ABSTRACT

Self-expanding metal stents (SEMSs) has been increasingly used for the palliation of metastatic colorectal cancer and as a bridge to surgery for obstructing neoplasms. Although there remain a few concerns regarding the use of stents as a bridge to surgical resection, use of SEMSs for palliation in patients with unresectable disease has been widely accepted. Advantages of colorectal stents include prompt restoration of luminal patency, avoidance of stoma creation, lower morbidity and shorter hospital stay. This review will discuss the technical aspects, general indications, clinical outcomes and complications of colorectal stents.

ABSTRACT

Objective To explore the correlation between serum cytokines and the depression degree in the patients with colorec-tal cancer. Methods 9 6 cases of colorectal cancer were selected and divided into 2 groups according to the patients health question-naire(PHQ-9):the depression group (52 cases)and the non-depression group (44 cases),and 40 healthy cases were recruited as the control.Serum interleukin-2 (IL-2),soluble interleukin-2 receptor (SIL-2R)and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay(ELSIA).The detection results were compared among various groups. And the correlation be-tween the expression of various cytokines and the depression degree was analyzed by the Pearson′s correlation coefficient. Results The levels of IL-2,SIL-2R and TNF-αhad statistical differences among 3 groups (P0.05),but which in these two groups was higher than that in healthy group (P<0.05);the IL-2 level in the depression group was significantly lower than that in the non-depression group(P<0.05);the SIL-2 R level in the depression group was obviously higher than that in the non-depression group (P<0.05), moreover, which in these two groups was higher than that in the healthy group (P<0.05).By correlation analysis, the depression degree in the patients with colorectal cancer was negatively correlated with the IL-2 level (r=-0.259,P<0.05)and positively correlated with the SIL-2R level(r=0.412,P<0.05).Conclusion The levels of IL-2 and SIL-2R have certain correlation with the depression degree in the patients with colorectal cancer,it is speculated that the above cytokines may play a mediating role in the pathogenesis process of depression in the colorectal cancer patients.

ABSTRACT

The incidence of colorectal cancer( CRC)increases rapidly in recent years. The role of dietary structure in CRC pathogenesis has caught much attention. This review summarizes the relationship between dietary fiber intake and risk of CRC. Accumulating evidence reveals that up to 90% CRC can be prevented by improvement of dietary structure, and high fiber intake(10-35 g/d)decreases 10%-17% of risk of CRC. All kinds of dietary fiber have preventive effect. So far, dietary fiber intake in Chinese people does not reach the recommended level. Optimization of dietary structure, increase intake of whole grains, vegetable and fruit, addition of fiber to daily food are effective approaches for prevention of CRC.

ABSTRACT

Background:Suppression of tumor suppressor genes plays a key role in the pathogenesis and progress of tumors. Some microRNAs may contribute to tumorigenesis by regulating tumor suppressor genes. Aims:To investigate the targeted regulatory effect of miR-483-3p on deleted in liver cancer 1(DLC1)gene in colorectal cancer. Methods:Sixteen patients with colorectal cancer admitted from October 2012 to April 2013 at Nanjing Drum Tower Hospital were enrolled. Expression of DLC1 in cancerous and adjacent noncancerous tissues was determined by Western blotting, and expression of miR-483-3p was determined by qRT-PCR. Dual luciferase reporter gene plasmid containing the 3’untranslated region(3’UTR)of DLC1 was constructed to validate the regulation of DLC1 by miR-483-3p in human colon cancer cell line HCT116. MiR-483-3p mimic was transfected into HEK293T cells and expression of DLC1 was determined by Western blotting;MiR-483-3p mimic was transfected into HCT116 cells and cell proliferation was measured by CCK-8 assay. Results:Expression of DLC1 was significantly lower in cancerous tissue than in noncancerous tissue, while expression of miR-483-3p was significantly higher in cancerous tissue than in noncancerous tissue(P<0. 05). MiR-483-3p mimic reduced the expression of DLC1 through directly binding to the 3’UTR of DLC1. Transfection of miR-483-3p mimic enhanced the proliferation of HCT116 cells significantly(P<0. 05). Conclusions:DLC1 is a target gene of miR-483-3p. MiR-483-3p might promote the development of colorectal cancer by down-regulating DLC1 expression at post-transcriptional level.

ABSTRACT

Background:Fanconi anemia( FA ),an autosomal or x-linked recessive inherited disease,is caused by gene mutation related to FA pathway of DNA damage and with the clinical features of congenital malformation, bone marrow failure and susceptibility to cancer. Aims:To investigate the expression of Fanconi anemia complementation group D2 (FANCD2)in colorectal cancer and its correlation with prognosis. Methods:Fifty-six surgical resected specimens of colorectal cancer tissue and para-cancer noncancerous tissue from May 2012 to September 2013 at Guangzhou First People’s Hospital were obtained. Ninety-three patients with colorectal cancer from January 2008 to April 2009 at Guangzhou First People ’s Hospital were enrolled and the specimens of colorectal cancer tissue were obtained. The expression of FANCD2 mRNA in 56 specimens of colorectal cancer tissue and para-cancer noncancerous tissue was determined by qPCR. The expression of FANCD2 protein in 49 of 56 specimens of colorectal cancer tissue and para-cancer noncancerous tissue and 93 specimens of colorectal cancer tissue was determined by immunohistochemistry. Follow up was conducted in 93 patients and the correlation between the expression of FANCD2 and prognosis was analyzed. Results:Expression of FANCD2 mRNA was significantly higher in colorectal cancer tissue than that in para-cancer noncancerous tissue [0.102(0.047,0. 163)vs. 0. 051(0. 025,0. 095)](P =0. 007). Expression of FANCD2 mRNA was significantly correlated with lymph node metastasis and Dukes stage( P <0. 05 ). Positivity rates of FANCD2 protein expression in colorectal cancer tissue and para-cancer noncancerous tissue were 77. 6%(38/49)and 22. 4%(11/49),respectively. Expression of FANCD2 protein was significantly correlated with lymph node metastasis and Dukes stage(P<0. 05). The overall 5-year survival rate was significantly lower in patients with positive expression of FANCD2 protein than that in patients with negative expression of FANCD2 protein(35. 5% vs. 71. 0%)(P<0. 01). Conclusions:FANCD2 is associated with malignant potential of colorectal cancer and could be a potential marker for prognosis.
