ABSTRACT

ABSTRACT: Gene expression of ErbB1 and ErbB2, and immunostaining of EGFR (Her1) and Her2 (c-erbB-2) were evaluated in this study to ascertain whether these receptors are involved in the evolution of canine premalignant and malignant prostatic lesions, as proliferative inflammatory atrophy (PIA) and prostatic carcinoma (PC). With regards to the intensity of EGFR immunostaining, there was no difference between normal prostatic tissue and tissues with PIA or PC. In relation to Her2 immunostaining, there were differences between normal prostatic tissue and those with PIA and PC, as also differences between prostates with PIA and PC. There was no correlation between EGFR and Her2 immunostaining. ErbB1 gene product was detected in two normal tissue samples, in one with PIA, and in all samples with PC. ErbB2 mRNA was recorded in two canine samples with PIA, in all with PC, but was not detected in normal prostatic tissue. It was concluded that EGFR and Her2 play roles in canine PIA and PC, suggesting that those receptors may be involved in canine prostatic carcinogenesis.


RESUMO: A expressão gênica de ErbB1 e ErbB2 e a imunomarcação de EGFR (Her1) e Her2 (c-erbB-2) foram avaliadas para verificar o envolvimento desses receptores em lesões pré-malignas e malignas da próstata canina, como a atrofia proliferativa inflamatória (PIA) e o carcinoma prostático (PC). Em relação à intensidade de imunomarcação para EGFR, não houve diferença entre o tecido prostático normal e com PIA e PC. Em relação a Her2, observou-se diferença de imunomarcação entre o tecido prostático normal e aqueles com PIA e PC e entre os com PIA e PC. Não houve correlação entre EGFR e Her2. O gene ErbB1 foi detectado em duas amostras normais, uma de PIA e em todas as amostras de PC. O gene ErbB2 foi detectado em duas amostras de PIA e em todas as amostras de PC, não sendo detectado no tecido prostático normal. Conclui-se que EGFR e Her2 atuam nas lesões de PIA e PC, sugerindo o envolvimento destes na carcinogênese da próstata canina.

ABSTRACT

Purpose To investigate the expression and clinical significance of DOG-1 and C-erbB-2 in papillary thyroid carcinoma.Methods Immunohistochemical SP method was used to detect the expression of DOG-1 and C-erbB-2 protein in 48 cases of papillary thyroid carcinoma and 30 cases of benign thyroid lesions (15 cases of nodular goiter and 15 cases of Hashimoto's thyroiditis).Results The DOG-1 positive rate of thyroid papillary carcinoma (27.08%,13/48) was significantly higher than that of benign lesions (0,0/30),the difference was significant (P < 0.05).The C-erbB-2 positive rate (39.58%,19/48) was significantly higher than that of benign lesions (3.33%,1/30),the difference was significant (P < 0.05).Positive expression of DOG-1 as well as C-erbB-2 correlated with advanced TNM and presence of lymph node metastasis.The expression of DOG-1 and C-erbB-2 in patients with multifocal carcinomas combined with lymphatic metastasis showed a significant positive correlation(r =0.503,P =0.024).Conclusion The data suggest that DOG-1 and C-erbB-2 contribute to the pathogenesis and progress of thyroid papillary carcinoma.Our results introduce DOG-1 combined with C-erbB-2 as a promising biomarker for recurrence prediction and target therapy.

