ABSTRACT

Objective To study stress relaxation behaviors of cartilage scaffolds under different degradation cycles by using finite element analysis combined with theoretical models. Methods Based on the established degradation theoretical model, the elastic modulus of the scaffold was calculated under different degradation cycles. The finite element model of cartilage scaffolds was established and stress relaxation simulation was performed to analyze the variation of scaffold relaxation stress with time. The stress relaxation constitutive model was established to predict mechanical properties of the scaffold. Results The elastic modulus of cartilage scaffolds at 14 th, 28th, 42nd, 56th day after degradation was 32. 35, 31. 12, 29. 91, 28. 74 kPa, respectively. The upper layer for cartilage scaffolds was the largest. The overall relaxation stress of the scaffold decreased rapidly with time and then tended to be stable. At 8th week after degradation, the stress which the scaffold couldwithstand was still within the physiological load range of the cartilage. The predicted results of the stress relaxation constitutive model were in good agreement with the finite element simulation results. Conclusions The elastic modulus of the scaffold gradually decreases with the increase of degradation time. The longer the degradation period is, the less stress the scaffold can withstand. At the same degradation period, the larger the applied compressive strain, the larger the stress on the scaffold. Both the finite element simulation and stress relaxation constitutive model can effectively predict stress variations of cartilage scaffolds under degradation

The karyotype and constitutive heterochromatin pattern of the white stork Ciconia ciconia samples obtained from Manzala lake, Dimiaat, Egypt was described. Somatic cells of Ciconia ciconia samples have diploid number 2n= 68 chromosomes. Out of 68 chromosomes, 11 pairs including sex chromosomes were macrochromosomes and the remaining pairs were microchromosomes. Of the 11 macrochromosome pairs, no.1, 2, 4 and 5 were submetacentric and pairs no. 6, 7 and 8 were described as metacentric. In addition, the autosome pair no.3 was subtelocentric, while autosome pair no.9 was acrocentric. Also, the sex chromosome Z represents the fourth one in size and it was classified as submetacentric while, W chromosome appeared as medium size and was acrocentric. Furthermore, C-banding pattern (constitutive heterochromatin) revealed variation in their sizes and occurrence between macrochromosomes. Pairs no. 7 and 8 of autosomes exhibited unusual distribution of heterochromatin, where they appeared as entirely heterochromatic. This may be related to the origin of sex chromosomes Z and W. However, there is no sufficient evidence illustrate the appearance of entirely heterochromatic autosomes. Therefore, there is no available cytogenetic literature that describes the C-banding and karyotype of Ciconia Ciconia, so the results herein are important and may assist in cytogenetic study and evolutionary pattern of Ciconiiformes.


O cariótipo e o padrão constitutivo de heterocromatina das amostras de cegonha-branca Ciconia ciconia obtidas no lago Manzala, Dimiaat, Egito, foram descritos. As células somáticas de amostras de Ciconia ciconia possuem número diploide 2n = 68 cromossomos. ABSTRACT

ABSTRACT Zephyranthes citrina is an ornamental American bulbous plant used as an ornamental garden crop for the aesthetic qualities of its yellow perigonium. The objective of this work was to characterize the species by classical chromosome staining and fluorochrome banding. A sporophytic chromosome number of 2n=8x=48 chromosomes was observed, being the karyotypic formula 20 m + 26 sm + 2 st. Satellites were detected in the short arm of metacentric chromosomes 8, 9, 11 and 12, which colocalized with constitutive heterochromatin CMA+/DAPI-/0 bands. The karyotype comprised chromosome pairs with terminal constitutive heterochromatin bands that included satellites and heteromorphic clusters indicating that it is an allooctoploid. These results will be used as a tool for monitoring genetic improvement, in interspecific crosses and its progenies and in biotechnological procedures by in vitro culture.


