ABSTRACT

Genetic variants in glioma tumor suppressor candidate region gene 1 (GLTSCR1)and ATM serine/threonine kinase (ATM) have been associated with various cancer risks.Epidemiological studies also revealed the association of variants of GLTSCR1 and ATM genes with different brain tumors.However,little is known about the relationship between both gene polymorphisms and lung cancer risk.We conducted a Chinese hospital-based casecontrol study involving 384 lung cancer cases and 387 cancer-free controls.No significant differences in the single polymorphism (GLTSCR1 rs1035938 and ATM rs11212592)association were found in five genetic models (co-dominant,dominant,recessive,overdominant and log-additive models) (adjusted by smoking duration).Join effect of three SNPs (PPP1R13L rs1970764,CD3EAP rs967591,GLTSCR1 rs1035938) on chromosome 19q 13.3 showed that the designated haplotype2 (rs 1970764<rs967591A-rs 1035938c) [OR (95% CI)=1.60 (1.11-2.32),P=0.012] and haplotype8 (rs 1970764G-rs967591G-rs 1035938T)[OR (95% CI)=2.45 (1.17-5.12),P=0.018] were associated with increased risk of lung cancer (adjusted by smoking duration).The analysis ofmultifactor dimensionality reduction revealed that two 3-way models were the best fit models in analyses of 2 loci (P<0.001)or 4 loci (P=0.015-0.016).The entropy-based analysis indicated the strongest synergistic effect between PPP1R13L rs1970764 and ATM rs11212592 in analysis of four genes.In conclusion,our study suggests that haplotypes consisting of PPP1R13L rs1970764-CD3EAP rs967591-GLTSCR1 rs1035938 on Chr19q13.3,interaction of smoking and GLTSCR1 rs1035938-ATM rs11212592,and synergistic action of PPP1R13L rs1970764 and ATMrs11212592 may associate with lung cancer risk in the Chinese population.

ABSTRACT

Objective·To construct effective shRNA-lentiviral vector targeting rat Eap1 gene and explore the effect of Eap1 on Kiss1 at cellular level. Methods·Four shRNA sequences targeting rat Eap1 mRNA and one negative control sequence were designed and synthesized, and then recombined with lentiviral vectors. After DNA sequencing identification, the shRNA recombinant vectors were co-transfected into 293T cells with pGag/Pol, pRev, pVSV-G to packaging lentiviral particles and the virus titers were determined. NRK-52E cells were transfected with all the four lentiviral-Eap1-shRNAs and the negative control lentivirus, and cells were harvested after 72 h. Real-time PCR and Western blotting was performed to detect the change of the mRNA and protein level of Eap1 gene. The most effective LV-Eap1-shRNA was used to infect NRK-52E cells [non-lentivirus and Eap1-shRNA(-) lentivirus as control groups], and then the level of Kiss1 mRNA expression was detected after infection for 72 h. Results·Seventy-two hours after transfection, the Eap1 mRNA and protein expression between blank group and LV-shRNA(-) group showed no significant difference; while Eap1 expression in all LV-Eapl-shRNA groups reduced significantly in comparing to the control groups. In addition, the lentiviral-Eap1-shRNA4 was the most effective in RNA interference. After blocking Eap1 expression in NRK-52E cells, the Kiss1 mRNA level was significantly increased comparing to the control groups. Conclusion·The effective Eap1-shRNA lentiviral vector is established successfully. The Kiss1 gene expression decreases after blocking Eap1 in NRK-52E cell line.

ABSTRACT

Se adaptó al contexto chileno la escala de Incremento de la Felicidad (HIS, de Tkach y Lyubomirsky, 2006) para medir estrategias conductuales de incremento de la felicidad, con el nombre de cuestionario de estrategias para aumentar los estados de Ánimo positivos (CEA-EAP) con 44 ítems en formato Likert de 4 puntos. Este, junto con la EFS (Lyubomirsky y Lepper 1999) fue respondido por 443 estudiantes universitarios (Cronbach global de .80), quienes se autoclasificaron como extra o introvertidos. El ANAFAC mostró una estructura diferente a la original de ocho factores y mayoritariamente con buena confiabilidad. Las estrategias de control mental y auto-cuidado o reevaluación positiva y reorientación positiva de la atención, las de sociabilización recreativa o festejar y, en menor medida pero también otras tres diferentes estrategias, se asociaron a felicidad (EFS), y a la autoclasificación de extrovertido. Ocio pasivo se asoció a introversión y no tuvo relación con la EFS.


