ABSTRACT

Guided by TNF-α secretion inhibitory activity assay, four taraxastane-type triterpenoids, including two new ones, 22-oxo-20-taraxasten-3β, 30-diol (1) and 22α-hydroxy-20-taraxasten-30β, 30-triol (2), have been obtained from an active fraction of the petroleum ether-soluble extract of the the medicinal and edible plant Cirsium setosum. Their structures were elucidated by spectroscopic data and CD data analysis. In the TNF-α secretion inhibitory activity assay, compounds 1 and 2 were active with the IC of 2.6 and 3.8 μmol·L, respectively. In addition, compounds 1 and 2 showed moderately selective cytotoxicity against the human ovarian cancer (A2780) and colon cancer (HCT-8) cell lines.

ABSTRACT

Cirsium setosum is used as both food and medicine for thousands of years. Recently, it is reported that the chemical constituents of C. setosum mainly contain flavonoids, terpenoids, phenylpropanoids, phenylethylglycosides, alkaloids, and phytosterols, which have the pharmacological activities of hemostasis, clotting, cardiovascular, anti-oxidation, anti-bacterial, anti-inflammatory and so on. This paper reviews the researches on the chemical constituents and pharmacological activities of C. setosum in the past 30 years at home and abroad to provide reference for futher study on the development and utilization of C. setosum.

ABSTRACT

To investigate the chemical constituents of ethyl acetate from Cirsium setosum, fifteen flavonoids were obtained by column chromatography on silica gel, MCI, Sephadex LH-20, and preparative HPLC. Their structures were identified as 4',5,6-trihydroxy-7-methoxyflavone(1), 4',5-dihydroxy-7,8-dimethoxyflavone(2), sorbifolin-6-O-β-glucopyranoside(3), kaempferol-7-O-α-L-rhamnoside(4), kaempferol(5), quercetin-3-O-β-D-glucosyl-7-O-α-L-rhamnoside(6), myricetin(7), myricetin-3-O-β-D-glucoside(8), 5,7- dihydroxy -3',4'- dimethoxyflavone(9), 3',4',5- trihydroxy-3,7-dimethoxyflavone(10), 3',3,4',5-tetrahydroxy-7-methoxyflavone(11), 3'-hydroxy-4',5,7-trimethoxyflavone(12), 7-hydroxy-3',4',5-trimethoxyflavone(13), 4',5-dihydroxy-2',3',7,8-tetramethoxylflavone(14), and 5-hydroxy-2',3',7,8-tetramethoxylflavone(15) by spectroscopic data analysis. All compounds were isolated from this plant for the first time.Compounds(1-15) were evaluated for their hypoglycemic activities by PTP1B enzyme model. Among them, compounds 2, 12, and 14 showed significant PTP1B inhibitory activities with IC₅₀ values of 2.54, 1.85, 2.11 μmol•L⁻¹, respectively.

AIM@#To study the chemical constituents of the aerial parts of Cirsium setosum (Willd.) MB..@*METHODS@#The chemical constituents were isolated and purified by various chromatographic techniques. Their structures were determined on the basis of physical properties and spectroscopic data.@*RESULTS@#A new megastigmane glycoside and six known compounds were obtained and identified as (7E, 9R)-9-hydroxy-5, 7-megastigmadien-4-one-9-O-α-L-arabinopyranosyl-(1→6)-β-D-glucopyanoside (1), (6R, 7E, 9R)-9-hydroxy-4, 7-megastigmadien-3-one-9-O-α-L-arabinopyranosyl-(1→6)-β-D-glucopyranoside (2), urolignoside (3), 4, 9, 9'-trihydroxy-3, 3'-dimethoxy-8-O-4$\prime $-neolignan-7-O-β-D-glucopyranoside (4), citroside A (5), salidrosidin (6), and adenosine (7).@*CONCLUSION@#Compound 1 is a new megastigmane glycoside, named as Xiaojiglycoside A.

ABSTRACT

Objective HPLC was used to establish a method for determination of rutin in herba crisii. Methods Lichrospher C18 column(4.6 mm?200 mm,5?m)was used with methoanol-acetic acid(35∶65) as mobile phase,flow rate being 1.0 mL/min,column temperature at 30℃,detection wavelength at 254 nm.Results The content of rutin in herba crisii was 0.5808 mg/g.Conclusions The method is proved to be simple,rapid,and accurate.It can be applied to content determination of rutin in herba crisii.

ABSTRACT

Objective To search for the method used in enriching caffeates in Cirsium setosum by macroporous adsorption resin, 23 types of macroporous adsorption resin were optimized, which could provide the industrial production of caffeates with the theory basis. Methods The adsorption and elution ratios, product purity and yield, as total indices, were comprehensively evaluatued by 23 types of macroporous adsorption resin in enriching the extracts in C. setosum. Results The product purity and yield of caffeates by the HPD-100 type macroporous adsorption resin were the highest and up to 52.2% and 87.6%. Conclusion The HPD-100 type macroporous adsorption resin shows better comprehensive adsorption property. It is available for the enrichment of caffeates in C. setosum.
