ABSTRACT

An intravascular ultrasound-enhanced thrombolysis excitation system with adjustable frequency, amplitude and duty cycle was designed based on FPGA (ZYNQ-7Z020). Firstly, the FPGA generated waveform amplitude binary data based on direct digital frequency synthesis (DDS) technology, and then the data was converted into burst signal through an external daughter card, which included D/A conversion circuit, active low-pass filter, power amplifier circuit and impedance matching circuit. The test results demonstrated that the output waveform reached the target with advantages of simple implementation and flexible control, the peak negative pressure generated from ultrasound transducer was doubled by means of an electrical impedance matching network. In vitro thrombus models were applied to verify the excitation system, it turned out that ultrasound cavitation effect generated could accelerate the penetration of urokinase and increase the thrombolysis rate by about 20%.

ABSTRACT

Objective To analyze the activity of diadenylate cyclase ( DAC) encoded by LA3304 gene of Leptospira interrogans ( L. interrogans) and to investigate the influence of CdaR encoded by LA3303 gene on DAC activity. Methods The LA3304 gene in L. interrogans serogroup Icterohaemorrhagiae serovar Lai strain Lai was amplified by PCR and inserted into a prokaryotic expression system for expressing DAC. The expressed recombinant protein, rDAC, was purified by Ni-NTA affinity chromatography. High Perform-ance Liquid Chromatography ( HPLC) was used to measure the synthesis of c-di-AMP from ATP by rDAC in vitro. Bacterial two-hybrid analysis was used to detect the interaction between CdaR and DAC. Prokaryotic co-expression system was constructed and used in combination with HPLC to analyze the role of CdaR in acti-vating DAC. Results The constructed prokaryotic expression system for LA3304 gene of L. interrogans strain Lai could highly express the rDAC upon the induction of IPTG. The purified rDAC showed high purity with a single protein band in gel as indicated by SDS-PAGE. rDAC could synthesize c-di-AMP from ATP in vitro. CdaR interacted with DAC and enhanced the activity of DAC (P<0. 05). Conclusion DAC encoded by LA3304 gene was a diadenylate cyclase that could convert ATP into c-di-AMP. CdaR promoted the acti-vation of DAC and formed a CdaR-DAC system with DAC. The system was involved in the synthesis of c-di-AMP in L. interrogans.

Brazil, Russia, India, China and South Africa are the five BRIC countries which have showed a rapid growth in economic development in recent years especially in international health assistance area, which has gradually become a force that cannot be ignored. This article compares and analyzes the current status and character-istics of the BRIC countries in the field of international health assistance and compares them with the Countries of the Economic Cooperation and Development Organization's Development Assistance Committee ( ECDO-DAC) . It will also summarize the new features, advantages and disadvantages of the health development assistance exerted by the BRICS countries;and ultimately provide suggestions for the future international health development aid policy of China.

ABSTRACT

Objective To study the effects of deoxycytidine (5-aza-2 deoxycytidine, DAC) on DNA Methylation state and expression of mRNA and protein of pl6 gene in human squamous lingual carcinoma SCC-9 cells in vitro. Methods The SCC-9 cells were divided into four groups, group 0, 1, 2 and 3 which processed using three gradients concentration of DAC. The group 0 without DAC was as the control group. Q-MSP was used to detect the state of methylation of the p16 in SCC-9 cells treated by DAC after 48 hours. Real-time fluorescence quantitative PCR was used to detect mRNA expression level changing of the p16 in SCC-9 cells treated by DAC. Immunohistochemical method was used to detect the expression of p16 protein. Results The hypermethylation and non-methylated p16 gene in SCC-9 was mixed with the results of Q-MSP. The results of Real-time PCR showed that mRNA expression of p16 in SCC-9 cells which treated by the different concentration of DAC for 48 hours was higher the control group. And difference was statistically significant (P < 0.05). The high expression of p16 protein was found in the experimental group with immunohistochemical method. Conclusion The p16 gene methylation states of SCC-9 may be suppresses and the recovery of mRNA and protein expression of p16 gene must be prompted by DAC.

