ABSTRACT

AIM: To evaluate the imaging characteristics of dense automatic real time B-scan optical coherence tomography angiography(DART-OCTA)in macular-involved branch retinal vein occlusion(BRVO)and the diagnostic value of capillary perfusion imaging in the macular area.METHODS: From June 2020 to December 2020, there were 51 cases of 51 eyes with BRVO diagnosed in Eye Hospital, Wenzhou Medical University. Imaging characteristics of the BRVO macular area were observed by fluorescein angiography(FA), optical coherence tomography angiography(OCTA)and DART-OCTA examination, respectively. According to the retinal capillary perfusion status, the included patients were divided into capillary and non-imaging groups, comparing the results of capillary perfusion imaging in the BRVO macular area among the three examination methods.Furthermore, quantitative analysis of capillary perfusion density in the lesion involved area and the lesion non-involved area was performed in DART-OCTA images. RESULTS: Patients with 51 eyes were included in this study, FA identified 10 eyes of capillary perfusion imaging, OCTA identified 14 eyes of capillary perfusion imaging, DART-OCTA identified 34 eyes of the capillary perfusion imaging.Comparison of the three test methods for capillary perfusion imaging findings in the BRVO macular area showed that DART-OCTA was more sensitive compared to FA and OCTA for capillary perfusion imaging in the ischemic area. In DART-OCTA examination, retinal capillary blood flow density was lower in the lesion-involved areas in both the capillary perfusion imaging group and the non-imaging groups(0.65±0.20/mm vs 1.16±0.31/mm,0.41±0.16/mm vs 1.06±0.38/mm, all P<0.0001).CONCLUSION: DART-OCTA can provide clearer tomographic imaging of retinal capillary perfusion. And the imaging with its observation of BRVO involving the macular area is least affected by macular hemorrhage and it is an important complementary method for BRVO patients with significant retinal hemorrhage.

Neste texto, traço a relação do tipo de interpretação da obra de arte proposto por J. A. Frayze-Pereira com o que se passa numa sessão de análise. Na sessão, a "obra" do paciente é a narração de seus conteúdos mentais em busca de uma forma simbólica. Nesse processo, sugiro que o que é evocado no analista, que se transforma numa interpretação ou comentário, é central na constituição de seu significado e parte dele. Daí considerarmos a transferência uma poiesis.


In this text I outline the relationship between the type of interpretation of the work of art proposed by the author, and what happens in an psychoanalytic session. In analytic sessions the patient's "work" (oeuvre) is his narrative of his emotional experience in search of a symbolic form. In this process I suggest that what is evoked in the analyst, which becomes an interpretation or comment, is the core to the formation of its meaning and part of it. Hence, we consider the transference as a process of poiesis.


En este texto trazo la relación entre el tipo de interpretación de la obra de arte, propuesta por el autor, con lo que sucede en una sesión de análisis. En la sesión, el "trabajo" del paciente es la narración de sus contenidos mentales en busca de una forma simbólica. En este proceso sugiero que lo evocado en el analista, que se convierte en interpretación o comentario, es central para la constitución de su significado y parte de él. Por tanto, consideramos la transferencia como una poiesis.


Dans ce texte, je trace la relation entre le type d'interprétation de l'Œuvre d'art, proposé par l'auteur, avec ce qui se passe lors d'une séance d'analyse. Dans la séance, le « travail ¼ du patient est la narration de son contenu mental à la recherche d>une forme symbolique. Dans ce processus, je suggère que ce qui est évoqué chez l>analyste, qui devient une interprétation ou un commentaire, est au cŒur de la constitution de son sens et d>une partie de celuici. Nous considérons donc le transfert comme une poïétique - poiesis en grec.

