ABSTRACT

Objective To analyze the effect of Compound Chaigui Prescription on the changes of serum metabolites in rats with depression by 1H-NMR metabolomics. Methods Rats model established were modeled by chronic mild unpredictable stress (CUMS) procedure. Compound Chaigui Prescription and positive drugs (venlafaxine) were used as intervention drugs. Rat serum was analyzed by nuclear magnetic resonance (NMR) method. After data pretreatment, SIMCA-P 14.1 software was introduced for multivariate statistical analysis to verify the antidepressant efficacy of Compound Chaigui Prescription from the perspective of small molecule metabolites, and find potential disease biomarkers and drug efficacy markers. Results The CUMS depression rat model was successfully replicated. The metabolomics results showed that the levels of isoleucine, trimethylamine oxide, and creatine in the serum of CUMS-depressed rats were significantly higher than those of the model group. The levels of N-acetyl-glycoprotein, choline, and glucose were decreased with a significant difference (P < 0.05, 0.01). Conclusion The serum metabolite levels in CUMS model rats were significantly corrected after Compound Chaigui Prescription intervention. This study can provide reference for the study of antidepression mechanism of Compound Chaigui Prescription.

ABSTRACT

Objective: To investigate the antidepressant effect of Chaigui Granules on the symptoms associated with depression. Methods: The chronic mild unpredictable stress (CUMS) model was used to simulate mild to moderate depression. The effects of Chaigui Granules on rats in Morris water maze test, the elevated plus maze test and the gastrointestinal function test were investigated. Results: In all doses of Chaigui Granules, the latency of platform exploration in Morris experiment was significantly reduced (P < 0.05) and the number of times entering the platform was increased (P < 0.05). The ratio of open-arms time and the frequency of open-arms entries in the elevated plus maze experiment were also increased (P < 0.05, P < 0.01). The gastric residual rate was significantly reduced (P < 0.05) in the low and middle dose group, and the propulsion rate of intestinum tenue was increased (P < 0.05, P < 0.01) in all group. Conclusion: The results showed that Chaigui Granules obviously improved the decreased ability of learning and memory, anxiety, gastrointestinal dysfunction caused by CUMS model in rats, indicating that it can significantly improve the depressive symptoms and alleviate its associated symptoms.

ABSTRACT

Objective: To study the antidepressant effect of Compound Chaigui Prescription and its mechanism of regulating 5-hydroxytryptamine (5-HT) metabolic pathway. Methods: SD rats were randomly divided into seven groups: blank control group, model group, western medicine positive control group (venlafaxine 0.035 g/kg), traditional Chinese medicine positive control group (Shugan Jieyu Capusle 0.15 g/kg), high-, mid-, and low-doses of Compound Chaigui Prescription (crude drug 16.6, 8.3, and 4.2 g/kg). And the rats were treated with chronic unpredictable mild stress (CUMS) for 4 weeks, the body weight, sugar preference rate and opening behavior were observed after administration; The levels of tryptophan (Trp), 5-HT, and 5-hydroxyindole acetic acid (5-HIAA) in serum were measured by LC-MS. Results: Compound Chaigui Prescription can significantly callback CUMS model rats abnormal level of body weight, sugar preference rate and opening behavior and the levels of Trp, 5-HT, and 5-HIAA in serum. Conclusion: The Compound Chaigui Prescription has significant antidepressant effect, which is equivalent to the antidepressant effect of venlafaxine and Shugan Jieyu Capsule, and the mechanism may be related to the regulation of 5-HT metabolic pathway.
