ABSTRACT

The eukaryotic genome is folded into higher-order conformation accompanied with constrained dynamics for coordinated genome functions. However, the molecular machinery underlying these hierarchically organized three-dimensional (3D) chromatin architecture and dynamics remains poorly understood. Here by combining imaging and sequencing, we studied the role of lamin B1 in chromatin architecture and dynamics. We found that lamin B1 depletion leads to detachment of lamina-associated domains (LADs) from the nuclear periphery accompanied with global chromatin redistribution and decompaction. Consequently, the inter-chromosomal as well as inter-compartment interactions are increased, but the structure of topologically associating domains (TADs) is not affected. Using live-cell genomic loci tracking, we further proved that depletion of lamin B1 leads to increased chromatin dynamics, owing to chromatin decompaction and redistribution toward nucleoplasm. Taken together, our data suggest that lamin B1 and chromatin interactions at the nuclear periphery promote LAD maintenance, chromatin compaction, genomic compartmentalization into chromosome territories and A/B compartments and confine chromatin dynamics, supporting their crucial roles in chromatin higher-order structure and chromatin dynamics.

ABSTRACT

SUMMARY: The objective of this study was to determine the occurrence of anatomical variants in the exocranial surface of the jugular foramen, specifically, the presence of single or double and complete or incomplete septation. A cross-sectional anatomical study was performed using 96 Brazilian dry human skulls (53 male and 43 female). One examiner determined the number (single or double) and type (i.e. complete or incomplete) of osseous septation at the outer surface of jugular foramens. Data went through statistical analysis on GraphPad Prism 6.01. Our results shown that Male individuals where more likely to present normal jugular foramens (male = 71.69%, female = 34.88%; p = 0.003). However, one incomplete septation occurred more often on the right side of female individuals (1 incomplete septation, male = 16.98%; 1 incomplete septation, female = 34.88%; p = 0.044). Similarly, one complete septation (i.e. the presence of two fully divided jugular compartments) also occurred more often on the right side of female individuals (1 complete septation, male = 9.43%; 1 complete septation, female = 25.58%; p = 0.038). Anatomical variants of the jugular foramen regarding single or double complete or incomplete septations were more likely to be found on the right side of female individuals, whose also presented a higher rate of jugular foramens with any type of septation than regular non-altered jugular foramens.

ABSTRACT

SUMMARY Body composition assessment at the molecular level is relevant for the athletic population and its association with high performance is well recognized. The four-compartment molecular model (4C) is the reference method for fat mass (FM) and fat-free mass (FFM) estimation. However, its implementation in a real context is not feasible. Coaches and athletes need practical body composition methods for body composition assessment, and the bioelectrical impedance analysis method (BIA) is usually seen as a useful alternative. The aim of this study was to test the validity of BIA (Tanita, TBF-310) to determine the FM and FFM of elite judo athletes. A total of 29 males were evaluated in a period of weight stability using the reference method (4C) and the alternative method (Tanita, TBF-310). Regarding the 4C method, total-body water was assessed by deuterium dilution, bone mineral by DXA, and body volume by air displacement plethysmography. The slops and intercepts differed from 1 (0.39 and 1.11) and 0 (4.24 and -6.41) for FM and FFM, respectively. FM from Tanita TBF-310 overestimated the 4C method by 0.2 kg although no differences were found for FFM. Tanita TBF-310 explained 21% and 72% respectively in the estimation of absolute values of FM and FFM from the 4C method. Limits of agreement were significant, varying from -6.7 kg to 7.0 kg for FM and from -8.9 kg to 7.5 kg for FFM. In conclusion, TBF-310 Tanita is not a valid alternative method for estimating body composition in highly trained judo athletes.

