Resumen Introducción. La Brucelosis bovina, es una enfermedad bacteriana e infecto contagiosa, causada por Brucella abortus. Se transmite a través de la ingestión de forrajes y aguas contaminadas con descargas vaginales infectadas, que conlleva a una patología del sistema reproductor en bovinos, que impactan la sanidad pecuaria y la economía de la agroindustria. Objetivo. Evaluar el comportamiento de la brucelosis bovina en el municipio de Aguazul, Casanare (Colombia) y los factores asociados al desarrollo de esta enfermedad. Metodología. Estudio descritptivo de 26.187 muestras de suero sanguíneo de ganado bovino evaluadas, que corresponden a 260 predios del municipio de Aguazul. Se empleó técnicas serológicascomo rosa de bengala, fluorescencia polarizada y ELISA competitiva para la evaluación de positividad a Brucella abortus y se evaluaron las pérdidas económicas asociadas a positividad en los ensayos de laboratorio. Resultados y conclusiones. La positividad a brucelosis bovina correspondió al 1%, que corresponde a hembras menores de 24 meses de edad y entre 37 a 48 meses y, machos entre los 57 a 68 meses de edad. Se sugiere consolidar esfuerzos en investigación para evaluar los factores que contribuyen a la seropositividad en el ganado y el riesgo para la propagación y mantenimiento de la enfermedad.


Abstract Introduction. Bovine brucellosis is a contagious bacterial and infectious disease caused by Brucella abortus. It is transmitted through the ingestion of forage and water contaminatedwith infected vaginal discharges, which leads to a pathology of the reproductive system inbovines, which impacts livestock health and the economy of agribusiness. Aim. To evaluatethe behavior of bovine brucellosis in the municipality of Aguazul, Casanare (Colombia) and the factors associated with the development of this disease. Methodology. Descriptive study of 26,187 bovine blood serum samples evaluated, corresponding to 260 farms in the municipality of Aguazul. Serological techniques such as rose bengal, polarized fluorescence, and competitive ELISA were used to evaluate positivity to Brucella abortus and the economic losses associated with positivity in laboratory tests were evaluated. Results and conclusions. The positivity to bovine brucellosis corresponded to 1%, which corresponds to females under 24 months of age and between 37 to 48 months and males between 57 to 68 months of age. It is suggested to consolidate research efforts to evaluate the factors that contribute to seropositivity in cattle and the risk for the spread and maintenance of the disease

ABSTRACT

Brucellosis is highly infectious zoonotic disease that causes huge economic losses to livestock farmers by affecting the reproductive potential of animals causing last trimester abortions and infertility. In the present study evaluation of different serological tests to diagnose the seroprevalence of brucellosis in bovines with history of abortion using various serological tests [Rose Bengal Plate Test (RBPT), modified rose bengal plate test (mRBPT), microtitre plate agglutination test (MAT) and indirect enzyme linked immunosorbent assay (i-ELISA)] was carried out. A total of 134 blood samples of cattle and buffalo with history of abortion were collected from organized and unorganized farms. Seroprevalence by mRBPT, RBPT, MAT and i-ELISA was 75.37%, 67.91%, 72.38% and 72.38%, respectively. In organized farms, prevalence of 78.12%, 81.25%, 78.12% and 81.25% while in unorganized farms prevalence of 64.70%, 73.52%, 70.58% and 69.60% was reported by RBPT, mRBPT, MAT and i-ELISA, respectively. The sensitivity and specificity of serological tests by keeping i-ELISA as gold standard were also calculated and the results revealed that sensitivities of RBPT, mRBPT and MAT were 91.75%, 97.94% and 96.91%, respectively, whereas specificities were 94.59%, 83.78% and 91.89%, respectively.

