ABSTRACT

The clinical data of 25 patients with acute exacerbations of chronic obstruction pulmonary disease (AECOPD) admitted from March 2009 to October 2010 were analyzed. The sputum samples were collected and the levels of inflammatory cell, interleukin-8 (IL-8) and eotaxin in sputum were determined. The lung function was measured. Twenty healthy subjects served as controls. The counts of neutrophils and eosinophils, and the levels of IL-8 and eotaxin in sputum of AECOPD were higher than those of health subjects. There was a positive correlation between the levels of IL-8 and the counts of neutrophils (P < 0. 01), and between the levels of eotaxin and the counts of eosinophils in sputum ( P < 0.01). The levels of IL-8 and eotaxin were negatively correlated with the lung function (FEV1% ).

ABSTRACT

Objective To investigate the plasma levels of regulated upon activation normal T cell expressed and secreted (RANTES), eotaxin, tumor necrosis factor (TNF)-α and leukotriene B4 (LTB4) in patients with chronic urticaria and their roles in the pathogenesis of chronic urticaria. Methods Forty-one patients with chronic urticaria were included into this study along with 20 normal human controls. Patients were graded into three groups, I.e. Mild group (n = 11), moderate group (n = 21) and severe group (n = 9), according to their symptom score. All patients were treated with mizolastine 10 mg per day for 4 weeks. ELISA was used to study the plasma levels of RANTES, eotaxin, TNF-α and LTB4 in normal controls and patients before and after treatment. Results The plasma levels of RANTES, eotaxin, TNF-α and LTB4 were (52.5 ± 10.2) g/L, (58.4 ± 16.1) g/L, (35.1 ± 9.6) ng/L and (109.4 ± 21.7) ng/L, respectively, in untreatedpatients with chronic urticaria, compared to (33.7 ± 9.4) g/L, (48.3 ± 13.6) g/L, (21.3 ± 8.9) ng/L and(77.8 ± 11.6) ng/L, respectively, in normal controls(P < 0.01, 0.05, 0.01, 0.01, respectively). Increased plasmalevels of RANTES, eotaxin, TNF-α and LTB4 were observed in patients with moderate or severe chronic urticaria compared with those with mild chronic urticaria (both P < 0.05), whereas there was no significant difference between patients with severe and mild chronic urticaria (P < 0.05). After treatment with mizolas-fine the plasma levels ofRANTES, eotaxin, TNF-α and LTB4 were (36.3 ± 8.9) g/L, (46.3 ± 10.2) g/L, (23.2 ± 7.5) ng/L and (83.1 ± 14.2) ng/L respectively, significantly lower than those in patients before treatment (all P < 0.01), but showed no difference from those in normal controls (all P > 0.05). Conclu-sions The plasma levels of RANTES, eotaxin, TNF-α and LTB4 are elevated in patients with chronic urticaria, and they exhibits a positive correlation tendency with disease activity. After treatment with mizolastine, a significant decrease is observed in the plasma levels of RANTES, eotaxin, TNF-α and LTB4, which hints that RANTES, eotaxin, TNF-α and LTB4 may play a certain role in the pathogenesis of chronic urticaria.

ABSTRACT

Objective To investigate the effect of immunoglobulin G (IgG) in bullous pempbigoid (BP) blister fluid on the secretion of chemokines by human keratinocytes. Methods IgG was obtained from the blister fluid of patients with bullous pemphigoid and sera of normal human controls, then purified by sequential precipitation with caprylic acid and ammonium sulfate. The immunological activity of blister fluid was tested before and after the purification by BP180 ELISA kit. Keratinocytes were isolated from the foreskin tissue of yong adults, and subjected to primary culture. After 3 passages, the primary keratinocytes were harvested and subcultured in the presence of purified IgG of 0.5, 1, 2, 4 and 8 g/L, respectively, for 24 hours, or IgG of 4 g/L for 3, 6, 12, 24 hours, respectively, followed by the detection of levels of eotaxin, monocyte chemoattractant protein (MCP)-1 and interleukin (IL)-8 in the supemate of keratinocytes by ELISA. Results The valence of IgG remained unchanged after the purification with caprylic acid and ammonium sulfate. Compared with IgG from the sera of normal controls, that from bullous pemphigoid blister fluid sig- nificantly enhanced the secretion of IL-8 by keratinocytes in a time- and dose-dependent manner (both P < 0.01 ). Neither eotaxin nor MCP-1 was detected in the supemate of control IgG-treated, BP IgG-treated or untreated keratinocytes. Condusions The IgG in BP blister fluid has been proved to stimulate the secretion of IL-8 by cultured human keratinocytes, which may be involved in the pathogenesis of BP.

ABSTRACT

Objective To investigate the molecular mechanism of inhibitory effect of Danshen injection on allergic airway inflamma-tion of asthma. Methods 30 SD rats were random divided into control group, asthma group and Danshen injection group. Bronchoalveolar lavage fluid (BALF) was collected, and cytology study was conducted. The lung tissue was obtained and pathologic analysis was done through HE staining, lnterleukin-13 (IL-13) and Eotaxin in lung tissue were measured by RT - PCR and immunohistochemistry SP method. Results Compared with the asthma group, less infiltration of inflammatory cells in lung tissues was observed in the Danshen injection group. Total cell number, the percentage of lymphocytes, neutrophils and eosinophils (Eos) in BALF of Danshen injection group were lower than that of asthma group (P<0.05, P<0.01). The expression of IL-13 and eotaxin in lung tissue of asthma group was higher than that of control group and Danshen injection group (P<0.05). The expression of IL-13 was negatively correlated with eotaxin (r=0.90, P< 0.05). Conclusion Danshen injection could suppress airway inflammation of asthmatic rats, which probably be through decreasing the ex-pression of IL-13 and eotaxin in the lung tissue of asthmatic rats.

ABSTRACT

AIM: To investigate the levels of IL-18, IL-16, IL-8, eotaxin and the chymase activity in the sputum of asthmatics. METHODS: IL-18, IL-16, IL-8 and eotaxin levels were detected with sandwich ELISA procedures and chymase activity was determined spectrophotometrically (410 nm) by the rate of hydrolysis of N-succinyl-L-Ala-L-Ala-L-Pro-L-Phe-p-nitroanilide (SAAPP). RESULTS: The specific chymase activities in the severe and moderate asthmatics were higher than that in controls. Native protease inhibitors ?_1-antitrypsin (?_1-AT) and soybean trypsin inhibitor (SBTI) inhibited 71.9% and 72.1% enzymatic chymase activity, respectively. The levels of IL-18, IL-16, IL-8 and eotaxin were significantly elevated in the sputum of patients with acute asthma. There were correlations between the levels of IL-8 and IL-16 (r=0.55, P
