ABSTRACT

Climate change has caused major changes in abiotic factors, with water stress as the greatest threat to agricultural production. The measures aimed at alleviating the problems caused by this limiting production factor have occurred through the adoption of sustainable strategies, especially microbial biotechnology, which uses the interactions between the microorganism and the plant, ensuring productive quality and inducing plant resistance to stresses biotic and abiotic. The objective of the present work was to evaluate the biological nitrogen fixation and the development of bean seedlings, with co-inoculation of two types of inoculants, which were subjected to water stress by different pot capacities. The experiment was conducted in a greenhouse, at Universidade Paranaense - UNIPAR, from April to June 2019. The experimental design was completely randomized (DIC), with 5 replications, 16 treatments and 80 experimental units. The cultivar used was SCS Riqueza. The parameters evaluated were pot capacity (25%, 50%, 75% and 90%); small, large and total nodules, shoot and root length, dry and fresh weight, total carbon and nitrogen. The evaluation of the morphological parameters of the bean seedlings indicated that the co- inoculation technique promoted beneficial effects for the dry mass parameters of shoot, nodule and root. The analysis of the percentage of carbon and nitrogen in the tissues of the seedlings provided an increase in the concentration of these elements in treatments that involved co-inoculation (Azospirillum brasilensis and Rhizobium tropici) with pot capacities of 25 and 75% (CV), demonstrating that the association of microorganisms is beneficial in the limiting water situation.(AU)


A mudança climática tem causado grandes mudanças nos fatores abióticos, sendo o estresse hídrico a maior ameaça à produção agrícola. As medidas destinadas a aliviar os problemas causados por este fator limitante de produção ocorreram através da adoção de estratégias sustentáveis, especialmente a biotecnologia microbiana, que utiliza as interações entre o microorganismo e a planta, garantindo a qualidade produtiva e induzindo a resistência da planta ao estresse biótico e abiótico. O objetivo do presente trabalho foi avaliar a fixação biológica de nitrogênio e o desenvolvimento de mudas de feijão, com co-inoculação de dois tipos de inoculantes, que foram submetidos ao estresse hídrico por diferentes capacidades de vaso. A experiência foi realizada em uma estufa, na Universidade Paranaense - UNIPAR, de abril a junho de 2019. O projeto experimental foi completamente randomizado (DIC), com 5 réplicas, 16 tratamentos e 80 unidades experimentais. A cultivar utilizada foi a SCS Riqueza. Os parâmetros avaliados foram a capacidade do vaso (25%, 50%, 75% e 90%); nódulos pequenos, grandes e totais, comprimento do rebento e da raiz, peso seco e fresco, carbono total e nitrogênio. A avaliação dos parâmetros morfológicos das mudas de feijão indicou que a técnica de co-inoculação promoveu efeitos benéficos para os parâmetros de massa seca do turião, nódulo e raiz. A análise da porcentagem de carbono e nitrogênio nos tecidos das mudas proporcionou um aumento na concentração destes elementos nos tratamentos que envolveram a co-inoculação (Azospirillum brasilensis e Rhizobium tropici) com capacidades de vaso de 25 e 75% (CV), demonstrando que a associação de microorganismos é benéfica na situação limite da água.(AU)