ABSTRACT

In general, European and North American countries, as well as Australia and New Zealand, have already eradicated or reached good levels of control of brucellosis and tuberculosis in cattle. In the rest of the world, however, the epidemiological situation of these two diseases is frequently poorly understood. In this review article, quantified data on these diseases in the South American countries are presented. Initially, the aspects that led the continent to host 25% of the world cattle population are presented, in addition to the aspects that placed the continent at a prominent position in the international meat market. Subsequently the continent was divided into three country groups, considering the size of the cattle population and how well the epidemiological situation of brucellosis and tuberculosis in cattle is quantified. It is argued that countries that do not generate high-quality quantitative epidemiological data on these diseases have serious limitations in outlining and managing control or eradication strategies. Thus, for successful outcomes, at least methodologies to estimate the prevalence of infected herds should be employed.(AU)


De maneira geral, os países da Europa e da América do Norte, além da Austrália e da Nova Zelândia, já erradicaram ou atingiram bons níveis de controle da brucelose e da tuberculose bovinas. Entretanto, no restante do mundo, raramente a situação epidemiológica dessas duas doenças é adequadamente conhecida. Neste artigo de revisão são apresentados dados de quantificação dessas importantes enfermidades nos países da América do Sul. Inicialmente são apresentadas as características que concorreram para que atualmente o continente tenha 25% do efetivo bovino mundial e uma posição de destaque no mercado internacional de carnes. Os países foram então alocados em três grupos, levando em consideração o tamanho da população bovina e a qualidade da quantificação referente à situação epidemiológica da brucelose e da tuberculose bovinas. Argumenta-se que países que não geram dados epidemiológicos quantitativos de alta qualidade em relação a essas doenças têm sérias limitações para traçar estratégias eficazes de combate e são incapazes de realizar a gestão dos processos. Conclui-se que os países que desejam ser exitosos no combate à brucelose e tuberculose bovinas deveriam ao menos utilizar metodologias para estimar a prevalência de focos.(AU)

ABSTRACT

Determinar a ocorrência da brucelose, da tuberculose e a percepção de riscos acerca de ambas as zoonoses foi o objetivo do estudo em rebanhos bovinos leiteiros do município de Paranaíba, Mato Grosso do Sul, Brasil. As visitas ocorreram entre novembro de 2012 e março de 2013, em 42 propriedades georreferenciadas, nas quais foram aplicados questionários aos produtores e realizadas as coletas de sangue para testes sorológicos e de tuberculinização. A ocorrência de focos, tanto da brucelose quanto da tuberculose, nas propriedades foi de 2,38% (1/42), porém, há um baixo conhecimento dos produtores sobre os fatores de risco para essas doenças.(AU)


The aim of these study was to determine the occurrences of brucellosis and bovine tuberculosis, and the perception of dairy farmers on the risk of these diseases in dairy cattle herds in the municipality of Paranaíba, Mato Grosso do Sul state, Brazil. The total of 42 properties were visited for the application of a questionnaire to dairy farmers, and blood sample collection for serological and tuberculin test, from November 2012 to March 2013. The occurrence of outbreaks, both of brucellosis and tuberculosis, in the properties was 2.38% (1/42). The data showed deficiency in risk perception of these diseases on the part of the farmers.(AU)

ABSTRACT

This paper aimed to determine the excretion period of B19 vaccine strain during a complete reproductive cycle (from estrus synchronization, artificial insemination, pregnancy and until 30 days after parturition) of dairy cows from 3 to 9 years old that were previously vaccinated from 3 to 8 months. Three groups were monitored with monthly milk and urine collection during 12 months: G1 with seven cows from 3 to 4 years old; G2 with three cows from 5 to 6 years old; and G3 with four cows from 7 to 9 years old. Urine and milk samples were submitted to bacteriological culture and urine and PCR reactions for detection of Brucella spp. and PCR-multiplex for B19 strain identification. Ring test (RT) was also performed in the milk samples, and serum samples were tested by buffered acidified plate antigen test (BAPA). All animals were serologically negative at BAPA and Brucella spp. was not isolated from both urine and milk samples. RT revealed 13/210 (6.2%) positive milk samples. PCR reactions detected DNA of Brucella spp. in 86/420 (20.5%) samples. In urine it was found a significantly higher frequency (35.2%; 74/210) than in milk (5.7%; 12/210), more frequently from the estrus to 150 days of pregnancy and after parturition (6.7%; 10/150), and from 150 days of pregnancy to parturition (3.4%; 2/60), and they were all identified as B19 strain. In three groups, intermittent excretion of B19 strain was detected mainly in urine samples, which confirmed its multiplication and persistence in cows for until 9 years.

