ABSTRACT

In this study, voltage was used as a disturbance factor to investigate the relationship between microbial community and methane (CH4) production flux in a microbial electrolytic cell coupled anaerobic digestion (MEC-AD). Metabolic flux analysis (MFA) was used to explore the relationship between the CH4 metabolic flux produced and the microbes. The results showed that both methane production flux and hydrogen production flux changed significantly upon voltage disturbance, while the voltage disturbance had little effect on acetic acid production flux. The maximum CH4 production flux under 0.6 V disturbance was 0.522±0.051, which increased by 77% and 32%, respectively, compared with that of the control group under 1.0 V (0.295±0.013) and under 1.4 V (0.395±0.029). In addition, an average of 15.7%±2.9% of H2 (flux) was used to reduce CO2 to produce CH4 and acetic acid, and an average of 27.7%±6.9% of acetic acid (flux) was converted to CH4. Moreover, the abundance of Lachnospiraceae significantly affected the flux of acetic acid. The flux of CH4 production is positively correlated with the abundances of Petrimonas, Syntrophomonas, Blvii28, and Acinetobacter, and negatively correlated with the abundances of Tuzzerella and Sphaerochaeta. The species that affected the flux of H2 and CH4 were similar, mostly belonging to Bacteroides, Clostridium, Pseudomonas and Firmicutes. Furthermore, the interspecies interaction is also an important factor affecting the MEC-AD methanogenesis flux.

ABSTRACT

This research deals with the investigation of electrocoagulation (EC) treatment of wastewater from a dairy plant using aluminum electrodes. Electrolysis time, pH, current density and distance between electrodes were considered to assess the removal efficiency of chemical oxygen demand (COD), total solids (TS) and their fractions and turbidity. Samples were collected from the effluent of a dairy plant using a sampling methodology proportional to the flow. The treatments were applied according to design factorial of half fraction with two levels of treatments and 3 repetitions at the central point. The optimization of parameters for treating dairy industry effluent by electrocoagulation using aluminum electrodes showed that electric current application for 21 minutes, an initial sample pH near 5.0 and a current density of 61.6A m-2 resulted in a significant reduction in COD by 57%; removal of turbidity by 99%, removal of total suspended solids by 92% and volatile suspended solids by 97%; and a final treated effluent pH of approximately 10. Optimum operating condition was used for cost calculations show that operating cost is approximately 3.48R$ m-3.

ABSTRACT

Ovomucoid (OMC) is the most prominent allergen causing hen's egg allergy, containing disulfide (S-S) bonds that may be responsible for its allergic action. As S-S bonds may be reduced during electrolysis, this study was undertaken to evaluate modulation of the allergic action of OMC after electrolysis. Electrolysis was carried out for 1% OMC containing 1% sodium chloride for 30 minutes with a voltage difference of 90 V, 0.23 A (30 mA/cm2). Protein assays, amino acid measurement, and mass spectrometry in untreated OMC and OMC on both the anode and cathode sides after electrolysis were performed. Moreover, 21 patients with IgE-mediated hen's egg allergy were evaluated by using the skin prick test (SPT) for untreated OMC and OMC after electrolysis. The allergic action of OMC was reduced after electrolysis on both the anode and cathode sides when evaluated by the SPT. The modifications of OMC on electrolysis caused the loss of 2 distinct peptide fragments (57E-63K and 123H-128R) as seen on matrix-associated laser desorption/ionization time-of-flight mass spectrometry. The total free SH groups in OMC were increased on the cathode side. Although the regions of S-S broken bonds were not determined in this study, the change in S-S bonds in OMC on both the anode and cathode sides may reduce the allergenic activity.

ABSTRACT

The synthesis and characterization of aromatic alcohols such as 1-(4-bromo phenyl) ethanol, 1-(4-Hydroxy-3-methoxyphenyl) ethanol, (4-Hydroxy-3-methoxy-benzyl alcohol) employing biotransformation (using whole cells of Baker’s Yeast in their free as well as immobilized form in mixtures of glycerol and water) and Electrochemical technique are reported. The electrochemical behavior of 4-bromoacetophenone, 4-Hydroxy-3-methoxyacetophenone, and 4-Hydroxy-3- methoxybenzaldehyde was analyzed using cyclic voltammetry at glassy carbon electrode (GCE) and constant current electrolysis. Effect of scan rate and pH on the reduction peaks has been calculated. The kinetic parameters were also calculated and the process was found to be diffusion controlled. The products obtained were purified & then characterized by spectroscopic techniques. All the compounds have been tested in vitro against a number of microorganisms in order to assess their antimicrobial properties. Biocatalytic and Electrochemical procedures were found to be more effective, safe, economical, environmental friendly, easy to handle. These green methodologies over conventional chemical methods provide new and improved synthetic routes to many valuable compounds.