ABSTRACT

Tobacco streak virus (TSV) is an important emerging virus belongs to the genus Ilarvirus and family Bromovidae. The Onion crop is infected by Tobacco streak ilar virus and it is major problem in different places of Andhra Pradesh in South India and also transmitted by thrips vector. TSV suspecting onion samples were identified by direct antigen coating enzyme linked immunosorbent assay using TSV polyclonal antiserum. The Replicase gene from each isolate was amplified using TSV replicase gene specific primers by using the RT-PCR. The ~530 bp product was amplified, cloned, sequenced and determined its length as 534 nucleotides and codes for 178 amino acids. The partial sequence of TSV-Rep shared identity of 87.6 -99.8% at nucleotide levels and 67.8- 99.4% at amino acid levels respectively with other reported TSV isolates. The phylogenetic tree relationship based on the nucleotide sequence of present study isolate (AP-Onion-Chittoor) from different geographical regions was also analyzed in this study.

OBJECTIVE@#To explore effect of 5-AZn-2 '-deoxycytidine on proliferation of human lung adenocarcinoma cell line A549 in vitro.@*METHODS@#Superoxide dismutase (SOD) activity was measured by hydroxylamine colorimetric method. Inhibition effect of 5-AZn-2' deoxycytidylic acid at different concentration and different time on growth of A549 cell was determined by MTT assay. Methylene dioxyamphetamine (MDA) was measured by thiobarbituric acid colorimetric method. Effect of 5-AZn-2' deoxycytidylic acid on apoptosis of A549 cell was determined by Hoechst 33258 dyeing detection.@*RESULTS@#5-AZn-2' deoxycytidylic acid had significant inhibition effect on proliferation of A549 cells in vitro, and the inhibition was notably dependent on time and dosage during 48-72 h; SOD level was significantly lower than those of control group (P<0.05, P<0.01), MDA level was significantly higher than those in the control group (P<0.05, P<0.01). A549 cells began to be in apoptosis after using 5-AZn-2'deoxycytidylic acid.@*CONCLUSIONS@#5- AZn-2' deoxycytidylic acid has significant inhibition effect on growth of A549 cell, and can lead the change of lipid peroxidation. It indicates that the mechanism has relationship with A549 cell cycle tissue and induction factor of apoptosis.

ABSTRACT

Introdução: No Brasil, aproximadamente, um terço da população adulta é fumantes ativos, sendo 16,7 milhões de homens e 11,2 milhões de mulheres. Porém, não existem dados específicos referentes à população com idade maior do que 65 anos. O objetivo deste estudo foi verificar a prevalências de doenças tabaco-relacionadas em indivíduos idosos fumantes com história tabagística maior do que 20 anos/maço. Métodos: Estudo longitudinal no qual foram avaliados 240 idosos fumantes antendidos pelo PSF-SP, todos os indivíduos responderam dois questionários, sendo um para obtenção de dados referentes ao estado de saúde e outro para avaliação do grau de dependência à nicotina, todos realizaram espirometria. Resultados: O cigarro industrial foi mais consumido do que cachimbo e cigarro de palha (p= 0,0004). Houve maior número de indivíduos com grau de dependência leve e moderada do que importante (p= 0,014 e p= 0,038), respectivamente. Dos indivíduos, 57 já tinham realizado exame de espirometria anteriormente. Entre os avaliados, 74 indivíduos tinham a relação VEF1/CVF menor do que 07 e menor do que 90% do previsto 228 indivíduos sabiam que tinham doenças relacionadas ao hábito do tabagismo, previamente diagnosticadas pelo seu médico, sendo a maior prevalência para a hipertensão arterial sistêmica (45,9%), seguida de doenças vasculares e pulmonares (27,1%), doenças cardíacas (25,6%), câncer (17,5%) e outros tipos de doenças (29,7%).