ABSTRACT

This paper describes the epidemiological characteristics of bovine brucellosis in Korea during January 2000~September 2004, which encompasses the period when the incidence of bovine brucellosis increased abruptly. Data from the National Animal Infectious Disease Data Management System were used for this study. A range of epidemiological measures was calculated including annual herd and animal incidence. During the study period, there were 1,183 outbreaks on 638 farms. In beef cattle, annual herd incidence increased from 0.2 (2000) to 11.5 (2004, to September) outbreaks per 10,000 and annual animal incidence varied between 3.4 (2000) and 105.8 (2004, to September) per 100,000, respectively. On 401 (62.9%) infected farms during this period, infection was eradicated without recurrence. Recurrence of infection was significantly higher on farms where abortion was reported (53.3%), compared to farms where it was not (30.0%). On beef cattle farms, infection was introduced most frequently through purchased cattle (46.2%). Based on the results of this study, the establishment and spread of brucellosis in the Korean beef cattle population were mainly due to incomplete or inappropriate treatment of aborted materials and the movement of infected cattle.

Esta investigação teve por objetivo validar o teste de polarização fluorescente (TPF) para o diagnóstico sorológico da brucelose bovina, determinando a sensibilidade (SE) e a especificidade relativas (SP) e verificando a reprodutibilidade do teste em quatro laboratórios no Brasil. Foram selecionadas 1.389 amostras de soro sanguíneo, as quais foram inicialmente submetidas aos testes do antígeno acidificado tamponado (AAT) e mercaptoetanol (2-ME). As mesmas amostras foram submetidas à reação de fixação de complemento (RFC) e ao TPF. Para a avaliação do TPF, foi adotada a combinação dos resultados do AAT, da RFC e do 2-ME, utilizados como população de referência (padrão-ouro). Para a determinação do ponto de corte do TPF que proporciona a melhor combinação de sensibilidade e especificidade, foi usada a análise TG-ROC. A concordância entre os resultados dos quatro laboratórios foi determinada com base no indicador kappa e no coeficiente de correlação de Pearson. Os pontos de corte do TPF situaram-se entre 85,2 e 93,6 mP, conforme o laboratório. A sensibilidade variou de 91,7 a 97,3 por cento, e a especificidade situou-se na faixa de 82,6 a 98,3 por cento. Na comparação entre os resultados do TPF dos quatro laboratórios, o indicador kappa ficou entre 0,69 e 0,95, o que indica, na maioria das situações, reprodutibilidade excelente, e o coeficiente de correlação variou entre 0,76 e 0,99. Os resultados indicaram que o TPF apresentou bom desempenho, na maioria das situações, com sensibilidade e especificidade elevadas. Em comparação com os testes convencionais, o TPF apresenta as vantagens de ser de execução mais rápida e mais fácil e não estar sujeito à ocorrência de prozona, como a RFC e o 2-ME, nem de atividade anticomplementar, como a RFC.


The purpose of this research was the interlaboratorial validation of the polarization fluorescence assay (PFA) for the serodiagnosis of bovine brucellosis, verifying the relative sensitivity, the relative specificity and the reproducibility of the test in four Brazilian laboratories. Serum samples from 1,389 bovines were selected and submitted to the rose Bengal (RBT) and 2-mercaptoethanol tests in one of the laboratories. The same samples were tested by the complement fixation (CFT) test and by the PFA in the four laboratories participating of the research. The reference population (golden standard) used to evaluate the PFA was the combination of the results of RBT, CFT and 2-ME. TG-ROC analysis was used to obtain the cut-off that provided the best combination of sensitivity and specificity. The agreement between laboratories was obtained by the kappa statistic and Pearson correlation coefficient (r). The PFA cut-off values were from 85.2 to 93.6. The sensitivity of the PFA assay varied from 91.7 percent to 97.3 percent, and the specificity values varied from 82.6 percent to 98.3 percent. When comparing PFA results from the four laboratories, the kappa values was between 0.69 and 0.95, which indicates, in most situations, excellent reproducibility, and the correlation coefficient varied from 0.76 to 0.99. The results showed that the PFA had a good performance, with high sensitivity and specificity. Compared to the conventional tests, the PFA has the advantages of being easy and quick to perform, and it is not prone to the occurrence of prozone, as the CFT or the 2-ME, nor to the occurrence of anti-complementary effect, as the CFT.

