ABSTRACT

Background: Skeletal muscles are susceptible for ischemic reperfusion injury especially in settings in order to achieve homeostasis in traumatic injury and vascular surgery. This study aimed at investigating the implication of induction of diabetes on generation of ischemic reperfusion injury in rat gastrocnemius muscle. In addition, the possible beneficial effect of metformin and co-enzyme q10 was investigated.Methods: About 80 male adult Sprague Dawley rats divided into 10 groups. Metformin was administrated as continuous oral dose for 28 days. Coenzyme q10 was administrated parenterally 2 and 24 hours before induction of ischemia in diabetic and non-diabetic animals.Results: In diabetic ischemic groups, tested drugs either singly or in combination significantly reduce HB1cA and plasma levels of muscle specific enzyme CPK and muscle myokin IL6, raised natural antioxidant GSH and reduced oxidative stress parameters (SOD and MDA), apoptotic (caspase-3) and inflammatory parameters TNF-? and TGF? were reduced. Continuous oral metformin for 28 days was more powerful than parenteral short-term coenzyme q10 as regards all tested parameters except for GSH and caspase-3 both drugs were equi-effective. Combined drugs have more powerful ameliorating effect than either drug singly except for HB1cA which was equi-effective with that of metformin. Regarding non diabetic ischemic groups, metformin was more powerful in reduction of caspase-3, IL6 and TNF- ? while coenzyme q10 was more powerful in elevating GSH.Conclusions: Co-enzyme q10 can be used as add on therapy with metformin in order to decrease the deleterious effects resulted from hind limb ischemia reperfusion in normal and diabetic rats.

ABSTRACT

Objective To explore the effect of ginseng co?enzyme Q10 suncream on the skin damage caused by ul?traviolet ( UV) radiation in mice. Methods 36 mice were randomly assigned to four groups. The mice were shaved on the back and the left untreated side was taken as control group, or was treated with UV as model group. Before treated with UV, the mice were painted with suncream containing ginseng co?enzyme Q10 , or octyl methoxycinnamate as positive con?trols. The mice were treated for 8 weeks. At the end of the experiment, blood samples of all mice were collected from the eyes, then subjected to cell counting or biochemical measurements, and skin samples were cut for pathological examina?tion. Results Compared with the control group, there was a significant increase in white blood cell counts ( P<0?05 ) and MDA content ( P<0?05 ) , and declined serum levels of SOD ( P <0?05 ) and GSH?Px ( P <0?05 ) in the model group, and the skin was rough and wrinkled with stratum corneum exfoliation. Compared with the model group, the mice of ginseng co?enzyme Q10 suncream group had significantly lower white blood cell count ( P<0?05 ) and MDA content ( P<0?05), and increased serum levels of T?SOD(P<0?05) and red blood cell counts (P<0?05). The skin had no rough? ness and wrinkles and without stratum corneum exfoliation. Compared with the model group, the positive control group showed significantly decreased white blood cell count (P<0?05) and MDA content (P<0?05), and increased serum lev?els of GSH?Px(P<0?05). The skin had no roughness and wrinkles and no stratum corneum exfoliation. However, there was no significant difference between the ginseng co?enzyme Q10 suncream group and positive control group. Conclusions Ginseng co?enzyme Q10 suncream shows satisfactory preventive effects on the UV radiation?induced skin damage in mice, similar to the preventive effects of the octyl methoxycinnamate?containing sunsream.

ABSTRACT

The study was undertaken to evaluate the efficacy of multivitamin and micronutrient supplementation in azoospermic patients with maturation arrest. A total of 35 azoospermic patients showing maturation arrest on testicular biopsy were recruited in this study. The patients were divided into two groups. Untreated group (n=11) without any treatment and treated group (n=24) who received multivitamins, micronutrients and co-enzyme Q10. The sperm concentration, motility and morphology were evaluated at monthly interval. The results showed reduction in liquefaction time and relative viscosity of the semen in the treated group. Further, in treated group there was appearance of spermatozoa (4.0 million/ml) exhibiting progressive motility (7%) and normal morphology (6%), even in the first follow up visit. The sperm count, motility and normal morphology increased significantly on subsequent visits. Within 3 months (3 visits) 2 pregnancies were reported. These observations indicate that multivitamin and micronutrient supplementation improve the qualitative and quantitative parameters of seminogram in patients with azoospermia of maturation arrest.
