ABSTRACT

BACKGROUND: Bone marrow mesenchymal stem cells have been induced into islet-like cell mass in vitro. However, little researches reported on the morphological changes of cells, the types of endocrine cells in the islet-like cell mass and their relationships. OBJECTIVE: To investigate the morphological changes of cells in the differentiation of bone marrow mesenchymal stem cells into islet-like structure and to explore the composition and distribution of endocrine cells. METHODS: Passage 3 bone marrow mesenchymal stem cells growing well were cultured and expanded until the cell colonies occupies 80% of the bottom of the culture bottle, and the pancreatic tissue lysate was added for continuous induction. Dithizone staining was used to screen the islet-like cell mass directly differentiated from bone marrow mesenchymal stem cells. The types and distribution of endocrine cells were identified by Mallory staining. Expressions of insulin, C-peptide, glucagon and somatostatin protein were detected by immunofluorescence cytochemical staining. RESULTS AND CONCLUSION: (1) Dithizone staining showed that the number of positive cells was increased over the induction time. (2) Mallory staining showed the red α-like cells were located in the periphery of the islet-like cell mass, the yellow β-like cells located in the center and periphery, and the light blue fibroblasts were distributed around the cell mass. (3) Immunofluorescence staining showed insulin, C-peptide, glucagon and somatostatin positive cells in the islet-like cell mass. To conclude, under certain microenvironment, bone marrow mesenchymal stem cells can differentiate into islet-like structures which containing α, β, δ-like cells, and are surrounded by fibroblast-like cells.

ABSTRACT

Background:It has been demonstrated that acid secretion-related gastric fundic structures do not degenerate in elderly individuals. As specimens obtained from human body are greatly influenced by environmental factors, the results need to be further verified by standard animals. Aims:To investigate the aging-related changes of histology of gastric fundus and ultrastructure of fundic gland in beagle dogs. Methods:Nineteen healthy beagle dogs were divided into three age groups,8 in younger group(aged 1-5 years),5 in junior elderly group(aged 6-8 years)and 6 in senior elderly group (aged ≥9 years). Gastric specimens obtained from the fundus were examined by light microscope, the thickness of lamina propria and percentage of parenchymal area of fundic gland in lamina propria were measured, and the parietal cells, chief cells and endocrine cells were counted. Ultrastructure of fundic gland cells were observed by electron microscopy, and the area percentages of related organelles were calculated. Results:The thickness of lamina propria in gastric fundus of beagle dogs showed a decline tendency with aging, but no significant differences were found among the three age groups(P >0. 05);in senior elderly group,a marked decrease in density of fundic gland was observed and with an increase of interstitial tissues. The amount of parietal cells and the area percentages of mitochondria and tubulovesicle in parietal cells showed no aging-related reduction(P > 0. 05),while the amount of chief cells, area percentages of zymogen granule in chief cells, and mucous granule in mucous neck cells were significantly decreased in senior elderly group(P < 0. 05). The amount of endocrine cells increased gradually with aging(P < 0. 05). Conclusions:Aging-related degeneration is not existed in acid secretion-related parietal cells and organelles in gastric fundus of beagle dogs. Furthermore, endocrine cells which secret histamine, gastrin, and somatostatin, etc. for stimulating acid secretion even increased. These changes might be a compensatory mechanism for acid secretion in elderly individuals.

ABSTRACT

Obesity is a multifactorial disorder often associated with many important diseases such as diabetes, hypertension and other metabolic syndrome conditions. Argyrophil cells represent almost the total population of endocrine cells of the human gastric mucosa and some reports have described changes of specific types of these cells in patients with obesity and metabolic syndrome. The present study was designed to evaluate the global population of argyrophil cells of the gastric mucosa of morbidly obese and dyspeptic non-obese patients. Gastric biopsies of antropyloric and oxyntic mucosa were obtained from 50 morbidly obese patients (BMI >40) and 50 non-obese patients (17 dyspeptic overweight and 33 lean individuals) and processed for histology and Grimelius staining for argyrophil cell demonstration. Argyrophil cell density in the oxyntic mucosa of morbidly obese patients was higher in female (238.68 ± 83.71 cells/mm2) than in male patients (179.31 ± 85.96 cells/mm2) and also higher in female (214.20 ± 50.38 cells/mm2) than in male (141.90 ± 61.22 cells/mm2) morbidly obese patients with metabolic syndrome (P = 0.01 and P = 0.02, respectively). In antropyloric mucosa, the main difference in argyrophil cell density was observed between female morbidly obese patients with (167.00 ± 69.30 cells/mm2) and without (234.00 ± 69.54 cells/mm2) metabolic syndrome (P = 0.001). In conclusion, the present results show that the number of gastric argyrophil cells could be under gender influence in patients with morbid obesity. In addition, gastric argyrophil cells seem to behave differently among female morbidly obese patients with and without metabolic syndrome.

