ABSTRACT

OBJECTIVE:To compare the chemical constituents of petroleum ether fraction from ethanol extract of Aconitum sinomontanum before and after processing. METHODS :After A. sinomontanum was purified with water ,the raw product decoction pieces were prepared ;the raw decoction pieces were steamed with licorice juice under high pressure to prepare processed decoction pieces of A. sinomontanum . The petroleum ether fractions of raw product and processed product were obtained after ultrasonic extraction with 95% ethanol. The chemical constituents in the samples were analyzed by GC-MS. NIST 2014 mass spectrometry database was used to compare and match the components . The peak area normalization method was used to determine the relative percentage content of each component. RESULTS :Before and after processing ,fatty acids and esters were the main components in the petroleum ether fraction from ethanol extract. Totally 18 chromatographic peaks were detected in the detection pieces of raw product,and 13 compounds were identified ,accounting for 94.60% of the total content of volatile components. The components with relatively high content were (Z,Z,Z)-9,12,15-octadecatrienoic acid (26.13%),hexadecanoic acid ethyl ester (25.27%), palmitoleic acid (10.84%),ethyl linoleic acid (10.67%),(Z,Z)-9,12-octadecenoic acid methyl ester (6.66%),pentadecanoic acid(5.11%)and so on. Totally 25 chromatographic peaks were detected in the decoction pieces of processed products,and 18 components were identified ,accounting for 82.40% of the total content of volatile components. The components with relatively high content were palmitoleic acid (18.95%),(Z,Z)-9,12-octadecenoic acid methyl ester (17.93%),hexadecanoic acid ethyl ester(11.94%),(Z,Z,Z)-9,12,15-octadecatrienoic acid (10.54%),(Z,Z)-9,12-octadecenoic acid (5.51%),(Z)-11-hexadecanoic acid(5.30%)and so on. After processing ,7 new components were added ,5 of which were identified as (-)-eucalyptus globulus alcohol,ethyl 2-methyltetrade-canoate,6-methyl-4-phenylcoumarin,β-sitosterol,heptadecane. After processing ,no components disappeared,and the content of some components increased or decreased. CONCLUSIONS :After processing ,the volatile components in the petroleum ether fraction from ethanol extract of A. sinomontanum are different ,and(-)-eucalyptus globulus alcohol and other components are added after processing.

ABSTRACT

OBJECTIVE:To study in vitro bacteriostatic activity of the extracts from Miao medicine Polygonum runcinatum. METHODS:Using chloramphenicol and fluconazole as positive control,the inhibitory effects of water extract,95% ethanol extract and its ethyl acetate and n-butanol fraction on 9 kinds of strains were determined by cup plate method, such as Staphylococcus aureus,Escherichia coli,Shigella flexneri,Salmonella typhi,Bacillus subtilis,Pseudomonas aeruginosa,type B Hemolytic streptococcus,Candida albicans and Cryptococcus neoformans. The parts with bacteriostatic activity and trial strains sensitive to drug were screened. Micro-broth dilution method and agar culture medium plate method were used to determine MIC and MBC of 95% ethanol extract and its fractions of P. runcinatum to sensitive strains. RESULTS:The water extract of P. runcinatum almost had no inhibitory effect. 95% ethanol extract showed different degrees of bacteriostatic activity to strains;bacteriostatic effects of it to 6 kinds of bacteria as S. typhi was better than that of type B H. streptococcus and 2 kinds of fungus. The ethyl acetate and n-butanol fraction of 95% ethanol extracts showed good bacteriostatic effect and the ethyl acetate fraction had stronger effect,while all the fractions of 95% ethanol extracts showed no inhibitory effect on fungus. MIC and MBC of 95%ethanol extract to above 6 kinds of bacteria were 6.25-12.5 and 12.5-25 mg/mL,respectively;MIC and MBC of ethyl acetate fraction were 3.13-6.25, 6.25-12.5 mg/mL; MIC and MBC of n-butanol fraction were 6.25-12.5, 12.5-25 mg/mL. CONCLUSIONS:95% ethanol extract,ethyl acetate and n-butanol fractions of Miao medicine P. runcinatum have obvious in vitro bacteriostatic activity to 6 common bacterias as S. typhi.

ABSTRACT

Objective To observe the change of cerebral oxygen metablism and the level of blood lactate in early stage of sepsis in rats.Method Sixty-four SD rats were randomly(random number)divided into septic group and control group.The sepsis model of rat was made by lipopolysaccharide (LPS,10 mg/kg)injected intra-abdominally,and rats of control group were treated with the same amount of physiological saline instead.And each group was further divided into 4 sub-groups of4 h,6 h,12 h and 24 h after treatment.At each interval,blood samples were obtained via jugular vein for detecting blood oxygen saturation (Sjv02)and blood lactate(LA).The blood oxygen saturation(Sa02)of ventral aorta was also measured.Arteriovenous oxygen content difference (AVD02) and oxygen extraction fraction (OEF) were studied.These four variables were analyzed and compared between two groups.Results The AVD02 and OEF in sepsis group were higher than those in control groups of 3 h,6 h and 12 h (P0.05).LA in sepsis group was higher than that in control group in each interval (P<0.05).Conclusions In early stage of sepsis,cerebral perfusion,cerebral oxygen supply and cerebral oxygen extraction increase.The global oxygen metabolic variables(Sjv02,AVD02 and OEF)and the level of LA can be used to objectively and accurately evaluate the cerebral oxygen metabolic dysfunction in early stage of sepsis.