ABSTRACT

Both cowpea and yard-long bean belong to Vigna unguiculata ssp. unguiculata but have diverged through human induced evolution in sub-Saharan Africa and Asia, respectively. To map the quantitative trait loci (QTLs) for yield associated traits and derive new lines that may combine the attributes of both types, we developed a F2:3 mapping population derived from a cross between cowpea line TVu2185 and yard-long bean line TVu6642. Using DArT markers, a total of 30 QTLs accounting for 1.8–13.0% phenotypic variation was detected for pod and seed traits. Some novel major QTLs for peduncle number per plant (qPeN2.2), pod length (qPoL3), seed breadth (qSB4), length (qSL7.2) and thickness (qST9) identified on chromosomes 2, 3, 4, 7 and 9, respectively, are particularly interesting and need to be validated. Moreover, we confirmed previously reported QTLs for pod length (qPoL8) and 100-seed weight (qSW8) on chromosome 8 and for seed number per pod (qSN9.2) on chromosome 9 suggesting usefulness for marker-assisted-selection purpose. Notably, some QTLs for these traits were clustered especially on chromosomes 5, 7, 8, 9 and 10 indicating the presence of the same QTL or linked loci in these regions. Moreover, the involvement of epistasis was observed for trait expressions, but compared with the main effect QTLs, the phenotypic effects of epistatic-QTLs detected were much less. The present QTL analysis may provide a useful tool for breeders to formulate efficient breeding strategy for introgression of the desirable alleles for yield related traits in cowpea using molecular markers.

ABSTRACT

ABSTRACT This experiment investigated the effect of number of visual demonstrations on the acquisition of discrete motor skill. Seventy-four college students of both sex participated in this study. The participants observed a visual model twice or ten times. In the acquisition phase, the participants were asked to throw a dart with postero-anterior movement into a circle target until reach a learning criterion. In transfer tests no difference between groups was found in the analysis of score. However, ten demonstrations were more effective than twice as regards performance of the correct pattern of movement and the number of subjects who reached the performance criterion. The results indicate a similar effect of the both number of demonstrations in the performance and dart throwing learning and both conditions influence subject's capacity to detect relevant information to perform the task.


RESUMO Este experimento investigou os efeitos do número de demonstrações na aquisição de uma habilidade motora discreta. Setenta e quatro universitários, de ambos os sexos, participaram desse estudo. Os participantes observaram um modelo duas ou dez vezes. Na fase de aquisição, os participantes foram requisitados a lançar um dardo por meio de um movimento antero-posterior em um alvo circular até atingir um desempenho critério. Não foram identificadas diferenças na aprendizagem entre os grupos na análise do escore. Contudo, dez demonstrações estimularam melhor do que duas demonstrações no que diz respeito ao desempenho do padrão correto e ao número de sujeitos que atingiram o desempenho critério. Os resultados indicam um efeito similar de ambos os números de demonstrações no desempenho e aprenizagem do arremesso de dardo de salão e que ambas as condições estimularam a capacidade dos aprendizes de detectarem informações relevantes para desempenhar a tarefa.

BACKGROUND: The restriction of wrist motion results in limited hand function, and the evaluation of the range of wrist motion is related to the evaluation of wrist function. To analyze and compare the wrist motion during four selected tasks, we developed a new desktop motion analysis system using the motion controller for a home video game console. METHODS: Eighteen healthy, right-handed subjects performed 15 trials of selective tasks (dart throwing, hammering, circumduction, and winding thread on a reel) with both wrists. The signals of light-emitting diode markers attached to the hand and forearm were detected by the optic receptor in the motion controller. We compared the results between both wrists and between motions with similar motion paths. RESULTS: The parameters (range of motion, offset, coupling, and orientations of the oblique plane) for wrist motion were not significantly different between both wrists, except for radioulnar deviation for hammering and the orientation for thread winding. In each wrist, the ranges for hammering were larger than those for dart throwing. The offsets and the orientations of the oblique plane were not significantly different between circumduction and thread winding. CONCLUSIONS: The results for the parameters of dart throwing, hammering, and circumduction of our motion analysis system using the motion controller were considerably similar to those of the previous studies with three-dimensional reconstruction with computed tomography, electrogoniometer, and motion capture system. Therefore, our system may be a cost-effective and simple method for wrist motion analysis.