ABSTRACT

INTRODUCTION: Ghrelin is a 28 amino acid peptide mainly secreted by endocrine cells of the gastric mucosa, which is believed to have a modulating effect on cell growth. OBJECTIVE: To assess the presence of ghrelin and its precursor preproghrelin molecule in endocrine hyperplasias associated with atrophic body gastritis (ABG). MATERIAL AND METHODS: Endoscopic biopsies from 54 patients with ABG were processed for immunohistochemistry and specific antibodies against ghrelin, preproghrelin and chromogranin were applied. We assessed the immunoreactive cells in endocrine hyperplasia from the atrophic mucosa and intestinal and pseudo-antral metaplasia areas. RESULTS: There was ghrelin expression in a variable number of hyperplastic endocrine cells from all patients studied. There was a statistically significant difference in the number of hyperplastic nodules with more than 50 percent immunostained cells for chromogranin and ghrelin and for chromogranin and preproghrelin. The mean number of hyperplastic nodules identified by chromogranin was 8.6 per patient. Most nodules were immunoreactive to ghrelin and preproghrelin. The presence of ghrelin and preproghrelin expression was uncommon in glands showing intestinal metaplasia: four (9.5 percent) and nine (21.4 percent) cases, respectively. In contrast, they were relatively frequent in pseudo-antral metaplasia areas: 37 (72.5 percent) and 26 (50.9 percent) cases, respectively. CONCLUSION: Ghrelin- and preproghrelin-immunoreactive cells are frequently present in endocrine hyperplasias associated with ABG. However, further studies are required to determine to what extent these hormones act as modulators of hyperplastic nodular growth and evolution.

ABSTRACT

The purpose of this study was to analyze the esophageal morphology of Caiman latirostris using histochemicaland immunohistochemical techniques. The mucosae layer is composed of a pseudostratified columnarepithelium with goblet cells that occasionally form intra-epithelial glands. The goblet cells showed differentaffinity for the histochemical techniques. The lamina propria is composed of loose connective tissue with bloodvessels. The smooth muscle fibers that compose the muscularis mucosa are thick and form a clear demarcationbetween the mucosa layer and adjacent submucosa. There are no glands in the esophageal submucosa. Themuscle layer is composed a circular internal layer and a longitudinal external one. The adventitia layer iscomposed of loose connective tissue and blood vessels. The serotonin immunoreactive (IR) cells were notpreferentially distributed in any region of the epithelium, ranging from the base of the intra-epithelial gland tothe connection to the lumen. Somatostatina, glucagon and CCK IR cells were not detected.

ABSTRACT

In this study, immunohistochemistry was used to examine the changes in the density of colonic endocrine cells - argyrophil and argentaffin cells, chromogranin A (CGA), serotonin, somatostatin and glucagon-containing cells in trinitrobenzene sulfonic acid (TNBS)-induced rat colitis. Ulcerative colitis was induced by the instillation of 10 mg of TNBS into the colonic lumen through the anus. To confirm the inducement of ulcerative colitis, the macroscopic and microscopic scores as well as the colonic myeloperoxidase (MPO) activities were monitored for 8 days after TNBS instillation in the colonic lumens. In addition, the number of argyrophil and argentaffin cells, CGA, serotonin, somatostatin and glucagon-immunoreactive cells were counted in the colonic mucosa, respectively. After TNBS instillation into the lumen of the colon from the anus in rats, increases in macroscopic and microscopic scores in the colon tissues were observed along with increases in the colonic MPO activities. Therefore, ulcerative colitis was relatively well induced by the TNBS instillations. Marked decreases in the number of colonic endocrine cells were detected in the TNBS-treated animal compared to the sham control. These results suggest that colonic endocrine cells were also disrupted by TNBS-induced ulcerative colitis.

ABSTRACT

In the present study, we investigated types of pancreatic endocrine cells and its respective peptides in the Brazilian sparrow species using immunocytochemistry. The use of polyclonal specific antisera for somatostatin, glucagon, avian pancreatic polypeptide (APP), YY polypeptide (PYY) and insulin, revealed a diversified distribution in the pancreas. All these types of immunoreactive cells were observed in the pancreas with different amounts. Insulin- Immunoreactive cells to (B cells) were most numerous, preferably occupying the central place in the pancreatic islets. Somatostatin, PPA, PYY and glucagon immunoreactive cells occurred in a lower frequency in the periphery of pancreatic islets.

ABSTRACT

Objective To clarify the morphological features, types, regional distributions and cell densities of gastrointestinal (GI) endocrine cells in various parts of the digestive track (DT) of Eumeces elegans. Methods Using immunohistochemical techniques of streptavidin-peroxidase(S-P) method. Results The endocrine cells found in the DT were round or oval, spindle, shuttle, pyramid, flask and bacilliform in shape. Three types of endocrine cells, namely 5-hydroxytryptamine (5-HT) positive cells, somatostatin (SS) positive cells and gastrin (GAS) positive cells were found. No substance P(SP)-, glucagon (GLU)-, pancreatic polypeptide ( PP)- and insulin(INS) positive cells were detected. 5-HT positive cells distributed throughout the whole DT at various densities and they were most predominant cell types in the DT. SS positive cells were detected only in stomach. GAS positive cells showed a restricted distribution and were only demonstrated in the pylorus and duodenum. In the DT of E. elegans the region with the highest degree of cell type heterogeneity was pylorus and all types of endocrine cell along the DT showed peak density in the pylorus as well. Conclusion Some common features of the distribution of different types of GI endocrine cells are found between E. elegans and other reptiles. This common trait may reflect the similarity in digestive physiology of various vertebrates. On the other hand, some species-dependent unique distribution features of endocrine cells in DT were also detected in E. elegans.