ABSTRACT

Objective To evaluate the relation between peripheral nutrient artery and expression of C-erbB-2 in breast invasive ductal carcinoma(IDC) . Methods The peak systolic velocity(PSV) and resistive index ( RI ) of peripheral nutrient artery of tumor in 122 cases with breast IDC and 138 cases with fibroadenoma which proved by operation and pathology were analyzed retrospectively . The C-erbB-2 was measured by immunohistochemical test ,according to the results of immunohistochemical examination ,the masses were divided into negative group( -) ,weakly positive group( + ) ,positive group( + + ) and strong positive group( + + + ) . The relationship between the classification of C-erbB-2 expression and the two factors of peripheral nutrient artery in breast tumor mass were analysed . Results The PSV of trophoblastic artery in IDC group was ( 20 .99 ± 8 .14 ) cm/s , RI 0 .66 ± 0 .07 , the PSV of trophoblastic artery in fibroadenoma group was (15 .56 ± 3 .68)cm/s ,RI 0 .66 ± 0 .07 ,there was significant statistically difference in peripheral nutrient artery of tumor between the breast IDC and fibroadenoma( P < 0 .001) . The RI and PSV of the peripheral nutrient artery in IDC were correlated to the classification of C-erbB-2 expression ( r=0 .323 ,0 .360 ,respectively) ,there was no correlation between the RI and PSV of peripheral nutrient artery in fibroadenoma and the classification of C-erbB-2 expression ( r = 0 .001 ) . Conclusions The spectrum form of peripheral nutrient artery in IDC is high speed and high resistance . The RI and PSV of the peripheral nutrient artery in IDC are correlated to the classification of C-erbB-2 expression .

ABSTRACT

Objective To study HB‐EGF ,C‐erbB‐2 test in differential diagnosis of ovarian benign and malignant tumor and de‐termine the meaning of a complex condition .Methods Totally 80 cases of patients with ovarian tumors were collected from August 2014 to July 2015 ,including 20 cases benign ovarian tumor ,and 40 cases epithelial ovarian cancer ,13 cases of Ⅰ period ,Ⅱ period in 14 cases ,Ⅲ ~ Ⅳ period 13 cases .At the same time ,normal endometrium ovarian tissue from 30 cases volunteers were collected .HB EGF ,C‐erbB‐2 expression in ovarian tissue were detected by using flow cytometry .The immune histochemical method was used to detect C‐erbB‐2 expression from 40 examples carcinoma ,30 cases of normal endometrium intrauterine membrane .Results The rate of positive cells of HB‐EGF ,C‐erbB‐2 was significantly higher than that of benign ovarian tumor(P< 0 .05) ;C‐erbB‐2 for lower ex‐pression in normal endometrium ,endometrial carcinoma in content was higher than the normal uterus express ,the difference was statistically significant(P< 0 .05) ;Endometrial cancer of different pathological staging ,C‐erbB‐2 expression was different also ,in‐cluding C‐erbB‐2 expression in endometrial carcinoma of stage Ⅰ ,Ⅱ significantly below Ⅲ ,Ⅳ period ,the difference was statistical‐ly significant(P< 0 .05) .Conclusion The high expression of C‐erbB‐2 protein is associated with the occurrence and development of endometrial carcinoma ,C‐erbB‐2 testing could help to clinically evaluate the biological behavior and prognosis of endometrial carci‐noma .

Objective To analyze the expressions and clinical significance of human epidermal growth factor receptor-2 (C-erbB-2) protein in sinonasal squamous cell cancer cells.Method Immunohistochemical and RT-PCR methods were used to detect the expression of C-erbB-2 in 62 cases of sinonasal squamous carcinoma tissues,30 cases of nasal polyps and 25 cases with normal nasal mucosa.The relationships between the expression of C-erbB-2 in sinonasal squamous carcinoma tissues and clinical pathological characteristics were analyzed.Results There was significant difference in the expression of C-erbB-2 in sinonasal squamous carcinoma tissues, nasal polyps and normal nasal mucosa tissues(P <0.05).The expression of C-erbB-2 was positively correlated with the clinicopathologic stage, tumor classification and lymph node involvement(P<0.05).Conclusion The expression of C-erbB-2 is correlated with tumorigenesis,invasion and metastasis of sinonasal squamous cell carcinoma.It can be used as an auxiliary diagnostic and prediction maker as well as a new therapeutic target of sinonasal squamous cell cancer.

