise por cromatografia líquida acoplada à espectrometria de massas em tandem, zimografia com gelatina, imunocoloração utilizando anticorpos específicos anti-proteinases, e glicoproteínas com afinidade pela concanavalina -A. Previous studies have demonstrated that the biological activities displayed by the venom of the snake Bothrops jararaca undergo a significant ontogenetic shift. In this investigation, we performed comparative proteomic, peptidomic and transcriptomic analyses of venoms and venom glands from newborn and adult specimens of B. jararaca and correlated the results with the evaluation of functional venom features. Venoms from 694 two-week old newborns and 110 adults from São Paulo state were milked and lyophilized for functional and proteomic/peptidomic analyses. Additionally, mRNA was obtained from the venom glands of 20 newborns and 10 adults and used for the construction of cDNA libraries and Expressed Sequence Tag (ESTs). We demonstrate that newborn and adult venoms have similar hemorrhagic activities, while the adult venom has a slightly higher lethal activity upon mice; however, the newborn venom is extremely more potent to kill chicks, a feature that might ensure protection against potential predators in early stages of B. jararaca life. Interestingly, the coagulant activity of the newborn venom upon human plasma is ten times higher than that of the adult venom and is contributed mainly by metalloproteinases. Differences in functional activities were clearly reflected in the venom different profiles of two-dimensional gel electrophoresis (2D-PAGE) and protein spot identification by in-gel trypsin digestion followed by liquid chromatography and tandem mass spectrometry (LC-MS/MS), gelatin zimography, immunostaining using specific anti-proteinase antibodies, and concanavalin A-binding proteins. The venom comparison by isobaric tag peptide labeling (iTRAQ) and ESTs analysis revealed clear differences in toxin levels. ABSTRACT

Esse estudo teve como objetivo determinar as alterações clínico-patológicas e laboratoriais em ovinos inoculados com a peçonha de Bothropoides jararaca e Bothrops jararacussu, no intuito de fornecer subsídios que possam facilitar o estabelecimento do diagnóstico e do diagnóstico diferencial dessa condição. Os venenos liofilizados foram diluídos em 1 ml de solução fisiológica e administrados a quatro ovinos por via subcutânea. Três ovinos foram a óbito e um que recebeu a dose de 0,5mg/kg (B. jararaca), recuperou-se. Os sinais clínicos tiveram início entre 7 minutos e 1 hora. O período de evolução variou de 7 horas 9 minutos a 21 horas 59 minutos. O quadro clínico, independentemente das doses, caracterizou-se por aumento de volume no local da inoculação, tempo de sangramento e de preenchimento capilar aumentados, taquicardia, dispnéia, mucosas hipocoradas e apatia. Os exames laboratoriais revelaram acentuada anemia normocítica normocrômica, trombocitopenia, acentuada redução de fibrinogênio e proteínas plasmáticas totais, hematócrito diminuído em dois animais, além de acentuado aumento de creatinaquinase e desidrogenase lática em todos os animais. À necropsia, os principais achados no local da inoculação e tecidos adjacentes eram extensas hemorragias no animal que recebeu o veneno de B. jararaca e edema e acentuado edema pulmonar agudo para os dois animais envenenados por B. jararacussu. Além de hemorragia e edema a principal alteração histopatológica verificada foi necrose das fibras musculares e de vasos, no local de inoculação e adjacências. A necrose tubular renal foi atribuída ao quadro de choque. Nos ovinos deste estudo, o aumento de volume observado no local de inoculação e adjacências era constituído predominantemente por sangue (B. jararaca) e por edema (B. jararacussu).


The purpose of this study was to establish the clinic-pathological and laboratory changes in sheep inoculated with Bothropoides jararaca and Bothrops jararacussu venom to provide subsidies for the differential diagnosis of snake bites. The liofilized venoms were diluted in 1 ml saline and administrated subcutaneously to four sheep. Three of the animals died, and the one that received 0.5mg/kg (B. jararaca venom) recovered. First symptoms were observed from 7 minutes to 1 hour after inoculation, and the clinical course varied from 7 hours and 9 minutes to 21 hours and 59 minutes. The symptoms, independent of the dosage, were swelling of the inoculation site, increased bleeding time and capillary filling, tachycardia, dyspnea, pale mucous membranes and diminished reaction to external stimuli. Laboratory tests revealed pronounced normocytic and normochromic anemia, trombocytopenia, slight reduction of fibrogen and total plasmatic protein, in two animals diminished hematocrit, besides pronounced increase of creatinaquinase and lactic dehydrogenase. At necropsy, the main findings at the inoculation site and adjacent tissues were extensive hemorrhages in the sheep inoculated with jararaca venom, and predominantly edema in the two animals inoculated with jararacussu venom. In two sheep which received jararacussu venom, acute pulmonary edema was observed. Hemorrhage and edema as the main histopathological changes, besides necrosis of muscle fibers and vessels at the inoculation site and adjacent tissue was observed. The renal tubular necrosis was attributed to shock. The volume increase at the inoculation site and surroundings was mainly due to hemorrhage (B. jararaca) or edema (B. jararacussu).

ABSTRACT

Recently, there has been an interest in the use of shed snake skin as alternative model biomembrane for human stratum corneum. This research work presented as objective the qualitative characterization of alternative model biomembranes from Bothrops jararaca and Spilotis pullatus by FT-Raman, PAS-FTIR and DSC. The employed biophysical techniques permitted the characterization of the biomembranes from shed snake skin of B. jararaca and S. pullatus by the identification of vibrational frequencies and endothermic transitions that are similar to those of the human stratum corneum.


Existe atualmente interesse no uso da muda de pele de cobra como modelos alternativos de biomembranas da pele humana. O presente trabalho apresentou como objetivo a caracterização qualitativa de modelos alternativos de biomembranas provenientes de mudas de pele de cobra da Bothrops jararaca e Spilotis pullatus por espectroscopia Raman (FT-Raman), espectroscopia fotoacústica no infravermelho (PAS-FTIR) e calorimetria exploratória diferencial (DSC). As técnicas biofísicas FT-Raman, PAS-FTIR e DSC permitiram caracterizar qualitativamente os modelos alternativos de biomembranas provenientes das mudas de pele de cobra da B. jararaca e S. pullatus e identificar freqüências vibracionais e transições endotérmicas similares ao estrato córneo humano.