ABSTRACT

Incorporation of green methodology via biocatalytic and electrochemical steps using Baker’s Yeast and electrons as reducing agent respectively have been employed as a novel and efficient route to furnish relevent chiral building blocks for fine chemicals and pharmaceuticals. Reduction of selected ketones such as 2-methylcyclohexanone, 3-methylcyclohexanone, 4-methylcyclohexanone and ethyl-2-oxocyclopentanecarboxylate have been carried out by biotransformation (using whole cells of Baker’s Yeast in their free as well as immobilized form in mixtures of glycerol and water) and via electrochemical method to the corresponding alcohols. Optimum conditions for electrochemical reduction like solvent, supporting electrolyte, reduction potential and pH were determined at glassy carbon electrode employing cyclic voltammetric technique. The effect of scan rate, pH were also studied. The electrochemical reduction was carried out at constant current using stainless steel (SS-316) electrodes. The products obtained were purified & then the results of both reduction routes (biocatalytic & electrochemical) were compared and then characterized by spectroscopic techniques.

ABSTRACT

The aim of this work was to investigate the disinfectant effect of electrolysis on chlorine-free water, artificially contaminated with Escherichia coli (CCT-1457) and to evaluate the bactericidal activity of electrolysis and kinetic behavior of a single-cell reactor, with a DSA (Dimensionally Stable Anode) electrode to develop a scaled-up system. A high-density E. coli suspension (10(6) CFU mL-1) was electrolyzed in this reactor at 25, 50 and 75 mA cm-2 for up to 60 min, at flow rates of 200 and 500 L h-1. Bacterial survival fell by 98.9 percent without addition of chlorinated compounds and a power consumption rate not more than 5.60 kWh m-3 at flow rate of 200 L h-1 and 75 mA cm-2. The process produced a germicidal effect that reached this inactivation rate within a relatively short contact time. Also, a solution of electrolyzed 0.08 M Na2SO4 added to the inoculum showed residual bactericidal effect. The efficiency of disinfection was regulated by both the contact time and current density applied, and a kinetic function for the survival rate was developed for the purpose of scaling up.

ABSTRACT

Introduction: With two consecutive experiments, we evaluated (1) the impact of electricity on chemotherapy response and on the lymphocyte proliferative response against the tumor (MTT assay) in a murine tumor model and (2) the effect of electricity alone and combined with chemotherapy in the treatment of spontaneous head and neck tumors of cats and dogs. Methods: (1) Five million cells of Ehrlich Ascitic Tumor were injected into the dorsal region of 35 male Balb/c mice. Seven days later, mice were divided into 7 groups, receiving different treatments combining Cisplatin or Bleomycin with electricity. Tumor progression in each mouse was measured daily and differences among the groups were assessed by ANOVA test. The immune impact of treatment was evaluated by the co-culture of lymphoid cells of treated and untreated mice with Ehrlich Tumor cells using co-culture with allogenic cells was used as a control. Following these results, we done a (2) sequential and non comparative study, in which dogs and cats presenting with a malignant neoplasm were clinically evaluated, staged and treated by electrolysis or electrochemotherapy in a veterinary hospital. Results: (1) The mean tumor area (+/- standard deviation) of mice receiving Cisplatin or Bleomycin with electricity were found significantly smaller if compared to that of controls (p<0.05). The co-culture results suggest that lymphocytes from mice treated by electricity promote elimination of Ehrlich Tumor cells in culture. In the study (2) 28 animals (14 dogs and 14 cats) were treated by electricity alone (15 cases) or combined to chemotherapy. The mean electrical charge/tumor volume was 38,50 C/cm3. By univariated analysis, factors impacting on survival were T stage (p=0.04) and charge/tumor volume (p<0.01). Conclusions: (1) Electricity enhanced the effect of both Cisplatin and Bleomycin and probably also stimulated an immune response against the tumor...

