ABSTRACT

Objective To investigate the protective effects of Coptis chinensis decoction on the metabolic index and renal function of rats with fructose-induced metabolism syndrome (MS).Methods From 56 male Wistar rats,6 were randomly selected as normal control group,the other rats were daily fed with 10% fructose water for 8 weeks to reproduce the MS model.Twenty-four rats were established successfully as MS model and randomly divided into 4 groups (6 each):model group,Coptis decoction group,tauroursodeoxycholic acid (TUDCA) group and berberine hydrochloride (BH) group.The body weight,visceral fat weight,fasting blood glucose (FBG),fasting insulin (FINS),insulin resistance index (HOMA-IR),cholesterol (TC),triglyceride (TG),systolic blood pressure (SBP),diastolic blood pressure (DBP),urinary N-acetyl-β-D-glucosidase (NAG),urine microalbumin (m-ALB),blood urea nitrogen (BUN) and serum creatinine (Scr) levels were compared between the groups,and the renal pathological changes were observed by light microscopy and electron microscopy.Results The levels of body weight,visceral fat weight,FINS,HOMA-IR,TC,TG,SBP,DBP and urinary NAG were obviously higher in the four MS model groups than in normal control group (P<0.05),while the levels of m-ALB,BUN and Scr showed no significant difference (P>0.05).After the intervention,the levels of body weight,visceral fat weight,FINS,HOMA-IR,TC,TG,SBP and DBP decreased significantly in Coptis decoction group than in model group (P<0.05).Compared with BH group and TUDCA group,the levels of FINS and HOMA-IR decreased obviously in Coptis decoction group (P<0.05),while the levels of body weight,TC,TG,SBP,DBP,urinary NAG,BUN and Scr showed no significant difference (P>0.05).It was observed in model group by light microscopy and electron microscopy that the glomerular swelled obviously,the capillary basement membrane of glomerular and renal tubules thickened,mesangial region and interstitial substances hyperplasia,podocytes swelled markedly and most of fusion,the epithelial cells of renal tubules serious vacuolar degeneration,and a lot of secondary lysosomes formed,autophagy of organelles occurred,and most of endoplasmic reticulum and mitochondria disappeared.The kidney pathological damage reduced obviously after treatment of Coptis decoction,but no significant difference when compared with that in BH group and TUDCA group.Conclusions Coptis decoction may decrease the body weight,reduce the levels of blood lipid and blood pressure,and better improve the insulin resistance when compared with BH and TUDCA.In addition,Coptis decoction can also reduce the renal pathological damage,and has the protective function for kidney.

ABSTRACT

Objective To study the effect of coptis decoction on rats with ulcerative colitis .Methods Rat ulcerative coli‐tis model was induced by 1 ,2‐dimethylhydrazine(DMH)(40 mg/kg s .c .)+3% dextran sodium sulfate solution(DSS)(freely drinking) .The drugs were given orally once a day for 7 day per week .After 5 weeks ,the body weight ,the number of colon ACFs and the number of colon cancer in rats ,as well as the morphological changes of rat colon tissues were evaluated . Results Coptis decoction significantly improved the dyscrasia induced by DM H+ DSS ,the both of body weight and general condition were better than those of control group(P<0 .05 ,P<0 .01) .Coptis decoction also significantly inhibited ACF in the rats treated by DM H+DSS for 10 weeks(P<0 .01) .The model group showed typical inflamed mucosa ,a nearly complete sub‐version of the normal surface architecture .Compared with the model group ,coptis decoction in each dosage group could im‐prove rat atypical hyperplasia ,mucosal hyperemia ,edema and cells in the crowded .Transmission electron microscope showed that ,in epithelial nuclei chromatin aggregation in the model group ,the cell surface microvilli arranged disorder ,fault phenome‐non .Compared with model group ,the coptis decoction had improved significantly .Conclusion Coptis decoction had significant therapeutic effect on rat ulcerative colitis .

ABSTRACT

Objective: To investigate the effects of particles in coptis (Coptidis Rhizoma)-decoction on the berberine absorption in intestine. Methods: The particles in copitis-decotion were separated by the high-speed centrifugation and weak-base anion-exchange resin. The characteristics of berberine absorption in intestine were evaluated by the in situ intestinal perfusion in rats. Results: Although each segment of small intestine exhibited obvious berberine absorption, the jejunum exhibited the highest absorption rate constant (Ka) of (3.587 9 ± 0.005 2) × 10-4/s and effective permeability coefficient (Peff) of (4.529 4 ± 0.009 7) × 10-5 cm/s. Particles P1 and P2, with similar size of (272.7 ± 25.2) and (264.8 ± 21.4) nm and different surface potential of (-6.85 ± 0.16) and (-18.20 ± 0.71) mV, were obtained from the copitis-decotion. Particle P1 could significantly improve the berberine absorption in intestine, with Ka of (5.853 6 ± 0.970 1) × 10-4/s and Peff of (8.082 4 ± 1.004 2) × 10-5 cm/s. Conclusion: Particles in coptis-decotion can improve the berberine absorption in intestine. The surface potential of the particles may be responsible for the improvement of berberine absorption.

ABSTRACT

Objective: To establish a method for determing the content of baicalin in Coptis decoction for detoxification. Methods: Baicalin as standard sample, a dual-wavelength UV spectrophotometric method was used to determine the baicalin content in coptis decoction for detoxification. The wavelengths for measurement and reference were 289 nm and 353 nm, respectively. Results: The linearity between concentration and absorbance of baicalin as standard was ranged from 6 to 18.0 μg/ml, the correlativity 0.999 6, the RSD 0.67%(intraday) or 1.11% (interday) , and the recovery 101.24%, respectively. Conclusion: The method is simple, rapid and accurate, and has potential application to the content determination of baicalin in Coptis decoction for detoxification.