El cambio climático ha provocado importantes cambios en los factores abióticos, siendo el estrés hídrico la mayor amenaza para la producción agrícola. Las medidas encaminadas a paliar los problemas causados por este factor limitante de la producción se han producido mediante la adopción de estrategias sostenibles, especialmente la biotecnología microbiana, que utiliza las interacciones entre el microorganismo y la planta, asegurando la calidad productiva e induciendo la resistencia de la planta a los estreses bióticos y abióticos. El objetivo del presente trabajo fue evaluar la fijación biológica de nitrógeno y el desarrollo de plántulas de frijol, con la co-inoculación de dos tipos de inoculantes, que fueron sometidos a estrés hídrico por diferentes capacidades de maceta. El experimento se realizó en un invernadero, en la Universidade Paranaense - UNIPAR, de abril a junio de 2019. El diseño experimental fue completamente al azar (DIC), con 5 repeticiones, 16 tratamientos y 80 unidades experimentales. El cultivar utilizado fue SCS Riqueza. Los parámetros evaluados fueron capacidad de maceta (25%, 50%, 75% y 90%); nódulos pequeños, grandes y totales, longitud de brotes y raíces, peso seco y fresco, carbono y nitrógeno total. La evaluación de los parámetros morfológicos de las plántulas de frijol indicó que la técnica de coinoculación promovió efectos beneficiosos para los parámetros de masa seca de brotes, nódulos y raíces. El análisis del porcentaje de carbono y nitrógeno en los tejidos de las plántulas proporcionó un aumento en la concentración de estos elementos en los tratamientos que involucraron la coinoculación (Azospirillum brasilensis y Rhizobium tropici) con capacidades de maceta de 25 y 75% (CV), demostrando que la asociación de microorganismos es beneficiosa en la situación de agua limitante.(AU)

ABSTRACT

Plant-growth-promoting bacteria are often used to enhance crop yield and for biological control of phytopathogens. Bacillus sp. CHEP5 is a biocontrol agent that induces systemic resistance (ISR) in Arachis hypogaea L. (peanut) against Sclerotium rolfsii, the causal agent of root and stem wilt. In this work, the effect of the co-inoculation of Bacillus sp. CHEP5 and the peanut nodulating strain Bradyrhizobium sp. SEMIA 6144 was studied on induction of both systemic resistance and nodulation processes. Bradyrhizobium sp. SEMIA 6144 did not affect the ability of Bacillus sp. CHEP5 to protect peanut plants from S. rolfsii by ISR and the priming in challenged-plants, as evidenced by an increment in phenylalanine ammonia-lyase enzyme activity. Additionally, the capacity of Bradyrhizobium sp. SEMIA 6144 to induce nodule formation in pathogen-challenged plants was improved by the presence of Bacillus sp. CHEP5.

ABSTRACT

Interactions between yeasts and lactic acid bacteria are strain specific, and their outcome is expected to change in simultaneous alcoholic -malolactic fermentations from the pattern observed in successive fermentations. One Oenococcus oeni strain Lalvin VP41TM was inoculated with two Saccharomyces cerevisiae strains either simultaneously, three days after the yeast inoculation, or when alcoholic fermentation was close to finish. Early bacterial inoculations with each yeast strain allowed for the growth of the bacterial populations, and the length of malolactic fermentation was reduced to six days. Alcoholic fermentation by Lalvin ICV D80® yeast strain left the highest residual sugar, suggesting a negative effect of the bacterial growth and malolactic activity on its performance. In sequential inoculations the bacterial populations did not show actual growth with either yeast strain. In this strategy, both yeast strains finished the alcoholic fermentations, and malolactic fermentations took longer to finish. Lalvin ICV D80® allowed for higher viability and activity of the bacterial strain than Fermicru UY4® under the three inoculation strategies. This was beneficial for the sequential completion of both fermentations, but negatively affected the completion of alcoholic fermentation by Lalvin ICV D80® in the early bacteria additions. Conversely, Fermicru UY4®, which was rather inhibitory towards the bacteria, favored the timely completion of both fermentations simultaneously. As bacteria in early inoculations with low or no SO2 addition can be expected to multiply and interact with fermenting yeasts, not only are the yeast-bacterium strains combination and time point of the inoculation to be considered, but also the amount of bacteria inoculated.

