ABSTRACT
OBJECTIVE：To establish the method for online rapid detection of related indexes in the concentration process of Huagai san extraction solution ，and to provide reference for judgment of concentration end point. METHODS ：Online diagram of 73 concentrated samples in the concentration process of Huagai san extraction solution were drawn by NIRS online collection equipment. The partial least squares （PLS）method was used to establish the NIRS quantitative calibration model of 5 related indexes （concentration density ， solid content ， the contents of amygdalin ， ephedrine hydrochloride and pseudoephedrine hydrochloride）. Another 15 samples were used to validate the model ，and the correlation of predicted value and measured value was analyzed. RESULTS ：The correlation coefficients （R2）of the concentration density ，solid content ，the contents of amygdalin ， ephedrine hydrochloride and pseudoephedrine hydrochloride in the quantitative calibration model were 0.982 5，0.999 9，0.998 3， 0.999 4 and 0.999 3，respectively；the root mean square errors of calibration （RMSEC）were 0.001 6，0.025 1，0.014 7，0.001 8 and 0.000 9；the root mean square errors of cross validation （RMSECV）were 0.002 1，0.035 8，0.033 6，0.006 3 and 0.001 3， respectively. After validation by 15 samples，root mean square errors of prediction （RMSEP）were 0.003 2，0.024 6，0.021 5， 0.007 7 and 0.005 9，respectively. CONCLUSIONS ：The established quantitative calibration model has good predictability and can provide basis for online judgment of concentration end point of Huagai san extraction solution.
ABSTRACT
Ganmaoling granule, with annual sale of over one billion yuan, is the first brand of domestic cold medicine sales. As the only traditonal Chinese medicine（TCM） quality control indicator of Ganmaoling granule, linarin is thermally unstable. Its content will be changed significantly during the production process, which would then affect the quality of the finished product. In this paper, the law of degradation of linarin was investigated. The experimental results showed that degradation reaction of linarin belongs to the first reaction characteristics. The effective methods to reduce the loss of linarin would be realized fortunately by strictly controlling the heating temperature or shortening the heating time.
ABSTRACT
OBJECTIVE: To investigate the application of near infrared spectroscopy in rapid and quantitative determination of multiple quality indicators in concentration process of Bufo bufo gargarizans Cantor extract. METHODS: NIR transmission spectra of concentrated solutions were collected in rectangular quartz cuvette with different optical pathlengths (2 and 5 mm). Calibration models of density, water content and indole alkaloids were developed by partial least squares regression (PLSR). The established models were applied to predict the unknown samples to test the performance of the models. RESULTS: The correlation coefficients of the density, water content and indole alka. The residual predictive deviations (RPD) were all above 3, and the relative standard errors of predictions(RSEP) were less than 10%, indicating the satisfactory model performance and predictive ability. Moreover, better calibration results were obtained with an optical path length of 2 mm for density and water content, and 5 mm for indole alkaloids. The results revealed that the optical path length has influence on the NIR analysis results, and the appropriate path length for the NIR analysis should be determined by comparison analysis. CONCLUSION: NIR spectroscopy technique provides a novel efficient and environmental friendly approach for the fast simultaneous determination of three key quality indicators (density, water content and indole alkaloids) in the concentration process of Bufo bufo gargarizans Cantor extract.