ABSTRACT

ABSTRACT: Understanding genetic variability in existing wheat accessions is critical for collection, conservation and use of wheat germplasms. In this study, 138 Chinese southwest wheat accessions were investigated by genotyping using two resistance gene makers (Pm21 and Yr26) and DArT-seq technique. Finally, about 50% cultivars (lines) amplified the specific allele for the Yr26 gene (Gwm11) and 40.6% for the Pm21 gene (SCAR1265). By DArT-seq analysis, 30,485 markers (6486 SNPs and 23999 DArTs) were obtained with mean polymorphic information content (PIC) value 0.33 and 0.28 for DArT and SNP marker, respectively. The mean Dice genetic similarity coefficient (GS) was 0.72. Two consistent groups of wheat varieties were identified using principal coordinate analysis (PCoA) at the level of both the chromosome 6AS and the whole-genome, respectively. Group I was composed of non-6VS/6AL translocation lines of different origins, while Group II was composed of 6VS/6AL translocation (T6VS/6AL) lines, most of which carried the Yr26 and Pm21 genes and originated from Guizhou. Besides, a model-based population structure analysis revealed extensive admixture and further divided these wheat accessions into six subgroups (SG1, SG2, SG3, SG4, SG5 and SG6), based on their origin, pedigree or disease resistance. This information is useful for wheat breeding in southwestern China and association mapping for disease resistance using these wheat germplasms in future.


RESUMO: O conhecimento da estrutura da população é essencial para o mapeamento de associação de resistência a doenças para a população de trigo. Neste estudo, a técnica de DART-seq™ foi usada para genotipar o genoma inteiro de cultivares de trigo. Finalmente, 30,485 marcadores (6486 SNPs e 23999 dardos) foram obtidos, e dois grupos de variedades de trigo foram identificados por meio de análise principal-coordenadas (PCoA) do nível de todo o genoma e o nível 6AS cromossomo. O grupo I foi composto por linhas não T6VS/6Al de diferentes origens, enquanto o Grupo II foi composto de linhas T6VS/6Al, sendo que da maioria destes realizados os genes Yr26 e PM21 originários de Guizhou.

In the evergreen forest of South America, as in many other scientific field expeditions, everything may suddenly go wrong, and nothing can prepare researchers to accidents that may occur in a succession of uncontrollable errors once the first mistake is done. This is what happened during an expedition in search for dendrobates by an experienced forest guide and naturalist. The authors decided to report the story, considering that it deserved to be brought to the attention of those interested in venomous animals and toxins, in order to illustrate the potential danger of dealing with these organisms.

ABSTRACT

A novel method for the simultaneous determination of 3-nitrotyrosine (NT) and 3-chlorotyrosine (CT) in human plasma has been developed based on direct analysis in real time-tandem mass spectrometry (DART-MS/MS). Analysis was performed in the positive ionization mode using multiple reaction monitoring (MRM) of the ion transitions at m/z 216.2/170.1 for CT, m/z 227.2/181.1 for NT and m/z 230.2/184.2 for the internal standard, d (3)-NT. The assay was linear in the ranges 0.5-100 μg/mL for CT and 4-100 μg/mL for NT with corresponding limits of detection of 0.2 and 2 μg/mL. Intra- and inter-day precisions and accuracies were respectively <15% and ±15%. Matrix effects were also evaluated. The method is potentially useful for high throughput analysis although sensitivity needs to be improved before it can be applied in clinical research.

ABSTRACT

Berberis petiolaris Wall. ex G. Don, an unexplored medicinal plant belonging to the family Berberidaceae, is a large deciduous shrub found in Western Himalaya between 1800-3000 m. Chemical profiling of fruit, leaf, root and stem was done by direct analysis in real time mass spectrometry followed by multivariate analysis for discrimination among the plant parts. The bioactive compounds, including magnoflorine, berberine, jatrorrhizine, thalifendine/berberrubine, demethyleneberberine, reticuline, 8-oxoberberine, N-methyltetrahydroberberine, tetrahydropalmatine, tetrahydroberberine and palmatine, were identified by their exact mass measurement and the corresponding molecular formula of each compound. A comparative study of distribution pattern for all these bioactive alkaloids showed qualitative and quantitative variations in different parts of B. petiolaris. Principal component analysis clearly dis-criminated each part of B. petiolaris plant.

