ABSTRACT

Tissue engineering is rapidly growing now and can become a promising alternative to transplantation of organs and tissues,as it is devoid of major shortcomings of transplantology, such as acute shortage, complexity of selection, delivery andstorage of donor material, lifelong immunosuppressive therapy. One of the most widely known methods of obtainingbiological scaffolds for the subsequent creation of tissue-engineered constructs of organs and tissues is decellularization.The evaluation of the quality of the obtained scaffolds, based on the study of the viability of cell structures in decellularizedand recellularized matrices, is one of the priorities of modern regenerative medicine worldwide. In this investigation, thebiophysical criteria of decellularization and recellularization of tissue-engineered constructs based on the evaluation of thegeneration of free radicals in native, decellularized and recellularized tissues by EPR spectroscopy and chemoluminescencein a complex assessment of the quality of biological matrixes obtained are considered using intrathoracic organs and tissuesof rats. It has been established that the intensity indices of free radical generation in native and recellularized tissues ofanimal organs, as well as in decellularized matrices, can serve as one of the express criteria for quantitative assessment ofcell structures viability

ABSTRACT

Objective To compare the analytical performance and results correlation of cortisol detection by the Beckman Access 2 Immunoassay System chemiluminescence system and the Roche COBAS E 411 electrochemical luminescence system .Methods The precision ,functional sensitivity ,reference range ,positive coincidence rate and negative coincidence rate of both Beckman Access 2 Immunoassay System and Roche COBAS E 411 systems for detecting cortisol were evaluated according to the method of ISO15198 .Results Both Beckman Access 2 Immunoassay System and Roche COBAS E 411 system exhibited better precision ,the functional sensitivity conformed to the lower limit of clinical detection set by the manufacturers ,after establishing the reference range of cortisol by the laboratory ,the cortisol values detected by these two systems were performed the linear regression analysis , the results indicated that the correlation between them was good ,the linear regression coefficient r2 =0 .977 ,and the positive and negative coincidence rates were 100% .Conclusion The two analytical systems have excellent performance for cortisol ,which all meet the clinical detection requirements .The methodological comparison shows good correlation between them ,and the detection re‐sults could be cited between these two systems by the regression equation .

ABSTRACT

OBJECTIVE To study the phenomenon of hepatitis B surface antigen(HBsAg) and antibody(HBsAb) both positive in blood samples.METHODS Using electro-chemo-luminescence immune system E170 to detect HBsAg and HBsAb in clinical samples and retest the both positive cases by another two reagents for HBsAg and another three reagents for HBsAb.RESULTS All 7332 samples showed 140 both HBsAg and HBsAb positive(1.9%) and 73 of them were retested for HBsAb by another three reagents which gave altogether 24 positive results,which was not related to each other.Those gave higher HBsAb value by E170 showed higher positive rate by Abbott reagent.CONCLUSIONS HBsAg and HBsAb both positive results are not rare.It may be due to the interactions between the patient and the infected hepatitis B virus.Besides,the quality of reagents and correct lab operating procedures may also affect the test results.