ABSTRACT

Avaliaram-se comparativamente as provas do antígeno acidificado tamponado (AAT), a combinação das provas de soroaglutinação lenta e 2-mercaptoetanol (2-ME) e a reação de fixação de complemento (RFC), provas preconizadas pelo Programa Nacional de Controle e Erradicação de Brucelose Tuberculose (PNCEBT). Para tanto, foram alisadas 1.061 amostras de soros bovinos. Os dados foram analisados pelo indicador kappa, adotando-se como ponto de corte o título 25 no 2-ME e 4 na RFC. Constatou-se sensibilidade relativa de 99,6%, 98,8% e 91,1%, respectivamente, para o AAT, a 2-ME e a RFC, e especificidade relativa de 83,9%, 96,2% e 100,0%. A comparação entre os testes adotados pelo programa apontou concordância boa entre o teste de triagem (AAT) e os testes confirmatórios (kappa: 2-ME = 0,80; e RFC = 0,73) e concordância ótima quando os testes confirmatórios foram comparados entre si (kappa = 0,86). No entanto foram encontrados soros com título elevado em um dos testes confirmatórios e resultado negativo no outro, o que reforça a ideia de que o diagnóstico sorológico da brucelose é mais confiável quando obtido por meio dos resultados de vários testes.


Serum samples from 1,061 bovine were analyzed by serological diagnostic techniques adopted by the Brazilian Program for Animal Brucellosis and Tuberculosis Control and Eradication in order to compare, as a screening test, the rose Bengal plate test (RBPT), and as confirmatory tests, the 2-mercaptoethanol plus standard tube agglutination test (2-ME), and the complement fixation test (CFT). Relative sensitivity of 99.6%, 98.8% and 91.1% were observed, respectively, for RBPT, 2-ME and CFT, and relative specificity of 83.9%, 96.2% and 100.0%. The agreement between tests was analyzed by the kappa statistic (2-ME cut-off = titer 25, and CFT cutoff = titer 4). RBPT showed a substantial agreement (kappa: 2-ME = 0.80; and RFC = 0.73) with the confirmatory tests, and excellent agreement between the confirmatory tests (kappa = 0.86) was observed. However sera with a negative result in one of the confirmatory tests and a high titer in the other were observed, reinforcing that the serological diagnosis of bovine brucellosis is more reliable when based on the results of several tests.

ABSTRACT

As estratégias de combate à brucelose bovina são bastante conhecidas e, até o momento, os resultados são divergentes. No Brasil, o Ministério da Agricultura, Pecuária e Abastecimento (MAPA) implementou, em 2001, o Programa Nacional de Controle e Erradicação da Brucelose e Tuberculose. Conhecer a situação epidemiológica da brucelose no início de um programa de controle permite: 1 - escolher as melhores estratégias de controle em função da frequência e padrão de distribuição da doença na população e 2 - acompanhar o programa com a finalidade de promover correções e evitar o desperdício de tempo e de recursos. Em razão disso, é necessário realizar estudos para dar suporte à escolha das melhores estratégias para os vários estados e regiões brasileiras e criar um mecanismo racional de verificação da efetividade das ações implementadas. Para tanto, o MAPA estabeleceu um Termo de Cooperação Técnica com a Faculdade de Medicina Veterinária da Universidade de São Paulo e envolveu também a Faculdade de Agronomia e Veterinária da Universidade de Brasília. Até o momento, foram concluídos os estudos de 15 unidades federativas, cujos resultados são apresentados nos artigos subsequentes. Além disso, há um 16º artigo que explora o impacto da vacinação de bezerras com a B19 na redução da prevalência da brucelose.


