ABSTRACT

Objective To explore the mechanisms on the damage in fetus liver,kidney and brain with administration of mifepristone and mesoproseol.Methods 17 specimens were obtained from the women who volunteered to terminate their pregnancy during 10~28 weeks.According to the fetus weight,they were divided into 3 groups.Immunohistochemistery was induced to investigate the expressions of pur-?,HSP70.Results The expressions of pur-? and HSP70 could be observed in both the treatment group and the control group.pur-? and HSP70 are the targets in detecting the damage of DNA.But pur-? was more sensitive than HSP70.The damage on group Ⅱ is the strongest.Conclusion Mifepristone can lead to the damage to lover,kidney and brain in mid-pregnancy.Its damage will be reducing as the growing of fetus.

ABSTRACT

This review focuses on the fetal origins of adult disease hypothesis put forward by David Barker and his colleagues,recent advances in epidemiological studies and experimental research in this field.Barker Hypothesis states that environmental factors,particularly intrauterine nutrition,as indicated by birth weight,operate in early life to program the risks for adverse health outcomes in adult life.A large growing body of reports described the association between the early development and adult diseases,such as diabetes,hypertension,coronary heart disease,abnormal lipids metabolism,obesity and cancer,etc.Experimental studies show that the changes of some key genes' expression,caused by epigenetic modifications,lead to a permanent alteration of cellular proliferation and differentiation and finally the genesis in key tissues and organs.These results bring about the impairment in structures and functions and the increased susceptibility to chronic diseases in adult life.The hypothesis provides a new perspective for the prevention and therapy of chronic diseases.

ABSTRACT

Objective To evaluate the relationship of small heterodimer partner(SHP) gene and birth weight in China. Methods A cohort study of 191 normal pregnant women was conducted. Both maternal and cord blood samples were collected. PCR-RFLP was used to detect the polymorphism of SNP-rs7504 of SHP. Results (1) The frequency of both neonatal and maternal C allele and (TC+CC) genotype increased significantly with birth weight (P=0.004, OR=3.168; P=0.005, OR=3.315; P=0.013, OR=2.495; P=0.013, OR=2.495). (2) The babies were heavier if they were C allele carrier. The average increase of birth weight was 246.3 g comparing the neonates with TC+CC genotype with those with TT genotype [(3658.7?400.94)g vs (3412.4?444.4)g, P=0.005]. The average birth weight of those maternal C allele carriers was 210.3 g heavier than those non-C allele carriers[(3628.9?405.5) g vs (3418.6?449.0 g]. (3) The fetal C allele was associated with maternal weight in pregnancy, prepregnant BMI, paternal height and weight. Women with C allele were heavier and had higher BMI without statistical significance comparing with those non-C allele carriers. Neither neonatal nor maternal SHP gene was associated with blood glucose and insulin level. (4) Multiple factors analysis showed that birth weight was related to maternal height, weight gain during pregnancy, prepregnant BMI, maternal and cord blood insulin level. After adjustment, the neonatal birth weight remained significantly correlated with cord blood SHP (P=0.0354), but not with maternal SHP gene (P=0.0711). Conclusions SHP gene is associated with newborns birth weight and may affect fetal growth.

ABSTRACT

Objective To compare the effects of vitrification with slow-freezing on the developmental ability of day 3 cleavage stage embryos.Methods Patients who had no less than 4 high quality embryos were included in this study.These embryos were cryopreserved using the methods of vitrification or slow-freezing.In the eryopreserved embryo transfer cycles,the embryos which were cryopreserved using one of the methods were chosen randomly.The developmental ability of embryos was compared between these two groups.Results A total of 80 patients were included in this study with 160 embryos.In the group of slow-freezing,73(91%)embryos were survived and achieved 15(38%)clinical pregnancies.Among these,3 were twins and the implantation rate was 25%(18/73).In the group of vitrification,71(89%)embryos were survived and achieved 19(48%)clinical pregnancies.Among these, 9 were twins and the implantation rate was 39%(28/71),which was significantly higher than the slow- freezing group(P

ABSTRACT

Recently, more and more attention have been paid to the impact of maternal thyroid hormones on neuropsychologic development of offspring. In the first 20 weeks of gestation, neuropsychologic development of the fetus is mostly dependent on thyroxine from the mother. Limited epidemiological data showed that full-scale WISC IQs averaged 7 points lower among children born to mothers with undiagnosed thyroid deficiency during pregnancy, in comparison with control children. The main cause of subclinical maternal hypothyroidism is autoimmune thyroiditis. Data from basic studies have confirmed the impacts of maternal thyroid hormones on the neurodevelopmental outcomes of offspring. In a Chinese female population, the positive rate of thyroid autoantibodies was 11.0%, and the prevalence of postpartum thyroiditis was 11.9%. Hence, it is necessary to set up a screening schedule for subclinical hypothyroidism and autoimmne thyroiditis in pregnant women, which is of great value for the health of both women and children.