RESUMEN Zephyranhtes citrina es una planta bulbosa americana, ornamental, utilizada en jardines por las cualidades estéticas de su perigonio amarillo. El objetivo de este trabajo fue caracterizar citogenéticamente la especie con tinción clásica convencional y bandeo cromosómico. Se observó un número cromosómico esporofítico de 2n=8x=48, siendo la fórmula cariotípica 20 m + 26 sm + 2st. Se detectaron satélites en el brazo corto de los cromosomas metacéntricos 8, 9, 11 y 12, que co-localizaron con bandas de heterocromatina constitutiva CMA+/DAPI-. El cariotipo comprendió pares de cromosomas con bandas de heterocromatina constitutivas terminales que incluyeron satélites y grupos heteromórficos que indican que es un alooctoploide. Estos resultados serán usados como herramientas en el monitoreo del mejoramiento genético, en análisis de cruzamientos interespecíficos y progenies y en procedimientos biotecnológicos de cultivo in vitro.

ABSTRACT

Objective To study influencing factors of renal blunt impact injury by using finite element (FE) method. Methods Based on CT images of the kidney, the kidney FE models for different age groups were constructed. The renal blunt impact test was reconstructed, and the influence of kidney material constitutive parameters, kidney tissue structure, kidney size, impact position and impact velocity on injury severity were analyzed. Results Under the same impact condition, the stress of renal cortex decreased with the kidney mass increasing, and increased with the impact velocity of the hammer increasing. The renal capsule had a certain energy absorption effect, so as to reduce the kidney stress. When the kidney was impacted, the stress of renal cortex under side impact was significantly higher than that under frontal impact. Conclusions Compared with viscoelastic constitutive model, Mooney Rivlin material constitutive model is more suitable for FE evaluation on renal injury severity. The renal injury decreases with the kidney mass increasing. The increase of impact velocity will intensify the renal injury severity. Renal capsule will reduce renal injury to a certain extent, so the existence of renal capsule structure must be considered in FE modeling of the kidney. Compared with frontal and rear impact, the renal injury severity is greater when the kidney is impacted from the lateral side.

ABSTRACT

Objective To study the constitutive model of adipose tissue at medium strain rate and its parameter inversion. Methods Based on experiments of adipose tissue mechanical properties, the compression experiment of adipose tissues was reconstructed by finite element method, and the parameters for characterizing constitutive models of adipose tissues were screened. Combined with the method of feasible direction (MFD) in optimization method, the reverse calculation for parameters of fat tissue constitutive model at medium strain rate was conducted. ResultsCompared with Ogden constitutive model, the viscoelastic constitutive model was more suitable for characterizing the mechanical response at medium strain rate (260 s-1). The parameters of the constitutive model suitable for simulation were obtained using the reverse method. Conclusions The viscoelastic constitutive model was more suitable for characterizing the mechanical response at medium strain rate. The results provide references for studying the influence of human adipose tissues on body injury in finite element simulation of vehicle collisions.

ABSTRACT

Objective On the basis of explicit dynamics calculation theory, a numerical model for calculating active and passive properties of muscles with high strain rate was proposed. Methods In the process of calculating the motion equation of muscle element with high strain rate, Hill’s three-factor muscle model with high strain rate was introduced into the noda force formula to modify the node force in each time step. Results As Hill’s three-factor muscle model was introduced in numerical calculation, the muscle element had the passive characteristics of the general structural constitutive model and its proprietary active characteristics. Conclusions The research findings will contribute to numerical calculation for dynamic response and damage of muscles with high strain rate.

