ABSTRACT

Objective By analysing the clinical features of Indigo Naturalis-associated ischemic lesion of colon mucosa to improve the precautionary and therapeutic level of the disease.Methods Thirteen patients diagnosed as Indigo Naturalis-associated ischemic lesion of colon mucosa in Peking University Third Hospital from 2005 to 2010 were reviewed.The endoscopic and clinical features were analysed.Results The 13 patients with an average age of(60.6±14.1)years old were prescribed Chinese traditional medicine containing Indigo Naturalis for psoriasis or idiopathic thrombocytopenic purpura(ITP).The ratio of males to females was 1:1.6.The typical manifestations were abdominal pain and bloody stool with watering diarrhea before bloody stool in 61.5%patients.Endoscopic and pathological characteristics were coincident with ischemic lesion and more like a chronic index.Vasodilatic medicine was effective and the average hemostatic time was(1.7±0.8)days.The prognosis was well and no recurrence was found during 3 months follow-up.Conclusions Patients having psoriasis or ITP treated with Chinese traditional medicine containing Indigo Naturalis have an inclination to colon mucosa lesions, even ischemic lesion. Careful assessment and observation before prescribing arc necessary in these patients.

ABSTRACT

Objective To investigate the effect of chronic stress on the mucin secretion and muc2 protein content in goblet cells of rat colonic mucosa. Methods 24 healthy Wistar rats were randomly divided into 2 groups. Control group moved freely, experimental group was forced to undergo 2-hour-long immobilization stress per day for 14 days. The number of goblet cells containing mucus was determined by AB/PAS histochemical staining. The expression of muc2 protein was detected by immuohistochemical staining. Results Chronic stress decreased the number of goblet cells containing mucus, and down-regulated the expression of muc2 protein in goblet cells in rat colonic mucosa. Conclusion Chronic stress damaged the colonic epithelial barrier and functions of colonic goblet cells.

ABSTRACT

An important property of the intestine is the ability to secrete fluid. The intestinal secretion is regulated by a number of substances including vasoactive intestinal peptide (VIP), ATP and different inflammatory mediators. One of the most important secretagogues is adenosine during inflammation. However, the controversy concerning the underlying mechanism of adenosine-stimulated Cl- secretion in intestinal epithelial cells still continues. To investigate the effect of adenosine on Cl- secretion and its underlying mechanism in the rabbit colon mucosa, we measured short circuit current (ISC) under automatic voltage clamp with DVC-1000 in a modified Ussing chamber. Adenosine, when added to the basolateral side of the mucosa, increased ISC in a dose-dependent manner. The adenosine-stimulated ISC response was abolished when Cl- in the bath solution was replaced completely with gluconate. In addition, the ISC response was inhibited by a basolateral Na-K-Cl cotransporter blocker, bumetanide, and by apical Clchannel blockers, dephenylamine-2-carboxylate (DPC), 5-nitro-2-(3-phenyl-propylamino)-benzoate (NPPB), glibenclamide. Amiloride, an epithelial Na+ channel blocker, and 4,4-diisothiocyanato-stilbene-2,2-disulphonate (DIDS), a Ca2+-activated Cl- channel blocker, had no effect. In the mucosa pre-stimulated with forskolin, adenosine did not show any additive effect, whereas carbachol resulted in a synergistic potentiation of the ISC response. The adenosine response was inhibited by 10 micrometer H-89, an inhibitor of protein kinase A. These results suggest that the adenosine-stimulated ISC response is mediated by basolateral to apical Cl- secretion through a cAMP-dependent Cl- channel. The rank order of potencies of adenosine receptor agonists was 5'-(N-ethylcarboxamino)adenosine(NECA) > N6-(R-phenylisopropyl)adenosine(R-PIA)>2-(p-(2-carbonylethyl)-phenyl-et hylamino)-5'-N-ethylcarboxaminoadenosine(CGS21680). From the above results, it can be concluded that adenosine interacts with the A2b adenosine receptor in the rabbit colon mucosa and a cAMP-dependent signalling mechanism underlies the stimulation of Cl- secretion.

ABSTRACT

Objective To observe the effect of selenium on the ACTH positive cells in the pars distalis of pituitary of rat with colon mucosa cancer induced by azoxymethane(AOM).Methods Twenty 3 weeks old Sprague Dawley male rats were randomly divided into 4 groups: the normal control group;the experimental control group;the selenium before AOM induced colon mucosa cancer group;the selenium after AOM induced colon mucosa cancer group.To induce rat colon mucosa cancer,3 weeks old SD male rats received(i.p.) injections of AOM at a dose of 15mg/kg once a week for two weeks.Selenium(Na_2SeO_3,4mg/L) began to be provided in the drinking water before and after AOM administration,and lasted for the whole experiment period.The rat pituitary tissues were excised from the 34 weeks old rats.The morphological changes of the ACTH positive cells in the pars distalis of pituitary were observed using immunohistochemical methods,and then the results were analyzed by image analysis system.Results Aberrant crypt foci(ACF) and aberrant crypt(AC) were showed in the colon mucosa of rats that received two weekly i.p.injections of AOM by light microscopy with methylene blue staining.The ACTH positive cells in the experimental group rat displayed a significant increase compared with those in the normal control group rat(P
