ABSTRACT

Colla Corii Asini(Ejiao)is an important Chinese medicine used in China for thousands of years, and is well known for its famous tonic properties. The herbalogical study was detailed carried out based on the naming, habitat, harvesting, processing, medicinal properties and clinical efficacy. The results showed that the name of Ejiao could be traced back to Shennong's Materia Medica, and various names of Lvpi Jiao, Penfu Jiao and Fuzhi Jiao were recorded in other ancient books. In the many intervening centuries, the main materials of Ejiao had been replaced from cow leather before Tang Dynasty to donkey skin in the middle to late Tang Dynasty. This phenomenon could be probably caused by complicated social factors of various periods and different efficacy of Ejiao made by all kinds of raw materials. Ejiao was merely processed with the simple methods before Tang Dynasty, which subsequently improved avariety of methods to enhance the supplementation action. Most importantly, Ejiao has a wide clinic application along with the development of processing theories and methods, which can be found in various Classics, especially in imperial medical case record in Qing Dynasty.

ABSTRACT

OBJECTIVE: To extract microamounts of animal-derived DNA from the products of Colla Corii Asini boiled at high temperature, establish and optimize a rapid identification method of donkey-derived components in Colla Corii Asini by polymerase chain reaction(PCR), and establish a new molecular biological method for assessing the quality of Colla Corii Asini. METHODS: The donkey-derived genomic DNA was extracted by DNA purification column instead of phenol, chloroform and other toxic organic solvents in SDS-PK method, and the SDS-PK method was optimized with the donkey-derived genomic DNA. RESULTS: The optimum sample size of Colla Corii Asini was 0.20 g. High quality genomic DNA of Colla Corii Asini could be obtained quickly after digestion in water bath for 1 h and then purified by DNA purification column. The purity ranged from 1.70 to 1.80, and the concentration of Colla Corii Asini could reach (187.8±0.56)ng•μL-1. PCR amplification, cloning, and sequencing were performed using specific primers, and the similarity to GenBank's registered Donkey species (MG931481.1) was 100%. CONCLUSION: This study provides animal-derived genomic DNA fragments from deep-processed Colla Corii Asini and Colla Corii Asini products within 90 min. The purity and concentration of extracted DNA can meet the requirements of molecular biological identification of Colla Corii Asini. The established PCR method can quickly identify the scorpion-derived components in Colla Corii Asini. The cloned donkey specific gene fragment can be used as a standard positive control to identify the authenticity of Colla Corii Asini. It is expected that it will be widely used in the quality supervision of Colla Corii Asini and related products.

ABSTRACT

Objective: The aqueous extracts of donkey-hide and its counterfeits (horse-hide and mule-hide) were identified using 1H-NMR metabolomics to provide the basis for the identification and quality control of donkey-hide gelatin. Methods: The water-soluble components of the hairless donkey-hide and its counterfeits were extracted and the 1H-NMR technique was applied to analyzing the components. Moreover, the NMR data were analyzed and the component belongings were recognized using relevant software for the multivariate statistical analysis. Results: Forty-two water-soluble components were identified in donkey-hide by 1H-NMR. There were more acetate, leucine, valine, isoleucine and less lactate, serine, pyroglutamate, and creatine in the horse-hide than in the donkey-hide; While there were more acetate, creatine, alanine, choline and less glycerol and acetate in mule-hide than in donkey-hide. And they were all in a significant difference (P 1H-NMR metabolomics was established for the first time, which provides the experimental references for the donkey-hide identification and quality control study.

ABSTRACT

Objective Colla Corii Asini,Colla Carapacis et Plastri Testudinis,Colla Cornus Cervi,three types of plastic bead microwave process parameters were optimized in order to standardize the production of the types of plastic beads and to enhance the stability of the quality of plastic beads.Methods Using a number of quality indicators to evaluate the quality of plastic beads and orthogonal test to optimize the plastic bead process parameters.Results The microwave process parameters of Colla Corii Asini,Colla Carapacis et Plastri Testudinis,Colla Cornus Cervi,three types of plastic beads were obtained through orthogonal optimization.Conclusion Different types of plastic type drugs,different production lot, and different manufacturers of similar plastic herbs have their own optimum process parameters.L4(23) orthogonal optimization can be used to screen out the plastic type drugs rapidly.