ABSTRACT

A 37-year-old woman was admitted to our hospital because of cervical and axillary lymphadenopathy that developed after delivery. An axillary lymph node biopsy revealed metastatic adenocarcinoma. Immunohistochemical staining indicated that the tumor cells expressed c-ErbB-2, but were negative for the estrogen and progesterone receptors. No definite evidence of breast cancer was detected. The patient underwent chemotherapy for suspected metastatic breast cancer. She complained of swelling in the left breast 22 months later, and a biopsy showed invasive ductal carcinoma. Here, we report a case of hormone receptor-negative occult breast cancer in a patient with cervical and axillary lymphadenopathy presenting as a cancer with an unknown primary site.

ABSTRACT

Objective To evaluate the relation of appearance of contrast-enhanced ultrasonography (CEUS) to Ki 67 and C-erbB-2 in breast invasive ductal carcinoma and ductal carcinoma in situ.Methods The appearance of CEUS in 29 patients with breast invasive ductal carcinoma and 12 patients with ductal carcinoma in situ,which was diagnosed by surgery or biopsy,was analysed retrospectively.The Ki-67 and CerbB-2 were measured by immunohistochemical test.The relation of the above two factors to appearance of contrast ultrasonography was analysed.Results The positive rate of Ki-67 was 68.97% (20/29) in patients with breast invasive ductal carcinoma and 16.67% (2/12) in patients with ductal carcinoma in situ.The positive rate of C-erbB-2 was 48.2% (14/29) in patients with breast invasive ductal carcinoma and 42.6% (5/12) in patients with ductal carcinoma in situ.The masses had a common appearance of high enhance in patients with breast invasive ductal carcinoma and ductal carcinoma in situ,which was not correlated to C-erbB-2.The Ki-67 was significantly higher in patients with ductal carcinoma in situ than that in patients with breast invasive ductal carcinoma,the appearance of CEUS was correlated to Ki 67.Conclusions The features of micro-vessels by CEUS were correlated to Ki-67 in patients with breast invasive ductal carcinoma and ductal carcinoma in situ.

Objective To discuss the application value of cancer gene c-erbB-2 and its protein products P185 in evaluation of the diagnosis and prognosis of breast cancer with lymph node metastasis,and to explore the relationship between c-erbB-2 expression and tumor pathology classification,relapse.Methods 112 cases with mammary disease were selected as the research subjects.According to different clinical diagnosis,they were divided into breast cancer lymph node metastasis group,breast cancer without metastasis group and mammary gland benign pathological change group.The c-erbB-2 gene and carcinoembryonic antigen(CEA) were detected by immunohistochemical method,enzyme-linked immunosorbent(ELISA) and electrochemical luminescence method.The c-erbB-2 protein(P185)and CEA of the three groups were compared,the sensitivity and specificity of c-erbB-2 expression and CEA in the diagnosis of breast cancer with lymph node metastasis were analyzed.At the same time,the relationship between cerbB-2 expression and tumor pathology classification,relapse was analyzed.Results P185 and CEA index of breast cancer lymph node metastasis group and breast cancer without metastasis group were significantly higher than those of mammary gland benign pathological change group(t =7.093,6.443,9.232,8.508,all P < 0.0 1),and P185 index of breast cancer lymph node metastasis group was significantly higher than without metastasis group (t =2.281,P <0.05),and there was no significant difference of CEA between the two breast cancer groups (t =0.406,P > 0.05).The sensitivity of c-erbB-2 expression in dignosis of breast cancer lymph node metastasis had no difference with CEA (x2 =0.632,P > 0.05),but the specificity was significantly higher than CEA(x2 =11.900,P < 0.01).The positive expression rate of c-erbB-2 and tumor pathology classification had no obvious relation:Postoperative recurrence rate of c-erbB-2 positive expression patients was significantly higher than the negative expression patients(x2 =5.141,P <0.05).Conclusion c-erbB-2 gene and its protein product has good sensitivity and specificity in the diagnosis of breast cancer lymph node metastasis,and has obvious correlation with postoperative breast cancer recurrence,has important reference value in evaluation of the diagnosis and prognosis of breast cancer.