The scale of happiness increase (HIS, of Tkach and Lyubomirsky, 2006) to measure behavioural strategies of increased happiness, was adapted to the chilean context with the name of questionnaire of strategies to increase positive moods (CEA-EAP), with 44 items in 4-point Likert format. This, together with the EFS (Lyubomirsky y Lepper 1999) was answered by 443 undergraduates (global Cronbach's alpha =.80), who self-rated as extra or Introverted. The ANAFAC showed an 8 factors structure -mostly with good reliability- that differed from the original. The strategies of mental and self-care control or positive re-evaluation and positive reorientation of attention, the recreational socialization or celebrate and, to a lesser extent but also other three different strategies were associated with happiness (EFS), and to the self-rating of Extrovert. passive leisure was associated with introversion and had no relationship with the EFS.

ABSTRACT

English for Academic Purpose (EAP), putting more emphasis on cultivating students ' ability in academic study, is an integral part of College English reform, This paper aims to explore how to carry out college English reform with the focus on EAP in universities of Traditional Chinese Medicine ( TCM ) , proposing practical approaches under the guidance of Content-based Language Teaching (CBLT). The suggestions include further implementation of EAP, the coherence of the General English courses, the construction of college English course system, the establishment of multiple as-sessment system and the development of suitable teaching material. The purpose is to advance the ap-plication and specialty of teaching content and make College English teaching serve TCM students' needs of academic studies and future careers more effectively.

ABSTRACT

O mecanismo de controle da secreção de GnRH inclui diversas vias neuronais. Estudos em modelos animais identificaram genes que codificam fatores de transcrição, tais como TTF-1 (thyroid transcription factor 1) e EAP1 (enhanced at puberty), que atuam no controle transcricional de genes codificadores de fatores excitatórios (KiSS1 e GnRH) e inibitórios (preproencefalinas) regulando a secreção de GnRH. Em primatas, a expressão de EAP1 e TTF-1 aumenta, no início da puberdade, nas regiões hipotalâmicas envolvidas na secreção de GnRH. Nos modelos animais, a deleção pós-natal de TTF-1 e o silenciamento do EAP1 provocam atraso puberal e prejuízo na função reprodutiva. TTF-1 também está envolvido na morfogênese diencefálica, por meio da via de sinalização da família Sonic-Hedgehog. Anormalidades na secreção de GnRH resultam em distúrbios puberais, que variam de puberdade precoce central (PPC) a hipogonadismo hipogonadotrófico. Hipotetizamos que anormalidades genéticas no TTF-1 e EAP1 estejam envolvidas na patogênese dos distúrbios puberais centrais. A PPC pode ser idiopática ou devido a causas orgânicas, sendo o hamartoma hipotalâmico, uma malformação congênita não neoplásica, a mais conhecida. Os pacientes com PPC devido a hamartoma hipotalâmico podem cursar com alterações neurológicas e cognitivas. Nossos objetivos foram: estudar as regiões codificadora do TTF-1 e do EAP1 e a região promotora do TTF-1 em pacientes com distúrbios puberais centrais; estabelecer a prevalência, taxa de penetrância e modo de herança da forma familial de PPC e caracterizar as manifestações neurológicas e neurocognitivas de pacientes com PPC devido a hamartoma hipotalâmico. Foram selecionados 133 pacientes com distúrbios puberais centrais - PPC idiopática (n=71), PPC devido a hamartoma hipotalâmico (n=15) e hipogonadismo hipogonadotrópico isolado normósmico (HHIn) (n=47) - e controles (n=53). GnRH secretion control involves multiple neuronal pathways. Animal studies have identified genes which codifies transcription factors, such as TTF-1 (thyroid transcription factor 1) and EAP1 (enhanced at puberty), that act in the transcriptional control of genes that codifies excitatory (KiSS1 and GnRH) and inhibitory factors (preproenkephalines) regulating GnRH secretion. In nonhuman primates, expression of EAP1 and TTF-1 are increased at the hypothalamic regions involved in GnRH secretion, at the beginning of puberty. In animal models, post-natal TTF-1 deletion and silencing of EAP1 lead to pubertal delay and damage of reproductive function. TTF-1 is also involved in diencephalic morphogenesis, through signalization via Sonic-Hedgehog family. Abnormalities in GnRH secretion are responsible for pubertal disorders, varying from central precocious puberty (CPP) to hypogonadotropic hypogonadism. We hypothesized that genetic anomalies at TTF-1 and EAP1 are involved in the pathogenesis of central pubertal disorders. CPP may be idiopathic or due to organic alterations and hypothalamic hamartoma, a non-neoplasic congenital malformation, is the most frequent known organic cause. Patients with CPP due to hypothalamic hamartoma may have neurological and cognitive disfunctions. Our aims were: to evaluated the codifying region of TTF-1 and EAP1 and the promoter region of TTF-1 in patients with central pubertal disorders; to establish the prevalence, penetrance rate and inheritance mode of familial CPP and to characterize neurologic and neurocognitive aspects of patients with CPP due to hypothalamic hamartoma. We selected 133 patients with central pubertal disorders idiopathic CPP (n=71), CPP due to hypothalamic hamartoma (n=15) and normosmic isolated hypogonadropic hypogonadism (nIHH) (n=47) - and controls (n=53). ABSTRACT