Although strategies for controlling and eradicating of bovine brucellosis are well known, the achievements vary widely. In 2001, the Brazilian Ministry of Agriculture, Livestock, and Food Supply (MAPA) started a new National Program for the Control and Eradication of Brucellosis and Tuberculosis (NPCEBT). In the beginning of a brucellosis control program, it is of utmost importance to gain insights into the epidemiological status of the disease in order to: (1) determine the sanitary measures according to the frequency and distribution patterns of the disease in the population; (2) monitor the development of the program and decide when to make adjustments, avoiding the waist of time and resources. As the epidemiological status of bovine brucellosis is not adequately known in Brazil, there was a case for undertaking cross-sectional studies targeted at identifying the best strategies for each state and region, as well as providing a basis for future appraisal of the NPCEBT. For this purpose, MAPA established a collaborative project with FMVZ-Universidade de São Paulo, which also included FAV-Universidade de Brasília. So far, 15 states completed the survey, whose results are described in the following papers. In addition, it is presented a paper that deals with modeling the impact of vaccination of heifers with strain 19 upon the reduction of brucellosis prevalence.

Se evalúa la capacidad operativa de las pruebas diagnósticas: anillo de la leche (PAL), inmunoensayo enzimático indirecto (ELISA-i), rosa de bengala (RB) y ELISA competitivo (ELISA-c), utilizadas tras 5 años (2003-2007) de vigilancia epidemiológica para brucelosis bovina (Brucella abortus) en el municipio Machiques de Perijá, partiendo de la revalorización de un conjunto de muestras de leche y suero probados positivos y negativos. La garantía de diagnóstico previamente positivo fue alcanzada a través de evidencias epidemiológicas y positividad comprobada en suero sanguíneo al ensayo con antígeno buferado en placa y confirmación mediante fluorescencia polarizada (FP); sólo se consideraron negativos probados, los sueros y leches globales provenientes de animales negativos a FP que a lo largo del estudio no presentaron antecedentes serológicos alguno. Los resultados en leche muestran superioridad de ELISA-i versus PAL, en sensibilidad, 98,55 contra 56,52 por ciento; especificidad 99,41 y 98,82 por ciento; predicción positiva 98,58 y 95,12 por ciento; predicción negativa 99,49 y 84,84 por ciento; y cocientes de probabilidad. El nivel de concordancia entre los resultados y el verdadero estatus de las muestras fue también preferente para ELISA-i, 97,8 contra 55,3 por ciento. Las evidencias sobre sueros probados dejan en duda la contribución de RB en el diagnóstico, control y erradicación de la enfermedad en el área de estudio, al demostrarse valores medios de validez (169,92 versus 197,21) y seguridad (171,74 versus 197,02), además de bajos coeficientes de verosimilitud entre sus resultados. ELISA-c, en cambio, mantuvo excelente capacidad diagnóstica.


The performance of the following diagnostic tests: Milk Ring Test (MRT), Indirect Enzyme-linked Immunosorbent Assay (iELISA), Rose Bengal Test (RBT) and Competitive ELISA (cELISA) used for 5 years (2003-2007) in the epidemiological surveillance system for bovine brucellosis (Brucella abortus) in Machiques de Perijá County was evaluated. For this purpose, milk and sera samples previously assessed through the abovementioned tests were reassessed to define their true condition (true positive or true negative). Samples were considered as true positive through epidemiological findings and a positive result in Buffered Plate Antigen Test, which was confirmed through Fluorescence Polarization Assay (FPA); samples were considered as true negative only when they were taken from FPA-negative animals that did not seroconvert during the study. Results from tests in milk showed a higher performance of iELISA versus MRT, in sensibility: 98.55 and 56.52 percent; in specificity: 99.41 and 98.82 percent; in positive predictive value: 98.58 and 95.12 percent; in negative predictive value: 99.49 and 84.84 percent; and in coefficients of probability, respectively. The level of agreement between test and true status of samples was higher in iELISA: 97.8 versus 55.3 percent. Results from true positive and true negative samples warrant a reappreciation of the contribution of RBT in diagnostics, control and eradication of brucellosis in the studied area, taking into account its medium values of validity (169.92 versus 197.21) and accuracy (171.74 versus 197.02), as well as its low coefficients of probability. By contrast, cELISA showed an excellent diagnostic performance.