Objective To investigate the roles of aberrant expression of c-myc,c-myb,and c-erbB-2 in the development of breast cancer and its clinical significance.Methods Immunohistochimical technique (S-P method)was used to detect the expression of c-myb,c-myc and c-erbB-2 protein in 150 cases of breast cancer and 30 cases of benign proliferative lesions.heir relation to clinical pathological parameters were analyzed.Results The protein expression levels of c-myc,c-myb,and c-erbB-2 in breast cancer were higher than those in benign proliferative lesions and the difference was significant(all P <0.05).Expression of c-myc protein had obvious relationship to histological grade,lymph nodes metastasis and pathological types(all P < 0.05).Expression of c-myb protein related significantly to histological grade (P < 0.05).c-erbB-2 was associated with the tumor size,grading,PR expression (all P < 0.05).The association among the three kinds of genetic proteins was observed.Conclusion Expressions of oncogenes c-myc,c-myb,and c-erbB-2 proteins are higher in breast cancer and lower in benign proliferative lesions.The difference is significant.They are related to the grading and some other prognostic factors such as tumor size,pathologic types,PR expression and lymph node metastasis.The results suggest that these oncogenes are activated in the development of breast cancer and provoke cancerous cells multiplication.Expressions of oncogenes c-myc,c-myb and c-erbB-2 mightbe considered to be the prognostic indicators of breast cancer.

El cáncer de mama es una de las enfermedades importantes en la mujer y se le dedican grandes esfuerzos científicos y económicos existiendo el propósito de la búsqueda constante de vías de mejor precocidad en el diagnóstico y con ello, mejoramiento del pronóstico. En los rangos etarios de mayor riesgo de presentar esta patología se han descrito varios factores desencadenantes como son los receptores de estrógenos (RE), progesterona (RP) y de la proteína c-erbB-2. Se utilizaron biopsias de cáncer mamario ductal y lobulillar de diagnóstico de rutina, sometidas a reacciones inmunohisto químicas con anticuerpos anti RE, anti RP y específicamente para anti c-erbB-2. El revelado fue a través de kit comercial HRP con AEC y DAB. Las imágenes fueron capturadas con microscopio Olympus CX31, cámara fotográfica digital incorporada y software Micrometrics SE Premium 2011. Los resultados permiten observar reacción positiva en color rojo intenso en las muestras reveladas con AEC y pardo cuando se utilizó DAB, en las células que expresan la marcación y que se distribuyen indistintamente en el sitio del tejido afectado por la patología. Estas técnicas son de uso clínico protocolizado en el diagnóstico de cáncer de mama, por lo cual su estandarización y visualización son de extrema importancia para el laboratorio de histopatología integrado a unidades de patologia mamaria.


Breast cancer is one of the major diseases in women and devote great economic and scientific eforts. The intention of continuing to find ways of better and early diagnosis, and improved prognosis. In the age ranges of increased risk for this disease have been described several triggers such as receptors: estrogen (ER), progesterone (PR), and protein c-erbB-2. Biopses were used ductal and lobular breast cancer diagnostic routine, subjected to immunohistochemical antibodies to ER, PR and specifically anti c-erbB-2. The development was through HRP AEC and DAB commercial kit. Images were captured with Olympus CX31 microscope and digital camera and software built Micrometrics SE Premium 2011. The results allow observed positive reaction in red color in the samples developed with AEC and brown when used DAB in cells expressing the bearing and which are distributed equally at the site of pathology tissue affected. These techniques are protocolized by clinical use in diagnosing breast cancer, so its standardization and visualization are of extreme importance to the histopathology laboratory of integrated units of breast disease.

