ABSTRACT

OBJECTIVE:To study the analgesic effects of 4 sources of Curcumae Radix decoction pieces (Curcuma wenyujin , C. kwangsiensis ,C. phaeocaulis and C. longa ),and compare the contents of curcumenol in their water extracts. METHODS : Using aspirin as positive control ,acetic acid writhing method was used to investigate the effects of 4 sources of Curcumae Radix decoction pieces water extract on writhing latency and times of writhing in mice. The moisture contents of 4 sources of Curcumae Radix decoction pieces (10 batches of each source ,the same below )were determined according to the drying method in 2015 edition of Chinese Pharmacopoeia (part Ⅳ). The yield of water extract in 4 sources of Curcumae Radix decoction pieces were investigated by hot dipping method ,the contents of curcumenol in water extract of 4 sources of Curcumae Radix decoction pieces were determined by HPLC ,and comparison was conducted. RESULTS :Compared with model group ,aspirin and water extracts of 4 sources of Curcumae Radix decoction pieces could significantly prolong the writhing latency of model mice ,and the water extracts of C. wenyujin and C. phaeocaulis could significantly reduce the writhing times of model mice (P<0.05 or P<0.01). For C. wenyujin ,C. kwangsiensis ,C. phaeocaulis and C. longa ,the contents of moisture were 7.39%-8.80%,7.88%-9.88%,7.66%- 10.5468/ogs.2019.62.6.382 9.86% and 7.68%-10.20%;the average yield of water extract were 46.30%,60.40%,38.65%,42.99%;the average contents of curcumenol in water extract were 0.271,0.066,0.310,0.058 mg/g. Except for a few batches ,the higher the yield of water extract,the higher the content of curcuminol in the same source of Curcumae Radix decoction pieces. CONCLUSIONS :Four sources of Curcumae Radix decoction pieces have analgesic effect. The contents of curcumenol in C. wenyujin and C. phaeocaulis were similar ,and the contents of curcumenol in C. kwangsiensis and C. longa about 1/5 of that in C. phaeocaulis and C. wenyujin .

ABSTRACT

OBJECTIVE:To establish a method for the content determination of diosgenin in Dioscorea zingiberensis,and to compare the contents of diosgenin in wild D. zingiberensis from different habitats. METHODS:HPLC method was adopted. The determination was performed on Inertsil ODS-3 column with mobile phase consisted of acetonitrile-water (50 ∶ 50,V/V) at the flow rate of 1.0 mL/min. The column temperature was set at 30 ℃. The detection wavelength was set at 203 nm,and sample size was 10 μL. Established method was used to determine the contents of diosgenin in wild D. zingiberensis from different habitats (Shaanxi Shangluo,Shaanxi Ankang,Henan Neixiang,Yunnan Xuanwei,Sichuan Deyang,Hubei Shiyan,Hunan Zhangjiajie,Hunan Changde). The differences of diosgenin content were compared. RESULTS:The linear range of diosgenin were 4.16-165.6 μg/mL (r=     0.999 9). RSDs of precision,reproducible and stability tests were all lower than 2% (n=5 or n=6). The average recovery of diosgenin was 101.18% (RSD=1.27%,n=6). The content of diosgenin in wild D. zingiberensis from different habitats were in descending order,i.e. Sichuan Deyang (1 405.36 μg/g)>Shaanxi Shangluo (1 201.79 μg/g)>Hunan Zhangjiajie (1 035.18 μg/g)>Shaanxi Ankang (632.64 μg/g)>Hunan Changde (598.64 μg/g)>Yunnan Xuanwei (425.34 μg/g)>Henan Neixiang (350.13         μg/g)>Hubei Shiyan (338.39 μg/g). CONCLUSIONS:Established method is simple,accurate and suitable for the content determination of diosgenin in D. zingiberensis. The contents of diosgenin in wild D. zingiberensis from different habitats are different significantly,and the content of diosgenin in the samples from Sichuan Deyang is the highest.

ABSTRACT

Objective: To optimize the extraction process of total flavonoid from different kinds of teas and com- pare the total flavonoid content in teas. Methods: The UV spectrophotometry was used to determine the total flavonoids in tea. The effects of ethanol concentration,material-liquid ratio,extraction temperature and extraction time on the ex- traction rate were investigated. The extraction processes were optimized by the orthogonal experiment and variance analy- sis,and the total flavonoid content was compared for different extraction processes. Results: The conditions for the opti- mal extraction processes were as follows:60% ethanol concentration,1:40(g/ml)material-liquid ratio,80℃ extraction temperature and 70 min extraction time for black tea;60% ethanol concentration,1:40(g/ml)material-liquid ratio, 90℃ extraction temperature and 70 min extraction time for oolong tea;and 70% ethanol concentration,1:40(g/ml)ma- terial-liquid ratio,80℃ extraction temperature and 70 min extraction time for green tea. Under the conditions of the opti- mized extraction processes,the content of total flavonoids in black tea was the highest,followed by the oolong tea,with the least content in green tea. Conclusion: The optimization of the extraction process could significantly increase the ex- traction of in tea,and the total flavonoid content obviously differed in the different kinds of teas. The present results pro-vide an experimental basis for future studies on the extraction of total flavonoid in teas to further explore the influencing factors to the extraction.

ABSTRACT

OBJECTIVE:To optimize the extraction technology of Tibetan medicine Pedicularis kansuensis and compare con-tent of verbascoside and isoverbascoside differences in P. kansuensis from various habitats. METHODS:Using verbascoside and iso-verbascoside and dry paste yield as comprehensive evaluation indexes,single factor test and orthogonal test were used to investigate the extraction solvent,solvent dosage,extraction time and times to optimize extraction technology,and the verification test was conducted. Contents of the 2 constituents verbascoside and isoverbascoside in P. kansuensis from Gansu,Qinghai and Sichuan were compared. RESULTS:The optimal extraction technology was as follows as 8-fold 50% ethanol,extraction for 3 times,90 min each time. The verification results showed that the average contents of verbascoside and isoverbascoside were 3.49%(RSD=1.28%,n=3),1.26%(RSD=1.32%,n=3),and average dry paste yields were 37.99%(RSD=1.97%,n=3). The contents of verbascoside and isoverbascoside in P. kansuensis from Qinghai were relatively higher. CONCLUSIONS:Optimized extraction tech-nology is reasonable,stable,feasible;the contents of index constituents in P. kansuensis from different habitats have certain differ-ences. The study can provide scientific evidence for the development and utilization of extraction,and the in-depth study of quality evaluation for medicinal material.

ABSTRACT

OBJECTIVE:To compare the contents of 2 index components in salt frying and salt steaming samples of Psoralea corylifolia,and select better processing technology. METHODS:Under the conditions that ratio of salt and medicinal materials was 1:50,4-5 times salt of adding water,brine run time was 2 h,the amounts of psoralen and isopsoralen in salt frying samples after 10,12,14 min of frying under 150 ℃,170 ℃,190 ℃ and in salt steaming samples after 1,1.5,2,2.5 h of steaming were re-spectively investigated,and the effects of 2 processing technology were compared. RESULTS:Under fixed condition,the contents of index components was the highest in salt frying samples by fried for 12 min under 150 ℃ and in salt steaming samples by steamed for 1 h;the content in salt steaming samples (1.49%) was higher than that in salt frying samples (1.29%)(P=0.011). CONCLUSIONS:According to the comparison of index components'contents in processing samples,salt steaming is superior to salt frying of P. corylifolia.