ABSTRACT

BACKGROUND: Accurate volume measurement is important in the management of patients with congestive heart failure or renal insufficiency. A bioimpedance analyser can estimate total body water in litres and has been widely used in clinical practice due to its non-invasiveness and ease of results interpretation. To change impedance data to volumetric data, bioimpedance analysers use equations derived from data from healthy subjects, which may not apply to patients with other conditions. Bioelectrical impedance vector analysis (BIVA) was developed to overcome the dependence on those equations by constructing vector plots using raw impedance data. BIVA requires normal reference plots for the proper interpretation of individual vectors. The aim of this study was to construct normal reference vector plots of bioelectrical impedance for Koreans. METHODS: Bioelectrical impedance measurements were collected from apparently healthy subjects screened according to a comprehensive physical examination and medical history performed by trained physicians. Reference vector contours were plotted on the RXc graph using the probability density function of the bivariate normal distribution. We further compared them with those of other ethnic groups. RESULTS: A total of 242 healthy subjects aged 22 to 83 were recruited (137 men and 105 women) between December 2015 and November 2016. The centers of the tolerance ellipses were 306.3 Ω/m and 34.9 Ω/m for men and 425.6 Ω/m and 39.7 Ω/m for women. The ellipses were wider for women than for men. The confidence ellipses for Koreans were located between those for Americans and Spaniards without overlap for both genders. CONCLUSION: This study presented gender-specific normal reference BIVA plots and corresponding tolerance and confidence ellipses on the RXc graph, which is important for the interpretation of BIA-reported volume status in patients with congestive heart failure or renal insufficiency. There were noticeable differences in reference ellipses with regard to gender and ethnic groups.

ABSTRACT

Objective Through the establishment of adjacent cabin blast injury model of Beagle dog,to investigate the pathophysiological changes in the experimental animals in this scenario, then speculate on the mechanisms of injury. Methods Several adjacent cabins were built in the same size with the real warship.Seven Beagle dogs were subjected to injuries from the explosion, from whom one was selected randomly to implant intracranial pressure transducers before blast, the others were tested on the pathophysiological changes after blast. The dogs were mounted on the platform of a cabinet in the adjacent cabin, subjected to injury from 650g bare TNT explosive blast. The transducers recorded the value of space and intracranial shock wave pressure.Following blast treatment, the serum levels of IL-6,IL-8,neuron specific enolase (NSE),brain and chest CT and pathological changes of the brain tissue were observed. Results Serum levels of IL-6,IL-8 and NSE were elevated to varying degrees after blast. All of them increased significantly at different time points after blast (P<0.05).Brain and chest CT examinations did not show any significant positive results.Pathological results showed that there was a little necrosis in the brain, some neurons had karyopycnosis, karyolysis or disappearance of the nucleoli, and the cell boundaries were blurred. The blast wave was blocked greatly by the scalp and skull (about 90%),but could still penetrate them and cause brain injuries. Conclusions Explosion in the adjacent cabin causes mainly mild traumatic brain injuries.Blast wave can be blocked by the scalp and skull greatly.

ABSTRACT

The aim of this study is to characterize the Pirapó, Paranapanema 3 and 4 Hydrographic Unit, emphasizing its physical attributes and processes of use and occupation, responsible for the structure of the current landscape and the state of its water resources. The recognition of the landscape’s spatial structure in the hydrographic unit and its drainage basins was obtained by integrated analysis of the main elements that compose it: geology, landforms (hypsometric and slope), soils, climate and land use. Analysis revealed that within each drainage basin several variations in the spatial structure of the landscape occur which produce an internal compartmentalization. Each compartment is defined by its own geo-ecological structure, physiognomic standards and dynamics, reflected in its potentialities and vulnerabilities and in the conditions of water resources in the wake of occupation and use over time.

ABSTRACT

The aim was to describe the different intraoral pressures that occur in the vestibular interocclusal space (IOS) and subpalatal space (SPS) under different oral functions in adult patients with maxillary edentulism. A descriptive and cross-sectional study was conducted. We selected 15 patients at the Dental Clinic of the Dentistry Faculty (CODA), Universidad de la Frontera (Temuco, Chile). Intraoral pressure measurement was performed using a digital manometer GMH 3156, by methods of cannulas (control) and oral shield. A calibration measurement was made for 15 minutes; then four functional measurements were performed: B1 (open-mouth condition, 0-25 s), B2 (semi-open compartment condition, 35-85 s), B3 (closed compartments condition; 95-145 s) and B4 (swallowing, 150-165 s). With the oral shield, the B1 phase in the IOS showed a constant pressure between 0-4 mBar and SPS between 1 and 3 mBar. In B2 phase, the IOS reached 10 mBar and SPS between 0 and 1 mBar . Phase B3 OIS oscillations showed between 0 and 20 mBar, and the SPS increased to 20 mbar. In B4 phase, the IOS showed a peak of 40 mbar, while the SPS showed a peak from 20 to 140 mbar. The formation of two closed compartments with negative intra-oral pressure also occurs in edentulous patients, where the SPS shows greater negative pressure changes than IOS. Regular patterns of intra-oral pressure in biofunctional compartments were observed in over 50% of cases.