ABSTRACT

A relação entre transferência de lipídeos, idade e aterogênese são complexas e ainda não estão claras. É possível que a troca de lipídeos esteja alterada com a avançar da idade e relacionada com a Doença Arterial Coronariana (DAC). O objetivo deste trabalho foi verificar a hipótese se em indivíduos mais jovens a habilidade da HDL de receber lipídeos é diferente de indivíduos mais velhos com e sem a evidência clínica da DAC. Dentro desses aspectos, foram determinados o diâmetro da partícula desta lipoproteína, a atividade da Paraoxonase (PON1) e sua capacidade de receber lipídeos. Para tanto, foram estudados 86 indivíduos divididos em quatro grupos: adulto jovem (25±4), meia-idade (42±8), idosos sem evidência clínica de DAC (75±6) e idosos com DAC (74±5). Uma nanoemulsão artificial rica em colesterol (LDE) marcada com 3H-TG e 14C-CL ou 3H-CE e 14C-FL foi incubada com plasma. Após a precipitação de outras lipoproteínas, o sobrenadante contendo HDL foi separado e em seguida, medida a radioatividade. O diâmetro da HDL foi medido por laser scattering (nm). Foram constatadas diferenças significativas entre as taxas de transferência de 3H-éster de colesterol (CE) entre os grupos: adulto jovem (3.7±1.0%); meia idade (4.1 ±0.7%) e idosos (5.3±1.8%);p= 0.024. Também ocorreu diferença entre as taxas de transferência do 14C-fosfolipídeo (FL): adulto jovem (18.7±4.6%), meia idade (18.3 ±4.0%) e idosos (20.6±5.3); p=0.0368. Com relação ao tamanho das partículas de HDL, foi encontrada diferença entre os grupos: os grupos adulto jovem (8.9± 0.3nm) e meia idade (8.9± 0.3nm) apresentaram menores diâmetros de HDL quando comparados ao do grupo de idosos sem evidência clínica da DAC (9.7± 1.6);p= 0,0444. As transferências de lipídeos foram expressas em % de radioatividade. A idade correlacionou-se positivamente com a taxa de transferência do 3H- éster de colesterol (r=0.3365; p=0.0036), com a concentração de colesterol total (r=0.4965; p=0.0001) e com a concentração de HDL colesterol (r=0.3559; p=0.0023). Também houve correlação positiva com o tamanho de HDL (r=0.3695; p=0.0013). Em princípio, os indivíduos idosos sem evidência clínica da DAC, aparentemente têm alguma proteção contra a mesma. Desse modo, com o intuito de saber se os resultados encontrados no presente trabalho sustentam a afirmação acima, foi realizada a comparação desse grupo com um grupo de idosos que apresentavam a DAC. O grupo com DAC apresentou menor tamanho de partícula de HDL (8,7±0,7). As taxas de transferência de 3H-CE e de 14C-FL também foram menores neste grupo (3H-CE=3,1 ±2,3 e 3H-TG= 5,1 ±1 ,6). Devido ao importante papel antiaterogênico da HDL, esses resultados podem ser relevantes para estabelecer novos mecanismos existentes entre os aspectos qualitativos dessa lipoproteína, o avanço da idade e a presença da DAC


The relationship between transfer of lipids, age and atherogenesis are complex and yet unclear and is possible that the shift of lipids to HDL may be altered by the aging process and related with coronary artery disease (CAD). We tested the hypothesis whether in younger patients the ability of HDL to receive lipids is different from that of elderly patients with or without CAD. Inside of these aspects, the HDL size, the activity of Paraoxonase (PON1) and its capacity to receive lipids was determined. It was studied, 25 younger, 25 middle age, 36 elderly patients with a coronariography and/or a perfusion scintilography on the last 6 months (11 with CAD, 74±5 yo; and 25 patients without proved CAD, 75±6 yo). An artificial cholesterol-rich nanoemulsion labeled with 3H-TG and 14C-FC or H-CE and 14C-PL was incubated, per 1 hour, with plasma. After chemical precipitation of apoB-containing lipoproteins and the nanoemulsion, the supernatant containing HDL was counted for radioactivity. The HDL diameter was measured by laser-light-scattering. Transfer of CE and PL to HDL was smaller in young patients than in the elderly patients without CAD, but the transfer of the other lipids are equal (CE: young= 3.7±1.0%; middle age= 4.1 ±0.7%; elderly without CAD= 5.3±1.8%; p= 0.024 and PL: young= 18.7±4.6%; middle age= 18.3 ±4.0%; elderly without CAD= 20.6±5.3; p=0.0368). The HDL size was greater in elderly group without CAD (9.7± 1.6nm) than in younger (8.9± 0.3nm) and middle age patients (8.9± 0.3nm); p=0,0444. Transfer of lipids to HDL was expressed as % of total incubated radioactivity. The age positively correlated with the transfer of CE (r=0.3365; p=0.0036), with the total cholesterol concentration (r=0.4965; p=0.0001) and with the HDL concentration cholesterol (r=0.3559; p=0.0023). Also had positive correlation with the size of HDL (; p=0.0013). In principle, the aged patients without CAD, have some protection against the same one. In this aspect, with intention to know if the results found in the present work support the affirmation above, was compared this group with a group of aged that presented the CAD. Comparing elderly patients without CAD with elderly patients with CAD, the transfer of CE and FL as well as HDL size was smaller in the CAD group (CE=3.1±2.3 and TG= 5.1±1.6; 8.7±0.7nm). Due to HDL important antiatherogenic roles, this result can be relevant to establish new mechanisms and risk factors in aging and in CAD

