ABSTRACT

Objective To explore and analyze the epidemiology of susceptibility to chronic obstructive pulmonary disease (COPD) among the elderly population in Liangjiang New Area of Chongqing based on CXC chemokine receptor 3 (CXCR3) gene polymorphism. Methods From January 2020 to September 2022, the Medical Laboratory Department of Chongqing Liangjiang New Area People's Hospital selected COPD patients and received treatment. Among the 276 patients who met the criteria were included in the study and included in the observation group. Among the 512 patients with healthy pulmonary function in the same period were included in the control group. The data of the two groups of patients were analyzed, and the genotypes were detected by SBaPhotoshot technology to analyze the relationship between gene polymorphism and the susceptibility and clinical characteristics of COPD. Results There was no significant difference between the two groups in age, sex, BMI and blood eosinophil granulocyte levels, which was comparable (P>0.05). There were significant differences in smoking history, pulmonary function index , MMP-9 and TIMP-1 levels (P0.05). In the observation group, the MMP-9 level of rs2280964 locus was significantly different (P=0.003), while the TIMP-1 level was not significantly different (P=0.187); There was no significant difference in MMP-9 and TIMP-1 levels among the three genes at rs34334103 locus (all P>0.05). The level of MMP-9 in homozygous TT patients with rs2280964 locus was significantly higher than that in homozygous CC patients (P=0.024). There were differences in FEV1/FVC of patients with CXCR3 rs34,334,103 gene distribution (P=0.008), among which there were significant differences in CC+CT and TT recessive models (P0.05). Conclusion CXCR3 gene polymorphism is significantly associated with the susceptibility to COPD, and also with the serum levels of MMP-9 and FEV1/FVC, which can be used as a new target for clinical research and treatment.

ABSTRACT

ObjectiveTo preliminarily explore the mechanism of Shufeng Tongluo prescription (SFTLP) in inhibiting airway inflammation in asthma mice by affecting the expression levels of eotaxin in the serum, CC type chemokine receptor 3 (CCR3), and extracellular signal-regulated kinase (ERK) phosphorylation in lung tissues. MethodSeventy C57BL/6 mice were randomly divided into a blank group, a model group, low-, medium-, and high-dose SFTLP groups (7.75, 15.5, 30 g·kg-1), a pertussis toxin (PTX) group, a CCR3 inhibitor (SB328437) group, a phosphoinositide 3-kinase (PI3K) inhibitor (LY294002) group, a p38 protein kinase antagonist inhibitor (SB203580) group, and an ERK inhibitor (PD98059) group. The asthma model was induced in mice by intraperitoneal injection of ovalbumin (OVA) and aluminum hydroxide [Al(OH)3] combined with OVA atomization (0.2 mL for all). After modeling, hematoxylin-eosin staining (HE staining) was used to observe the inflammatory infiltration of lung tissues in mice. Enzyme-linked immunosorbent assay (ELISA) was used to detect the serum levels of eotaxin [CC chemokine ligand (CCL) 11 and CCL24) in each group. Western blot was used to detect the levels of ERK phosphorylation and CCR3 in lung tissues. ResultCompared with the blank group, the model group showed obvious bronchial constriction, lumen stenosis, damaged alveolar structure, massive inflammatory cell infiltration in lung tissues, mucous plug in the bronchus, edema in the submucosal tissues of the trachea, increased folds, increased serum levels of CCL11 and CCL24 (P<0.01), and increased expression of CCR3 protein in lung tissues (P<0.05). The ERK levels in lung tissues of the model group and the PTX group increased (P<0.05). The level of p-ERK in lung tissues of the model group and the low-dose SFTLP group increased (P<0.05). As revealed by pathological results, compared with the model group, the high-dose SFTLP group showed relieved lung lesions. The high-dose SFTLP group and the SB328437 group showed reduced CCL11 content (P<0.05). The low- and high-dose SFTLP group, the PTX group, the SB203580 group, the PD98059 group, and the SB328437 group showed decreased CCR3 protein expression in lung tissues (P<0.05). The high-dose SFTLP group and the PD98059 group showed reduced p-ERK level (P<0.05). The PD98059 group showed reduced ERK level (P<0.05). ConclusionSFTLP can inhibit airway inflammation in asthma, and the mechanism may be related to the inhibition of eosinophil activation by down-regulating CCR3 and CCL11 expression and ERK phosphorylation.