ABSTRACT

A simple modification converts an electrospray ion source to an ambient-pressure helium plasma ionization source without the need of additional expensive hardware. Peaks for active ingredients were observed in the spectra recorded from intact pharmaceutical tablets placed in this source. A flow of heated nitrogen was used to thermally desorb analytes to gas phase. The desorption temperatures were sometimes as low as 50 1C. For example, negative-ion spectra recorded from an aspirin tablet showed peaks at m/z 137 (salicylate anion) and 179 (acetylsalicylate anion) which were absent in the background spectra. The overall ion intensity increased as the desorption gas temperature was elevated. Within the same acquisition experiment, both positive- and negative-ion signals for acetaminophen were recorded from volatiles emanating from Tylenol tablets by switching the polarity of the capillary back and forth. Moreover, different preparations of acetaminophen tablets could be distinguished by their ion-intensity thermograms.

ABSTRACT

Ultraviolet (UV) radiation is a major environmental factor that leads to acute and chronic reactions in the human skin. UV exposure induces wrinkle formation, DNA damage, and generation of reactive oxygen species (ROS). Most mechanistic studies of skin physiology and pharmacology related with UV-irradiated skin have focused on proteins and their related gene expression or single- targeted small molecules. The present study identified and analyzed the alteration of skin metabolites following UVB irradiation and topical retinyl palmitate (RP, 5%) treatment in hairless mice using direct analysis in real time (DART) time-of-flight mass spectrometry (TOF-MS) with multivariate analysis. Under the negative ion mode, the DART ion source successfully ionized various fatty acids including palmitoleic and linolenic acid. From DART-TOF-MS fingerprints measured in positive mode, the prominent dehydrated ion peak (m/z: 369, M+H-H2O) of cholesterol was characterized in all three groups. In positive mode, the discrimination among three groups was much clearer than that in negative mode by using multivariate analysis of orthogonal partial-least squares-discriminant analysis (OPLS-DA). DART-TOF-MS can ionize various small organic molecules in living tissues and is an efficient alternative analytical tool for acquiring full chemical fingerprints from living tissues without requiring sample preparation. DART-MS measurement of skin tissue with multivariate analysis proved to be a powerful method to discriminate between experimental groups and to find biomarkers for various experiment models in skin dermatological research.

ABSTRACT

The genus Stylogaster Macquart (Diptera, Conopidae) is represented by 14 species in the Afrotropical region, five of them endemic to Madagascar and two recorded from South Africa, one of them endemic. Fifteen specimens of Muscidae, 14 from Madagascar and one from South Africa, belonging to 10 species, impaled with Stylogaster eggs were examined. Among them, three new muscid host species were found. The dissected eggs were all similar, suggesting the presence of the same species in both localities, what is not recorded until now. The available data on the dipteran Stylogaster eggs hosts in Africa is summarized. Color photos of the material examined are presented.


O gênero Stylogaster Macquart (Diptera, Conopidae) é representado por 14 espécies na região Afrotropical, cinco delas endêmicas para Madagascar e duas assinaladas na África do Sul, uma delas endêmica. Quinze exemplares de Muscidae, 14 de Madagascar e um da África do Sul, pertencentes a 10 espécies, empalados com ovos de Stylogaster foram examinados. Entre eles, três novas espécies hospedeiras de muscidae foram encontradas. Os ovos dissecados eram todos similares, sugerindo a presença da mesma espécie nas duas localidades, o que ainda não está registrado. As informações disponíveis sobre hospedeiros dípteros de ovos de Stylogaster na África são resumidas. Fotos coloridas do material examinado são apresentadas.

