ABSTRACT

The origin of motor neuronal cell bodies innervating heart in the cat was investigated using CTB (Cholera toxin B subunit) as neuronal tracer. The neural tracer was injected into subepicardial layer and myocardium of the right atrium, left atrium, right ventricle and left ventricle, respectively. Labeled parasympathetic motor neuronal cell bodies were found in DMV (dorsal motor nucleus of vagus nerve) and anterolateral part of NA (nucleus ambiguus) of brainstem. Heavier labeled neuronal cell bodies were found in the NA when the neural tracers were injected into left ventricle and DMV when injected into left atrium and left ventricle. These results may provide a neuroanatomical data on origin of motor neuronal cell bodies innervating the heart of the cat.

ABSTRACT

These studies were performed to identify the localization, and neuronal function of calcitonin gene-related peptide[CGRP] in the neural axis of rat stomach by retrograde tracing and immunohistochemical techniques. After injection of pseudorabies virus Bartha strain[PRV] as tracer between serosa and muscle layer of stomach, the rats were perfused and the brains were removed. PRV-immunoreactive cells were observed in central nucleus of amygdaloid, insular cortex, subfornical organ, bed nucleus of stria terminalis, paraventricular nucleus, organum vasculosum of terminalis, suprachiasmatic nucleus, lateral hypothalamic area, K lliker-Fuse nucleus, parabrachial nucleus, locus ceruleus, A1 noradrenaline area, A5 noradrenaline area, area postrema, dorsal motor nucleus of vagus nerve, nucleus tractus solitarius and raphe nuclei. CGRP-immunoreactive cells are observed in insular cortex, bed nucleus of stria terminalis, paraventricular nucleus, lateral hypothalamic area, parabrachial nucleus, area postrema, nucleus tractus solitarisu, neucleus ambiguus, facial nucleus, hypoglossal nucleus and raphe nuclei. The dobule immunofluorescent study was carried out to examine the coexistence of CGRP and PRV in several nuclei : insular cortex, bed nucleus of stria terminalis, paraventricular nucleus, later hypithalamic area, parabrachial nucleus, area postrema, nucleus tractus solitarius and raphe nuclei. At the results of double immunofluorescent study, we could not observe the double immunoreactive neurons CGRP and PRV in those nuclei but raphe nuclei. These results suggest that CGRP should not have a neural functions in the neurons in nuclei projecting to rat stomach except raphe nuclei.

ABSTRACT

The present investigation was performed in fifteen adult rats using four fluorescent tracers, including 4'-6-diamidino-2-phenylindol. 2 HC1 (DAPI), nuclear yellow (NY), propidium iodide (PI), and fast blue (FB), which were divided into four sets: PI-DAPI, PI-NY, DAPI-NY, and FB-NY. In each set two fluorescent tracers were used separately to label gastric corpus and pylorus antrum. In the medulla oblongata, the labelled cells in the dorsal motor nucleus of vagus (DmnX) were observed in every animal, but those in the nucleus ambiguus (nA) were only seen. in 11 cases and in the nucleus tractus solitarius (nTs) in 5 cases. In addition, labelled cells were also found in laminae V of the posterior grey columns of the thoracic (T6-9) spinal cords, spinal ganglia and sympathetic trunks. The results of counting and analysing the labelled cells of DmnX of both sides of 75 sections in four cases are as follows: 1233 cells on the left, including 1143 labelled cells of gastric corpus (92.7%), 63 labelled cells of pylorus antrum (5.1%), 27 double labelled cells (2.2%); 1007 cells on the right, including 930 labelled cells of gastric corpus (92.3%), 41 labelled cells of pylorus antrum (4.1%), and 36 double labelled cells (3.7%). The results indicate that PI is not fitted for 'long distance labelling in peripheral nervous system; the density of vagus innervation of gastric corpus is larger than that of pylorus antrum; a part of the nerve endings of vagus postganglionic neurons are distributed both in gastric corpus and in pylorus antrum; a part of postganglionic neurons of sympathetic fibres are located at the sympathetic trunk.

ABSTRACT

By using a highly sensitive HRP-TMB method, the present study found parasympathetic preganglionic neurons innervating rat stomach originated from the dorsal motor nucleus of the vagus(DMV), ambiguus nucleus(AM), reticular formation between DMV and AM(RF). rostroventrolateral reticular nucleus (RVL), lateral parvocelluler reticular nucleus (LRP), paraependymal nucloeus of the vagus (PEV), intercalated nucleus(IC) and retroambiguus nucleus(RA). There was a specific representation area of the stomach in the DMV between 0.3-0.6mm rostral to obex, and the different parts of the stomach had topographical organization within this area. The lateral portion of the DMV mainly innervated the forestomach, the medial and medial-middled portion of the DMV predominantly projected to the antrum/pylorus and corpus, respectively. The right and the left DMV did not show difference in innervating the forestomach and antrum/pylorus, but the corpus was mainly innervated by the right DMV. RF only projected to the forestomach and the corpus. RVL and RA only innervated the forestomach. AM, PEV and IC innervated all the three parts. The relationship between the different neuronal types of the DMV and the different parts of the stomach was first discussed in this paper. The forestomach, corpus and antrum/pylorus received their inputs mainly from the large and small neurons, respectively. The dentrites of the DMV contacting CSF were confirmed.