ABSTRACT

Orthodontic tooth movement is achieved by remodeling periodontal ligament, alveolar bone and other periodontal tissues in response to mechanical loading. The periodontal ligament is the fibrous connective tissue connecting cementum and alveolar bone, which plays an important role in the transmission, absorption and dispersion of supporting teeth and biting force. When orthodontic forces are applied to the teeth, load transfer occurs from the tooth through the periodontal ligament to the alveolar bone. The remodeling of periodontal ligament interacts with the alveolar bone formation and absorption alternately. As the initial factor of orthodontic tooth movement, the stress/strain of periodontal ligament is involved in the process of alveolar bone remodeling and affects the direction and speed of tooth movement. Therefore, the biomechanical properties of the periodontal ligament have received extensive attention. In this paper, the related researches on the structural changes and biomechanical characteristics of the periodontal ligament during orthodontic tooth movement were reviewed as well as the researches on the mechanical constitutive model of the periodontal ligament. The mechanical response and biomechanical mechanism of the periodontal ligament were discussed in order to provide a theoretical basis for accurate numerical simulation of orthodontic tooth movement.

ABSTRACT

Objective Aiming at the problem that mechanical properties for the continuum of muscle tissues cannot be considered in active and passive behaviors of different structurally coupled muscles, a method of passive and active coupling in the same constitutive equation was proposed to construct ahyperelastic active and passive constitutive model of skeletal muscle continuum. Methods In order to calibrate parameters of the passive constitutive model, the uniaxial tensile experiment method and conditions were given, and through theoretical derivation, the specific method of using experimental data to solve the passive model parameters was introduced. In order to verify effectiveness of the active model, the model was verified with an example. Results The curves predicted by the model were in good agreement with the experimental output stress-stretch ratio curves. At the same strain, the maximum error of passive stress and total stress were only 20 kPa and 40 kPa. Conclusions The continuum hyperelastic constitutive model can better simulate active and passive behavior of skeletal muscles, which is beneficial for modeling and simulation of human muscles in further study.

ABSTRACT

Genetically engineered plants have varied applications in agriculture for enhancing the values of food and feed.Genetic engineering aims to introduce selected genetic regions with desirable traits into target plants for bothspatial and temporal expressions. Promoters are the key elements responsible for regulating gene expressionsby modulating the transcription factors (TFs) through recognition of RNA polymerases. Based on theirrecognition and expression, RNA polymerases were categorized into RNA pol II and pol III promoters.Promoter activity and specificity are the two prime parameters in regulating the transgene expression. Since theuse of constitutive promoters like Cauliflower mosaic virus (CaMV) 35S may lead to adverse effects on nontarget organisms or ecosystem, inducible/tissue specific promoters and/or the RNA pol III promoters providemyriad opportunities for gene expressions with controlled regulation and with minimum adverse effects.Besides their role in transgene expression, their influence in synthetic biology and genome editing are alsodiscussed. This review provides an update on the importance, current prospects, and insight into the advantagesand disadvantages of promoters reported thus far would help to utilize them in the endeavour to developnutritionally and agronomically improved transgenic crops for commercialization.

ABSTRACT

Helicoverpa armigera Hübner, 1805 and Spodoptera frugiperda Smith, 1797 are polyphagous pests of great agricultural importance in subtropical and temperate climate regions. The usual management of production areas in the southern region of Brazil occurs after the harvesting of summer crops, and the areas are sown with pasture. Thus, forages recommended for grazing are azevém (Lolium multiflorumLam.) and cornichão (Lotus corniculatus L.) due to their nutritional benefits, good palatability, regrowth, and hardiness. Considering the high degree of polyphagia of H. armigera and S. frugiperda, and the impact of maintaining continuous feeding areas (green bridges) in the management of these species, this work aimed to evaluate the development of H. armigera and S. frugiperda fed leaves of azevém and cornichão under laboratory conditions. The insects were collected in the city of Capão do Leão/RS in corn and soybean plantations. For each forage species, 130 newly hatched caterpillars were each placed in autoclaved glass tubes, with one-third of the tube length containing forage. The tubes were capped with waterproof cotton and placed in a climate-controlled room at 25 ± 1°C, 70 ± 10% relative humidity, and a 12-h photophase. The food was replenished daily until the caterpillars entered the pupae stage. The pupae were sexed and weighed, and the newly emerged adults were placed in pairs in PVC cages, lined with white A4 sulfite paper. The papers used as a laying substrate were removed and changed daily, and eggs were counted. The number of instars was determined by the linearized Dyar rule model. The complete randomization design was employed for the variables biological cycle length and viability of egg, caterpillar, pre-pupa, pupa, adult, and pre-oviposition phases and weight of caterpillars on the 14th day and pupae after 24 h. Based on the results obtained, a fertility life table was prepared. H. armigera did not complete the cycle, with only three instars and a duration of 22.1 and 24.6 days in azevém and cornichão, respectively. When evaluated in S. frugiperda caterpillars, development in forage species was observed, with five and six instars and duration of 51.7 and 45.1 days in azevém and cornichão, respectively. The azevém was distinguished by interference in the development cycle of the species, reducing the effect of the green bridge. In addition, surviving insects were susceptible to the effects of the agroecosystem due to the low-quality food source.