ABSTRACT

Background: Gallbladder cancer (GBC) is a lethal malignancy presenting at an advanced stage. The pathogenesis is not well categorized, and surgery is the only treatment available at the early stage of the disease. There have been few reports on role of growth factor receptors in GBC. C-erbB2 is one such receptor whose over-expression is being explored in GBC as one of the factors involved in carcinogenesis and possible target for therapy. Materials and Methods: One hundred and four consecutive cases of GBC were retrospectively studied with regard to clinical features, histological type, grade and stage of tumor. Immunohistochemistry for C-erbB2 was done and expression was correlated with different clinic-pathological parameters and survival. Results: C-erbB2 overexpression was seen in 9.4% cases with complete staining and both complete and incomplete staining (2+ and 3+) was seen in 13.4% cases. Eighty percent of the C-erbB2 over-expressed cases were well differentiated and in stage II to stage IV disease. Dysplasia adjacent to carcinoma did not show any expression. No correlation was found with tumor grade, stage, gall stones, and patient survival. Xanthogranulomatous inflammation was inversely correlated with C-erbB2 over-expression. Median survival was 30 months in C-erbB2 over-expressed cases, and 12 months in C-erbB2 negative cases. Conclusion: We found complete membranous staining of C-erbB2 in 9.4% of GBC which was frequent in well differentiated and stage II to stage IV tumors. C-erbB2 tumors had longer median survival than C-erbB2 negative tumors. C-erbB2 is not involved early in the carcinogenetic process as none of the dysplasia showed expression. C-erbB2 over-expression may be considered as target for therapy in advanced stage of GBC.

ABSTRACT

The effect of human cytomegalovirus (HCMV) on invasive capability of early pregnant extravillous cytotrophoblasts (EVTs) was investigated in vitro.Primary EVTs were obtained by complex phosphoesterasum digestion and gradient centrifugation from villous tissue aseptically taken from healthy pregnant women.Cytokeratin7 (CK7),vimentin (Vim) and c-erbB-2 were immunocytochemically detected to identify source of cells,and HCMVpp65 antigen was assayed to determine the infection state of primary EVTs by immunocytochemical staining.The EVTs were divided into two groups:control group and HCMV group,and the expression of c-erbB-2,matrix metalloproteinase-2 (MMP-2) and MMP-9 proteins was detected in two groups by immunocytochemistry and Western blotting.Enzymic activity changes of MMP-2 and MMP-9 were tested by gelatin zymography in primary EVTs infected with HCMV.The invasion of primary EVTs was detected by cell invasion assay in vitro after they were infected by HCMV.The cell source identification showed that the cells obtained were highly-pure primary EVTs,and primary EVTs could be infected by HCMV.Primary EVTs could express c-erbB-2,MMP-2 and MMP-9 proteins,and as compared with control group,the protein expression was decreased significantly in HCMV groups (P<0.05).Primary EVTs could secrete active MMP-2 and MMP-9 in vitro,and the activity of two MMPs was decreased significantly in HCMV groups (P<0.05).The in vitro cell invasion assay showed that the number of primary EVTs permeating Matrigel in HCMV group was decreased (P<0.05).We are led to conclude that HCMV can infect primary EVTs and inhibit their invasion capability,suggesting that the impaired EVT's invasion capability might be related to the abnormal expression of c-erbB-2,MMP-2and MMP-9 proteins.

ABSTRACT

Objective To investigate the expression of c-erbB-2 and cathepsin D (cath-D) in primary breast carcinoma, and to analyze its relationship to the patients' clinicopathological status and its prognostic significance. MethodsThe immnohistochemical test were used to detect the expression of c-erbB-2 and cath-D in 128 patients with primary breast carcinoma. All data were statistically analyzed.ResultsThe positive expression rate of c-erbB-2 and cath-D was 36.5 % and 40.9 %, relatively. Cath-D was correlated with tumor size (x2 =21.0, P <0.0001) and lymph node status (35.29 % vs 63.83 %, P <0.0001). The Kaplan-Meier curve and log rank test indicated that the positive c-erbB-2 expression was a significant prognostic factor for patients disease free survival (P<0.01). Conclusionc-erbB-2 and cath-D show a clinicopathological invasive characteristic. They are prognostic factors for patients with primary breast carcinoma, and also can be prognostic indicator for patients.