ABSTRACT

OBJECTIVES@#This study aimed to determine whether a correlation existed between CXC chemokine ligand 10 (CXCL10)-CXC chemokine receptor 3 (CXCR3) and CC chemokine ligand 17 (CCL17)-CC chemokine receptor 4 (CCR4) in the pathogenesis of oral lichen planus (OLP).@*METHODS@#Peripheral blood of OLP patients (non-erosive and erosive groups) and healthy controls were collected, and T cells were isolated and purified. T cells were co-cultured with three groups: blank, anti-CXCR3, and anti-CCR4. CXCR3 and CCR4 expression were detected by flow cytometry, and CXCL10 and CCL17 were detected by enzyme-linked immunosorbent assay, respectively.@*RESULTS@#The purities of T cells were all >95% in the three groups (@*CONCLUSIONS@#Two axes interact with each other in the pathogenesis of OLP and may play different roles in its occurrence and development.

ABSTRACT

@#AIM: To investigate the expression of vascular endothelial growth factor(VEGF)and chemokine receptor 3(CCR3)in pterygium.<p>METHODS: The expression of VEGF and CCR3 were detected in 18 patients(18 eyes)with pterygium and compared with the normal conjunctiva epithelium in other 14 cases(14 eyes). The histological differences between the two groups were compared by hematoxylin and eosin(HE)staining. The expression of VEGF and CCR3 in pterygium and normal conjunctival tissues were analyzed by immunofluorescence and immunohistochemical. The mRNA expression of VEGF and CCR3 were assessed by Real-time PCR.<p>RESULTS: Compared with normal conjunctival tissue, the epithelial layer of pterygium was obvious thickening, the stromal layer was more disordered, and the expression levels of VEGF(0.69±0.0875 <i>vs</i> 0.05±0.0024)and CCR3(0.45±0.0248 <i>vs</i> 0.03±0.0074)were significantly elevated(all <i>P</i><0.01). The expression level of VEGF mRNA in pterygium was about 12 times that of normal conjunctival tissue(12.33±2.84 <i>vs</i> 1.00±0.08), and the expression level of CCR3 mRNA was about 160 times as many as conjunctival tissue(159.60±34.15 <i>vs</i> 1.00±0.09).<p>CONCLUSION: The expression of VEGF and CCR3 in pterygium was significantly increased, suggesting that they may be involved in the occurrence of pterygium and promote the development of the disease.

ABSTRACT

ObjectiveTo explore the expression of monokine induced by interferon- γ(Mig) in serum and chemokine receptor 3(CXCR3)on lymphocytes of peripheral blood in children with bronchiolitis.MethodsIn this study, 55 patients with bronchiolitis in our hospital were randomly recuited, and were divided into two groups: atopic group and non-atopic group. Of the same age 26 healthy children had been enrolled randomly as control group. The level of CXCR3 expression (CD183) on lymphocytes of peripheral blood was detected by lfow cytometry in all children. The level of Mig in serum was assayed by ELISA.ResultsThe level of CD183 expression on the CD3+CD4+T lymphocytes in atopic group and non-atopic group(16.39±4.13%,14.39±3.74 %)were higher than that of control group(11.17±3.13%,P<0.05),CD183+CD4+/CD4+% in atopic group were higher than that in non-atopic group(P<0.05). The level of CD183 expression on CD3+CD8+T lymphocytes in atopic group and non-atopic group(67.18±10.57 %, 61.44±11.46 %)were higher than that of control group(51.19±5.42 %, P<0.05),CD183+CD8+/CD8+% in atopic group were signiifcantly higher than that in non-atopic group(P<0.05). The level of Mig in serum of children with bronchiolitis in atopic group and non-atopic group(99.67±35.77ng/L, 120.28±32.28ng/L)were signiifcantly higher than that in control group(63.90±15.82 ng/L,P<0.05). The level of Mig in non-atopic group was higher than that in atopic group, there was signiifcant difference(P<0.05).ConclusionsMig and CXCR3 are involved in the pathogenesis of bronchiolitis, and CXCR3 may relate to allergic factors.

