ABSTRACT

Object To develop a high-performance capillary electrophoresis method to determine ferulic acid concentrations in serum of rats treated with Buyang Huanwu Decoction (BHD). Methods Capillary zone electrophoresis was applied for ferulic acid assay, quantitative determination was based on internal standard and detection was carried on by direct UV. The electrolyte buffer was composed of 25 mmol/L borax-methanol (85∶15). Capillary electrophoresis was performed using a 52 cm (30 cm to detector)?50 ?m fused-silica capillary tube. Separation voltage was 20 kV, sampling time was 3 s, detection wavelength was 320 nm, and the temperature was 30 ℃. Results Ferulic acid was successfully separated within 6 min, the recoveries were 97.2% in serum and 103.28% in BHD, respectively. RSD were 3.73% and 0.91% (n=3), respectively. Conclusion This method can supply reference for the determination of ferulic acid in serum samples and BHD.

ABSTRACT

Objective To study the effect of Buyang Huanwu Decoction (BHD) on adhesion molecules in vascular endothelial cells (VECs) of blood stasis rats. Methods Expression of ICAM-1, VCAM-1, PECAM-1, iNOS in vascular endothelial cells of blood stasis rats receiving different doses of BHD was observed by immunohistochemical method and RT-PCR.Results The expression of ICAM-1, VCAM-1, PECAM-1, iNOS was increased in model group, while was decreased by BHD. There was an increase tendency on the expression of molecules and significant dose-effect relationship along with the decrease of the BHD doses. Conclusion The increased expression of adhesion molecules in vascular endothelial cells of blood stasis rats could be reduced by BHD in a dose dependent manner.

Objective To investigate the effects of Buyang Huanwu Decoction(BHD) and two kinds of fractions extracted from BHD on expression of caspase-1,caspase-3,and caspase-8 mRNA after cerebral ischemia-reperfusion in rats.Methods The model of cerebral ischemia-reperfusion was induced by the middle cerebral artery occlusion(MCAO) in rats,via string ligation of arteria carotis interna.The expression of caspase-1,caspase-3,and caspase-8 mRNA in cerebral ischemic tissues was assessed by reverse transcription-polymerase chain reaction(RT-PCR).Results The expression of caspase-1,caspase-3,and caspase8 mRNA in cerebral tissues was significantly increased after 22 h reperfusion following 2 h cerebral ischemia in model group(P

ABSTRACT

Objective To investigate the antithrombosis effects of Buyang Huanwu Decoction(BHD) and its active fractions on cultured vascular endothelial cells(VEC) following the stimulation of thrombin and their mechanism.Methods The human umbilicus vein endothelial cells(ECV 304) were cultured in media containing 10 U/mL thrombin and different dosese of drugs.The levels of tPA and PAI-1 were detected by ELISA,the expression of TF,TFPI,and TM mRNA was measured by RT-PCR,and the expression of PKC? was examined by immunohistochemical staining 24 h later.Results Compared to the control without any treatment,the release of tPA was increased evidently(P0.05).Compared to the data after the mere stimulus of thrombin,each dose of BHD increased the release of tPA(P0.05);And glycoside(1.25 mg/mL) increased the release of tPA(P