Após a colheita das culturas de verão no sul do Brasil, as áreas são utilizadas como pastagem. Com isso, as forrageiras recomendadas ao pastoreio são cornichão (Lotuscorniculatus L.) e azevém (Loliummultiflorum Lam.) por apresentarem benefícios nutricional, palatabilidade, rebrota e rusticidade. Devido ao alto grau de polifagia de H. armigera e S. frugiperda e o impacto de áreas continuas de alimento (pontes verdes) no manejo destas espécies, este trabalho objetivou-se avaliar o desenvolvimento de H. armigera e S. frugiperda alimentadas com azevém e cornichão em condições de laboratório. Os insetos foram coletados no município de Capão do Leão/RS em plantios de milho e soja. Para cada espécie forrageira foram individualizadas 130 lagartas recém-eclodidas em tubos de vidro autoclavados, com um terço (1/3) do seu comprimento contendo a respectiva forrageira. Os tubos foram tampados com algodão hidrófugo e levados para a sala climatizada a 25 ± 1°C, UR de 70 ± 10% e 12 horas de fotofase. Diariamente, o alimento foi reposto até que as lagartas entrarem em estágio de pupa. As pupas foram sexadas e pesadas e os adultos recém-emergidos foram individualizados em casais em gaiolas de PVC, forradas com papel sulfite A4 branco no seu interior. Os papéis utilizados como substrato de postura foram retirados e trocados diariamente e os ovos foram contabilizados. O número de instares foi determinado pelo modelo linearizado da regra de Dyar. O delineamento utilizado foi inteiramente casualizado (DIC) para as variáveis duração do ciclo biológico e viabilidade das fases de ovo, lagarta, pré-pupa, pupa, adultos e pré-oviposição, peso de lagartas ao decimo quarto (14°) dia e pupas após 24 horas. Com base nos resultados obtidos, foi elaborada tabela de vida de fertilidade. H. armigera não completou o ciclo, com apenas três instares e duração de 22,1 e 24,6 dias em azevém e cornichão, respectivamente. Quando avaliado em lagartas de S. frugiperda observou o desenvolvimento nas espécies forrageiras, com 5 e 6 instares e duração de 51,7 e 45,1 dias em azevém e cornichão, respectivamente. Contudo, considerando o critério que evite a formação de ponte verde, azevém destacou-se por interferir no ciclo de desenvolvimento das espécies. Onde, mesmos os que sobrevivem, estão suscetíveis aos efeitos do agroecossitema devido a fonte alimentar ser de baixa qualidade.

ABSTRACT

This research aimed to evaluate the biological aspects and the feeding behavior of Aphis gossypii in watermelon cultivars submitted to silicon application. The experiment was conducted at the Institute of Education, Agriculture and Environment of the Federal University of Amazonas, Humaitá, Brazil. The experimental design was completely randomized in a 2×3 factorial (with and without silicon; cultivars Crimson Sweet, Fairfax and Charleston), with ten replications. The application of silicic acid (1%) was carried out directly on the substrate using dose equivalent to 1 ton SiO2·ha-1, 25 days after sowing. The rearing of aphids was kept in cucumber plants, cultivar Caipira. Insect biology tests were conducted to evaluate the duration of the prereproductive, reproductive and postreproductive periods, longevity, number of nymphs, and feeding behavior using the honeydew secretion technique. Analysis of variance was performed using the statistical program SISVAR and the means were compared by the F and Scott­Knott test (p ≤ 0.05). The silicon application to watermelon plants affects the reproduction and feeding of A. gossypii. The watermelon plants cultivar Crimson Sweet treated with silicon has high resistance to feeding by A. gossypii.(AU)


