ABSTRACT

Objective To explore the expressions and correlation of COX-2, bcl-2 and Caspase-3 in diffuse large B-cell lymphoma(DLBCL). Methods The expression of COX-2, bcl-2 and Caspase-3 was studied in 29 cases of DLBCL and 10 cases of reactivited lymphoid tissue with immunohistochemistry of PowerVisionTM. Results The expression of COX-2 and bcl-2 are seperately 68.9 % and 72.4 %. The expression of COX-2 had correlation with Ann Arbor stage. The expression of COX-2 was positively correlated with that of bcl-2, the expression of bcl-2 was negatively correlated with that of Caspase-3, the expression of COX-2 was negatively correlated with that of Caspase-3. Conclusion COX-2, bcl-2 and Caspase-3 may be involved in the carcinogenesis of DLBCL, and their expression may be helpful in estimating the histology grade and prognosis of DLBCL.

ABSTRACT

Objective To investigate the role of cyclooxygenase-2 (COX-2) in sinusoidal capillarization in liver cirrhotic rats. Methods The SD rats were intraperitoneally injected with carbon tetrachloride (CCl4) twice a week for 8 weeks to induce liver cirrhosis. The rats were randomly divided into three groups: normal control group (n= 10), model control group (n= 15) and rofecoxib treated group (received 10 mg/kg of rofecoxib daily, n = 15). Liver histopathology was examined by light microscopy, and sinusoidal ultrastructure was observed by transmission electron microscopy. Furthermore, the level of basement membrane proteins (collagen type Ⅳ, laminin) and their localizations in liver were determined by Western blotting and immunohistochemistry, respectively, and the microvessel density was detected following vWF (yon Willebrand factor) immunolabeling on liver tissue sections. Results Fibrotic areas were reduced in rofecoxib treated group compared with that in model group (30.7±8.9 vs 23.5±6.5,P<0. 05). The light and electron microscopy showed that the pathologic changes including loss or reduction in number of sinusoidal endothelial fenestrate, the accumulation of extracellular matrix in the Disse's space and development of subendothelial basal lamina (basement membrane formation) were more severe in model group than those in rofecoxib treated group. Compared with model group, administration of rofecoxib resulted in significant decrease in microvessel density (11.3 ± 1.6 vs. 6.4 ±0. 7, P<0. 01). Rofecoxib could significantly decrease the expression of type Ⅳ collagen and laminin at protein levels (3.0±0.5 and 3.0±0.5, respectively) when compared with model group (3.8±0.4 and 3.7±0. 5, respectively). Conclusion The results indicate that early administration of rofecoxib may reduce sinusoidal capillarization.

ABSTRACT

Objective To study the expression of eyelooxygenase-2(COX-2)in invasive duetal carcinoma of breast and its relationship with cell prolireration and angiogenesis. Methotis Expression of COX-2 and Ki-67 was detected in 52 cages of breast cancer tissue by immunohistochemistry. CD34 marking vascular endothelial cell was used to measure the miemvascular density(MVD).Results The positive COX-2 expression was 57.7%in invasive duetal carcinoma of breast. The Ki-67 index was (56.07±17.37)%in COX-2 positive group while(27.32±16.28)%in the negative group, and the MVD W88 32.46±12.59 in COX-2 positive group while 25.04±10.48 in the negative group. The positive group of COX-2 protein had higher Ki-67 and MVD than that in negative group(P<0.05).Conclusion The overexpression of COX-2 protein Were found in invasive ductal carcinoma of breast. To stimulate cell prolireration and angiogenesis may be one of the pathways of COX-2 protein to contribute to the invasion and metastasis in human breast cancer.

ABSTRACT

Objective To investigate photo-protective effect of NS398 on above cells damaged from UV irradiation. Methods HaCaT cell and HaCaT keratinocytes were incubated in the culture medium supplemented with different concentration of NS398 for 2h before diffierent dosages of UV irradiation.MTT assay was used to detect cell proliferation and cellular activity:Fluorospectrophotometer assay was used to measure the change of ROS level in the cells which were UV-mediated.Resuits After irradiation with UV,proliferation and cellular activity of HaCaT cells were decreased(P<0.05);NS398 reduced the generation of ROS in cells induced by UV irradiation apparently and the cell apoptotic rate was dependent on the concentration of NS398. Conclusion NS398 might play an important role in the treatment of UV irradiation injury by decreasing the generation 0f ROS in cells after UV irradiation and increasing the survival ability of HaCaT cells.