ABSTRACT

ABSTRACT

The regional distribution and relative frequency of some endocrine cells in the pancreas of the carp, Cyprinus carpio Linnaeus, belonging to the family Cyprinidae in the order Cypriniformes, were observed using specific mammalian antisera against insulin, glucagon, somatostatin and human pancreatic polypeptide (hPP) by peroxidase antiperoxidase (PAP) method. The pancreas was divided into four regions (principal and secondary islets, exocrine and pancreatic duct regions). In addition, the pancreatic islet regions were further subdivided into three regions (central, mantle and peripheral regions) and the pancreatic duct regions were subdivided into two regions (epithelial and subepithelial regions). Spherical to spindle or occasionally round to oval shaped immunoreactive (IR) cells were demonstrated in the pancreatic islets, exocrine and pancreatic duct. In the principal islet regions, some cells were also detected in the other regions, most of insulin- and somatostatin-IR cells were located in the central regions, and glucagon- and hPP-IR cells were situated in the peripheral regions. In this regions, insulin-IR cells were most predominant cell types and then, glucagon, somatostatin and hPP in that order. In the secondary islet regions, the regional distribution and relative frequency of these four types of endocrine cells were quite similar to those of the principal islets except for cell clusters consisted of hPP-IR cells that were situated in the peripheral to mantle regions. In the pancreatic duct regions, all four major pancreatic endocrine cells were demonstrated in the inter-epithelial cells and/or basal regions of the epithelial linning. In addition, cell clusters composed of numerous insulin-, moderate glucagon- and somatostatin-IR cells of low frequency were also observed in the subepithelial regions of the pancreatic duct. In the exocrine regions, insulin-, glucagon-, somatostatin- and hPP-IR cells were located in the inter-acinus regions with rare, a few, moderate and moderate frequencies, respectively. In conclusion, the regional distribution and relative frequency of four major pancreatic endocrine cells, insulin-, glucagon-, somatostatin- and hPP-IR cells, in the pancreas of the carp showed general patterns which were observed in other stomachless teleost. However, some species- dependent different distributional patterns and/or relative frequencies were also demonstrated.

ABSTRACT

Endocrine cells have been observed from 13 cases of human fetal lungs through the using of Grimelius silver staining technique, fluorescence microscopy and ultrastructural examinations. They were found in great number in the small bronchi than in any other part of bronchial tree. Most of them were round or oval in shape, and usually rest on the basement membrane of epithelium. They were stained by Grimelius silver stain. After incubation with L-Dopa they emitted a green glyoxylic acid-induced fluorescence. Typical feature of these cells was the numerous electron dense neurosecretory-type granules, 168-312 nm in diameter, which can be seen throughout the cytoplasm.

Results Seven kinds of gastrointestinal endocrine cells were found positive in the digestive tracts of Canis lupus and Canis familiaris. The endocrine cells were mainly located between the epithelia cells of gastric gland, intestinal villus and intestinal gland. There were also some endocrine cells distributed in the lamina propria and submucosa. The endocrine cells found in the digestive tract were in various shapes. Most of the endocrine cells had long cell processes run ning out to the lumen or the nearby cells.Among these endocrine cells,5-hydroxytryptamine(5-HT) positive cells had a widest distribution, followed by vasointestinal poplypetide(VIP) positive cells. The distributions of other cells were limited. From the comparison of the characters of endocrine cells of Canis lupus and Canis familiaris, most of the endocrine cells had similar morphological features and regional distributions except pancreatic polypeptide(PP) positive cells and substance P(SP) positive cells. There were differences in cell densities of some endocrine cells. Conclusion The results of Canis lupus and Canis familiaris reflect the similarity of mammals in the characteristics of the morphological features, types, regional distributions and cell densities of gastrointestinal endocrine cells,as well as the similarity in digestive physiology of canine animals. At the same time, the results of Canis lupus also exhibit characteristics of gastrointestinal endocrine cells of wild animals.

ABSTRACT

The distribution of dopamine-immunoreactive endocrine cells and nerves in the digestive tract of guinea-pigs was examined with the ABC immunostaining technique. Dopamine-immunoreactive endocrine cells were observed in the pyloric antrum, small intestine and colon. The highest density of the positive ceils was found in the duodenum. From the jejunum to the colon the number of immunoreactive cells gradually decreased. A similar population of the positive cells was seen in the antrum and colon. Dopamine-immunoreactive nerve fibres were mainly located in the lamina propria and submucosa of the digestive tract. There were more positive fibres in the small intestine than in the stomach and colon. The dense networks of immunoreactive fibres were demonstrated in the villi. Some positive nerve fibres penetrated into the superficial and glandular epithelia. This representation indicated that the epithelial cells were directly innervated with the dopamine containing nerves.