Nesta pesquisa objetivou-se avaliar os aspectos biológicos e o comportamento alimentar de Aphis gossypii em cultivares de melancia submetidas à aplicação de silício. O experimento foi conduzido no Instituto de Educação, Agricultura e Ambiente da Universidade Federal do Amazonas, Humaitá, Brasil. Utilizaram-se o delineamento experimental inteiramente ao acaso e o esquema fatorial 2×3 (sem silício e com silício; cultivares Crimson Sweet, Fairfax e Charleston), com dez repetições. A aplicação do ácido silícico (1%) foi realizada diretamente no substrato, com dose equivalente a 1 ton SiO2·ha-1, 25 dias após a semeadura. Os pulgões da criação foram mantidos em plantas de pepino, cultivar Caipira. Foram conduzidos ensaios de biologia do inseto para avaliação da duração dos períodos pré-reprodutivo, reprodutivo e pós-reprodutivo, longevidade, número de ninfas e comportamento alimentar por meio da técnica de secreção de honeydew. Realizou-se a análise de variância dos dados utilizando-se o programa estatístico SISVAR e as médias foram comparadas pelo teste de F e Scott­Knott (p ≤ 0,05). A aplicação de silício em plantas de melancia afeta a reprodução e a alimentação de A. gossypii. Plantas de melancia do cultivar Crimson Sweet tratadas com silício apresentam alta resistência à alimentação por A. gossypii.(AU)

ABSTRACT

Coronary artery diseases (CAD) have always been serious threats to human health. The measurement, constitutive modeling, and analysis of mechanical properties of the blood vessel wall can provide a tool for disease diagnosis, stent implantation, and artificial artery design. The vessel wall has both active and passive mechanical properties. The passive mechanical properties are mainly determined by elastic and collagen fibers, and the active mechanical properties are determined by the contraction of vascular smooth muscle cells (VSMC). Substantial studies have shown that, the two-layer model of the vessel wall can feature the mechanical properties well, and the circumferential, axial and radial strain and stress are of great significance in arterial wall mechanics. This study reviewed recent investigations of mechanical properties of the vessel wall. Challenges and opportunities in this area are discussed relevant to the clinical treatment of coronary artery diseases.

ABSTRACT

Pregnane X receptor (PXR) is the major regulator of xenobiotic metabolism. PXR itself is controlled by various signaling molecules including glucocorticoids. Moreover, negative feed-back regulation has been proposed at the transcriptional level. We examined the involvement of the 3'-untranslated region (3'-UTR) of mRNA and microRNAs in PXR- and glucocorticoid receptor (GR)-mediated regulation of gene expression. PXR ligands were found to significantly downregulate mRNA expression in a set of 14 human hepatocyte cultures. Similarly, PXR was downregulated by PCN in the C57/BL6 mice liver. In mechanistic studies with the full-length 3'-UTR cloned into luciferase reporter or expression vectors, we showed that the 3'-UTR reduces PXR expression. From the miRNAs tested, miR-18a-5p inhibited both expression and gene induction. Importantly, we observed significant upregulation of miR-18a-5p expression 6 h after treatment with the PXR ligand rifampicin, which indicates a putative mechanism underlying negative feed-back regulation in hepatic cells. Additionally, glucocorticoids upregulated expression not only through the promoter region but also 3'-UTR regulation, which likely involves downregulation of miR-18a-5p. We conclude that miR-18a-5p is involved in the down-regulation of expression by its ligands and in the upregulation of mRNA expression by glucocorticoids in hepatic cells.