Objective To evaluate the mental health of medical workers, and develop an EAP suitable for heahhcare sector. Methods A 1-year EAP was implemented in a hospital in Beijing, in which SF-36 and JSS scales were used to study the health-related quality of life and job satisfaction of these workers. Results Work pressure is found to be the main source of pressure for medical workers, accounting for 63.7%. Scores of the group receiving EAP service are found to be significantly higher (P<0.01) in all dimensions (excepting work nature and supervisor dimension) of both SF-36 and JSS. Conclusions Medical workers are exposed to high pressure of work, having strong need for EAP service as result. EAP is an effective means to maintain mental health and improve quality of life and job performance, if applied widely.

ABSTRACT

OBJECTIVE: Employee Assistance Programs have recently been introduced to Korea as a potential solution to relieve workers's stress and improve their mental health. This study was conducted to determine the needs of EAPs among manufacturing industry workers in Gyeongsang-do Korea. METHODS: Responses to structured survey questionnaires were received from 556 workers at 7 mediumsized manufacturing factories. RESULTS: Among the 556 workers who responded to the questionnaire 'Prevention & management of work stress', 'Mental health & management of daily life stress', 'Physical exercise health promotion program'. 'Elderly care', 'Outplacement', 'Financial support for leisure activities', and 'Ensuring leisure time'were raised as the main issues and services that they expected to be provided by the company. CONCLUSION: EAPs have recently been introduced in Korean companies. The study results will be helpful to clarify the needs of individual Korean workers and to determine what EAP services should be provided.

ABSTRACT

In the present paper, we examined differences in therapeutic effectivenesses of venous methods of acupuncture, ranging from filiform needle to EAP, LFEA, laser and SSP, in cases with shoulder stiffness. The immediate and delayed effect of each method, comfortableness during treatment session, changes in temperature, blood flow, and the oxygen density at the Jianjing and Hegu point were examined.<br>Significant improvement was seen in every therapeutic method. Electrical acupuncture using direct current was most effective. No difference were evident among SSP, low-frequencey electro-acupuncture and filiform needle. There was a correlation between confortableness and the improvement rate in electrical acupuncture using direct current and laser acupuncture. Deep temperature, blood flow and the degree of oxygen saturation of the stimulated part tended to increasd in each case.

The 340 blood samples from Guangdone area were detected by ultra thin poly-acrylamide gel isoelectric focusing for EAP phenotypes.The phenotypes were20 EAP A,119 EAP BA,201 EAP B.The gene frequencies of EAP were as foll- ows p~(?)0. 2338,p~(?)0. 7662. The 110 bloodstain samples of the cloth kept at roomtemperature for 7 weeks could be phenotyped correctly,The 21 bloodstain samp-les on the porcelain plate kept at room temperature for 9 weeks could be phen-otyped correctly.When the blood volume of bloodstain was equal to or over5?l EAP could be phenotyped.6 out of 20 mouldy bloodstains,the EAP BAphenotype were changed to EAP B.In blind trial,20 bloodstain samples kept atroom temperature for 7 weeds could be phenotped correctly.

ABSTRACT

We have treated 117 patients, who visited the Pain Clinic of our Department complaining some pain 1973-1975, applying “in situ” needle, electrical acupuncture and low frequency electrical acupuncture therapy. And we have examined the immediate effects during 3 days after the treatment through patients' subjective evaluation, at what degree the state of pain was improved as compared with the state before the treatment on a scale of 1-10.<br>As the results, we classified the appearing ways of effects in 4 types: continuous type (the effect lasts 3 days right after the treatment), decreasing type (the effect gradually disappears immediately after the treatment), increasing type (it slowly produces effect) and invariable type (during 3 days a slight effect or no effect is observed). And it became clear that for the duration of effect and the efficiency of the therapy the low frequency electrical acupuncture therapy is most execellent.<br>Discussion of Results:<br>Immediate good results from directly following therapy to the 2nd day after therapy were obtained in more than 70% of the cases and were especially remarkable with LFEA therapy as compared to EAP or LN therapies.
