ABSTRACT

Lung cancer is the most commonly diagnosed cancer and the leading cause of cancer death. Although great progress has been made in chemotherapy, radiotherapy and targeted therapy, the emergence of acquired drug resistance hinders the efficacy of clinical treatment. Studies have shown that tumor is a class of diseases with damaged cell cycle regulation mechanism, in which checkpoint kinase (Chk) plays a core role, Chk1 and Chk2 are very important protein kinases in the checkpoint. In recent years, it has been found that the regulation of Chk1 and Chk2 plays an important role in the clinical treatment and drug resistance mechanism of lung cancer. This article reviews the mechanism of cell cycle checkpoint kinase and drug resistance of lung cancer, and expounds the effective therapeutic targets and methods of lung cancer.

ABSTRACT

Acute kidney injury(AKI) has emerged as a major public health problem that leads to decreased survival.In AKI,cell hyperplasia,hypertrophy and apoptosis are related to the cell cycle control.Two supervisory restriction points,G1/S and G2/M checkpoints,are responsible for cell-cycle control.Furthermore,cell cycle regulatory proteins are also involved in the regulation of the cell cycle in AKI.This review focuses on the cell cycle regulation mechanism of AKI.

ABSTRACT

To review the recent research advances on the cell cycle checkpoint kinase 1 (Chk1) inhibitors. METHODS: Through searching and summarying literatures of domestic and abroad, the research progress of the Chk1 inhibitors were introduced comprehensively. RESULTS AND CONCLUSION: Chk1 inhibitors combining with DNA damaging agents have better anti-tumor efficacy. With the deepening studies on the Chk1 inhibitors and their structure-activity relationship, Chk1 inhibitors will have broader development prospects and play more important roles in the treatment of cancer.

ABSTRACT

Cell cycle checkpoints are protective mechanism responding to DNA damages originated from externalor internal factors. When cells are exposed to genotoxic stress or when nutrition crisis occurs, cell cycleprogression is usually stopped or slowed down by cell cycle checkpoints to allow for DNA repair or for handlingthe crisis. Besides, recent studies suggest that some cell cycle checkpoint proteins are also involved in regulatingphysiological DNA replication via controlling the rate of DNA replication. Cell cycle checkpoint proteins ATR,9-1-1 complex, Chk1, Cdc25A and CDK2 may participate in this process. This kind of regulation is supposed to bevery important for ensuring accurate DNA replication and maintaining genomic stability.