Predictive biomarkers such as oestrogen (ER) and progesterone (PR) receptors and c-erbB-2 oncoprotein have become a staple in breast cancer reports in the country as they increasingly play an important role in the treatment and prognosis of women with breast cancers. This study reviews the practice of histopathology reporting of these biomarkers in a Malaysian tertiary hospital setting. Retrospective data on demographic, pathological and biomarker profi les of patients with invasive ductal carcinoma who had undergone mastectomy or lumpectomy with axillary node clearance from 2005 to 2006 were retrieved from the Department of Pathology, Penang Hospital and analysed. The prevalence of ER positivity (55.8%), PR positivity (52.5%), c-erbB-2 oncoprotein overexpression (24%) and triple negativity (ER negative, PR negative, c-erbB-2 negative) (15%) by immunohistochemistry were comparable with other studies. Notably, c-erbB-2 overexpression was equivocal (2+) in 15% of cases. Since about a quarter of equivocal (2+) cases usually show amplifi cation by FISH, a small but certain percentage of patients would miss the benefi t of anti-cerbB- 2 antibody therapy if FISH is not performed. New ASCO/CAP guidelines on the quantitation of ER and PR will probably increase the prevalence of ER/PR positivity, invariably leading to signifi cant ramifi cations on the management of patients as more patients would be deemed eligible for endocrine therapy, as well as categorisation of triple negative breast cancers.

PURPOSE: We investigated the correlation between expression of c-erbB-2 and p53 proteins and clinicopathologic features of gastric cancer to reveal prognostic factors. METHODS: 125 patient records under going curative resection for gastric carcinoma at our institution from January 2000 to June 2003 were collected. Surgical specimens embedded in paraffin block were evaluated for p53 and c-erbB-2 protein as detected by immunohistochemistry. RESULTS: Samples from 30 cases (24.0%) of 125 patients with gastric adenocarcinomas demonstrated positive staining for c-erbB-2 and 72 patients (57.6%) showed positive nuclear staining for p53 protein. c-erbB-2 stained tumors were significantly associated with the depth of tumor invasion (P=0.022), lymph node metastasis (P=0.004) and lymphatic invasion (P=0.019). In a subgroup of patients with gastric carcinoma not exposed to serosa (n=91), expression of both c-erbB-2 and p53 significantly related with poor disease-free survival (OR=5.107) and survival (OR=4.449) in multivariate analysis. CONCLUSION: When patients with gastric adenocarcinoma showed expression of c-erbB-2 with p53, they were associated with aggressive pathologic features. In the subgroup of patients with gastric adenocarcinoma that did not involve serosa, expression of c-erbB-2 combined with p53 could become a predictive factor for recurrence and survival in multivariate analysis.

ABSTRACT

In carcinoma ex pleomorphic adenoma (CXPA), pleomorphic adenoma (PA) and diverse carcinoma components showing luminal (ductal) or non-luminal (myoepithelial) differentiation coexist. To elucidate the clinicopathological implications of cellular differentiation in CXPA and the potential role of p53, vascular endothelial growth factor (VEGF), c-erbB-2, c-kit, and glucose transporter 1 (Glut-1) in carcinogenesis, we analyzed 11 CXPAs with luminal differentiation (CXPAs-LD) and 6 CXPAs with non-luminal differentiation (CXPAs-NLD) and compared protein expressions in residual PAs and carcinomas by immunohistochemistry. Among the CXPAs-LD, 5 were invasive and 8 were histologically high-grade tumors. The 5-year survival rate was 72.7%. P53, c-erbB-2, VEGF, and Glut-1 were more immunoreactive in carcinoma components than in PAs (P = 0.008, 0.004, 0.002, and 0.024, respectively); c-erbB-2 overexpression was associated with high histological grade (P = 0.024). Carcinoma components frequently lacked c-kit expression (P = 0.009). CXPAs-NLD were all low-grade and invasive with a larger mean tumor size (5.2 cm) than CXPAs-LD (3.3 cm) (P = 0.040). The patients remained disease-free without significant immunohistochemical expression. The immunoprofiles and clinical course of CXPA differed according to cellular differentiation. Therefore, it is important to report the histological subtype and to assess potential biomarkers in diagnostic and therapeutic trials.

