ABSTRACT

Objective: To evaluate the clinical significance of detection of serum glypican 3 (GPC3) on the diagnosis of hepatocellular carcinoma. Methods: Mouse anti-GPC3 (25-358 aa) monoclonal antibody and rabbit anti-GPC3 (379-393 as) antibody were used to establish the sandwich ELISA method for detecting serum GPC3 level in 364 HCC patients, 96 patients with chronic hepatitis, and 106 healthy subjects. Serum alpha-fetoprotein (AFP) level was detected by radioimmunoassay. Results: The mean concentration of serum GPC3 was 86.96 ± 42.2 ng/mL, 11.7 ± 12.23 ng/mL, and 6.04 ± 9.21 ng/mL in HCC patients, chronic hepatitis patients, and healthy subjects, respectively. The difference between HCC patients and chronic hepatitis patients and healthy subjects was highly significant (P 0.05). When the cutoff value was set at 30 ng/mL, the sensitivity and specificity of GPC3 in the diagnosis of hepatocellular carcinoma was 40% and 93%, respectively. Meanwhile, the positive rate of AFP was 49% and the positive rate of GPC3 was 40% in 151 HCC serum samples. The detection rate of HCC increased to 72% by combined detection of AFP with GPC3. Conclusion: Serum GPC3 can be used as a marker for the early diagnosis of HCC in clinic.

ABSTRACT

Objective:To study the levels of soluble interleukin-2 receptor(sIL-2R) in patients with head and neck cancer.Methods:Using the method of sandwich ELISA, the sIL-2R in 60 cases of patients with head and neck cancer, 60 with benign tumor and 50 of healthy subjects were measured.Results:The concentration(U/ml) of serum sIL-2R in patients with head and neck cancer was significantly higher than that in those with benign tumor(571.9?294.2 vs 305.9?104.8, P0.05).Conclusion:sIL-2R may be related to head and neck cancer.

ABSTRACT

Objective To finally diagnose the outbreak of acute respiratory infection caused by Adenovirus 3 in students of the north of Jiangsu province by serologic study. Methods An enzyme-linked immunosorbent assay (ELISA) has been used to detect the adenovirus IgM in the serum of acute phase and the adenovirus IgG in the serum of recovery phase. A neutralization test has been used to detect the neutralization antibody in the serum of acute and recovery phase from cases and in the serum from the control. The SPSS11.0 has been applied to the statistical analysis of the data. Results The positive rate of IgM, IgG and recovery phase neutralization antibody of cases are 3.7%, 44.4% and 59.5% respectively, while those of control are 0%, 8.3% and 33.3% respectively, and the P values of Chi-Square are 0.510, 0.018 and 0.226 respectively. The neutralization test of paired serum of 6 in 9 cases showed that the antibody titers was obviously increased. The concordance between IgG detected by ELISA and neutralization antibody detected by neutralization test is 61.4% and the P value of Kappa is 0.070. Conclusion By the serologic study, we can finally diagnose that the outbreak of acute respiratory infection was caused by Adenovirus 3.