ABSTRACT

In order to define the effect of nutrient enrichment on trophic webs in an Andean mountain stream we performed an experiment using stable isotope ratios (δ15N and δ13C) to analyze different trophic compartments: 1) basal level: CPOM and biofilm; 2) primary consumers - macroinvertebrates: collector-gatherers (Heterelmis sp., Thraulodes sp. and Trichorythodes sp.), and collectorfilterers (Simulium sp.); 3) predators - fish (Oncorhynchus mykiss and Trichomycterus bogotensis). The average fractionation of nitrogen among the primary consumers with respect to CPOM was 4.7 ‰, and 1.7 ‰ with respect to biofilm. Predators incremented their δ15N signal by 5.9 % with respect to primary consumers. A depletion of δ15N was observed in Impact with respect to Control reach after fertilization in different compartments (biofilm, Heterelmis, Simulium andTricorythodes), while depletion was not significant for top predators. In most cases, the δ13C signal of biofilm overlapped with that of primary consumers, but a clear enrichment was observed with respect to CPOM. Macroinvertebrate gut contents showed fine detritus to be their most abundant food, and that in general there were no changes in diet as a consequence of nutrient enrichment. The only exception was Heterelmis, who increased its consumption in the Impact reach.

Objective To dynamically observe and evaluate the position and movement of pelvic floor organs in vaginal delivery primipara with translabial two-dimensional(2D) ultrasound, and to provide the imaging basis for the effects of pregnancy and vaginal delivery on the pelvic floor support structures. Methods Translabial 2D ultrasound was performed on 52 vaginal delivery postpartum women (postpartum group) and 43 normal women as control group to observe the position and movement of pelvic floor organs at rest,on pelvic muscle contraction and during maximum Valsalva maneuver. Results The location of urethrovesical junction and cervix were significantly lower in postpartum group compared with the control group at three stages. The movement of urethrovesical junction and cervix in vertical direction was greater in postpartum group than control group during maximum Valsalva maneuver, while was smaller on pelvic floor contraction. Conclusions Translabial 2D ultrasound could dynamically observe and evaluate the position and movement of pelvic floor organs.Pregnancy and vaginal delivery produce certain effect on pelvic floor support structures, which manifest as weak, defective and dysfunction. The impact was more obvious in anterior and middle compartments.

ABSTRACT

Objective To investigate the abnormality of fluid distribution and its effect on blood pressure (BP) in hemodialysis patients. Methods One hundred and six healthy volunteers as normal controls were divided into two groups(male group and female group).ECV%?ICV%(% means a percentage of weight) and ECV/ICV were measured by bioelectrical impedance spectroscopy and calculated in 2 groups respectively.Sixty-seven stable HD patients who had been on HD for more than 2 months in our hospital were selected. Patients were divided into 4 groups (hypotension prone group, normotensive group, hypertensive group and refractory hypertensive group). ECV%, ICV% and ECV/ICV were measured and calculated as above immediately before and after HD session. The measurement was conducted by Xitron 4200 HYDRA ECF/ICF Bio-Impedance Spectrum Analyzer. Results In patients on HD, a significant decrease of ECV% and ECV/ICV, and a significant increase of ICV% were seen post-HD. The pre-HD ECV% and ECV/ICV were significantly higher than those of normal controls but dropped close to normal level after hemodialysis. Post-HD ECV% was still higher than that of normal controls[(31.6?1.0)% vs (25.3?1.6)%, P 0.05]and post-HD ECV% of these patients was significantly lower than that of normal controlsand post-HD ECV% of these patients was significantly lower than that of normal controls[(19.3?1.6)% vs (22.6?1.4)%,P
