ABSTRACT

Long noncoding RNAs (LncRNAs), a class of noncoding RNAs greater than 200 bases in length, are widely involved in the initiation, progression and glycolytic processes of many tumors, and can act as competitive endogenous RNA sponges to absorb miRNAs. LncRNAs can also inhibit miRNA expression, thereby regulate the glycolysis of tumor cells, affects cell proliferation, invasion and other biological activities. This review explores the roles of LncRNAs and glycolysis in digestive system tumors (DST), a representative group of malignant tumors. Extending the LncRNA role in the diagnosis, treatment and prognosis of other tumors, we conclude that LncRNAs have the potential to be new candidate genes for tumorigenesis and serve as tumor biomarkers, which provides new insight into morbidity and mortality decrease of DST and other tumors.

ABSTRACT

Objective To investigate the inhibition effects of "Phrul sByor Chem Mo (PBCM)" against different common digestive system tumor cell lines in vitro, including human gastric carcinoma cell line MGC803, colorectal carcinoma cell line LoVo, and hepatocellular carcinoma cell line HepG2. Methods The digestive system tumor cell lines in the logarithmic growth phase were incubated with different concentrations of PBCM for 48h. The cell viability was investigated by MTT assays, apoptosis was detected by Hoechst 33342 assays, and the level of Bcl-2 and Bax was estimated by ELISA tests. Results MTT assays proved that the proliferation of digestive system tumor cell lines was significantly inhibited by PBCM in vitro. Fluorescent staining demonstrated that PBCM was able to promote apoptosis of the cell lines. ELISA tests illustrated that the level of proapoptotic Bax was elevated, while the value of antiapoptotic Bcl-2 was decreased. Conclusion PBCM is able to inhibit proliferation and induce apoptosis of digestive system tumor cell lines in vitro. The mechanism may be related with Bcl-2 family proteins.

ABSTRACT

MicroRNA(miRNA) is a kind of small non-coding RNA which contents 18-25 nucleotide (nt). MiRNA plays an important role in human tumor of digestive system. Recently lots of research demonstrates that miRNA can act either as oncogenes or as tumor suppressor genes or sometimes as both. And many families of miRNAs'targets have been found. So the study of miRNA will supply a new way to the diagnosis and therapy of digestive diseases.

ABSTRACT

With the quick development revelant techniques, proteomics is a very active subject in modern biological fields. The key techniques of proteomics are two dimensional electrophoresis, mass spectrometry and bioinformatics. From the new point of view, proteomics may play a vital role in the early detection of neoplasm by looking for tumor specific protein and elucidating the relationship between the change of expression and different phases of tumorigenesis. The article briefly review the relevant technique, strategy and application for the early diagnosis of in digestive system tumors.
