ABSTRACT

Objective:To observe the efficacy of benefiting Qi for warming Yang and activating blood circulation for inducing diu-resis method in the treatment of patients with chronic heart failure( CHF) and its effect on the expression of circulating miRNAs ( miR-423-5p, miR-210-3p). Methods:Totally 80 CHF patients were randomly divided into two groups, the control group with 40 patients was treated with the conventional anti-heart failure therapy, and the treatment group with 40 patients was treated with benefiting Qi for warming Yang and activating blood circulation for inducing diuresis decoction on the basis of the control group, and the treatment course was 2 weeks. The clinical efficacy and syndrome integral of traditional Chinese medicine ( TCM) were evaluated. Left ventricular ejec-tion fraction ( LVEF) and left ventricular end diastolic diameter( LVEDD) were measured by echocardiography;the plasma expression levels of circulating miRNAs(miR-423-5p, miR-210-3p) were measured by real-time PCR in the two groups before and after the treat-ment. The adverse drug reactions were observed as well. Results:The total clinical effective rate of the treatment group was significant-ly higher than that of the control group (87. 50% vs 65. 00%, P <0. 05); the syndrome integral of TCM, indices of LVEF and LVEDD and the expression of circulating miRNAs were improved in both groups(P<0. 05);the total clinical effective rate, syndrome integral of TCM, indices of LVEF and LVEDD and the expression of circulating miR-423-5p of the treatment group were better than those of the control group(P<0. 05); there was no significant difference in adverse drug reactions between the groups (P>0. 05). Conclusion:The benefiting Qi for warming Yang and activating blood circulation for inducing diuresis method can improve the clinical effects in the treatment of CHF, and improve the expression of circulating miRNAs.

ABSTRACT

Objective To identify the circulating miRNAs which can be used to evaluate the diagnosis and prognosis of sepsis by microarray and quantitative real-time PCR,and to predict target genes of miR-519c-5p by bioinformatics analysis. Methods Three sepsis patients,3 septic shock patients and 3 normal controls were enrolled in this study. Plasma RNA was extracted, and was used for hybridized by miRCURY LNATMmicroRNA Array. The signals were scanned and used to conduct differential expression profilings and cluster analysis.Further-more,we performed qRT-PCR to confirm the expression of miRNAs chosen from microarray screening. We used the miRanda and Targetscan databases to predict target genes of the concerned miRNAs and used KEGG database to analyze the related pathways. Results Fifty-seven and 11 miRNAs were observed significantly upregulated in sepsis and septic shock patients, respectively(fold change≥2.0;P<0.05).qRT-PCR results showed that miR-519c-5p was significantly upregulated in patients with sepsis or patients with septic shock compared with the healthy normal controls(P<0.05).Twenty-nine target genes of miR-519c-5p were predicted by the bioinformatics analysis, and 7 potential target genes participate in the sepsis-related pathways.MiR-519c-5p might be a potential positive regulator for the critical cell cycle control gene of MAP2K4,contributing to the vascular endothelial cells apoptosis via MAPK signaling pathway. Conclusions We demonstrated that the plasma level of miR-519c-5p can be used for the diagnosis of sepsis and miR-519c-5p may be a potential therapeutic target for sepsis.

ABSTRACT

Objective To study the expression profiles of circulating microRNAs(miRNAs)in newly-developed advanced schistosomiasis patients. Methods Ten healthy controls(Group 1),ten patients with schistosomiasis history(without advanced schistosomiasis,Group 2)and ten patients with newly-developed advanced schistosomiasis(Group 3)were randomly selected from Jiangsu Province. The expression profiles of 570 human related circulatingmiRNAs were measured and analyzed with the Agilent Human microRNAs microarray Rel 12.0. Results Compared to the healthy controls, four miRNAs were up-regulated, while 16 miRNAs were down-regulated in the patients with newly-developed advanced schistosomiasis. Moreover ,six miRNAs were up-regulated,while 21 miRNAs were down-regulated in the patients in Group 2. Among of them, the expression level of miR-383 was significantly decreased by 4.23 and 11.82 folds in Group 2 and Group 3,respectively. Conclusions There are significant differences among the healthy controls, patients with schistosomiasis history(without advanced schistosomiasis)and the patients with newly-developed advanced schistosomiasisin the expression profiles of circulatingmiRNAs. Moreover ,circulat-ing miR-383 might be involved in the development of newly-developed advanced schistosomiasis.

ABSTRACT

Non-small cell lung cancer is a major leading cause of cancer-related death and its late diagnosis/prognosis, accounting for the high rate of mortality. Presences of circulating microRNAs (miRNAs) in body fluids represent stable and reproducible biomarkers for several solid tumors, including non-small cell lung cancer. Micro RNAs have been categorized as oncogenic microRNAs and “tumor suppressor micro RNAs” and miRNAs activities may provide exciting opportunities for early cancer detection. There is an urgent need to find a less invasive and a more reliable biomarker which can increase the probability of early non-small cell lung cancer detection.