ABSTRACT

The green and black poison-dart frog Dendrobates auratus exhibits high intraspecific variation in hue color and pattern throughout its range, making it a very popular species in the pet trade. We analyzed the correspondence between color variation and molecular variation of D. auratus from Costa Rica using RAPD analysis. Twenty-six random primers were analyzed for variation in 99 individuals from seven populations. Color pattern was scored from digital images of the dorsal and ventral views. In general, frogs from the Caribbean coast had significantly more light coloration than black color but cannot be grouped by population based only on hue pattern. Only 3 RAPD primers were found to be polymorphic, representing a total of 16 loci. Most of the molecular variation encountered here occurs within populations, thus making unclear the degree of population structure and differentiation. Further examination of COI mtDNA sequences from our samples also supports these results. Partial Mantel correlations suggested that the pattern of molecular variation is not congruent with the variation in color pattern in this species, an outcome that is discussed in terms of phenotypic evolution. La rana venenosa Dendrobates auratus posee una gran variación intraespecífica en tonos y patrones de coloración a lo largo de toda su distribución, lo que la hace una especie muy reconocible entre las especies de dendrobátidos. Analizamos la correspondencia entre variación de coloración y variación molecular de D. auratus de Costa Rica empleando análisis de RAPDs. La variación resultante en veintiséis "primers" aleatorios fue analizada en 93 individuos de siete localidades en Costa Rica. El patrón de coloración fue evaluado de imágenes digitales del dorso y vientre para los mismos individuos. En general, las ranas provenientes de localidades en la costa Caribe tienen significativamente una coloración más clara, con menos proporción de color negro que las de localidades en la vertiente Pacífica, pero no pueden ser agrupadas por localidad basadas simplemente en el patrón de coloración. Solamente tres RAPD "primers" fueron encontrados polimórficos, representando un total de 16 loci. Mucha de la variación molecular encontrada habita dentro de poblaciones, lo que hace difícil determinar el grado de estructura poblacional y diferenciación. La reexaminación posterior de secuencias del gen mitocondrial CO1 también apoya estos resultados. Correlaciones parciales de matrices (test de Mantel) sugieren que el patrón de variación molecular no es congruente con la variación en el patrón de coloración en esta especie, un resultado que es discutido en términos de evolución fenotípica.

El estudio químico del extracto polar de hojas de Calliandra calothyrsus.Meissner (Mimosaceae) (accesión CIAT 22316) condujo a la identificación de dos flavonoides glicosilados: 3-O-a-L-ramnopiranosil- 7-metoxi-5,3’,4’-trihidroxiflavona y 3-O-a-L-ramnopiranosil-5,7, 3’,4’-tetrahidroxiflavona; sus estructuras fueron elucidadas por métodos espectroscópicos IR y RMN (1D y 2D). Adicionalmente, por espectrometría de masas por las técnicas DART y ESI de alta resolución, se identificaron derivados fenólicos mayoritarios, tales como ácido gálico y su dímero (PM 339,032), dihidroxicumarina, trihidroxicumarina, quercetina, dihidroquercetina, 3-O-a-Lramnopiranosil- 7-metoxi-5,3’,4’-trihidroxiflavona, 3-O-a-ramnopiranosil- 5,7,3’,4’-tetrahidroxiflavona y un dímero de flavonoide como principales constituyentes químicos.


Chemical investigation of the polar extract from Calliandra calothyrsus. Meissner (Mimosaceae) (accession CIAT 22316) leaves, afforded two known glycosylflavonoids: 3-O-a-L-rhamnopiranosyl-7-metoxi-5,3’,4’-trihydroxyflavone and 3-O-a-L-rhamnopiranosyl -5,7, 3’,4’-tetrahydroxyflavone; their chemical identities were established by IR and 1D and 2D-NMR experiments. Furthermore gallic acid, dihydroxycumarine, trihydroxycumarine, dihydroquercetin, 3-O-a-L-rhamnopiranosyl-7-methoxy-5, 3’,4’-trihydroxyflavone, 3-O-a- L-rhamnopiranosyl- 5,7,3’,4’-tetrahydroxy flavone, quercetin, a flavonol and a gallic acid dimmers were identified using high resolution mass spectrometry by DART and ESI methods.


Os estudos químicos do extrato polar das folhas de Calliandra calothyrsus conduzem à identificação de dois flavonóides dos glicosilados: 3-O-a-L-ramnopiranosil- 7-metoxi-5,3’,4’-triidroxiflavona e 3-O-a-L-ramnopiranosil-5,7,3’,4’-tetraidroxiflavona; suas estruturas foram elucidadas pelos métodos de espectroscopia de absorção na região de infravermelho (IV) e espectroscopia de ressonância magnética nuclear RMN (1D e 2D). Também, por espectrometria de massas pela técnica DART e ESI identificaram- se derivados fenólicos tais como: ácido gálico, diidroxicumarina, triidroxicumarina, 3-O-a-L-ramnopiranosil-7- metoxi-5,3’,4’-triidroxiflavona, 3-O-a- L-ramnopiranosil-5,7,3’,4’-tetraidroxiflavona, quercetina, dihidroquercetina, um dímero do flavonóide e um dímero do ácido gálico como principais constituintes químicos.
