ABSTRACT

The bone marrow hematopoietic niche is the place where hematopoietic stem cells and progenitors survive. It is not only related to the hematopoiesis function, but also its steady-state disorders and abnormalities can induce a variety of hematologic diseases, which will even be adapted by malignant cells such as leukemia stem cells, so as to cause seriously poor prognosis. Therefore, it is of great significance to the regulation and construction of the bone marrow niche. In this review, we outline the composition, physiological and pathological basis of bone marrow hematopoietic niche, from the regulation of Chinese materia medica components and TCM formula to graphically analyze the target and effect of drug efficacy. Then, combined with the clinical practice for integrating and summarizing decentralized research, aiming to provide new directions and perspectives for the TCM research and treatment of hematologic diseases.

ABSTRACT

The quality of Chinese materia medica (CMM) has become an important factor affecting the inheritance and development of traditional Chinese medicine. However, due to the complexity of CMM ingredients and the specificity of prescription application, the current quality standards of CMM are difficult to guarantee clinical efficacy. Academician Changxiao Liu proposed the concept of the quality marker (Q-marker) of CMM and its basic attributes, which provided a direction for the study of CMM quality standards. Chinmedomics, integrating serum pharmacochemistry and metabolomics technology, under the conditions of correspondence of prescription-syndrome with therapeutic effect, can be used to find the Q-markers of CMM which are related to the clinical efficacy, and reflect the compatibility of prescriptions, and trace the in vivo metabolism and preparation process. The research strategy based on Chinmedomics is an effective method for discovering the Q-markers of CMM, it has been applied to discovery Q-markers of Yinchenhao Decoction, Liuwei Dihuang Pill, NanShi Capsule, AS1350, and other CMM formula as well as the health products.

ABSTRACT

Chinese materia medica (CMM) formula granule is made from CMM extract powder and suitable pharmaceutical excipients, powder properties of CMM extract powder such as the particle size, hydroscopicity, and fluidity directly affect thepelleting quality of CMM formula granule. Drying is one of the indispensable operating units for preparations of CMM extract, which has great influence on the powder properties of CMM extract powder. Therefore, there is a certain correlation between the drying process and pelleting quality of CMM formula granule. Through the literature research, this paper summarizes the engineering principle and characteristics of each main granulation method, analysis of the evaluation factors of pellet quality, and combines with experimental research to explore the correlation between the drying process of CMM extract and pelleting quality of CMM formula granule in the perspective of powder properties. Finally, we puts forward that more attention should be payed to study the drying process of CMM extract, in order to further improve the quality of pelleting quality of CMM formula granule.

ABSTRACT

Recently, the Chinese materia medica (CMM) compatibility theory research was focused on aspect of chemical composition, which is an important strategy to explain the principle of prescriptions. There are various chemical compositions in a compound prescription of CMM, including alkaloids, glycosides, organic acids, tannins, protein, micro elements and so on. Complex interactions always inevitably occur during the water decoction process of CMM prescription after the dissolution of active ingredients, most significantly, it also generates the precipitation. Clinically, traditional Chinese medicines (TCM) treat diseases with both precipitation and decoction. Precipitation generated process might contain the scientific connotation of CMM compatibility. Thus, the scientists had been studying the precipitation from this process. Through summarizing the results of related researches in recent years, this review introduces different interactions of various active ingredients in compound prescriptions and research progress of precipitation in decoction, aiming to lay a foundation for further study on the precipitation from CMM decoction and provide a new idea to explain the theory of CMM compatibility.

The connectivity map (CMAP) database is established initially to connect biology, chemistry, and clinical conditions, which helps to discover the connection of disease-gene-drug. The CMAP approach has been applied in the field of drug discovery and development, which is widely recognized. In recently years, CMAP analysis has been applied in the research on Chinese materia medica (CMM). The study of CMM is facing a wide range of challenges, such as complicated ingredients, multiple targets, multiple pathways of action and complex functioning mechanism. The idea of employing CMAP in the CMM research has brought a new perspective for researchers and provides a systematic method for elucidating the mechanism of CMM.

ABSTRACT

As the fundamental rule to construct multi-herb prescriptions of Chinese materia medica (CMM), formula compatibility is the key to study the principles of CMM prescriptions. Modern scientific connotation on the formula compatibility could be investigated on the aspects of chemistry, pharmacokinetics, and pharmacodynamics, according to the interactions between CMM prescriptions and the organism. The chemistry of CMM prescriptions is a prerequisite to investigate formula compatibility, and chemical alterations, including quantitative and qualitative changes,induced by formula compatibility are especially emphasized. Formula compatibility can modify the pharmacokinetic behavior of drugs by regulating key steps of pharmacokinetic process to influence the effect and toxicity of CMM prescriptions. Ingredients of a formula affect its therapeutic actions;to elucidate the therapeutic mechanisms of formula compatibility, a new method named "combination of drug targets guided by the pathophysiology of diseases" is proposed to investigate the phenomenon that multi-components of a formula act on multi-target in the organism. It sheds light on understanding the mechanisms of improving effect/reducing toxicity of formula compatibility and provides the reference to enhance the modern scientific research of CMM prescriptions.

ABSTRACT

The development of Chinese materia medica (CMM) formula granule is a great reform on CMM. Compared with the traditional herbal pieces, CMM formula granule is not only convenient and rapid-acting as boiling-free, but also advanced in producing techniques, and easily handled in both storage and quality-control. It could perfectly meet the basic reqirement on modern drugs and have a broad market prospect both at home and abroad. The popularization and application of CMM formula granule could be a new way to promote the internalization of CMM. Through the analysis of the pushing effects, international competitive advantages, and problems on accessing to the international market, this essay tries to put forward some effective countermeasures for CMM formula granule with expecting to provide a new idea and a new route for the internationalization of CMM.

ABSTRACT

Objective To investigate the chemical constituents of Baoyuan Decoction.Methods Many chromatographic techniques and spectral analysis means were employed for isolation and identification of the constituents.Results Fifteen flavonoids were isolated from Baoyuan Decoction and their structures were identified as licuraside(1),formononetin-7-O-?-D-glucoside(2),formononetin(3),isomucronulatol(4),davidigenin(5),2',4'-dimethoxy-3'-hydroxyisoflavan-6-O-?-D-glucoside(6),(6aR,11aR)9,10-dimethoxypterocarpan-3-O-?-D-glucoside(7),calycosin(8),liquiritigenin(9),5-dehydroxykaempferol(10),liquiritin(11),isoliquiritin(12),isoliquiritigenin(13),7,3'-dihydroxy-5'-methoxyisoflavone(14),and odoriflavene(15).Conclusion All fifteen compounds are isolated from Baoyuan Decoction for the first time.
