ABSTRACT

Objective To evaluate the expression of epithelial cadherin (E-cad) and proliferating cell nuclear antigen (PC-NA) in various stages of tongue carcinogenesis and explore its relevance.Methods SP immunohistochemical method was employed to detect the expression of E-cad and PCNA protein in 82 rat tongue carcinogenesis specimens which induced by 4-nitroquinoline 1-oxide(4NQO).Chi square test for trend and the spearman correlation were used to analysis the correlation between E-cad and PC-NA.Results In normal mucosa,epithelial hyperplasia,mild dysplasia,severe dysplasia and squamous cell carcinoma,the positive rate of E-cad were 100%,95.24%,92.86%,80%,68.75%,the differences were statistically significant(P<0.05);The positive rate of PCNA were 9.52%,14.29%,35.71%,50%,56.25%,the difference were statistically significant(x2 =16.676,P<0.05).The expression of E-cad and PCNA has negative correlation(r=-0.614,P<0.01).Conclusion E-cad and PCNA may be one of the biomarkers of carcinogenesis of tongue mucosa.

ABSTRACT

Objective To further clarify the pathogenesis of different types of Kaposi′s sarcoma (KS) by measuring the protein expressions of caspase-3 and E-cad in tumor tissues of Xinjiang Uygur patients with acqured immunodeficiency syndrome (AIDS)-Kaposi′s sarcoma (KS) and classical KS.Methods From July 2011 to October 2014, 38 patients with KS at the First Affiliated Hospital of Xinjiang Medical University and Urumqi Infectious Disease Hospital were enrolled, among whom 28 were male and 10 were female, and all of them were uygur.Immunohistochemical and Western blot methods were used to detect the expressions of caspase-3 and E-cad proteins in 22 cases of AIDS-KS patients and 16 cases of classic KS.The quantitative data of normal distribution were analyzed by t test, while count data were compared with χ2 test with R × C table.Results KS lesions in patients with classic KS were confined to the skin, without mucosal, lymph node or visceral involvement.Lesions in AIDS-KS patients were not only confined to the skin and superficial lymph nodes, but also oral mucosa involved in 12 cases and internal organs involved in 7 cases.Liver and lung involvement was more common.The CD4+T lymphocyte count in patients with AIDS-KS was (200.8±166)/μL.All 15 AIDS cases with CD4+ T cell count less than 200/μL developed opportunistic infections.CD4+ T lymphocyte count of patients with classic KS was (562.52±222.66)/μL and the 16 patients with CD4+T lymphocyte count greater than 350/μL had no opportunistic infections.The results of immunohistochemistry showed that the positive expression rate of caspase-3 protein in KS tissues in patients with AIDS-KS was 68.2%, in patients with classic KS was 100.0%, with significant difference between two groups (χ2=7.37, P=0.01).The positive expression rate of E-cad protein in KS tissues in patients with AIDS-KS was 72.7%, in patients with classic KS was 100.0%, with significant difference between two groups (χ2=5.18, P=0.03).Western blotting showed that the gray value of caspase-3 in the KS tissue of patients with AIDS-KS was 0.55±0.36, and that in patients with classic KS was 0.86±0.56, with significant difference between two groups (t=-2.070, P<0.05).The gray value of E-cad in the KS tissue of patients with AIDS-KS was 0.54±0.41, and that in patients with classic KS was 0.85±0.45, with significant difference between two groups (t=-2.060,P<0.05).Conclusions There are differences in the protein expressions of caspase-3 and E-cad in tumor tissues of patients with AIDS-KS and classical KS in Xinjiang Uygur patients with Kaposi's sarcoma, which may correlate with a faster progression and a higher mortality rate for AIDS-KS.

ABSTRACT

Objective To investigate the relationship between expression of E -cad and IGF -1R and the recurrence of patients with primary hepatocellular carcinoma (HCC)who underwent liver resection.Methods The clinical data of 91 cases of primary HCC underwent liver resection from January 2010 to December 2012 were analyzed retrospectively.The expression of E -cad and IGF -1R was detected by RT -PCR,and the relationship between expression of E -cad and IGF -1R and the recurrence of patients was analyzed.Results The expression of E -cad was obviously lower in HCC tissues than that in normal liver tissues,with the expression percentage of 38.7% vs. 75.0%.The recurrence rate of E -cad negative group was higher(71.1%).The TNM Ⅱ -Ⅳ stage (χ2 =7.161, P =0.009)and liver capsule invasion (χ2 =5.144,P =0.036)in E -cad negative group were higher than E -cad positive group.The expression of IGF -1R was obviously higher in HCC tissues than that in normal liver tissues,with the expression percentage of 66.1% vs.20.0%.The recurrence rate of IGF -1R negative group was higher(68.3%). The TNMⅡ-Ⅳ stage (χ2 =4.195,P =0.014),liver capsule invasion (χ2 =5.144,P =0.036),non -tumor capsularin (χ2 =7.201,P =0.012)and PVTT(χ2 =6.538,P =0.032)in E -cad negative group were higher than E -cad positive group.And there was negative correlation between the expression of E -cad and IGF -1R(χ2 =14.329,P =0.000).Conclusion The mRNA expression of E -cad in the tissues of HCC is obviously lower than normal liver tissues.The mRNA expression of IGF -1R in the tissues of HCC is obviously higher than normal liver tissues.The expressions of E -cad and IGF -1R are associated with the degree of local invasion and malignant.The E -cad and IGF -1R expression is correlated in the tissues of HCC,which means that E -cad and IGF -1R play synergistic effect in HCC genesis.

