ABSTRACT

OBJECTIVE:To conduct the abnormal toxicity test and study on the in vitro hemolytic reaction of Gluconate enoxa-cin injection, and provide reference for its safe use. METHODS:According to"Test for Abnormal Toxicity"in Chinese Pharmaco-poeia(2015 edition,Vol.Ⅱ),half lethal dose(LD50)of Gluconate enoxacin injection and setting of abnormal toxicity limit were carried out in mice. According to"Test for Haemolysis and Agglomeration"in Chinese Pharmacopoeia (2015 edition,Vol.Ⅱ), study for the in vitro hemolytic and agglomeration reaction was carried out in rabbits. RESULTS:The LD50 of Gluconate enoxacin injection for mice was 248.9 mg/kg;the setting limit of abnormal toxicity test was 28.4 mg/kg,and the results derived from the test using this limit met requirements. The haemolysis and agglomeration results derived from the test using clinical maximum con-centration(0.2 g/100 mL)met requirements. CONCLUSIONS:In order to reduce the clinical ADR and feasibility of ensuring quali-ty standard, the abnormal toxicity limit of Gluconate enoxacin injection should be set as 28.4 mg/kg.

ABSTRACT

Objective:To establish a method for the determination of related substances in enoxacin gluconate injection by HPLC with gradient elution. Methods:HPLC was performed on a DiamonsilTM C18 column (250 mm × 4. 6 mm, 5 μm), the mobile phase A was 0. 025 mol·L-1 phosphonic acid solution (adjusting pH to 3. 0 with triethylamine)-methanol- acetonitrile (80∶10∶10) and the phase B was 0. 025 mol·L-1 phosphonic acid solution (adjusting pH to 3. 0 with triethylamine)-methanol-acetonitrile (350∶325∶325) with gradient elution at a flow rate of 1. 1 ml·min-1 , the detection wavelength was 269 nm and 284 nm, the injection volume was 20μl, and the column temperature was 40℃. Results:Under the HPLC conditions, the samples had good stability and separation. The calibration curve was linear within the concentration range of 19. 80 ng·ml-1-19. 80 μg·ml-1(r=0. 999 9) for 5-hydroxymethylfur-fural with the detection limit of 0. 18ng, and the average recovery was 99. 68% with RSD of 0. 12% (n=9). Conclusion:The method is accurate, sensitive, specific and reproducible, and can be used in the determination of related substances in enoxacin gluconate in-jection.

ABSTRACT

Objective To evaluate the efficacy and safety of enoxacin in the treatment of lower respiratory tract bacterial infection in the old patients. Methods 48 cases were given enoxacin and 46 cefotaxime. Results There was no significant difference between the two groups in cure rate, effective rate, bacterial clearance and side effects incidence. Conclusion Enoxacin is effective and safe in the treatment of lower respiratory tract bacterial infection of the old patients.

ABSTRACT

Gram-negative toeweb infections have clinical manifest, ations of variable severity. In its mildest form, the affected tissue is damp, softened, boggy and white. But. more severe form may progress to marked denudation of second, third and fourth toewebs with a profuse serous or purulent discharge, and its treatment is difficult. We described a 39-year-old man who had ulcerated arid oozing patches on several toewebs. Pseudomonas aeruginosa was detected in bacterial cultures. We successfullg treated with split-thickness skin graft, and enoxacin on first, visit and enoxacin with gentamicin ointment, for 2 weeks on second visit.

ABSTRACT

A new synthetic pyridone carboxylic antimicrobial agent, Enoxacin (B-Mark(TM)) was administered orally in 37 cases of genitourinary tract infection and following results were obtained. 1. Among 12 cases of acute simple cystitis, the cure rate was 91.7%. 2. Among 6 cases of chronic complicated urinary tract infections, the cure rate was 66.7 %. 3. Among 9 cases of gonorrheal urethritis, the cure rate was 88.9%. 4. Among 10 cases of nongonorrheal urethritis, the cure rate was 60%. There was no serious side effects but mild G-I symptoms such as nausea, indigestion and fatigability were found. The results suggest that Enoxacin (B-Mark(TM)) is a useful antibacterial agent in the treatment of urinary tract infections, especially in acute simple cystitis and gonorrheal urethritis.

ABSTRACT

Effect of Enoxacin (Flumark), an oral pyridone carboxylic acid, in the urinary tract infection was evaluated clinically and susceptibility of 292 strains of microorganism isolated from urine to the various antimicrobial agents was tested and compared. Thirty six patients with urinary tract infection were given with 300 to 600mg of Enoxacin daily for 7 to 14 days according to severity of urinary tract infection. The clinical symptoms and urinary findings improved in 30 patients (83.3%) and no clinical response was achieved in 6 patients (16.7%), but revealing 100% response in 9 patients of acute uncomplicated cystitis. In the antimicrobial susceptibility test, Enoxacin was much superior to aminoglycoside, especially against Pseudomonas aeruginosa. Serratia spp., Enterobacter spp. and other Pseudomonas spp.. Enoxacin is a very effective and safe antimicrobial agent in treatment of the urinary tract infection.