ABSTRACT

O Modelo de Avaliação e Orientação de Carreira proposto por Donald Super (C-DAC Model) traduz a sua teoria do desenvolvimento de carreira em termos práticos. Após décadas de pesquisas teóricas, Donald Super criou um modelo de avaliação e orientação que utiliza os principais conceitos de sua teoria, tais como, a maturidade de carreira, saliência dos papéis e autoconceitos, contribuindo para que os orientadores profissionais possam auxiliar as pessoas a resolverem os seus problemas de carreira de forma integradora. Este modelo auxilia na compreensão dos determinantes objetivos e subjetivos do desenvolvimento de carreira, superando o modelo tradicional de orientação de carreira que se baseia fundamentalmente nos modelos de combinação (matching models). O presente artigo tem como objetivo descrever as características e as etapas deste modelo de avaliação e orientação de carreira, além de discutir as suas implicações práticas para os orientadores de carreira.


The Career Developmental Assessment and Counseling model proposed by Donald Super (C-DAC Model) reflects his career development theory in practice. After decades of theoretical research he developed important concepts such as: career maturity, role salience in work and self concept. Donald Super created a new career assessment and counseling model that is based on the main concepts of his theory so helping career practitioners and counselors to guide their clients in dealing with their career problems in an integrating way. This model helps to understand the objective and subjective determinants of career development, reaching beyond the traditional models that are mainly based on matching models. This paper aimed at describing and discussing the principal characteristics and procedures proposed by Super´s Model and its practical implications for career counseling.


El modelo de evaluación y orientación de Carrera propuesto por Donald Super (C-DAC Model) pone en términos prácticos su teoría de desarrollo de carrera. Después de décadas de investigaciones teóricas Donald Super creó un modelo de evaluación y orientación que utiliza los principales conceptos de su teoría, tales como madurez de carrera, destaque de los papeles y autoconceptos, contribuyendo para que los orientadores profesionales puedan auxiliar a las personas a resolver sus problemas de carrera de forma integradora. Este modelo facilita la comprensión de los determinantes objetivos y subjetivos del desarrollo de carrera, superando al modelo tradicional de orientación de carrera que se basa fundamentalmente en los modelos de combinación (matching models). Este trabajo se propone describir las características y las etapas de este modelo de evaluación y orientación de carrera, además de discutir sus implicaciones prácticas para los orientadores de carrera.

ABSTRACT

The interaction of the oligopeptides Ala-Gln-GIn-Leu-Ala-Gly-OH and Gln-Leu- Ala-Gly-OMe corresponding, respectively, to the sequence 53–58 and 55–58 of lac repressor protein with four polynucleotides was studied. The two peptides did not interact with poly dA. poly dT, poly d(A-T).poly d(A-T) or poly d(A-G).poly d(C-T). But they interacted in a characteristic way with poly d(A-C). poly d (G-T), the sequences of which are in abundance in the lac operator region. Both the peptides stabilised the melting of poly d (A-C). poly d (G-T) at a peptide to nucleotide ratio (P/N) of 4; at lower ratios, they destabilised the DNA slightly. The circular dichroism of the alternating polynucleotide with CAC/GTG sequences was perturbed by both the oligopeptides. The hexapeptide at a P/N of 4 caused the transformation of the Bform circular dichroism spectrum to a new state, characterised by strong 220 and 240 nm bands, and a rather weak long wavelength spectrum.