ABSTRACT

Oxygen-derived free radicals have been implicated in many important functions in the biological system. Electrical field stimulation (EFS) causes arterial relaxation in animal models. We found that EFS applied to neither muscle nor nerve but to Krebs solution caused a relaxation of rat aorta that had been contracted with phenylephrine. In the present study, therefore, we investigated the characteristics of this EIRF (electrolysis-induced relaxing factor) using rat isolated aorta. Results indicated that EIRF acts irrespective of the presence of endothelium. EIRF shows positive Griess reaction and is diffusible and quite stable. EIRF-induced relaxation was stronger on PE-contracted aorta than on KCl-contracted one, and inhibited by the pretreatment with methylene blue. Zaprinast, a cGMP-specific phosphodiesterase inhibitor, potentiated the EIRF-induced relaxation. NG-nitro-L-arginine, NO synthase inhibitor, did not inhibit the EIRF-induced relaxation. Deferroxamine, but not ascorbic acid, DMSO potentiated the EIRF-induced relaxation. These results indicate that electrolysis of Krebs solution produces a factor that relaxes vascular smooth muscle via cGMP-mediated mechanism.

ABSTRACT

BACKGROUND: Reactive oxygen free radicals have been implicated as an important factor in the development of ischemia-reperfusion injury of heart. Captopril, a SH-containing angiotensin converting enzyme(ACE) inhibitor has been reported to provide the protective effect in different models of myocardial ischemia and reperfusion injury of animal hearts. It is postulated that the myocardial protective effect may be related to a potential anti-free radical effect independent of ACE inhibition. The present study was designed to elucidate the myocardial protective mechanism of Captopril by investigating the drug effect on the experimentally induced oxygen free radical-mediated myocardial injury in isolated hearts of rats. METHODS: The heart isolated from rat was perfused retrogradly by Langendorff method. Myocardial dysfuntion was induced by oxygen free radicals generated by electrolysis of the perfusing solution(Kreb-Henseleit) with 2mA direct current for 45 sec. The cardiac functions(left ventricular pressure, dP/dt, heart rate, coronary flow) and the ventricular content of a lipid peroxidation product, malondialdehyde(MDA) were measured under presence of absence of Captopril and the compairing drugs(enalaprilat, cysteine and dithiothreitol). RESULTS: Electrolysis of oxygen-saturated Krebs-Henseleit perfusing solution led to the production of superoxide anion increasingly with intensity and duration of the applied electric current. The hearts perfused with the electroyzed solution demonstrated significant decrease in left ventricular pressure, dp/dt, heart rate, coronary folw and increase in myocardial MDA content. The depression of myocardial function as well as the increase of MDA content and oxygen radical production were reversed by Captopril(0.75~2mM) dose-dependently. Enalaprilat, a non-SH containing ACE inhibitor, however, showed no protective effect at all. Cysteine and dithiothreitol, the SH-containing agents without ACE inhibitory action showed also protective effects on the myocardial depression induced by electrolysis. CONCLUSION: It is suggested that Captopril may exert protective effect on oxygen radical-mediaed myocardial injury probably by its antioxidative and anti-free radical mechanism related to SH-group.

ABSTRACT

In order to investigate the mechanism of antibacterial effect of electrical stimulation on bone and joint infection, 5, 50 and 500 µA constant currents were passed through the silver electrodes and stainless electrodes on the Brain Heart Infusion agar culture plates which contained Staphylococcus aureus, Pseudomonas aeruginosa, and Lscherichia coli. After 24 hours of incubation, the widths of inhibitory zone around the electrodes were measured. The broth dilution susceptibility test was also used to verify the antibacterial effects of electrically generated silver in the Brain Heart Infusion broth. The results were as follows. 1. Silver anodes using constant currents manifested antibacterial effects definitely on the three bacterias, but stainless anodes showed none. 2. Antibacterial effects of silver anodes on the three bacterias increased in proportion to current density. However, antibacterial effects were negligible when current density was under 2 nA/mm2. 3. When current density was between 6.6 and 660 nA/mm2, there were no statistically significant differences in the antibacterial effects among 5, 50 and 500µA constant currents. 4. The culture broth containing silver ions generated by constant current electrolysis also manifested antibacterial effects. The retentate showed stronger antibacterial effects than the filtrate. Form these results, following conclusions were drawh. 1. The antibacterial eff ct of silver electrode using constant current was due to silver ions rather than electricity itself. 2. The substance which showed most effective antibactericity effects was silver complex larger than 2,000 m.w. and heat-unstable.