ABSTRACT

Se evalúa la evolución durante 5 años (2003-2007) de la brucelosis bovina (Brucella abortus) en el municipio Machiques de Perijá. El estudio se realizó identificando en leche, tres veces por año, fincas reactivas a la prueba de anillo (PAL) y positivas a un ensayo inmuno enzimático indirecto (ELISA-i). Paralelamente, fueron analizadas muestras de sangre de la población bovina adulta, procedente de las explotaciones problemas, para identificar animales positivos a Rosa de Bengala (RB) y confirmados, por un ensayo inmuno enzimático competitivo (ELISA-c). Adicionalmente, se analizan los alcances de la enfermedad a nivel de un matadero municipal. Los resultados del monitoreo epidemiológico permiten evidenciar disminución progresiva en la ocurrencia de la problemática. Así, en el 2003, se detectó la existencia de 32,3% (209/648) de fincas reactivas a la PAL y 62,9% (404/583) positivas a ELISA-i, y se constató, la presencia y beneficio de animales positivos a RB/ELISA-c sin la identificación pertinente, lo cual repercute sobre la salud de los trabajadores. Para el 2007, se identifican 11,4% de rebaños reactivos a la PAL y 27,2% a ELISA-i; con una positividad de 0,38% (158/41630) de animales infectados según criterio diagnóstico seriado de RB/ELISA-c, aunque se demuestra sesgo importante en esa medición. Además, se identificaron 178 explotaciones (27,0%) sin evidencia de la enfermedad, que pudieran optar por una clasificación o estatus de libres de brucelosis. En el futuro próximo se hace necesario extender el monitoreo hacia fincas proveedoras de queseras artesanales, evaluar las contribuciones individuales de PAL y ELISA-i, como ensayos primarios en la vigilancia de la problemática, implementar providencias coercitivas de control y explorar nuevas técnicas diagnósticas para la identificación rápida de animales infectados. Estas medidas contribuirán, en mediano plazo, a la erradicación de esta enfermedad en esa región del país.


Bovine brucellosis (Brucella abortus) behavior in Machiques of Perijá County was evaluated for 5 years (2003-2007). The survey was undertaken through Ring Test and indirect ELISA (iELISA) performed on milk samples collected 3 times per year from farms located in Machiques de Perijá County. Moreover, serum samples from the adult bovine population of those farms were analyzed to detect reactors to Rose Bengal Test, which were subsequently confirmed by competitive ELISA (cELISA). Brucellosis prevalence was determined at a county slaughterhouse as well. Epidemiological surveillance results suggest a progressive decrease in brucellosis prevalence. In 2003, 32.3% (209/648) of farms were Ring Test reactors and 62.9% (404/583) were iELISA reactors; also, RBT/cELISA positive animals were slaughtered without having been branded adequately according to the law, which negatively affects slaughterhouse workers’ health. In 2007, 11.4% of herds were Ring Test reactors and 27.2% were iELISA reactors; 0.38% (158/41630) of animals were infected according to RBT/cELISA results, there is an important bias in this serial methodology though. Furthermore, 178 farms (27.0%) did not have reactors to the methods used in this survey, which could be candidates to a brucellosis-free status. In the near future, it would be necessary to extend the epidemiological surveillance for brucellosis to farms supplying traditional cheese making factories; to replace Ring Test for iELISA as the main test for milk; to implement compulsory control measures and to explore newer diagnostic tools for rapid identification of infected animals. These measures will contribute, in a short term, to brucellosis eradication in this region of Venezuela.