ABSTRACT

ObjectiveTo study the relationship between C-erbB-2 and estrogen (ER) and progesterone (PR) receptors, and the relationship between C-erbB-2, ER, PR with histologic grade. MethodsTo detect ER, PR and C-erbB-2 states by using immunohistochemical analysis and fluorescence in situ hybridization for C-erbB-2 in 163 unselected invasive breast carcinomas. ResultsC-erbB-2, ER ,PR were expressed in 21.5% ,64.4% ,44.2% of 163 cases respectivly . 5 pure mucinous carcinomas , 3 tubular carcinomas and 1 micropapillary carcinoma were ER + ( 100.0% ) 、C-erbB-2 - ( 100.0% ) and PR + (40.0% ,66.7%, 100.0% ). C-erbB-2 was positive in 22.3% of grade Ⅱ and 27.0% of grade Ⅲ invasive ductal carcinomas and negative in all grade Ⅰ invasive ductal carcinomas.ER and PR expression were decreased significantly in C-erbB-2 + tumors compared with C-erbB-2 - tumors( ER,25. 7% vs 75.0% ; PR,25.7% vs 49.2% ). Although ER or PR expression is decreased in C-erbB-2 + tumors, a substantial proportion of them still express ER or PR. ConclusionC-erbB-2 overexpression or amplifcation was limited to a minority of invasive breast carcinomas. Tumour grade was an independent predictor for ER expression. ER was expressed in small number of high-grade and in large number of grade Ⅰ invasive ductal carcinomas. C-erbB-2 overexpression or amplification essentially was limited to grades Ⅱ and Ⅲ ductal carcinomas and correlated inversely with ER or PR expression.

Methods: Totally 104 patients with IDCs confirmed pathologically were involved in this study. ER, PR and c-erbB-2 expression in the IDC specimens was determined by immunohistochemical staining technique. The correlation between the ultrasonographic features and the positive expression of ER, PR or c-erbB-2 was analyzed. Results: The positive expression rate of ER and PR in the group with tumor spiculation sign and posterior acoustic attenuation was higher than that in the group without. The positive expression rate of ER differed significantly (P 0.05). The over-expression rate of c-erbB-2 in the group of microcalcification, sufficient blood flow and axillary lymph node metastasis was higher than that in the group of non-microcalcification, deficient blood flow or without axillary lymph node metastasis (P 0.05). Conclusion: The expression of ER and c-erbB-2 is closely related to the ultrasonographic characteristics of IDC, which may, to some extent, reflect the expression level of ER and c-erbB-2.

ABSTRACT

Objective: To apply chromogenic in situ hybridization (CISH for detection of HER2 gene amplification in breast cancer tissues and to discuss some modifications of the CISH method. Methods: HER2 gene amplification was detected by CISH in 60 breast cancer specimens with an immunohistochemical score over 2+. The correlation between the results of IHC and CISH was analyzed. Our experience in CISH manipulation was summarized and optimization to CISH was discussed. Results: CISH identified gene amplification in 91% (40/44) specimens with an IHC score of 3+ and in 50% (8/16) specimens with an IHC score of 2+. The total concordance rate between IHC and CISH was 80% (48/60, P<0.01). The thickness of sections should be controlled within 4-5 μm; the denaturation should be complete; and the post-hybridization washing temperature and time were also very important and the temperature should be controlled at 70-75 °C. The dyeing time of hematoxylin should also be restrictedly controlled. Positive control should be set up in the experiment for high quality of the experiment. Conclusion: CISH has high concordance rate with IHC in examining HER2 amplification and it may be a new method for detection of HER2 gene. The thickness of the sections, the post hybridization washing temperature and time, and the time of hematoxylin dyeing should be strictly controlled.