ABSTRACT

Objective:To investigate the effects of CCR3 on Muc5ac in the airway of asthmatic mice.Methods: Fifty clean BALB/c mice were randomly divided into control group,asthmatic group,dexamethasone treatment group,SB328437 treatment group, vehicle-control group.Total cells and differential inflammatory cells were counted in BALF, the levels of IL-4 and TNF-αwere determined by ELISA,lung tissue HE staining the expressions of Mucin5ac(Muc5ac) and CCR3 in lung tissue were detected by immu-neohistochemical staining and RT-PCR.Results:The total cells,eosinophil,monocytes and lymphocyte cells in BALF,the levels of IL-4,TNF-αin BALF ,the goblet cell of airway wall,the expression of Muc5ac and CCR3 positive staining IOD in the airway and the expression of Muc5ac and CCR3 mRNA lung tissue obviously decreased in SB328437 treatment group and DEX treatment group which compared with asthmatic mice model group ( P<0.05 ) .Conclusion: SB328437 can inhibit the expression of CCR3 in pulmonary tissue,furtherly inhibit the expression and secretion of Muc5ac and control the airway inflammation.

ABSTRACT

Objective Toexplore the expressions of interleukin-16 (IL-16), interferon-γ (IFN-γ), CXC chemokine receptor 3 (CXCR3), and CRP and their clinical significance in acute exacerbation chronic obstructive pulmonary disease by observing the changes in these factors in patients with AECOPD. Methods 103 patients with AECOPD and 20 healthy controls were collected. According to the 2013 GOLD guideline, all the patients with AECOPD were divided into4 groups(group A of 21 patients, B of 30, C of 27, andD of 25). Results As compared withthe control group, plasma concentrations of IL-16, IFN-γ, CXCR3. and CRP were significantly increased in the patients with AECOPD (P < 0.01), and as the severity of the disease was elevating, these expression levels were significantly increased.While the expression levels of IL-16, IFN-γ, CXCR3, and CRP levels were significantly reduced after treatment, but they were still higherthan those in the control group (P < 0.05). The expression levels of serum IL-16, IFN-γ, CXCR3, and CRP were significantly correlated in patients with AECOPD. Conclusions Expressions of IL-16, IFN-γ and CXCR3 are significantly increased in AECOPD, which is correlated with disease severity and decreased after treatment, suggesting that these three factors may be associated with the occurrence and development of COPD.

ABSTRACT

Chemokine receptor 3 (CCR3) and its ligands eotaxin are inflammatory factors,and they were thought to be related to asthma and allergic diseases before.Recent studies showed that CCR3 plays significant roles in many eye diseases,such as choroidal noevascularization (CNV),allergic cunjunctivititis,age-related macular degeneration (AMD),central retinal vein occlusion and retinal degeneration etc.The biological function study suggested that CCR3 and eotaxin participate in not only promoting the transportation of eosinophilic granulocyte and mastocyte but also inflammatory response.To understand the roles of CCR3 and eotaxin in eye diseases has a very important significance for the recognition and treatment of eye diseases.The structure and function of CCR3 and its ligands,and the relationship between CCR3 and some eye diseases were reviewed.

ABSTRACT

Serum levels of CXC chemokine ligand 10 (CXCL10) and chemokine receptor 3 (CXCR3) were determined in 50 patients with type 1 diabetes mellitus (T1 DM) and 30 normal control subjects by ELISA method.The results showed that serum levels of CXCL10 and CXCR3 in T1DM patients were significantly higher than those in normal subjects [(258.17 ± 39.12 vs 96.47 ± 26.91) ng/L,(851.87 ± 70.04 vs 441.82 ± 72.24) pg/ml,both P<0.05].Serum level of CXCL10 in patients dropped sequentially with durations of diabetes <6 weeks,≥6 weeks or <3 years,and ≥ 3 years,being statistically significant between groups (P<0.05).These results suggest that serum levels of CXC10 and CXCR3 may reflect the immune activity in T1DM.
