ABSTRACT

PURPOSE: Activated leukocyte cell adhesion molecule (ALCAM), a member of the immunoglobulin superfamily, is highly expressed on dendritic cells. ALCAM and its receptor CD6 are co-stimulatory molecules in the immunological synapse; their interaction is required for T cell activation. While atopic dermatitis (AD) is recognized as a T helper 2 (Th2)-mediated allergic disease, the role of ALCAM in its pathogenesis is unclear. METHODS: ALCAM levels were measured in the serum of AD patients and AD-induced murine model by ovalbumin treatment. We next investigated transepidermal water loss, clinical score, Th2-immune responses, skin barrier gene expression and T-cell activation using wild-type (WT) and ALCAM deficiency mice. An oxazolone-induced AD-like model was also established and analyzed using WT- and ALCAM-deficient mice. RESULTS: We found that serum ALCAM levels were elevated in pediatric AD patients as well as WT AD mice, whereas Th2-type cytokine production and AD symptoms were suppressed in ALCAM-deficient mice. In addition, CD4+ effector T-cell counts in murine skin and skin-draining lymph nodes were lower in ALCAM-deficient mice than in their WT counterparts. ALCAM deficiency was also linked to higher expression of skin barrier genes and number of lamellar bodies. CONCLUSIONS: These findings indicate that ALCAM may contribute to AD pathogenesis by meditating a Th2-dominant immune response and disrupting the barrier function of the skin.

ABSTRACT

Objective:To study the clinical significance of tumor tissue of patients with liver metastasis of colorectal cancer CD44,CD166 expression.Methods:Retrospective analysis clinical data in our hospital from Octobor 2008 to October 2013,these data from 76 patients with rectal cancer was divided into the test group ( liver metastasis group, n=40 ) and control group ( without liver metastases group, n=36 ) .All of specimens were identified with qualitative immunohistochemical analysis.Observation the positive expression of CD166/CD44, age, sex, clinical stage, invasion depth, lymph node metastasis, Dukes staging, pathological tissue differentiation parameter correlation and the survival rate.Results: In test group,the positive expression of CD44,CD166 groups were respectively 72.50%,85%;in the control group,the positive expression rates of CD44 and CD166 were 36.11%,41.67%.The two groups have significant difference(P0.05) with age,gender, histological differentiation.However it showed significant difference with the depth of invasion, Dukes ( P<0.05 ) .In addition,the survival ratio of the test group was significant lower than control group(P<0.05).Conclusion: In rectal cancer,CD166 and CD44 are a hallmark of liver metastasis,rectal cancer,which could determine the process of development.To explore the expression of CD44 and CD166 is helpful to early diagnosis and treatment scheme of rectal cancer.

ABSTRACT

Objective To study the expression of activated leukocyte cell adhesion molecule (ALCAM/CD166) in serous ovarian tumors and its significance.Methods The expression level of CD166 protein,estrogen receptor,progestogen receptor were detected by immunohistochemical staining in 56 cases of malignant tumors,30 cases of borderline tumors and 10 cases of benign tumors,the relationship was investigated between the expression of CD166 and clinicopathologic information and its correlation with the expression of estrogen receptor (ER) and progestogen receptor (PR).Results The positive rates of CD166 protein in malignant,borderline and benign epithelial ovarian tumors were 87.5 % (49/56),56.7 % (17/30) and 10.0 % (1/10) respectively,there were significant differences (x2 =7.463,31.054,10.563,all P < 0.01).There were also significant differences in positive rates of CD166 protein among different clinical stages,pathologic grades and lymph node metastasis of epithelial ovarian tumors (x2 =41.205,Z =-4.663,Z =-2.981,all P < 0.01).There was a negative correlation between the expression of CD166 protein and the leves of ER and PR (x2 =23.36,21.441,both P < 0.05).Conclusion CD166 takes part in development and progression of serous ovarian carcinoma.The expression of CD166 increase significantly in ovarian serous carcinoma.