ABSTRACT

The endophytic strain Zong1 isolated from root nodules of the legume Sophora alopecuroides was characterized by conducting physiological and biochemical tests employing gfp-marking, observing their plant growth promoting characteristics (PGPC) and detecting plant growth parameters of inoculation assays under greenhouse conditions. Results showed that strain Zong1 had an effective growth at 28 ºC after placed at 4-60 ºC for 15 min, had a wide range pH tolerance of 6.0-11.0 and salt tolerance up to 5% of NaCl. Zong1 was resistant to the following antibiotics (µg/mL): Phosphonomycin (100), Penicillin (100) and Ampicillin (100). It could grow in the medium supplemented with 1.2 mmol/L Cu, 0.1% (w/v) methylene blue and 0.1-0.2% (w/v) methyl red, respectively. Zong1 is closely related to Pseudomonas chlororaphis based on analysis the sequence of 16S rRNA gene. Its expression of the gfp gene indicated that strain Zong1 may colonize in root or root nodules and verified by microscopic observation. Furthermore, co-inoculation with Zong1 and SQ1 (Mesorhizobium sp.) showed significant effects compared to single inoculation for the following PGPC parameters: siderophore production, phosphate solubilization, organic acid production, IAA production and antifungal activity in vitro. These results suggest strains P. chlororaphi Zong1 and Mesorhizobium sp. SQ1 have better synergistic or addictive effect. It was noteworthy that each growth index of co-inoculated Zong1+SQ1 in growth assays under greenhouse conditions is higher than those of single inoculation, and showed a significant difference (p < 0.05) when compared to a negative control. Therefore, as an endophyte P. chlororaphis Zong1 may play important roles as a potential plantgrowth promoting agent.

ABSTRACT

This study evaluated the synergism between several P-solubilizing fungi isolates and arbuscular mycorrhizal fungi to improve clover ( Trifolium pratense) growth in the presence of Araxá apatite. Clover was sown directly in plastic pots with 300g of sterilized washed sand, vermiculite and sepiolite 1:1:1 (v:v:v) as substrate, and grown in a controlled environment chamber. The substrate was fertilized with 3 g L-1 of Araxá apatite. A completely randomized design, in 8×2 factorial scheme (eight P-solubilizing fungi treatments with or without arbuscular mycorrhizal fungi)and four replicates were used. The P-solubilizing fungi treatments consisted of five Brazilian P-solubilizing fungi isolates (PSF 7, 9, 20, 21 and 22), two Spanish isolates ( Aspergillus niger and the yeast Yarowia lipolytica) and control (non-inoculated treatment). The greatest clover growth rate was recorded when Aspergillus niger and PSF 21 were co-inoculated with arbuscular mycorrhizal fungi. Aspergillus niger, PSF 7 and PSF 21 were the most effective isolates on increasing clover growth in the presence of arbuscular mycorrhizal fungi. Greater mycorrhizal colonization resulted in greater clover growth rate in most PSF treatments. PSF 7 was the best isolate to improve the establishment of mycorrhizal and rhizobia symbiosis.


Este estudo avaliou o sinergismo entre diversos isolados defungos solubilizadores de fosfato e micorrízicos arbusculares para beneficiar o crescimento de trevo ( Trifolium pratense) na presença de apatita de Araxá. A cultura foi semeada diretamente em potes plásticos com 300 g de substrato esterilizado formado por areia lavada, vermiculita e sepiolita 1:1:1 (v:v:v) e cultivada em câmara climática. O substrato foi fertilizado com 3 g L-1 de apatita de Araxá. O experimento foi instalado em delineamento completamente casualizado, esquema fatorial 8×2 (oito tratamentos de inoculação de fungos solubilizadores de fosfato com ou sem fungos micorrízicos arbusculares) e quatro repetições. Os tratamentos de fungos solubilizadores de fosfato consistiram em cinco isolados brasileiros de fungos solubilizadores de fosfato (FSF 7, 9, 20, 21 e 22), dois isolados procedentes da Espanha ( Aspergillus niger e a levedura Yarowia lipolytica) e o controle (tratamento não inoculado). A maior taxa de crescimento da cultura foi obtida quando Aspergillus niger e FSF 21 foram co-inoculados com fungos micorrízicos arbusculares. Aspergillus niger, FSF 7 e o FSF 21 foram os isolados mais efetivos para incrementar o crescimento de trevo na presença de fungos micorrízicosarbusculares. A maior taxa de colonização micorrízica resultou em alta taxa de crescimento de trevo na maioria dos tratamentos com fungos solubilizadores de fosfato. O isolado FSF 7 foi o melhor para favorecer o estabelecimento das simbioses com fungos micorrízicos e com rizóbio.