ABSTRACT

Purpose To investigate the expression of the Cx43 and E-cad protein in esophageal carcinoma and normal esophageal tis-sue and to discuss their relationship. Methods SP immunohistochemical and Western blot analysis were used to detect the expression of Cx43 and E-cad in esophageal carcinoma and normal esophageal tissue. The relation between their expression and esophageal carci-noma was investigated. Results Immunohistochemical assay and Western blot analysis showed the expression levels of Cx43 and E-cad in esophageal carcinoma were significantly lower than that in normal esophageal tissue. The expression of Cx43 had correlation with lymph node metastasis. The expression of E-cad had correlation with degrees of pathological differentiation. There was a positive rela-tionship between the expression of Cx43 and E-cad in same sample. Conclusion Expression of Cx43 and E-cad protein is low in e-sophageal carcinoma, which can be helpful in the prediction of the biological behavior and prognosis of esophageal carcinoma.

ABSTRACT

Objective To investigate the role and prognostic impact of E-cadherin( E-cad)on in-vasion and metastasis of gastric carcinoma by analyzing E-cadherin expression at DNA,RNA and protein levels. Methods RNA and protein expression of E-cad were tested in 60 cases of gastric carcinoma and adjacent tissues by streptavidin peroxidase( SP )immunohistochemistry and in situ hybridization. DNA methylation of E-cad was tested with methylation-specific polymerase chain reaction( MSP). Results The positive rates of E-cadherin protein and mRNA in gastric carcinoma were significantly lower than those in adjacent tissues. DNA methylation of E-cad negatively correlated with the E-cad protein and RNA expres-sion in gastric carcinoma respectively(r= -0. 355,r = -0. 271). DNA methylation of E-cad negatively correlated with the 5-year survival rate of gastric carcinoma(r = -0. 381,P ﹤0. 01). However,E-cad protein positively correlated with the 5-year survival rate of gastric carcinoma(r=0. 378,P﹤0. 01). Con-clusion The decreased expression of E-cad is one of the most important biological features in gastric car-cinoma. Methylation of E-cad DNA may be an important agent causing low expression of E-cad protein and mRNA and it can be used as an important indicator in predicting invasion and metastasis of gastric cancer.

ABSTRACT

Previous researches showed that the expression level of E-Cad in most infiltrating cancer cells was reduced or negative.This study explored whether 4HPR restrained the infiltration of bladder cancer cells through regulating the expression of E-Cad.The infiltrating bladder cancer cells T24 were cultured,and then treated by a proper dosage of drug.Their viability was a determined by MTT method.Western blotting and RT-PCR were adopted to detect the changes of E-Cad gene expression at both protein and mRNA levels.Moreover,immunofluorescent staining and confocal fluorescence microscopy were employed for the observation of the expression of E-Cad.The result showed that,at both mRNA and protein levels,the expression level of E-Cad in T24 cells treated by 4HPR was significantly higher than that of control group,while the β-Cat expression was also relocated from the cell nucleus to cytoplasm.Our findings suggested that the regulatory function of 4HPR on infiltration of bladder cancer cells T24 is at least partly achieved by regulating the expression of E-Cad.

ABSTRACT

Objective: To study the expression and significance of VEGF-C, D2-40, and E-Cad in Human breast carcinoma. Methods: S-P immunohistochemical method was used to assess the expression of VEGF-C in 88 cases of infiltrating ductal carcinoma of the breast (with intact clinicopathologic and follow-up in-formation) and 54 cases of different intraductal hyperplastic lesions. The relationship of VEGF-C expression with D2-40 and E-Cadherin in infiltrating ductal carcinoma of the breast was analyzed. Results: VEGF-C ex-pression was higher in the invasive group than that in the dysplasia group and benign lesions. VEGF-C ex-pression in the metastatic group was significantly higher than that in the group without lymph node metasta-sis, indicating that VEGF-C plays an important role in the growth and metastasis of breast cancer. VEGF ex-pression in breast cancer group was higher than that in the dysplasia group and benign lesions. LVD count in the group with lymph node metastasis was significantly higher than that in the group without lymph node me-tastasis. The positive rate of E-Cad expression in the invasive group was significantly lower than that in the dysplasia group and benign lesions. VEGF-C expression was positively correlated with D2-40 expression. VEGF-C expression was negatively correlated with E-Cad expression. Conclusion: VEGF-C and D2-40 could be used for the detection of early lymphatic metastasis of breast cancer. VEGF-C and E-Cad can be used as important indices for the evaluation of the prognosis of breast cancer.
