ABSTRACT

Objective: To study lignans and terpenes of Moringa oleifera Lam. Methods: The compounds were isolated and purified by various column chromatographic techniques and High Performance Liquid Chromatography (HPLC). The structures of the compounds were identified through the combined analysis of physicochemical properties and spectroscopic evidence. The antineoplastic activity, α-glucosidase and acetylcholinesterase inhibitory activity of compounds were evaluated by MTT method, PNPG method and Ellman colorimetric method, respectively. Results: Twelve compounds were isolated from M. oleifera by various chromatographic methods and were identified as lariciresinol (1), 3-(α,4-dihydroxy-3-methoxybenzyl)-4-(4-hydroxy-3- methoxybenzyl) tetrahydrofuran (2), (7S,8R)-dihydrodehydrodiconiferyl alcohol (3), macadiol (4), diethyl pinoresinol (5), pinoresinol (6), evofolin B (7), erythro-guaiacylglycerol-β-O-4’-dihydroconiferyl alcohol (8), tricyclohumuladiol (9), 9α-hydroxy-2β-methoxyclovane (10), 3β-hydroxy-oleana-11,13 (18)-dien-28-oic acid (11), oleanolic acid (12). Compounds 12 showed antineoplastic activity. Compound 1 and 2 exhibited α-glucosidase inhibitory activity. Conclusion: Compounds 1-11 are separated from Moringa Adans for the first time. This plant has the potential of developing functional product for their antineoplastic and α-glucosidase inhibitory activity.

ABSTRACT

Objective To study the chemical constituents of the stems and leaves of Mappianthus iodoides. Methods The chemical constituents of M. iodoides were separated and purified by silica gel, ODS, Sephadex LH-20 gel column chromatographies, and preparative HPLC. Their structures were identified by physicochemical properties, spectroscopic analysis, as well as comparisons with the data reported in literature. Results Sixteen compounds were isolated from the 90% ethanol extract of the stems and leaves of M. iodoides, which were identified as 9-hydroxy-4,6-megastigmadien-3-one (1), 9-hydroxy-4,7-megastigmadien-3-one (2), blumenol A (3), 9,10-dihydroxy-4,7-megastigmadien-3-one (4), 5,12-epoxy-9-hydroxy-7-megastigmen-3-one (5), 5,12-epoxy-6,9-hydroxy-7- megastigmen-3-one (6), loliolide (7), lariciresinol (8), isolariciresinol (9), 5′-methoxylariciresinol (10), olivil (11), dehydrodiconiferyl alcohol (12), chushizisin I (13), 3,3-didemethoxyverrucosin (14), 4-epi-larreatricin (15), and glycerol monolinoleate (16). Conclusion All compounds are isolated from the genus Mappianthus for the first time.

ABSTRACT

Objective: To establish a quantitative analysis of multi-components by single mark (QAMS) for determining the content of eight components in Ganneng Dripping Pills (GDP) and its active pharmaceutical ingredients (APIs), including dehydrodiconiferyl alcohol, herpetotriol I, herpetolide A, herpetrione, herpetin, herpetetrone, herpetone, and herpetenone. Methods: Based on HPLC method, the relative correction factors (fk/m) were established with the herpetrione as the internal reference, and the content of other seven components was calculated. Moreover, the content of eight components in five batches of GDP and its APIs were determined by the external standard method (EMS), and the difference between the calculated value and the measured value were compared to verify the accuracy and feasibility of QAMS. Results: The fk/m value of dehydrodiconiferyl alcohol, herpetotriol I, herpetolide A, herpetin, herpetetrone, herpetone, and herpetenone with reference to herpetrione were 0.908, 0.728, 0.516, 0.720, 0.461, 0.623, and 0.410, respectively; And it had good reproducibility under different conditions. There were no significantly differences in the content of eight components between EMS and QAMS. Conclusion: Using GAPDH as an internal standard, the QAMS for determining the contents of eight components in GDP and raw material medicine was reliable and accurate and could be applied to control the quality of GDP.

ABSTRACT

Objective: To study the chemical constituents in the twigs and leaves of Ervatamia hainanensis. Methods: The chemical constituents were isolated by silica gel, ODS, Sephadex LH-20 column chromatographies, and preparative HPLC. The structures were elucidated on the basis of physicochemical properties and spectral analysis. Results: Four lignans were isolated and identified as threo-7-O-ethylguaiacylglycerol-8-O-4'-(coniferyl alcohol) ether (1), threo-guaiacylglycerol-8-O-4'-(coniferyl alcohol) ether (2), erythro-guaiacylglycerol-8-O-4'-(coniferyl alcohol) ether (3), and dehydrodiconiferyl alcohol (4). Conclusion: Compound 1 is a new 8-O-4'-neolignan and named as hainanervaligan A (1), and compounds 2-4 are obtained from the plants of Ervatamia (A. DC.) Stapf for the first time.

ABSTRACT

Objective: To investigate the chemical constituents from the stems of Clausena emarginata. Methods: The compounds were isolated by macroporous resin, silica gel, ODS column chromatography, Sephadex LH-20, reversed-phase MPLC, and then purified by preparative HPLC. Their structures were determined by the analysis of ultraviolet spectrum, mass spectrum, and NMR spectrum. Neuroprotective activities of compounds 11 and 12 were initially investigated. Results: Sixteen compounds were isolated from the petroleum ether and acetone fractions of 95% ethanol extract of the stems of Cl. emarginata, and their structures were identified as 1H-Indole-3-carboxaldehyde (1), E-N-benzoiltiramine (2), dehydrodiconiferyl alcohol (3), tortoside A (4), zhebeiresinol (5), evofolin B (6), 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucopyranoside (7), echipuroside A (8), 3-methylcarbazole (9), murrayafoline A (10), clausine Z (11), indizoline (12), clausenaline B (13), mafaicheenamine A (14), dictamnine (15), and honokiol (16). Conclusion: Compounds 1-8 are isolated from the plants of genus Clausena L. for the first time, compounds 1-16 are isolated from this plant for the first time. Compounds 11 and 12 show the neuroprotective activity against rotenone induced PC12 cell damage.

ABSTRACT

Objective: To investigate the chemical constituents of Daphne gracilis. Methods: The constituents were separated and purified by chromatographic methods and their structures were elucidated by spectroscopic methods and chemical analyses. Results: Fifteen compounds were obtained and identified as β-sitosterol (1), friedelin (2), stigmast-4-ene-3-one (3), stigmast-4-ene-3, 6-dione (4), lupeol (5), stigmasterol (6), 1, 3-distearate glycerol (7), glycerin-docosanate (8), octadecanol (9), phytol (10), dehydrodiconiferyl alcohol (11), epoxyconiferyl alcohol (12), oleodaphnone (13), 14-gingerol (14), and matairesinol (15). Conclusion: All the compounds are isolated from the plants of D. gracilis for the first time. Among them, compound 2-4, 7-9, 11, 12, and 14 are isolated from the plants in Daphne L. for the first time; Compounds 11, 12, and 14 are isolated from the plants of family Thymelaeaceae for the first time.