Abstract This study proposes in vivo tests and design of experiments to determine the influence of experimental factors on the mechanical response of the soft tissue. The experimental factors considered are: room temperature (A), indentation velocity (B), indenter temperature (C), pump pressure (D) and muscle activation (E). An inverse method was developed to obtain the constants for constitutive equations of a multilayer biological model (skin, hypodermis, and muscle) through the use of indentation tests in combination with a finite element method. For each combination of the experimental factors, two groups of constants were established from the inverse method. Sixteen combinations of experimental conditions and their corresponding constants for the Mooney-Rivlin constitutive equations were obtained to be used in further numerical models. The factor D and factor interactions ADE, CDE, and ACDE were statistically significant with respect to skin mechanical response. Therefore, it can be concluded that there is not a current equation able to represent the mechanical properties of the skin under all the experimental conditions considered in this study


Resumen Este estudio propone pruebas in vivo y diseño de experimentos para determinar la influencia de los factores experimentales en la respuesta mecánica de tejidos blandos. Los factores experimentales considerados son: temperatura ambiente (A) velocidad de indentación (B), temperatura del indentador (C), presión de bombeo (D) y activación muscular (E). Se desarrolló un método inverso con el fin de obtener las constantes para las ecuaciones constitutivas de un modelo biológico de multicapa (piel, hipodermis y músculo) a través del uso de pruebas de indentación en combinación con el método del elemento finito. Para cada combinación de los factores experimentales, se establecieron dos grupos de constantes del método inverso. Se obtuvieron dieciséis combinaciones de condiciones experimentales y sus correspondientes constantes para las ecuaciones constitutivas de Moorney-Rivlin, que se pueden usar en futuros modelos numéricos. El factor D y las interacciones de los factores ADE, CDE y ACDE fueron estadísticamente significativas con respecto a la respuesta mecánica de la piel. En consecuencia, se puede concluir que no hay actualmente una ecuación capaz de representar las propiedades mecánicas de la piel bajo las condiciones experimentales consideradas en este estudio.

ABSTRACT

In order to study the mechanical behavior of degeneration and nucleotomy of lumbar intervertebral disc, compression experiments with porcine lumbar intervertebral discs were carried out. The lumbar intervertebral discs with trypsin-treated and nucleus nucleotomy served as the experimental group and the normal discs as the control group. Considering the effects of load magnitude and loading rate, the relationship between stress and strain, instantaneous elastic modulus and creep property of intervertebral disc were obtained. The creep constitutive model was established. The results show that the strain and creep strain of the experimental group increase significantly with the increase of compression load and loading rate, whereas the instantaneous elastic modulus decreases obviously, compared with the control group. It indicates that the effect of load magnitude and loading rate on load-bearing capacity of intervertebral disc after nucleotomy is larger obviously than that of normal disc. The creep behavior of the experimental group can be still predicted by the Kelvin three-parameter solid model. The results will provide theoretical foundation for clinical treatment and postoperative rehabilitation of intervertebral disc disease.

ABSTRACT

Aiming at the problem of the influence of preloading force on its mechanical response in soft tissue compression experiments, an elimination method of preloading force based on linear loading region is proposed. Unconfined compression experiments under a variety of different preloading forces are performed. The influence of the preloading force on the parameters of constitutive model is analyzed. In the preload phase, the mechanical response of the soft tissue is taken as a linear model. The preloading force is eliminated by taking the preloading phase into account throughout the response process. According to five different preloading forces of the unconfined compression experiments, the elimination method is validated with two different constitutive models of soft tissue, and the error between the models obtained by the preloading force elimination method and the traditional method with the experimental results is compared. The results show that the error obtained by preloading force elimination method is significantly smaller than the traditional method. The preloading force elimination method can eliminate the influence of preloading force on mechanical response to a certain extent, and constitutive model parameters which are closer to the true properties of soft tissue can be obtained.
